        cDNA array 방법을 이용한 망간에 노출된 흰쥐 뇌기저핵의 유전자발현 분석

        Objectives: This study investigated the gene expression profile in basal ganglia of manganese-exposed rats based on cDNA array analysis. Methods: For cDNA array, 25 male Sprague-Dawley rats (250±25 g) were intraperitoneally injected with 25 ㎎/㎏ B. W./day of MnCl2 (0.3 ㎖) for 10 days. For dose-related gene expression analysis, rats were intraperitoneally injected with 0.2, 1.0, and 5.0 ㎎/㎏ B. W/day of MnCl2 for 10 days. Control rats were injected with an equal volume of saline. RNA samples were extracted from brain tissue and reverse-transcribed in the presence of [α^(32)P]-dATP. Membrane sets of the Atlas Rat 1.2 array Ⅱ and Toxicology array 1.2 kit (Clontech, Palo Alto, CA) were hybridized with cDNA probe sets. Northern blot hybridization method was employed to assess the dose-related gene expression. Results: Fifty-two genes showed significant changes in expression of more than two-fold. Twenty-eight were up-regulated and 24 were down-regulated in the manganese-exposed group compared to the control. Among the 52 genes, 28 genes including nuclear factor I-X1 (NF1-X1), neuroligin 2 and 3, mitochondrial stress-70 protein (MTHSP70), neurodegeneration-associated protein 1 (Neurodap 1), multidrug resistance protein (MDR), and endoplasmic reticulum stress protein 72 (ERP72), were reported for the first time related to the manganese-induced neurotoxic-metabolism in the rat basal ganglia. According to the dose-related gene expression analyses, MTHSP70, Neurodap 1 and ERP72 genes were up-regulated compared to the control even in the group exposed to low manganese dose ( 0.2 ㎎/㎏ B.W./day). Conclusions: Twenty-eight genes detected for the first time in this study were closely related to the manganese-induced neurotoxic-metabolism in the rat basal ganglia and further study of these genes can give some more useful information about the manganese metabolism.

      • Antioxidant and Anti-Inflammatory Effects of Rhei Rhizoma and Coptidis Rhizoma Mixture on Reflux Esophagitis in Rats

        <P>The purpose of this study was to investigate the antioxidant and anti-inflammatory effects of the combined extract of Rhei rhizoma and Coptidis rhizoma (RC-mix) in experimental model of acute reflux esophagitis. The antioxidant activity was assessed by<I> in vitro</I> 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays. RC-mix was given at 100, 200, and 400 mg/kg body weight 2 h prior to induction of reflux esophagitis (RE). After 5 h, the effects of RC-mix treated rats were compared with those of normal and control rats. The representative flavonoid contents of RC-mix, such as sennoside A, epiberberine, coptisine, palmatine, and berberine, were detected using HPLC. The elevated esophageal mucosa damage was markedly ameliorated by RC-mix treatment in a dose-dependent manner. Furthermore, the administration of RC-mix reduced the increase of serum reactive oxygen species (ROS) and peroxynitrite (ONOO<SUP>−</SUP>). The improvement of superoxide dismutase (SOD) and heme oxygenase-1 (HO-1) levels were marked in the group given RC-mix. Moreover, the elevation of inflammatory mediators and cytokines by nuclear factor-kappa B (NF-<I>κ</I>B) activation in control rats decreased by RC-mix pretreatment. These results indicate that RC-mix treatment reduces the pathological states of esophagitis<I> via</I> regulating NF-<I>κ</I>B mediated inflammation related to oxidative stress. </P>

        Glomerular filtration rate (GFR)검사는 사구체의 여과기능을 나타내는 중요한 지표로서 신장기능과 그 기능의 정도를 검사하기 위하여 시행되고 있다. 사구체의 혈액 여과 작용은 신장의 중요한 기능 중 하나로 여과 장벽을 구성하여 체내에 필요한 혈구와 단백질은 빠져나가지 못하게 하고 크기가 작은 물질이나 수분, 노폐물은 자유롭게 통과시킨다. 하지만 사구체의 여과기능이 제대로 이루어 지지 않을 경우 혈뇨 및 단백뇨가 검출된다. 이와 같이 사구체의 기능에 문제가 있을 경우 급성 사구체 신염, 만성 사구체 신염 등 신장질환이 발생하게 되고 GFR검사를 통해 사구체 기능의 정도를 판단하고 있다. GFR검사는 $^{51}Cr$-EDTA으로 방사선의약품을 만들어 정맥주사를 하고 경과시간에 따라 EDTA Tube에 채혈하여 Plasma 내 방사능을 측정하여 결과 값을 도출해 낸다. 따라서 진료과 및 병동에 통보하여 동위원소를 사용하는 PET-CT, Bone Scan과 혈액에 영향을 주는 투석 및 수혈은 GFR검사와 병행하지 않도록 사전에 통보하고 있다. 검사방법으로는 먼저 $^{51}Cr$-EDTA 2 mL와 생리식염수 5 mL를 잘 혼합하고 혼합된 용액 중 5 mL로 환자에게 주입 할 방사성 의약품을 만든다. 먼저 주사 전 주사무게를 측정하고 환자의 정맥에 주사를 시행하고 난 뒤 주사 후 주사무게를 측정한다. 채혈은 총 2번 시행되고 성인의 경우에는 주입 후 3시간, 5시간 뒤, 소아의 경우 주입 후 2시간, 5시간 뒤 EDTA Tube에 5 mL채혈한다. 채혈 된 검체는 3300rpm에 5분간 원심분리 시키고 Plasma $500{\mu}L$를 Test tube에 분주하여 Sample rack에 준비해 둔다. Standard용액 제조방법은 200 mL Volumetric flask에 표시된 눈금까지 Diluent water를 채워주고 난 후 $500{\mu}L$을 버리고 $^{51}Cr$-EDTA $500{\mu}L$를 주입한 후 잘 혼합해준다. 제조 된 Standard 용액은 2개의 Test tube에 각각 $500{\mu}L$씩 분주하고 측정대상인 환자의 Plasma와 함께 Gamma Counter로 계측한다. 계측으로 얻은 CPM과 환자의 신장, 체중, 주사 전 후 무게 등을 GFR함수식에 입력하여 결과 값을 도출해 낸다. Reference range는 남성은 $119.5{\pm}30.3ml/min/1.73m^2$ 이고 여성은 $125.2{\pm}28.2ml/min/1.73m^2$ 설정되어 있지만 본원에서는 성별에 관계없이 $97.0ml/min/1.73m^2$ 이상일 경우 신장이식이 가능하다고 판단하고 있다. GFR검사 결과 값에 따라 신장기능의 정도가 반영 되기 때문에 병행하지 않아야 할 검사들과 주의사항을 준수한다면 보다 더 신뢰할 수 있는 결과 값을 얻을 수 있을 것이라 생각한다. Purpose The glomerular filtration rate (GFR) test is an important indicator of glomerular filtration and has been used to test renal function and the extent of its function. The GFR test is performed by intravenous injection of radioactive medicines made of $^{51}Cr$-EDTA, and blood concentration is measured by taking blood according to the elapsed time. also, PET-CT, bone scan, transfusion and so on will affect the outcome. Therefore, we will improve the quality of the test by providing guidelines for the GFR test for more accurate testing. Materials and Methods 5 mL of physiological saline solution and 2 mL of $^{51}Cr$-EDTA solution are used to make 5 mL of the radiopharmaceutical solution to be injected into the patient. First, the syringe weight is measured before the injection, and then the radioactive medicine is injected into the patient's vein and the syringe weight is measured after the injection. Blood sampling is performed twice in total. In adults, blood is collected 3 hours / 5 hours after injection and in children 2 hours / 5 hours after injection. The blood sample is centrifuged at 3300 rpm for 5 minutes. Standard solution is prepared by filling diluent water up to the scale indicated in the 200-mL volumetric flask, discarding $500{\mu}L$, injecting $500{\mu}L$ of GFR reagent and mixing well. $500{\mu}L$ each of the standard solution is dispensed into two test tubes, and $500{\mu}L$ of each of the plasma samples collected in time is dispensed into two test tubes and measured with a Cobra Counter. Results At present, the reference range applied in this study is $119.5{\pm}30.3ml/min/1.73m2$ for males and $125.2{\pm}28.2ml/min/1.73m^2$ for females. Conclusion The GFR test is conducted using radioactive medical products. GFR testing is performed as a scheduled test, but PET-CT, dialysis and transfusion, which may affect GFR testing, may be scheduled during GFR testing. Therefore, we could get accurate GFR test results by notifying the ward and department beforehand when booking.

      • A STUDY ON THE DETERMINANTS OF FRANCHISING IN KOREAN FOOD SERVICE INDUSTRY

        Purpose This study, for the first time attempts to investigate the determinants of the Korean foodservice franchising employing various theories. In addition this study attempts to consider distinguishable foodservice characteristics such as the segment of foodservice and the market demand. This study expects to contribute to the theoretical underpinnings of the foodservice franchising by incorporating distinguishable foodservice characteristics to franchising theories. Method The sample for this study came from uniform franchise offering circular based on Korea Fair Trade Commission. There were 1,598 food service franchisors in uniform franchise offering circular 2009-2011 listing. However, 596 were excluded because of data irregularities remaining 1,002 available for sample. Based on the literature review, we choose seven determinants. The seven determinants were measured as follows. Firm age was measured as two proxy variables. One was measured as the years of franchising business. The other was measured as the years of business. Firm size was measured as total sales. Capital scarcity was measured as the ratio of total debt to total asset. Start-up costs were measured as initial investment. Monitoring costs were measured as the number of major cities franchised units operates. Royalty was measured as sales of a franchised unit. Finally, brand name was measured as advertising expenditures. Results The first proposition (firm age is related to the use of franchising) was supported by the signaling theory and the transaction costs theory instead of the resource scarcity theory. The age (measured as the years of franchising business) had a statistically significant positive relationship with the use of franchising. The second proposition (firm age is related to the use of franchising) was supported by the logic of the resource scarcity. The age (measured as the years of business) had a statistically significant negative relationship with the use of franchising. The third proposition (firm size is related to the use of franchising) was supported by the signaling theory instead of the resource scarcity theory. The firm size had a statistically significant positive relationship with the use of franchising. The fourth proposition (capital scarcity is related to the use of franchising) was not supported by the resource scarcity theory. The capital scarcity had not a statistically significant positive relationship with the use of franchising. The fifth proposition (start-up costs is related to the use of franchising) was not supported. The start-up costs had not a statistically significant either positive or negative relationship with the use of franchising. The sixth proposition (monitoring costs is related to the use of franchising) was supported by the agency theory. The monitoring costs had a statistically significant positive relationship with the use of franchising.. The seventh proposition (royalty is related to the use of franchising) was not supported by the agency theory. The royalty had not a statistically significant positive relationship with the use of franchising. The eighth proposition (brand name is related to the use of franchising) was not supported by the agency theory. Brand name had not a statistically significant negative relationship with the use of franchising. Conclusions Five propositions were found to be statistically significant from eight propositions. The four determinants such as firm age, firm size, monitoring costs, and brand name can affect the use of franchising for the foodservice industry in South Korea. Relating the significant propositions to the diverse theories, there was no one theory that dominated all propositions. The signaling theory was more appropriate for both firm age (the years of franchising business) and firm size while the transaction costs theory was more appropriate for age (the years of franchising business) as well. In addition, the resource scarcity theory was more appropriate for firm age (the years of business). Finally, the agency theory was appropriate for monitoring costs although this theory needed to be ameliorated for brand name.

      • Assessment of IL-6, IL-8 and TNF-α levels in the gingival tissue of patients with periodontitis

        <P>In periodontal diseases, inflammatory mediators, including interleukin (IL)-6, IL-8 and tumor necrosis factor-α (TNF-α), may promote the degeneration of inflamed periodontal tissues. In previous studies, levels of these three cytokines were demonstrated to be elevated in inflammatory gingival tissues and gingival crevicular fluid. The aim of the present study was to quantify IL-6, IL-8 and TNF-α levels in the human gingival tissues of patients with periodontitis and to assess the correlation of these three cytokines with each other. In this study, human gingival tissues from 19 patients with periodontitis (male, n=14; female, n=5) were collected. The tissues were homogenized, centrifuged and the protein in the supernatant was quantified. Enzyme-linked immunosorbent assay (ELISA) was used in the measurement of the IL-6, IL-8 and TNF-α levels, and the mean levels were observed to be 8.41±0.25, 34.01±1.09 and 20.70±0.31 pg/ml, respectively. The mean levels of IL-8 were higher than those of the other two cytokines. In each sample, the level of TNF-α expression was consistently high, with little difference between the results, which contrasted with the fluctuations in IL-6 and IL-8 levels. The expression of the two ILs (IL-6 and IL-8) showed a positive correlation (r=0.932, P=0.01), whereas TNF-α levels were not correlated with IL-6 or IL-8 levels. These results suggest that IL-6, IL-8 and TNF-α may be relevant in the pathophysiology of periodontitis, and the measurement of these cytokines may be beneficial in the identification of patients with periodontitis.</P>

      • Layer-by-layer assembled multilayers of charged polyurethane and graphene oxide platelets for flexible and stretchable gas barrier films

        <P>With the advent of the era of consumer-oriented displays and mobile devices, the importance of barrier film coatings for securing devices from oxygen or moisture penetration has become more salient. Recently developed approaches to generate gas barrier films in a combination of polyelectrolyte multilayer matrices and incorporated inorganic nanosheets have shown great potential in outperforming conventional gas barrier films. However, these films have the intrinsic drawback of vulnerability to brittleness and inability to stretch for flexible device applications. To overcome this issue, we present a method in which we prepare multilayered films of complementarily charged polyurethane and graphene oxide platelets using spin-assisted, layer-by-layer self-assembly to obtain well-stacked film structures. As a result, the multilayered, thin films deposited on a poly(ethylene terephthalate) (PET) substrate can exhibit significantly reduced oxygen penetration properties (∼30 cc m<SUP>−2</SUP> day<SUP>−1</SUP> for the oxygen transmission rate) while still demonstrating large bending or stretching deformations. Therefore, the proposed approach in this study is anticipated to be extensively utilized for surface coating and protection of flexible and stretchable devices under various operating conditions.</P>

        Metformin use and cardiovascular outcomes in patients with diabetes and chronic kidney disease: a nationwide cohort study

        Background: Metformin has recently been shown not to increase the risk of lactic acidosis in patients with chronic kidney disease (CKD). Thus, the criteria for metformin use in this population has expanded. However, the relationship between metformin use and clinical outcomes in CKD remains controversial. Methods: This study considered data from 97,713 diabetes patients with an estimated glomerular filtration rate of <60 mL/min/1.73 ㎡. The primary outcome was major adverse cardiac and cerebrovascular events (MACCE), and the secondary outcomes were all-cause mortality and incident end-stage renal disease (ESRD). Results: Metformin users had a significantly higher risk of MACCE than non-users (hazard ratio [HR], 1.20; 95% confidence interval [CI], 1.14-1.26; p < 0.001). However, metformin users had a lower risk of all-cause mortality (HR, 0.78; 95% CI, 0.74-0.81; p < 0.001) and ESRD (HR, 0.44; 95% CI, 0.42-0.47; p < 0.001) during follow-up than non-users did. The relationships between metformin use and clinical outcomes remained consistent in propensity score matching analyses and subgroup analyses of patients with adequate adherence to anti-diabetes medication. Conclusion: Treatment with metformin was associated with an increased risk of MACCE in patients with diabetes and CKD. However, metformin users had a lower risk of all-cause mortality and ESRD during follow-up than non-users did. Therefore, metformin needs to be carefully used in patients with CKD.
