      • Crocetin Induces Cytotoxicity in Colon Cancer Cells Via p53-independent Mechanisms

        Li, Cai-Yan,Huang, Wen-Feng,Wang, Qun-Li,Wang, Fan,Cai, E.,Hu, Bing,Du, Jia-Cheng,Wang, Jing,Chen, Rong,Cai, Xiao-Jing,Feng, Jing,Li, Hui-Hui Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.8

Objective: Crocin has been proposed as a promising candidate for cancer chemoprevention. The purpose of this investigation was to investigate the chemopreventive action and the possible mechanisms of crocin against human colon cancer cells in vitro. Methods: Cell proliferation was examined using MTT assay and the cell cycle distribution fractions were analyzed using fow cytometric analysis after propidium iodide staining. Apoptosis was detected using theTUNEL Apoptosis Detection Kit with laser scanning confocal microscope. DNA damage was assessed using the alkaline single-cell gel electrophoresis assay, while expression levels of p53, cdk2, cyclinA and P21 were examined by Western blot analysis. Results: Treatment of SW480 cells with crocetin (0.2, 0.4, 0.8 mmol/L) for 48 h signifcantly inhibited their proliferation in a concentration-dependent manner. Crocetin (0.8 mmol/L) signifcantly induced cell cycle arrest through p53-independent mechanisms accompanied by P21 induction. Crocetin (0.8 mmol/L) caused cytotoxicity in the SW480 cells by enhancing apoptosis and decreasing DNA repair capacity in a time-dependent manner. Conclusions: This report provides evidence that crocetin is a potential anticancer agent, which may be used as a chemotherapeutic drug.

Predictive Significance of Claudin-3 for Epithelial Barrier Dysfunction in Chronic Rhinosinusitis With Nasal Polyps

        Huang Zhi-Qun,Ye Jing,Liu Jing,Sun Li-Ying,Ong Hsiao Hui,Wei Yong-Hao,Fu Shu-Cai,Hu Xiao-Xun,Xu Yu,Wang De-Yun 대한천식알레르기학회 2023 Allergy, Asthma & Immunology Research Vol.15 No.4

Purpose: The abnormal expression of tight junction (TJ) plays a vital role in the pathogenesis of chronic rhinosinusitis with nasal polyps (CRSwNP). However, there is no appropriate tool to distinguish and diagnose epithelial barrier defects in clinical practice. This study aimed to evaluate the predictive value of claudin-3 for epithelial barrier dysfunction in CRSwNP. Methods: In this study, TJ protein levels were evaluated by real-time quantitative polymerase chain reaction, immunofluorescent, and immunohistochemistry staining in control subjects and CRSwNP patients. The receiver operating characteristic (ROC) curve was created to assess the predictive value of TJ breakdown in clinical outcomes. In vitro, human nasal epithelial cells were cultured at the air-liquid interface to analyze the transepithelial electrical resistance (TER) level. Results: The expression levels of occludin, tricellulin, claudin-3, and claudin-10 were decreased (all P < 0.05), and those of claudin-1 was increased (P < 0.05) in CRSwNP patients as compared to healthy subjects. Additionally, claudin-3 and occludin levels were negatively correlated with the computed tomography score in CRSwNP (all P < 0.05), and the ROC curve indicated that the claudin-3 level had the most predictive accuracy in evaluating epithelial barrier disruption (area under the curve = 0.791, P < 0.001). Finally, the time-series analysis showed the highest correlation coefficient between TER and claudin-3 (cross-correlation function = 0.75). Conclusion: In this study, we suggest that claudin-3 could be a valuable biomarker for predicting nasal epithelial barrier defects and disease severity in CRSwNP.

Finite element analysis and axial bearing capacity of steel reinforced recycled concrete filled square steel tube columns

        Jing Dong,Hui Ma,Changming Zou,Yunhe Liu,Chen Huang 국제구조공학회 2019 Structural Engineering and Mechanics, An Int'l Jou Vol.72 No.1

This paper presents a finite element model which can simulate the axial compression behavior of steel reinforced recycled concrete (SRRC) filled square steel tube columns using the ABAQUS software. The analytical model was established by selecting the reasonable nonlinear analysis theory and the constitutive relationship of material in the columns. The nonlinear analysis of failure modes, deformation characteristics, stress nephogram, and load-strain curves of columns under axial loads was performed in detail. Meanwhile, the influences of recycled coarse aggregate (RCA) replacement percentage, profile steel ratio, width thickness ratio of square steel tube, RAC strength and slenderness ratio on the axial compression behavior of columns were also analyzed carefully. It shows that the results of finite element analysis are in good agreement with the experimental results, which verifies the validity of the analytical model. The axial bearing capacity of columns decreased with the increase of RCA replacement percentage. While the increase of wall thickness of square steel tube, profile steel ratio and RAC strength were all beneficial to improve the bearing capacity of columns. Additionally, the parameter analysis of finite element analysis on the columns was also carried out by using the above numerical model. In general, the SRRC filled square steel tube columns have high bearing capacity and good deformation ability. On the basis of the above analysis, a modified formula based on the American ANSI/AISC 360-10 was proposed to calculate the nominal axial bearing capacity of the columns under axial loads. The research conclusions can provide some references for the engineering application of this kind of columns.

New Non-uniformity Correction Approach for Infrared Focal Plane Arrays Imaging

        Qu Hui-Ming,Gong Jing-tan,Huang Yuan,Chen Qian 한국광학회 2013 Current Optics and Photonics Vol.17 No.2

Although infrared focal plane array (IRFPA) detectors have been commonly used, non-uniformity correction (NUC) remains an important problem in the infrared imaging realm. Non-uniformity severely degrades image quality and affects radiometric accuracy in infrared imaging applications. Residual non-uniformity (RNU) significantly affects the detection range of infrared surveillance and reconnaissance systems. More effort should be exerted to improve IRFPA uniformity. A novel NUC method that considers the surrounding temperature variation compensation is proposed based on the binary nonlinear non-uniformity theory model. The implementing procedure is described in detail. This approach simultaneously corrects response nonlinearity and compensates for the influence of surrounding temperature shift. Both qualitative evaluation and quantitative test comparison are performed among several correction technologies. The experimental result shows that the residual non-uniformity, which is corrected by the proposed method,is steady at approximately 0.02 percentage points within the target temperature range of 283 K to 373K. Real-time imaging shows that the proposed method improves image quality better than traditional techniques.

      • GSTP1, ERCC1 and ERCC2 Polymorphisms, Expression and Clinical Outcome of Oxaliplatin-based Adjuvant Chemotherapy in Colorectal Cancer in Chinese Population

        Li, Hui-Yan,Ge, Xin,Huang, Guang-Ming,Li, Kai-Yu,Zhao, Jing-Quan,Yu, Xi-Miao,Bi, Wen-Si,Wang, Yu-Lin Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.7

Aim: Platinum agents have shown to be effective in the treatment of colorectal cancer. We assessed whether single nucleotide polymorphisms (SNPs) in GSTP1, ERCC1 Asn118Asn and ERCC2 Lys751Gln might predict the overall survival in patients receiving oxaliplatin-based chemotherapy in a Chinese population. Methods: SNPs of GSTP1, ERCC1 Asn118Asn and ERCC2 Lys751Gln in 335 colorectal cancer patients were assessed using TaqMan nuclease assays. Results: At the time of final analysis on Nov. 2011, the median follow-up period was 37.7 months (range from 1 to 60 months). A total of 229 patients died during follow-up. Our study showed GSTP1 Val/Val (HR=0.44, 95% CI=0.18-0.98), ERCC1 C/C (HR=0.20, 95% CI=0.10-0.79) and ERCC2 G/G (HR=0.48, 95% CI=0.19-0.97) to be significantly associated with better survival of colorectal cancer. GSTP1 Val/Val, ERCC1 C/C and ERCC2 G/G were also related to longer survival among patients with colon cancer, with HRs (95% CIs) of 0.41 (0.16-0.91), 0.16 (0.09-0.74) and 0.34 (0.16-0.91), respectively. Conclusion: GSTP1, GSTP1, ERCC1 Asn118Asn and ERCC2 Lys751Gln genotyping might facilitate tailored oxaliplatin-based chemotherapy for colorectal cancer patients.

A Modified PiggyBac Transposon System Mediated by Exogenous mRNA to Perform Gene Delivery in Bovine Mammary Epithelial Cells

        Guangdong Hu,Jing Wang,Hui Huang,Fusheng Quan,Jian Kang,Yongyan Wu,Yuanpeng Gao,Feng Su,Minghao Shao,Yong Zhang 한국생물공학회 2014 Biotechnology and Bioprocess Engineering Vol.19 No.2

Transposons are widely used for geneticengineering in various model organisms. Recently, piggyBac(PB) has been developed as a transposable and efficientgene transfer tool in mammalian cells. In the present study,we developed three types of PB transposon systemscontaining a dual plasmid system (DPS), a single plasmidsystem (SPS), and a DNA-mRNA combined system (DRPS)and characterized their basic properties in HEK293 cells. The basic elements of the donor plasmid included aselectable-reporter gene expression cassette, two loxP sitesin the same orientation, a multiple cloning site, and twochicken β-globin insulator core elements. We further identifiedthe function of the selectable-reporter and examined PBintegration sites in the human genome. Moreover, wecompared the transposition efficacy and found that SPStransposed more efficiently, as compared to DPS; integrationinto the host genome was determined by measuring PBaseactivity. Results discovered the loss of PBase activity in theDRPS, indicating that this system is much more biologicallysafe, as compared to DPS and SPS. Finally, we employedthe DRPS to successfully perform a gene delivery intobovine mammary epithelial cells (BMECs). Taken together, the information from this study will improve the flexibilityof PB transposon systems and reduce the genotoxicity ofPBase in genetic engineering.

      • Structure of Penta-Alanine Investigated by Two-Dimensional Infrared Spectroscopy and Molecular Dynamics Simulation

        Feng, Yuan,Huang, Jing,Kim, Seongheun,Shim, Ji Hyun,MacKerell, Alexander D.,Ge, Nien-Hui American Chemical Society 2016 The Journal of physical chemistry B Vol.120 No.24

<P>We have studied the structure of (Ala)(5), a model unfolded peptide, using a combination of 2D IR spectroscopy and molecular dynamics (MD) simulation. Two different isotopomers, each bis-labeled with C-13=O and C-13=O-18, were strategically designed to shift individual site frequencies and uncouple neighboring amide-I' modes. 2D IR. spectra taken under the double-crossed <pi/4, -pi/4, Y, Z > polarization show that the labeled four-oscillator systems can be approximated by three two-oscillator systems. By utilizing the different polarization dependence of diagonal and cross peaks, we extracted the coupling constants and angles between three pairs of amide-I' transition dipoles through spectral fitting. These parameters were related to the peptide backbone dihedral angles through DFT calculated maps. The derived dihedral angles are all located in the polyproline-II (ppII) region of the Ramachandran plot. These results were compared to the conformations sampled by Hamiltonian replica-exchange MD simulations with three different CHARMM force fields. The C36 force field predicted that ppII is the dominant conformation, consistent with the experimental findings, whereas C22/CMAP predicted similar population for alpha(+), beta, and ppII, and the polarizable Drude-2013 predicted dominating beta structure. Spectral simulation based on MD representative conformations and structure ensembles demonstrated the need to include multiple 2D spectral features, especially the cross-peak intensity ratio and shape, in structure determination. Using 2D reference spectra defined by the C36 structure ensemble, the best spectral simulation is achieved with nearly 100% ppII population, although the agreement with the experimental cross-peak intensity ratio is still insufficient. The dependence of population determination on the choice of reference structures/spectra and the current limitations on theoretical modeling relating peptide structures to spectral parameters are discussed. Compared with the previous results on alanine based oligopeptides, the dihedral angles of our fitted structure, and the most populated ppII structure from the C36 simulation are in good agreement with those suggesting a major ppII population. Our results provide further support for the importance of ppII conformation in the ensemble of unfolded peptides.</P>
