        Paricalcitol attenuates lipopolysaccharide-induced inflammation and apoptosis in proximal tubular cells through the prostaglandin E<sub>2</sub> receptor EP4

        Background: Vitamin D is considered to exert a protective effect on various renal diseases but its underlying molecular mechanism remains poorly understood. This study aimed to determine whether paricalcitol attenuates inflammation and apoptosis during lipopolysaccharide (LPS)-induced renal proximal tubular cell injury through the prostaglandin E<sub>2</sub> (PGE<sub>2</sub>) receptor EP4. Methods: Human renal tubular epithelial (HK-2) cells were pretreated with paricalcitol (2 ng/mL) for 1 hour and exposed to LPS (1 μg/mL). The effects of paricalcitol pretreatment in relation to an EP4 blockade using AH-23848 or EP4 small interfering RNA (siRNA) were investigated. Results: The expression of cyclooxygenase-2, PGE<sub>2</sub>, and EP4 were significantly increased in LPS-exposed HK-2 cells treated with paricalcitol compared with cells exposed to LPS only. Paricalcitol prevented cell death induced by LPS exposure, and the cotreatment of AH-23848 or EP4 siRNA offset these cell-protective effects. The phosphorylation and nuclear translocation of p65 nuclear factor-kappaB (NF-κB) were decreased and the phosphorylation of Akt was increased in LPS-exposed cells with paricalcitol treatment. AH-23848 or EP4 siRNA inhibited the suppressive effects of paricalcitol on p65 NF-κB nuclear translocation and the activation of Akt. The production of proinflammatory cytokines and the number of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling-positive cells were attenuated by paricalcitol in LPS exposed HK-2 cells. The cotreatment with an EP4 antagonist abolished these anti-inflammatory and antiapoptotic effects. Conclusion: EP4 plays a pivotal role in anti-inflammatory and antiapoptotic effects through Akt and NF-κB signaling after paricalcitol pretreatment in LPS-induced renal proximal tubule cell injury.

        The purpose of this study is to investigate the relation to stress level, time management strategies and home managerial satisfaction of caregivlng wives, and to find out the most influential factor in them. For the purpose of this study, it was distributed to nursing of patients' families hospitalized patients in Kyung-sang Nam-do. The scales are to measure stress, time management strategies and home managerial satisfaction. The analysis of the data used SPSS progiam. The significant results are as follows : 1. The stress level in patient nursing wiles in the families background varlables differed significantly from nursing wives's age, a housing size, suitable nursing charges, nursing helper-wanted method, family's nursing help, nursing wives's health, the source of paitents' hospital charges, household burden of hospital charges, patients' level of desease, nursing equipment being or nothing after out of hospital. The stress level showed the correlation nursing wives age, household burden degree. 2. The time management strategies and home managerial satisfaction on the stress level of nursing wives were higher stress level, lower time strategied and nursing wives were higher stress level, lower home managerial satisfaction. 3. Home managerial satisfaction on the time management strategies of nursing wives home managerial satisfaction was high according to time management strategies because system strategies, compromise strategies of time management strategies had significant diffrent. 4. The variables predicting nursing wives' home managerial satisfaction were for; they were placed according to household income, household budget, stress level, caregiving wives' health condition.

      • 좋은 죽음의 개념 분석

        Purpose: Death is not an option but a necessity in our life. However, there is little understanding of what a good death means. This study was conducted to analyze the concept of a good death by exploring attributes, antecedents, consequences and empirical criteria for a good death in literatures. Method : Reviewed materials were collected by searching for web cites such as PubMed, National Library of Korea, National Assembly Library. Walker and Avant's method(2004) was used for this concept analysis. Results : The findings showed that a good death includes seven attributes: dignity (personal respect and privacy), self-control (the right of choice, independence, clear consciousness), comfort (physical, psychological, spiritual). optimal relationships (supportive environment, proper care, restoration of relationships), appropriateness (sequence and proper age of death), preparation for death (completing unsettled tasks, leaving a legacy, positive attitude toward life, awareness and acceptance of death) and minimizing burden. Conclusion : This results will provide health professionals and people with a preliminary understandings of a good death. In addition, there is the need for further study to develop objective measurement of a good death and effective nursing intervention for the dying.

      • 12-O-Tetradecanoylphorbol-13-Acetate에 의한 HL-60 세포 분화유도중 Vimentin 유전자 전사조절에 대한 AP-1의 역할

        Purpose: To gain insight on the role of AP-1 in transcriptional regulation of vimentin gene during differentiation of HL-60 cells by 12-O-tetradecanoylphorbol-13-acetate (TPA), the levels of vimentin mRNA and AP-1 have been investigated with Northern blot hybridization and DNA mobility shift assay. Materials and Methods: HL-60 cells were grown in RPMI 1640 medium supplemented with 10% heat-inactivated fetal calf serum and antibiotics in a humidified 5% CO_2 at 37℃. Total RNA was prepared by a modification of the method of Karlinsey et al. Northern blot hybridization was performed by the method of Virca et al. EcoRI fragment of pVIM-GEM was used as probe for vimentin mRNA. DNA mobility shift assay was performed by the method of Lim et al. End labeled DNA probe(Upper strand, 5'-CGCTTGATGAGTCAGCCG- 3') for AP-1 binding activity was mixed with nuclear extracts in a 20 μl reaction volume containing 300 mM KCI, 60 mM HEPES, pH 7.9, 25 mM MgCI_2, 1 mM EDTA, 1 mM DTT, 60% glycerol, and 2 ㎍ of poly[dI-dC]. Results: TPA increased vimentin mRNA levels, with maximal stimulation reached at 24 hr. The level of vimentin mRNA was induced in proportion to the concentration of TPA. TPA-induced vimentin mRNA was almost reduced by actinomycin-D pretreatment. TPA-induced stimulation of vimentin gene was completely reduced by staurosporin pretreatment. In DNA mobility shift assay, AP-1 newly appeared at 24 hr during TPA-induced differentiation and was almost not detected after the pretreatment of staurosporin. Conclusions: These results suggest that the induction of vimentin mRNA during TPA-dependent differentiation in HL-60 cells may be mediated by protein kinases C signal transduction and AP-1 is important to transcriptional regulation.

      • Genistein, a soy phytoestrogen, prevents the growth of BG-1 ovarian cancer cells induced by 17β-estradiol or bisphenol A via the inhibition of cell cycle progression.

        <P>An endocrine disrupting chemical (EDC) is a global health concern. In this study, we examined the effects of genistein (GEN) on bisphenol A (BPA) or 17β-estradiol (E2)-induced cell growth and gene alterations of BG-1 ovarian cancer cells expressing estrogen receptors (ERs). In an in vitro cell viability assay, E2 or BPA significantly increased the growth of BG-1 cells. This increased proliferative activity was reversed by treatment with ICI?182,780, a well-known ER antagonist, while cell proliferation was further promoted in the presence of propyl pyrazole triol (PPT), an ERα agonist. These results imply that cell proliferation increased by E2 or BPA was mediated by ERs, particularly ERα. BPA clearly acted as a xenoestrogen in BG-1 ovarian cancer cells by mimicking E2 action. In contrast, GEN effectively suppressed BG-1 cell proliferation promoted by E2 or BPA by inhibiting cell cycle progression. E2 and BPA increased the expression of cyclin D1, a factor responsible for the G1/S cell cycle transition. They also decreased the expression of p21, a potent cyclin-dependent kinase (CDK) inhibitor that arrests the cell cycle in G1 phase, and promoted the proliferation of BG-1 cells. As shown by its repressive effect on cell growth, GEN decreased the expression of cyclin D1 augmented by E2 or BPA. On the other hand, GEN increased the p21 expression downregulated by E2 or BPA. Collectively, our findings suggest that GEN, a dietary phytoestrogen, has an inhibitory effect on the growth of estrogen-dependent cancers promoted by E2 or BPA.</P>

        일부 중,고등학교 리듬체조 선수와 지도자의 영양정보원 및 영양지식에 관한 연구

        The purpose of this study was to investigate the nutrition information sources and the nutrition knowledge in 41 rhythmic gymnasts aged 14.8±1.5 years and 17 coaches by questionnaires. Information about weight control, nutritional knowledge and its source, and food intake were collected by individual interview. Food intake was investigated with the 24 hours recall and two days dietary record methods. The results were as follows. All rhythmic gymnasts had experienced weight control. Most of them (97.6%) preferred `food restrict` as methods for weight control. Only fifty-one percent of rhythmic gymnasts had ever received the nutritional information. Their major information sources were parents & friends (76.2%). Eighty-two percent of rhythmic gymnasts wanted to obtain the nutritional information from the mass media and nutritionist & dietitian by the methods such like internet or nutrition counseling. Seventy-seven percent of coaches had ever counseled rhythmic gymnasts on nutrition, especially on the diet for weight control (92.3%). Their major information source were the mass media and curriculum. All coaches wanted to get the nutritional information from the nutritionist & dietitian and specialized books & dictionary by the methods such like internet or nutrition counseling. Total nutritional knowledge score of rhythmic gymnasts was 28.4 of 50 scores. Categorical score was the highest at the knowledge on `food source` and the lowest at the knowledge on `food exchange`. Total nutrition knowledge score of coaches was 37.6 of 50 scores, and higher than score of rhythmic gymnasts. Categorical score was the highest at the knowledge on `food sources` and the lowest on `food exchange` like rhythmic gymnasts. All nutrient intakes except vitamin C were higher in the group have received nutrition information compared with the group have not. In conclusion, it is very necessary to give rhythmic gymnasts and their coaches professional nutrition information by the information sources and methods they want. It could be helpful for improving the nutrition knowledge and enhancing performance of rhythmic gymnasts.
