Celiac disease (CD) is an immune-mediated enteropathy of small intestine diagnosed in both childhood and adulthood. CD is caused by gluten, which produces gliadorphin during its digestion. The enzyme dipeptidyl peptidase-4 (DPP4) breaks gliadorphin down nevertheless the last tripeptide remains and eventually inhibits DPP4, thus slowing down its process. Therefore, the idea is to produce an additional DPP4 enzyme which is crucial. Consequently, the functional DPP4 gene was cloned into pCDNA3 intermediate (FLAG+DPP4) vector and finally a recombinant plasmid pSB1C3 (Andersons promoters+FLAG+DPP4) was constructed using synthetic biology. Normally, a DPP4 inhibitor is used as a cure for diabetes. Another important concern was overexpression of DPP4, which might lead to diabetes, accordingly the work was also performed for the regulation of the DPP4 gene expression. In this regard, three types of Anderson promoters (strong, moderate and weak) were utilized to study the control overexpression. This is the first report of idealistic trial for control the exogenous DPP4 gene-expression by molecular biologic tools.

<P>Astragalus extract mixture HT042 is a standardized ingredient of health functional food approved by Korean FDA with a claim of “height growth of children.” HT042 stimulates bone growth rate and increases local IGF-1 expression in growth plate of rats which can be considered as direct stimulation of GH and its paracrine/autocrine actions. However, it remains unclear whether HT042 stimulates circulatory IGF-1 which also plays a major role to stimulate bone growth. To determine the effects on circulatory IGF-1, IGF-1 and IGFBP-3 expressions and phosphorylation of JAK2/STAT5 were evaluated in the liver after 10 days of HT042 administration. HT042 upregulated liver IGF-1 and IGFBP-3 mRNA expression, IGF-1 protein expression, and phosphorylation of JAK2/STAT5. HT042 also increased bone growth rate and proliferative zonal height in growth plate. In conclusion, HT042 stimulates bone growth rate via increment of proliferative rate by upregulation of liver IGF-1 and IGFBP-3 mRNA followed by IGF-1 protein expression through phosphorylation of JAK2/STAT5, which can be regarded as normal functioning of GH-dependent endocrine pathway. </P>
Predicting Three-Dimensional Gait Parameters with a Single Camera Video Sequence

Human gait reflects biomedical conditions and thus can potentially be used for identification. With the increasing utility of CCTVs for surveillance, there have been various attempts to recognize persons using gait image sequences from a single camera. We investigated the accuracy of estimating body segment lengths and joint angles during gait calculated from a video sequence using a gait database. We recruited 30 subjects and collected motion capture data during walking and extracted the trajectories of 17 body points. Principal component analysis (PCA) was applied to the collected gait. We implemented full gait cycle-based (FGC) PCA and gait-phase-specific (GPS) PCA. Three-dimensional poses were estimated from gait event frames using FGC-PCA and GPS-PCA. The estimated poses in discrete gait event frames were interpolated to estimate motion during a full gait cycle. The body pose from GPS-PCA was less sensitive to camera angles and smaller errors compared to FGC-PCA. The segment lengths of the upper arm (r=0.79), lower arm (r=0.63), upper leg (r=0.86), and lower leg (r=0.81) were highly correlated with the lengths obtained from the motion capture data. Three-dimensionally reconstructed human motion can reveal personal biometric information and has the potential to be used for human identification.
Pituitary Apoplexy After Leuprolide Therapy in a Breast Cancer Patient: A Case Report

Pituitary apoplexy (PA) is a clinical syndrome resulting from sudden hemorrhage and/or infarction of the pituitary gland. Recent reports documented the development of PA secondary to treatment with gonadotropin- releasing hormone (GnRH) agonists for prostate cancer. A 52-year-old woman visited our emergency room with a severe headache, occurred 1 day prior. She underwent breast-conserving surgery for breast cancer 1 month prior. She was currently undergoing radiation and hormone therapy, consisting of leuprorelin. Brain contrast-enhanced MRI revealed a pituitary adenoma with internal hemorrhage in the sellar and suprasellar areas. Pachymeningeal enhancement was observed along the retroclival and bilateral frontal areas. The patient was diagnosed with PA and aseptic meningitis. The patient underwent total excision via transsphenoidal surgery 8 days after admission. The patient was pathologically diagnosed with a pituitary adenoma with necrosis. On immunochemical staining, the tumor was positive for follicle-stimulating hormone. The follow-up MRI revealed no evidence of residual tumor or an improved pachymeningeal enhancement. She is currently undergoing follow-up at the neurosurgery and endocrinology outpatient departments with no noted complications. In breast cancer patients receiving GnRH agonist therapy, PA may be rare complication.

This study investigated the effect of center of mass in the passive bipedal walker. We made a SimMechanics model of a passive bipedal walker and compared it with experimental data of the real physical bipedal walker. In this simulation, the effect of gravity, contact force and joints were implemented. We obtained the foot clearance and knee angular velocity in accordance with the center of gravity location in simulation. The foot clearance during swing phase increased as the center of mass of thigh and shank move lower and higher, respectively. Angular velocity of the knee decreased as the center of mass of thigh and shank move higher and lower, respectively.

Gait is important motion that contains biological information. Individual gait pattern is determined by subject’s habit, body type and diseases. Motion capture system is used to measure joint motion but is limited to capture within a laboratory. Optical camera is portable, simple to configure and take photographs of movement of a person, while it produces only two-dimensional information. The purpose of the study was to estimate three-dimensional pose and joint angles from two-dimensional sequential images of walking motion. Three-dimensional trajectories of body markers from three subjects and five motions were pooled together and their mean pose and principal components were obtained. One of the motion capture data was projected to a camera plane by assuming a fixed camera position. Optimizations to calculate a weight matrix of the principal components of body pose were performed with and without a constraint of body segment length to match the plane projected marker positions. Mean three-dimensional errors (standard deviation) of estimated marker trajectory were calculated using measured marker trajectory. Three-dimensional errors with and without constraint of body segment length were 3.44(±0.65) cm and 3.25(±0.80) cm, respectively. Two-dimensional errors (standard deviation) of projected marker to image were calculated using original image. Two-dimensional errors were 1.63(±0.41) pixel and 1.40(±0.38) pixel respectively. Flexion and extension angle of knee was estimated as RMS error, 2.54degree and 2.28degree respectively.
