        The effects of BRL-50481 on ovalbumin-induced asthmatic lung infl ammation exacerbated by co-exposure to Asian sand dust in the murine model

        Asian sand dust (ASD), which mainly originatesin China and Mongolia in the spring and blows into Korea,can exacerbate respiratory and immunological diseases. This study aims to observe eff ects of co-exposure to ASD onovalbumin (OVA)-induced asthmatic lung infl ammation andof treatment with a phosphodiesterase 7 (PDE7) inhibitor ina mouse model. The challenge with OVA increased airwayhyperresponsiveness (AHR) and infl ammatory cell infi ltrationinto the lung tissue. Interleukin (IL)-13, tumor necrosisfactor-alpha, monocyte-protein-1, mucin, and antigen-specific IgE and IgG1 production increased in mouse serum. The co-exposure of ASD signifi cantly exacerbated theseeff ects in this asthma model. Notably, the administration ofa PDE7 inhibitor, BRL-50481 (BRL), signifi cantly reducedAHR, infi ltration of infl ammatory cells into the lungs, andthe levels of type 2 T helper cell-related cytokines, antigenspecific immunoglobulins, and mucin. Thus, the administrationof BRL ameliorated OVA-induced allergic asthmaticresponses exacerbated by co-exposure to ASD. This studysuggests that PDE7 inhibition can be a therapeutic strategyfor infl ammatory lung diseases and asthma via the regulation of T lymphocytes and reduction of IL-13, and, consequently,mucin production.

        Pulmonary inflammation caused by silica dioxide nanoparticles in mice via TXNIP/NLRP3 signaling pathway

        Background Silica dioxide nanoparticles (SiONPs) have been used for various medical applications, including therapeutics and imaging, and the use of SiONPs has increased gradually over the years. However, despite an increase in the use of SiONPs, not much is known about mechanism of action of SiONPs and their pulmonary toxicity. Objective The present study investigated the pulmonary toxicity of SiONPs and explored the underlying mechanism of action, primarily focusing on thioredoxin-interacting protein (TXNIP)/NOD-like receptor pyrin domain-containing 3 (NLRP3) in SiONPs-treated mice. We investigated the toxic effects of SiONPs in the lung of BALB/c mice administered 5, 10, and 20 mg/kg SiONPs for 3 days. Results Exposure to SiONPs markedly increased inflammatory cell counts, including those of neutrophils and macrophages, and levels of inflammatory mediators, such as interleukin (IL)-1β, IL-6, and tumor necrosis factor-α in a dose-dependent manner in the bronchoalveolar lavage fluid. Moreover, the inflammation was verified upon histopathological analysis. In addition, exposure to SiONPs increased the expression of TXNIP in a dose-dependent manner and, in turn, upregulated NLRP3 inflammasome proteins, which subsequently induced IL-1β production. Conclusion Collectively, exposure to SiONPs induced inflammation in the lungs of mice, which resulted in the activation of IL-1β production via the TXNIP-NLRP3 axis. Our results provide useful information on the pulmonary toxicity induced by SiONPs and provide insights into the underlying mechanism of action.

      • A genome-wide by PM<sub>10</sub> interaction study identifies novel loci for lung function near <i>BICD1</i> and <i>IL1RN-IL1F10</i> genes in Korean adults

        <P><B>Abstract</B></P> <P>Although several genome-wide interaction studies (GWIS) have been performed in specific European populations to understand the missing link between genetic and environmental factors for lung function, GWIS of Asian samples remain rare. Therefore, we performed a GWIS of exposure to air pollution to identify loci for lung function in Korean adult men. A total of 1826 adult men recruited from two health check-up centers were included in the analysis and the annual mean concentrations of ambient particulate matter with an aerodynamic diameter ≤10 μm (PM<SUB>10</SUB>) were used. In case of forced vital capacity (FVC), one SNP (rs12312730) that passed our genome-wide threshold of <I>p</I>int < 1 × 10–5 was detected in the intronic region of the <I>BICD1</I> gene on chromosome 12. In addition, we found two variants (rs6743376 and rs17042888) located near the <I>IL1RN-IL1F10</I> gene that were involved in the inflammatory response and associated with decreased FVC via interaction with PM<SUB>10</SUB> exposure. A stratified association analysis according to these SNP genotypes showed that PM<SUB>10</SUB> concentrations in subjects with one or two of the risk alleles, compared with those with the non-risk allele, were significantly correlated with a reduction in FVC. This pattern was replicated in another 892 Korean adult samples. The current study reports the first GWIS discovery in an Asian population: the <I>BICD1</I> and <I>IL1RN-IL1F10</I> genes may contribute to the decrease in FVC levels by interacting with PM<SUB>10</SUB> exposure.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Significant interactions between <I>BICD1 or IL1RN-IL1F10</I> and PM<SUB>10</SUB> for FVC were found. </LI> <LI> The several SNPs in these genes were more susceptible to FVC decline by PM<SUB>10</SUB>. </LI> <LI> For FVC, these interaction effects were reproducible in another sample. </LI> </UL> </P>

        BMS 수리모형하에서 시스템의 수리효과 및 고장강도함수의 모수추정

        Estimation problems of parameters of the failure process and the repair effect in repairable systems are considered. We propose estimation procedures in repairable systems without preventive maintenances. The fallure process is modeled by a proportional age reduction model (Brown. Mahoney, Sivazlian [5]) which is able to consider both aging and repair effects. Maximum likelihood method is used to estimate the repair effect and parameters of intensity function simultaneously. Simulations are performed to evaluate the accuracy of estimators. A numerical example is also presented.

        This paper deals with a new approach to the application of the LTIM(Low Thermal Inertia Molding) Concept for molding a plastic product. Thermoelectric modules are used to form a mold cavity. By controlling the polarity of power input to the thermo-electric modules, a mold cavity surface can be rapidly heated and cooled down. An experiment was conducted on the thermoelectric modules to test the feasibility of using these modules as a mold cavity. A prototype mold was designed for the experiment and the modules were controlled by a temperature control system designed in Goldstar Co. A general injection molding machine was interfaced with the temperature control system. Some tests were conducted to compare the LTIM mold with a conventional mold.

        This study was designed to examine the in vitro antibacterial activities and pharmacokinetics of enrofloxacin acetate (ENFXA) and enrofloxacin (ENFX). Two enrofloxacins had wide spectrum antibacterial activities against Gram positive and negative bacteria, and mycoplasma, showing potent antibacterial activities with low MICs (0.05-3.33 ㎍/㎖ for Gram positives, 0.002 ㎍/㎖ for Gram negatives and 0.02 ㎍/㎖ for mycoplasma). In order to investigate the profiles of two formulations of enrofloxacin we studied comparative pharmacokinetics of them in rabbits. Five rabbits were given 5 ㎎/㎏ body weight of each enrofloxacin formulation intravenously (i.v.) and orally (p.o.) in a cross-over study. Pharmacokinetic parameters of both formulations were calculated by the use of PCNONLIN, a computer program. Their bioavailibility in rabbits, the means of the area under the curve (AUC). After oral administration of each formulation in rabbits, the mean elimination half-lives (t_1/2,ke) were 3.25h (ENFX) and 4.32 h (ENFXA), and mean AUC 4.27 ㎍·h/㎖ (ENFX) and 6.21 ㎍·h/㎖ (ENFXA). Both enrofloxacin formulations seemed to have good tissue distribution and penetration as indicated by large volume of distribution: 4.76 1/㎏ for ENFX and 7.23 1/㎏ for ENFXA. With the results obtained in this study, ENFXA could be used in place of ENFX in rabbits.

        Comparative blook analysis was carried out to investigate the difference of blood constitient in four types of physical constitution. Glycine, valine, leucine, pheny, lalanine, histidine, arginine, lysine were measured. The results obtained were summarized as follows: 1. The value of glycine on the constitution didn't shown significant difference. 2. The value of valine on the constitution didn't shown significant difference. 3. The value of leucine on the constitution didn't shown significant difference. 4. The value of phenyalalnine on the constitution didn't shown significant difference, but between the value of SO-YANG-IN and TAE-EUM-IN showed significant difference. 5. The value of histidine on the constitution showed significant difference, and between the value of SO-YANG-IN and TAE-EUM-IN, of SO-YANG-IN and SO-EUM-IN showed significant difference. 6. The value of arginine on the constitution didn't shown significant difference, but between the value of SO-YANG-IN and TAE-EUM-IN showed significant difference. 7. The value of lysine on the constitution didn't shown significant difference.

        The transient left ventricular apical ballooning syndrome, also known as takotsubo cardiomyopathy is a recently described novel cardiac syndrome. This syndrome is characterized by transient asynergy of the ventricular apex or mid-ventricle in the absence of obstructive epicardial coronay artery disease. This report concerns two types of morphologic difference about left ventricular apex and mid-ventricle. In this case, we report two types of stress-induced cardiomyopathy with review of literatures.

        The nortropinones 11 (tropinone lla, N-isopropylnortropinone 11b, N-Carbethoxynortropinone 11c, N-furfurylnortropinone 11d, N-(p-methoxyphenyl)nortropinone 11e) were respectively synthesized by the treatment of acetonedicarboxylic acid 8 with, 2, 5-dimethoxytetrahydrofurane 9 in various amines 10 (methylamine 10a, N-isopropylamine 10b, ethylcarbamate 10c, furfurylamine 10d, p-anisidine 10e). The nortropane spirohydantoins 14 (tropane spirohydanttoin 14a, N-isopropylnortropane spirohydantoin 14b, N-Carbethoxynortropane spirohydantoin 14c, N-furfurylnortropane spirohydantion 14d, n_(p-methoxyphenyl) nortropane spirohydantoin 14e) were respectively synthesized by the treatment of synthesized nortropinones 11 (tropinone 11a 54%, N-isopropylnortropinone 11b 50%, N-Carbethoxynortropinone 11c 58%, N-furfurylnortropinone 11d 31%, N-(p-methoxyphenyl) nortropinone 11e, 70%) with potassium cyanide 12, ammonium carbonate 13.

        TALP-35 purified from human term placenta is known to increase microtubule polymerization and stabilize the polymerized microtubule. To examine the effect of TALP-35 on immune system this study was performed. MTT assay was performed to investigate the effect of TALP-35 on the proliferation of RAW264.7 cells. TALP-35 dose dependently suppress the proliferation of RAW264.7 cells at high concentration (above 1 μM) in unstimulated cells, in case of 10 μM TALP-35 treated cells the suppression was 25% but in stimulated cells it was only 15%. Cosedimentation assay was carried out to investigate whether TALP-35 can bind to tubulin of RAW264.7, monocyte/macrophage lineage of mouse, and polymerize it. TALP-35 polymerize the tubulin of RAW264.7 cells and sedimented in dose-dependent manner. To investigate the effect of TALP-35 on the expression of iNOS protein western blotting was performed. The expression level of iNOS was decreased dose dependently in high concentration of TALP-35 treatment. To examine the activity of iNOS, secreted NO was determined by method based on Griess reaction. Interferon-γ and LPS-stimulated production of NO from RAW264.7 cells was decreased dose dependently above 0.1 μM concentration of TALP-35 and 50% is decreased at 10μM of it. This study shows TALP-35 can control cytokine induced-iNOS expression therefore it might control inflammatory diseases.
