        SIRT5-related desuccinylation modification of AIFM1 protects against compression-induced intervertebral disc degeneration by regulating mitochondrial homeostasis

        Mitochondrial dysfunction plays a major role in the development of intervertebral disc degeneration (IDD). Sirtuin 5 (SIRT5) participates in the maintenance of mitochondrial homeostasis through its desuccinylase activity. However, it is still unclear whether succinylation or SIRT5 is involved in the impairment of mitochondria and development of IDD induced by excessive mechanical stress. Our 4D label-free quantitative proteomic results showed decreased expression of the desuccinylase SIRT5 in rat nucleus pulposus (NP) tissues under mechanical loading. Overexpression of Sirt5 effectively alleviated, whereas knockdown of Sirt5 aggravated, the apoptosis and dysfunction of NP cells under mechanical stress, consistent with the more severe IDD phenotype of Sirt5 KO mice than wild-type mice that underwent lumbar spine instability (LSI) surgery. Moreover, immunoprecipitation-coupled mass spectrometry (IP-MS) results suggested that AIFM1 was a downstream target of SIRT5, which was verified by a Co-IP assay. We further demonstrated that reduced SIRT5 expression resulted in the increased succinylation of AIFM1, which in turn abolished the interaction between AIFM1 and CHCHD4 and thus led to the reduced electron transfer chain (ETC) complex subunits in NP cells. Reduced ETC complex subunits resulted in mitochondrial dysfunction and the subsequent occurrence of IDD under mechanical stress. Finally, we validated the efficacy of treatments targeting disrupted mitochondrial protein importation by upregulating SIRT5 expression or methylene blue (MB) administration in the compression-induced rat IDD model. In conclusion, our study provides new insights into the occurrence and development of IDD and offers promising therapeutic approaches for IDD.

        Effects of supplementing sweet sorghum with grapeseeds on carcass parameters, and meat quality, amino acid, and fatty acid composition of lambs

        Objective: Sweet sorghum is an important forage crop for ruminants, especially in low rainfall areas. Grapeseeds are an abundant by-product of wine-making and contain bioactive substances that can improve the antioxidant capacity of meat. We examined the effect of sweet sorghum forage with supplementary grapeseeds on carcass and meat quality in lambs. Methods: Twenty-eight Small-tailed Han lambs (body weight = 19.1±1.20 kg), aged 3 to 4 months, were penned, and fed individually. The lambs were divided into four groups (n = 7 each) and were offered one of four diets: i) sweet sorghum silage; ii) sweet sorghum silage + grapeseeds; iii) sweet sorghum hay; and iv) sweet sorghum hay + grapeseeds. The grapeseeds were added to the concentrate at 6% DM and the diets were fed for 100 d. Results: Sweet sorghum silage tended (p = 0.068) to increase hot carcass weight, while grapeseeds tended (p = 0.081) to decrease dressing percentage without affecting other carcass parameters. Lambs consuming supplementary grapeseeds increased (p<0.05) meat redness and tended to decrease (p = 0.075) concentration of methionine in meat. Lambs consuming sweet sorghum silage increased (p<0.001) water content of the meat and had a lower (p<0.05) concentration of n-6 polyunsaturated fatty acids (PUFA) and n-6:n-3 PUFA ratio than lambs consuming sweet sorghum hay. Saturated fatty acids content in meat was lowest (p<0.05) in lambs consuming sweet sorghum silage with grapeseeds. Lambs with supplementary grapeseeds tended (p<0.10) to increase eicosapentaenoic acid and docosahexaenoic acid and have a lower thrombogenic index than lambs not consuming grapeseeds. Conclusion: It was concluded that sweet sorghum with supplementary grapeseeds fed to lambs; i) improved the color of the meat to be more appetizing to the consumer; ii) tended to improve the fatty acids composition of the meat; and iii) lowered thrombogenic index of the meat.

        On-line identifying stator winding short-circuit approach for a submersible motor based on faulty current monitoring

        To monitor, diagnose, and suppress the inter-turn short-circuit faults (ITSCFs) of a submersible motor, an approach for the on-line identification of its winding faults has been proposed based on monitoring the stator current. First, an ITSCF model with the global leakage referred to the stator is given. With this model, the detection parameter, which is equal to the ratio of the turns of the fault windings to the total turns of the windings in the healthy phase, can be derived. Second, a faulty model described by the fourth order state-space equation of the motor with a winding fault has been given. Based on sampled stator voltage and stator current, the detection parameter has been solved and used to estimate the location and the turns of inter-turn short-circuit windings of the motor in real time. The accuracy and the robustness of the proposed approach has been illustrated with a 1.5 kW motor that is fed by a 10 kW inverter. Experiment shows that the identification accuracy in terms of the number of the ITSCF windings of the motor stator is less than 3. It can give a reference for the on-line diagnose the ITSCFs of the stator windings of a submersible motor that works in 2 km deep well.

        The Impact of Foreign Ownership on Stock Volatility in Indonesia

        This study documents the negative relationship between foreign ownership and the future volatility of Indonesian stocks. The calming effect of foreign ownership is present before, during, and after the Asian financial crisis. It is independent of gross and net foreign trading and the stock’s historical volatility. The effect increases with the level of foreign holdings. The findings are contrary to the volatility impact of institutional ownership in developed markets, and indicate the presence of different economic mechanisms leading to the opposite volatility impact from foreign ownership and foreign trading.

        Estimating the Effects of Multipath Selection on Concurrent Multipath Transfer

        Multi-mode device which combines multiple access technologies into a device will offer more cost-effective solution than a sole access implementation. Its concurrent multipath transfer (CMT) technology can transmit media flows over multiple end-to-end paths simultaneously, which is essential to select at least two paths from all available paths. At real networks, different paths are likely to overlap each other and even share bottleneck, which can weaken the path diversity gained through CMT. Spurred by this observation, it is necessary to select multiple independent paths as much as possible to avoid underlying shared bottleneck between topologically joint paths. Recent research in this context has shown that different paths with shared bottleneck can weaken the path diversity gained through CMT. In our earlier work, a grouping-based multipath selection (GMS) mechanism is introduced and developed. However, how to estimating the selection is still to be resolved. In this paper, we firstly introduce a Selection Correctness Index (SCI) to evaluate the correctness of selection results in actual CMT experiment. Therefore, this metric is helpful to discuss and validate the accuracy of the output paths. From extensive experiments with a realized prototype, the proposed scheme provides better evaluation tool and criterion in various network conditions.

        Shape Memory Effect of TiNi-based Springs Trained by Constraint Annealing

        This study investigated shape memory effect (SME) behaviors of TiNi-based shape memory alloys springs trained by constraint annealing treatments. The results indicated that SME behaviors were determined by constraint annealing sequences and the chemical composition of alloys. Springs made by Ni-rich TiNi binary alloys showed both single-way SME (SW-SME) and two-way SME (TW-SME) depending on the constraint annealing sequences, while those made by TiNiCu alloys only showed SW-SME. The mechanism and stability of TW-SME induced by constraint annealing were discussed. The occurrence of TW-SME was found to be associated with oriented internal coherent strain caused by preferential Ti3Ni4 precipitates. A special method,which could achieve a stable post-inherent TW-SME and size selectivity, was proposed to provide an effective approach for devising actuators in Ni-rich alloys.

        Research progress of single-atom coating and its application prospect in protective coatings

        Coating technology is the most economical and efficient approach to retard metal corrosion, and newcomposite coatings prepared by adding monoatomic fillers to a polymer matrix have excellent corrosionresistance and good mechanical properties. The single-atom coating formed by the immobilization ofmetal atoms alone on the matrix or filler represents the maximum utilization of atoms at the presentstage, which has unique coordination properties, active sites, and synergistic effects with the support. Compared with traditional fillers, single-atom fillers not only have the advantages of supporting fillersbut can also provide monatomic metals to form passivation films, promote the progress of catalytic passivationreactions, and combine organic compounds to generate new composite materials. This paperfocuses on the preparation methods and mechanisms of single-atom coatings in the field of batteriesand catalysts as well as the application of composite materials prepared by combining single-atom materialswith polymer matrices in the field of metal protection. The preparation methods, application prospects,and coating characteristics of various single-atom composites were summarized. In addition,we discuss the potential applications of single-atom coatings in the design of future composite coatings.

        Recombinant Goat VEGF164 Increases Hair Growth by Painting Process on the Skin of Shaved Mouse

        To detect goat vascular endothelial growth factor (VEGF)-mediated regrowth of hair, full-length VEGF164 cDNA was cloned from Inner Mongolia cashmere goat (Capra hircus) into the pET-his prokaryotic expression vector, and the recombinant plasmid was transferred into E. coli BL21 cells. The expression of recombinant $6{\times}his-gVEGF164$ protein was induced by 0.5 mM isopropyl thio-${\beta}$-D-galactoside at $32^{\circ}C$. Recombinant goat VEGF164 (rgVEGF164) was purified and identified by western blot using monoclonal anti-his and anti-VEGF antibodies. The rgVEGF164 was smeared onto the dorsal area of a shaved mouse, and we noted that hair regrowth in this area was faster than in the control group. Thus, rgVEGF164 increases hair growth in mice.

        fects of Dietary Soy Intake on Maternal Thyroid Functions and Serum Anti-Thyroperoxidase Antibody Level During Early Pregnancy

        Soy and its isoflavones have been suggested to suppress thyroperoxidase (TPO), induce goiter, inhibit deiodinase, and modulate immune functions. This study initially investigated the effects of dietary soy consumption on maternal thyroid functions and anti-TPO antibody (TPOAb) production during early pregnancy. Data were collected through questionnaire from 505 women enrolled during early pregnancy by random sampling in Shenyang, China. Based on soy intake frequency, the subjects were divided into three groups (frequent [three or more times per week], conventional [more than twice per month but less than three times per week], and occasional [two or fewer times per month]). Serum thyrotropin (TSH), free thyroxine (FT_4), and TPOAb were measured by chemiluminescence immunoassay. Additionally, the concentrations of two primary isoflavones (daidzein and genistein) and creatinine were assessed in the spot urine samples from representative subjects (about 20%) randomly selected from the three groups. The percentages of frequent, conventional, and occasional consumers were 18.6%, 62.6%, and 18.8%, respectively. No difference was found in age, medical records, family history of thyroid diseases, serum FT_4, TSH, and TPOAb levels, TPOAb-positive percentages, or prevalence of thyroid dysfunctions among the groups. Both urinary daidzein and genistein levels were significantly higher in the frequent consumers compared with the other two groups. No correlations were found between urinary isoflavone levels and serum FT_4 or TSH. Urinary isoflavone levels were not significantly different between TPOAb-positive and -negative women among the randomly selected representative subjects. On the whole, our findings suggest dietary soy consumption during early pregnancy is not associated with the development of thyroid dysfunction or autoimmunity.

        Topology-aware Virtual Network Embedding Using Multiple Characteristics

        Network virtualization provides a promising tool to allow multiple heterogeneous virtual networks to run on a shared substrate network simultaneously. A long-standing challenge in network virtualization is the Virtual Network Embedding (VNE) problem: how to embed virtual networks onto specific physical nodes and links in the substrate network effectively. Recent research presents several heuristic algorithms that only consider single topological attribute of networks, which may lead to decreased utilization of resources. In this paper, we introduce six complementary characteristics that reflect different topological attributes, and propose three topology-aware VNE algorithms by leveraging the respective advantages of different characteristics. In addition, a new KS-core decomposition algorithm based on two characteristics is devised to better disentangle the hierarchical topological structure of virtual networks. Due to the overall consideration of topological attributes of substrate and virtual networks by using multiple characteristics, our study better coordinates node and link embedding. Extensive simulations demonstrate that our proposed algorithms improve the long-term average revenue, acceptance ratio, and revenue/cost ratio compared to previous algorithms.
