        An Efficient System for the Expression and purification of Yeast Geranylgeranyl Protein Transferase Type Ⅰ

        To purify the geranylgeranyl protain transferase type Ⅰ(GGPT-I) efficiently, a gene expression system using the pGEX-4T-1 vector was constructed. The cal1 gene, encoding the βsubunit of GGPT-I, was subcloned into the pGEX-4T-1 vector and co-transformed into E. coli cells harboring the ram2 gene, the α subunit gene of GGPT-I. GGPT-I was highly expressed as a fusion protein with glutathione S-transferase (GST) in E. coli, purified to homogeneity by glutathione-agarose affinity chromatography, and the GST moiety was excised by thrombin treatment. The purified yeast GGPT-I showed a dose-dependent increase in the transferase activity, and its apparent K?? value for an undecapeptide fused with GST (GST-PEP) was 0.66 μM and the apparent K?? value for geranylgeranyl pyrophosphate (GGPP) was 0.071 μM.

        유아교육기관에서의 미세먼지 관련 교육현황 및 교사인식

        본 연구는 유아교육기관에서 현재 이루어지고 있는 미세먼지 관련 교육현황을 살펴보고, 이를 교육하는 교사의 미세먼지 교육에 대한 인식을 알아보는 데 그 목적이 있다. 이를 위해 2018년 7월에 예비조사를 하고 8~9월 두 달간 전라도에 소재한 유아교육기관 담임교사 302명을 연구대상으로 본조사를 실시하였다. 연구결과, 80% 이상의 유아교육기관에서 미세먼지로부터의 대응방법에 중점을 두고 교육을 실시하고 있었고 교육은 주로 미세먼지가 심한 날, 이야기 나누기 형식으로, 연관되는 단원에 따라 부분적으로 언급하며 실시하는 것으로 나타났다. 또한 체계적인 계획보다는 즉흥적이고 한정된 내용과 방법으로 교육이 진행되고 있음을 알 수 있었다. 한편, 교사들의 미세먼지 교육에 대한 인식을 살펴보았을 때, 교사들은 미세먼지에 대한 심각성을 인식하고 있었으며, 체계적인 미세먼지 관련 교육 또한 매우 필요하다고 느끼고 있다는 결과가 나타났다. 그러나 미세먼지 관련 교육에 대한 내용과 방법에 대한 지식의 부족과 관련 교재 교구의 부족으로 인해 미세먼지 교육에 어려움을 느끼고 있었다. 이에 앞으로의 효과적인 미세먼지 관련 교육을 위해서는 적절한 교사교육 실시와 관련 교재 교구의 개발 및 지원이 필요함을 알 수 있었다. The purpose of this study was to find out the status of fine dust education in educational institutes for young children and the awareness of teachers about fine dust education. For this, 302 teachers of kindergarten and day cre centers were participated in this study. The results of this study are summarized as follows. First, the current status of education on fine dust has been found to focus on how to deal with fine dust, and to partially mention that way depending on the unit involved in a day with high dust level. Moreover, the detailed items of fine dust education are more biased toward one response, indicating that education is being conducted in more impromptu manner and content rather than systematic planning. Second, the results of recognizing fine dust related education show that teachers are aware of the seriousness of fine dust in Korea and feel that education related fine dust is very necessary. In addition, in response to difficulties in fine dust education, the respondents said they lacked knowledge of the contents, methods of effective education and the necessary teaching materials for fine dust education. In other words, the results showed that teachers education on fine dust and the development of teaching materials related to fine dust education are needed for the future education against fine dust.

        “Gattaca” and the Problem of Genetic Enhancement

        Andrew Niccols's movie Gattaca (1997) inspired the formulation of the “Gattaca Argument” concerning the negative outcome of biotechnology, which has since been critiqued especially in the context of transhumanism and posthumanism. According this argument the development of genetic enhancement will produce a genetic discrimination and lead us to serious form of inequality. However, in particular transhumanists deny that here are reasons to worry and advocating instead the transformation of human condition in terms of genetic enhancement. Moreover, they question that genetic enhancement will necessarily lead to social inequality. In what follows, we will reexamine the Gattaca Argument and its critiques based on the movie in order to reassess the role the movie plays in the subsequent scholarly discussion. We will argue that existing critiques fall short of capturing the problem posed in the movie – the problem of the inhumane. Based on a hermeneutic approach to the movie we will both reconstruct the arguments and evaluate the transhuman counterarguments in terms of modern history of philosophical ideas.

        Quercetin-3-glucuronide 함유 약용 식물의 pentobarbital 유도 수면모델에서 수면증진 활성

        본 연구에서는 GABA 수용체에 작용하여 진정, 불안 완화 및 항경련 그리고 수면 활성에 효과가 있다고 보고된 Q3G를 함유한 식물 소재를 탐색하여 pentobarbital 유도 수면 실험에서 수면증진 활성을 평가하였다. Q3G는 GABA<SUB>A</SUB>-BZD 수용체에 높은 결합 활성을 보이는 것으로 확인되었으며, Q3G를 20 mg/kg 투여하였을 때 정상 대조군과 비교하여 수면 잠복 시간과 수면 시간에서 유의적인 차이를 보였다. 국내에 자생하는 식물 중 Q3G를 다량 함유하고 있는 식물을 탐색한 결과, 병풍쌈, 단풍터리풀, 며느리배꼽이 각각 10.95, 14.10, 45.37 mg/g으로 높은 Q3G 함량을 보였다. 세 가지 식물 추출물 모두 정상 대조군과 비교하여 유의적인 수면의 증가를 보였으며, 며느리배꼽 추출물이 가장 우수한 수면 증진 활성을 보였다. 며느리배꼽 추출물의 수면 증진 활성은 GABA<SUB>A</SUB>-BZD 수용체와의 상호작용에 기인한 것으로 사료된다. 며느리배꼽 추출물은 수면 증진 활성을 가지며, 라디칼 소거활성 또한 우수한 소재이므로 향후 수면 장애를 개선하는데 도움을 줄 수 있는 기능성 원료로서의 활용 가능성이 높다. The purpose of this study was to evaluate the sleep-promoting activities of plant extracts by screening domestic plants with a high quercetin-3-glucuronide (Q3G) content. The Q3G (20 mg/kg) group had a significantly improved sleep latency time (p<0.001) and sleep duration time (p<0.05) compared to that of the normal group. In addition, Q3G has a high binding affinity to the benzodiazepine site of the GABA<SUB>A</SUB> receptor and thus promotes sleep through GABAergic action. Extracts of Filipendula multijuga Max. (14.10 mg/g), Cacalia firma Kom. (10.95 mg/g), and Persicaria perfoliate (45.37 mg/g) had higher Q3G contents compared to that of other domestic plants. The Persicaria perfoliate extract exhibited the highest sleep-promoting activity among the three extracts. Furthermore, it had a significantly higher polyphenol (p<0.05) and flavonoid (p<0.05) contents among the three extracts and showed excellent radical scavenging abilities. Therefore, the Persicaria perfoliate extract has great potential as a functional material for improving sleep disorders.

        보건소 보건의료직 종사자들의 감정노동, 자기효능감 및 소진의 관계

        Purpose: The purpose of this descriptive, cross-sectional study was to examine relationships among emotional labor, self-efficacy, and burnout of employees in public health centers. Factors that influence burnout of workers were also assessed. Methods: One hundred sixty six workers in public health centers completed a pack of self-report questionnaires. Collected data were analyzed using descriptive statistics, t-test, one-way ANOVA along with $Scheff{\acute{e}}$ test, and stepwise multiple regression. Results: Employees in public health centers seemed to experience emotional labor to some extent. There were statistically significant differences in burnout depending on the age of workers. Employees' burnout had a positive relationship with emotional labor and a negative relationship with self-efficacy. Factors influencing burnout of employees in public health centers were emotional dissonance, surveillance & monitoring of organization, age, and self-efficacy. Conclusion: Considering the findings from this study, health care providers need to develop effective interventions which increase the level of self-efficacy and decrease certain types of emotional labor for employees in public health centers and to help them effectively manage burnout.
