        흰쥐 골격근의 노화성 위축에 대한 운동훈련의 영향 : III. 노화 흰쥐에 적용한 단기간의 운동훈련의 영향 III. Effect of short Term Exercise Traning for Senile Rat

        본 실험은 골격의 위축이 일어난 노화 흰쥐를 사용하여 단기간동안 트레드밀 훈련과 웨이트 트레이닝을 시켰을 때 골격근에 나타나는 변화들을 고찰하고자 고안되었다. 출생 후 24개월 된 수컷 흰쥐를 사용하여 대조군과 트레드밀 훈련군, 웨이트 트레이닝군으로 구분하여 각 군별로 4주간 훈련시켜 각 군의 골격근은 조직 및 조직화학, 미세구조 및 세포화학적 연구 및 입체해석학적 연구에 적용하였다. 각 훈련 기간동안 개체의 체중과 근육의 무게는 큰 변화가 없었고, 근섬유의 분포양상은 트레드밀 훈련군에서는 ⅡB형과 ⅡC형이 증가를 한 반면 ⅡA형은 감소를 하였고 Ⅰ형의 근섬유 조성은 변화가 없었다. 웨이트 트레이닝 후에는 ⅡA와 ⅡB형이 증가를 한 반면 ⅡC형은 감소하였다.그러나 모든 근섬유 조성의 변화에는 유의성이 없었다. 노화 대조군의 조직구조 및 미세구조의 특징은 노화색소의 출현, 약한 수축대의 출현, 근섬유들의 분할현상 등이 관찰되었다. 트레드밀 훈련 후에는 손가락형의 돌출부를 포함한 근초의 불규칙성이 나타났으며, 밝은띠의 확장으로 인한 Z-선의 불규칙한 배열, 공포화, 장축과 평행한 근섬유의 분할현상이 나타났으며, 웨이트 트레이닝 후에는 많은 용해소체들과 함께 포식소자를 함유한 자가용해소체들이 나타났으며 심한 수축대가 나타났고 근원섬유들의 배열이 와해되는 현상도 나타났다. 조직화학 및 세포화학적인 측면에서 succinic dehydrogenase와 acid phosphatase는 활성도의 변화가 거의 없었으며, Mg++-ATPase의 활성은 훈련 후 약간 감소하는 경향이었다. 미세구조의 입체해석학적 분석에서는 측량한 구조물들의 체적밀도와 수밀도는 변화가 거의 없었고 사립체 내막의 표면밀도만 트레드밀 훈련군에서 증가되었다. 이상의 결과에서 볼 때 단기간의 지구력 훈련을 실시할 경우는 노화 골격근은 잘 적응하여 골격근의 위축 방지에 효율적이었으나 웨이트 트레이닝은 노화 골격근의 위축방지에 영향을 주지 못하였다. The present study was designed to examine effect of short term treadmill and weight-training on aging atrophy in the rat skeletal muscle. Male rats of 24 months old were used. Each groups included control, treadmill and weight-training for 4 weeks by using treadmill apparatus and body press apparatus. The histo and cytochemical, ultrastructural and stereological changes in senile skeletal muscles of the rat were observed in the present study. During the training period the body weight and muscular weight in all groups remained constant. The volume density of muscle fiber type ⅡC and ⅡB were increased, that of type ⅡA was decreased, but type Ⅰ remained constant in treadmill-training group. In weight-training rat, the density of type ⅡA and ⅡB were increased, but those of type ⅡC was decreased. But, all changes of muscle fiber type is not significant. Senile control group some usual formation of mild contraction band, lipofuscin pigment and muscular splitting were observed. After treadmill-training, histological and ultrastructural changes occured in the muscle fiber, such as irregularity of the sarcolemma, interfibrillar vacuolization, longitudinal splitting, and widened Ⅰ-band. After weight-training, the changes occured in the trained muscle fiber, such as apearance of many lysosomes and autophagic vacuoles, severe contraction band, and breakup of myofibrils. Histo and cytochemical studies showed that the activities of succinic dehydrogenase and acid phosphatase remained constant, activities of Mg++-ATPase decrease with training. Stereological changes were not observed in the volume and numerical density of all subject component, but the surface density of mitochondrial inner membrane was increased with treadmill-training. These experimental results suggested that the endurance training during short-term may result in the adaptible response in senile skeletal muscles. On the other side, weight-training is bad for senile skeletal muscle.

      • 어육단백질의 냉동변성에 관한 연구 : Ⅲ. 동결 저장중 방어 보통육과 혈합육 근원섬유 단백질의 단백질 상호작용, 용해도 점도 및 Ca-ATPase활성의 변화 Ⅲ. Changes of Protein-Protein Interaction, Solubility, Viscosity and Ca-ATPase Activity of Myofibrillar Protein from Ordinary and Dark Muscle in Yellowtail during Frozen Storage

        The denaturation mechanism of the protein during freezing of myofibrillar protein extracted from the ordinary and dark muscle of yellowtail (Seriola quinqueradits) were investigated by analyzing the Protein-Protein interaction, Solubility, relative viscosity and Ca-ATPase activity. The rate of protein-protein interaction and inactivation of Ca-ATPase were the first reaction against the frozen storage time. The rate of protein-protein interaction in ordinary myofibrillar protein was 1.14-2.24 times as large as that of dark one, but Ca-ATPase was conversely. A similar relation-ship between ordinary and dark myofibrillar protein was recognized in relative viscosity and solubility, but the extent of decreased relative viscosity was large than solubility. The exposure of hydrophobic residures and protein-protein interaction were significantly related with the freezing denaturation of myofibrillar protein.

      • 烏貝散이 HCI-aspirin 으로 유발된 십이지장궤양에 미치는 조직학적 영향

        오패산이 HCI-aspirin tm으로 유발된 rat 십이지장궤양에 미치는 조직학적 영향을 관찰하기 위하여, 오패산을 1,3 및 5일 동안 경구투여하고 십이지장의 조직학적 변화를 관찰하였던 바 다음과 같은 결과를 얻었다. 실험 전 기간동안 대조군에서는 십이지장 융모의 위축과 붕괴가 관찰되었으며, 조직학적으로는 점막상피의 변성 및 탈락이 관찰되었고 점막고유층에서는 염증세포의 침윤(infilteration)의 소견이 관찰된 반면 오패산 투여군의 투여 1일에서는 대조군과 유사하게 관찰되었으나, 투여 3일후부터 점막상피의 첨부에서 경미한 카타르성 물질이 관찰된 이외에 정상군과 유사한 소견을 나타내었다. 이상에서 오패산이 십이지장궤양의 치유를 촉진시키는 것으로 관찰되었다. In order to study, the histological effects of Opeaesan on the duodenal ulcer induced by HCI-aspirin tm in rate, experiments were done by oral administration and observed histological profiles of the ulcerative lesions. Ulcerative lesions were unmerously detected in control groups especially in pylorus-duodenal junctions. Histologically, very severe injury to duodenal epithelium wer observed in control groups and these injuries were increased with time intervals. But in the Opaesan-administration groups, no gross lesion of ulcer were demonstrated and histologically minor injury to the mucosal epithelium were observed. Among the Opaesan treatment groups, most slight injuries to duodenal mucosa were detected in 5 days after administration when the histological profiles were similar to that of normal groups. According to these result, it is suggested that Opaesan should be used in treatment of the duodenal ulcer but the funcitonal mechanisms were unknown.

        <P>Mesh-based computer generated hologram enables realistic and efficient representation of three-dimensional scene. However, the dark line artifacts on the boundary between neighboring meshes are frequently observed, degrading the quality of the reconstruction. In this paper, we propose a simple technique to remove the dark line artifacts by matching the phase on the boundary of neighboring meshes. The feasibility of the proposed method is confirmed by the numerical and optical reconstruction of the generated hologram.</P>

      • Application of Artificial Neural Network method for deformation analysis of shallow NATM tunnel due to excavation

        Currently an increasing number of urban tunnels with small overburden are excavated according to the principle of the New Austrian Tunneling Method (NATM). For rational management of tunnels from planning to construction and maintenance stages, prediction, control and monitoring of displacements of and around the tunnel have to be performed with high accuracy. Computational method tools, such as finite element method, have been and are indispensable tool for tunnel engineers for many years. It is, however, a commonly acknowledged fact that determination of input parameters, especially material properties exhibiting nonlinear stress-strain relationship, is not an easy task even for an experienced engineer. Use and application of the acquired tunnel information is important for prediction accuracy and improvement of tunnel behavior on construction. Artificial Neural Network (ANN) model is a form of artificial intelligence that attempts to mimic behavior of human brain and nervous system. The main objective of this paper is to perform the deformation analysis in NATM tunnel by means of numerical simulation and artificial neural network (ANN) with field database. Developed ANN model can achieve a high level of prediction accuracy.

        Protective Effects of Chlorella vulgaris on Liver Toxicity in Cadmium-Administered Rats

        The biochemical mechanisms of Chlorella vulgaris protection against cadmium (Cd)-induced liver toxicity were investigated in male Sprague-Dawley rats (5 weeks of age, weighing 90–110 g). Forty rats were randomly divided into one control and three groups treated with 10 ppm Cd: one Cd without Chlorella (Cd-0C), one Cd with 5% Chlorella (Cd-5C), and one Cd with 10% Chlorella (Cd-10C) groups. The rats had free access to water and diet for 8 weeks. Body weight gain and relative liver weight were significantly lower in the Cd-0C group than in Cd-5C and Cd-10C groups. Rats in the Cd-0C group had significantly higher hepatic concentrations of Cd and metallothioneins (MTs) than in the Cd-5C or Cd-10C group. The hepatic MT I/II mRNA was expressed in all experimental rats. MT II was more expressed in the Cd-5C and Cd-10C groups than in the Cd-0C group. Morphologically, a higher level of congestion and vacuolation was observed in the livers of the Cd-0C group compared to those of the Cd-5C and Cd-10C groups. Therefore, this study suggests that C. vulgaris has a protective effect against Cd-induced liver damage by reducing Cd accumulation and stimulating the expression of MT II in liver. However, the details of the mechanism of C. vulgaris on liver toxicity remains to be clarified by further studies.

        가열조리가 복어 추출물 함질소화합물의 조성에 미치는 영향

        수산동물조직 추출물중의 함질소화합물은 생체 중의 대사에 중요한 역할을 할 뿐만 아니라 맛을 좌우하는 성분으로도 잘 알려져 있다. 본 연구는 복어를 시료로 하여 냉동복어의 알맞은 해동조건을 검토하고 복어추출물중의 함질소화합물을 분석하므로서 그 독특한 정미효과와 관련있는 성분을 검색하기 위하여 몇 가지 조리방법별로 아미노산, 핵산관련 화합물 및 유기염기의 조성을 비교 분석하였기에 보고한다. 1) 해동방법별로 냉동복어의 일반성분, drip량, pH를 측정한 결과, 해동소요 시간이 가장 짧은 유수(15± 0.5℃) 해동이 가장 효과적이었다. 2) 생시료와 냉동시료의 추출물중외 총질소의 양은 거의 차이가 없었으며 가열함으로써 그 양은 증가하였다. 조리방법으로는 물을 가하여 가열한 것이 물을 가하지 않고 가열한 것보다 더 높은 질소의 양을 보였다. 3) 추출물중의 유리아미노산의 조성을 검토한 결과, taurine, lysine, glycine, alanine이 전체 아미노산 중 약 70%을 차지하였으며, 냉동시료가 생시료에 비하여 높았고, 가열 조리함으로서 추출물중의 유리아미노산의 함량은 증가하였다. 그리고 가열방법으로는 microwave에 의해 가열한 것에 비하여 직화가열한 것이 유리아미노산 함량이 더 많았다. 4) 추출물중에 함유되어 있는 핵사관련화합물중에는 IMP와 ADP의 함량이 높았고, 이 두 성분은 조리방법에 따라 가열함으로서 감소하는 경향을 나타내었다. 5) 총 creatinine의 함량은 총질소의 양에 대해 약50%를 차지하였으며, glycinebetaine, TMA, TMAO는 미량에 불과하였다. 6) 복어의 추출물중에 분포하는 함질소 화합물의 조성에 비추어, creatinine계 화합물, taurine과 lysine, glycine 및 alanine등의 유리아미노산 및 IMP등이 복어의 맛에 기여하는 중요한 성분임을 알았다. Globefish, Fugu xanthopterus, known to have a severe toxin, is one of the favorite food in Korea and Japan when the toxic part is removed. In this paper, the effect of cooking on the composition of nitrogenous components in the extractives from globefish cooked by various methods such as boiling, steaming and microwave oven cooking has been investigated and the changes of the taste compounds originated from the nitrogenous components in the extractives were discussed. When the sample fish was analysed for general composition, drip amount and pH by the different methods of thawing, the method effective method was the running water thawing. Total nitrogen content in raw globefish and the frozen globefish was not different, and the nitrogen content was increased with the heat treatment. It seemed that the nitrogen content was higher in the extract from the boiled globefish than that of the steamed globefish. Taurine, lysine, glycine and alanine were occupied about 70% of the total free amino acids. Total free amino acid content was higher in the extracts from the frozen sample than those from the raw sample. The amount of free amino acids was increased when the globefish soup cooked under the direct-heat cooking than in the microwave oven-heat cooking. Among nucleotides in the extracts from the thawed and cooked fishes, IMP and inosine contents were increased, and the both components were decreased with the heating time and by the heating method. The content of total creatinine-nitrogen were 50% of the total nitrogen content of the extracts, but the concentration of glycinebetaine, TMA and TMAO were only a few amounts. It could be concluded that total creatinine components, including free amino acids such as taurine, lysine, glycine and alanine, and IMP might be the important components contributing to the taste of the cooked globefish.

        LED 디스플레이용 유연 보드의 자동 펀칭 시스템 연구

        본 논문에서는 플라스틱 평판에 핀 홀을 생성하여 문자나 이미지를 표현하는 디스플레이 보드를 자동으로 제작하는 펀칭시스템을 개발하였다. 핀 홀을 생성한 평판은 가장자리에 설치한 다양한 색상의 발광다이오드 빛을 반사하여 미려한 디스플레이 보드에 사용된다. 펀칭시스템은 정밀한 위치 및 깊이 생성을 위해 총 4개의 엑추에이터를 사용하였다. 디스플레이 보드 상의 글자나 이미지의 반사효과와 빠른 생산성을 위하여 정밀한 구동 기구부를 개발하고 PID 제어 알고리즘을 적용하였다. 그리고 사용자가 반사되는 글자의 모양이나 이미지를 구성하는데 도움을 줄 수 있는 GUI 프로그램과 PC기반의 제어시스템을 구성하였다. 또한 펀칭속도와 생성된 핀 홀의 깊이를 검사하여 본 시스템의 성능을 검증하였다. In this paper, a new automatic punching system that generates pinholes expressing texts or images on a plastic plate is developed. The pin-holed plate is used as a new glamorous display board reflecting colourful lights from the light emitting diode (LED) installed on the edge side of the plate. The punching system has four actuators which work together to make multiple holes with accurate position and depth on the plastic plate. For even reflection of the lights from texts or images on the board and fast production of the pin-holed boards, we developed an accurate actuating structure of the system cooperating with a PID control algorithm. We also built a GUI-based integrated control system to help users easily design luminous texts or images on the plastic plate. Also, we conducted a performance test of the system to verify the punching speed and depth control of the pin holes on the plastic plate.
