Metastasis associated genomic aberrations in stage II rectal cancer

Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

The simple and effective analytical method for the quality control of a novel scalp tonic formulation has been developed and optimized in terms of HPLC conditions and sample preparation method, meanwhile, the optimization of preparation condition was using response surface methodology (RSM) based on central composite design (CCD). Oleanolic acid was selected as marker compound because of its bioactivities for alopecia therapy. The developed analytical method and extraction condition were successfully qualified. Coefficient of determination ($r^2$) for the calibration was 0.9997 with a line passing through the origin point in the range of 0.1-100 mg/mL. The limit of detection (LOD) and the limit of quantitation (LOQ) were 17.5 ng/mL and 55.0 ng/mL, respectively. The intra-day and inter-day precision of the method were 0.5-1.4 % and 0.7-1.8 % in relative standard deviation, respectively, while those accuracy were 99.5-100.9 % and 100.0-102.2 %, respectively. The repeatability of oleanolic acid in samples ranged of 0.3-1.9 % based on peak area and 0.3-0.7 % for retention time. Recoveries from samples were 95.0-99.4 % with lower than 1.8 % in relative standard deviation. Overall, the developed analytical method will be used for quality control of this commercial scalp tonic products successfully.

Semi-rigidity Connection Model for Unstiff ened CHS X-type Joints Subjected Out-of-plane Bending

Mechanical performance of unstiff ened circular hollow section (CHS) multi-planar X-joints subjected two diff erent directions (sagging and hogging) of out-of-plane bending moment (OPBM) was analyzed. A seven-parameter exponential function model, which describing semi-rigidity behavior (moment-rotation curves) of the X-joints subjected sagging and hogging OPBM, was proposed, and then the method to obtain the values of each parameters in the seven- parameter model was also proposed. Based on the results of FE parametric analysis, formulas for parameters in the seven-parameter model were established combined with multiple nonlinear regression analysis. FE and test results were used to verifi ed the seven-parameter model. The results indicated that: the seven-parameter model can well refl ect the semi-rigidity behavior of CHS multi-planar X-joints subjected to OPBM.

This study investigates the potential of using biochar as a substrate component to replace pine bark (PB) to produce horticultural crops in containers. Biochar was incorporated in PB at 0, 20, 40, 60, 80 and 100% (by volume) to grow chrysanthemum, tomato, lettuce and basil plants in containers. The responses of plant growth to the percentage of biochar combined in PB mixtures varied among the different crops. Chrysanthemum plants grown in PB mixtures with 80% and 100% biochar had significant higher shoot fresh weight (FW) and dry weight (DW) than plants grown in PB alone, while the same treatments resulted in reduced shoot FW, DW, growth index measured at 19 and 33 days after transplanting and root rating of tomato plants. Growing lettuce plants in 100% biochar reduced the FW of the second crop compared to plants grown in PB substrates supplemented with 0, 20, 40, 60, and 80% biochar. For basil, although biochar alone caused lower root rating, it did not adversely influence the fresh basil yield. Generally, no negative effect on plant growth was observed in PB mixes with biochar as high as 60%, which is probably the outcome of similar physical properties of the biochar to the commonly used PB.

<P>In this work, we designed a process to assemble gold nanoparticles onto a three-dimensional (3D) polymer surface, which can then be monitored using surface-enhanced Raman scattering (SERS). This work is the first demonstration of the assembly of gold nanoparticles on a filter film and in situ measurement with Raman spectroscopy. Herein, a polyhexamethylene adipamide (Nylon66) film embedded in the organic filter film was used as a template to fabricate a tunable SERS-active substrate. A 'hotspot''-rich gold-nanoparticle-decorated polymer substrate for SERS was prepared; this substrate exhibited high sensitivity in trace detection of targets. The study was conducted using 4-mercaptobenzoic acid as a probe molecule with the aim of comparing the scattering efficiency and the homogeneity of the Raman signal on selected substrates. In addition, we used the gold-decorated polymer film to detect a biotin-avidin complex. The most powerful advantage of the proposed microanalytical device is the in situ SERS application. The 3D nanoporous structures described in this work hold strong potential for use in various applications such as environmental monitoring and biomolecule detection.</P>
Acute Liver Failure Associated with Occupational Exposure to Tetrachloroethylene

Tetrachloroethylene is a chlorinated solvent that is primarily used in dry cleaning and degreasing operations. Although the hepatotoxicity caused by tetrachloroethylene has been well documented in literature, it is rarely considered as a cause of acute liver failure. We report a case of a 39-yr-old man who was admitted to our hospital for acute liver failure due to tetrachloroethylene exposure. Histological examination of the liver revealed massive hepatic necrosis, prominently, in zone 3 of the hepatic lobules. The patient underwent supportive treatment along with 3 sessions of plasmapheresis, and consequently, he presented a favorable outcome. Repeat liver biopsy performed 6 months after the patient’s discharge showed architectural distortion with postnecrotic cirrhosis. Physicians should be aware of the possibility of acute liver failure induced by tetrachloroethylene. Early plasmapheresis can be effective for individuals with sufficient capacity for hepatocyte regeneration.

Background: Ginseng (G) and Ligustrum lucidum Ait (LLA) are core traditional Chinese medicines intreating myelosuppression formula. The present study was designed to profile effect of G and LLA herbpair (G-LLA) on myelosuppressed mice. Methods: The mice myelosuppression model was established by intraperitoneal (i.p.) injection ofcyclophosphamide (Cy). Hematopoietic function of bone marrow was measured by hemopoietic progenitorcell culture and peripheral blood count, and serum hemopoietic factors were tested by enzymelinkedimmunosorbent assay. Bone marrow cell cycle was performed by flow cytometry. HPLC was usedto measure 20 potential chemical components related to myelosuppression, including ginsenoside Rg1,Re, Rb1, Rc, Rb2, Rb3, Rd, Rk3, Rh4, 20 (S)-Rg3, 20 (R)-Rg3, Rk1, Rg5, salidroside, and so on. Results: G, LLA, and G-LLA improved the amount of peripheral blood cells and bone marrow cellsof myelosuppressed mice (P < 0.01). They significantly increased the colony quantity of colony-formingunitegranulocyte macrophage, burst-forming unit erythroid, colony-forming unit erythroid, and colony-forming unit megakaryocyte and amount of G2/M and S phase cells (P < 0.01). They also significantlydecreased the amount of hematopoiesis-related cytokines (P < 0.01). The content of chemicalcomponents in G-LLA changed, and the change of rare saponin was the most obvious. Conclusion: These results show that G-LLA herb pair might produce synergistic or complementarycompatibility effects on bone marrow suppression after chemotherapy. It suggests that the substancebasis of G-LLA for treating bone marrow suppression may be effective chemical components.
