      • Enhanced antitumor immunotherapeutic effect of B-cell-based vaccine transduced with modified adenoviral vector containing type 35 fiber structures

        For successful clinical tumor immunotherapy outcomes, strong immune responses against tumor antigens must be generated. Cell-based vaccines compromise one strategy with which to induce appropriate strong immune responses. Previously, we established a natural killer T-cell (NKT) ligand-loaded, adenoviral vector-transduced B-cell-based anticancer cellular vaccine. To enhance tumor antigen delivery to B cells, we established a modified adenoviral vector (Ad-k35) that encoded a truncated form of the breast cancer antigen Her2/neu (Ad-k35HM) in which fiber structure was substituted with adenovirus serotype 35. We observed increased tumor antigen expression with Ad-k35HM in both human and murine B cells. In addition, an Ad-k35HM-transduced B-cell vaccine elicited strong antigen-specific cellular and humoral immune responses that were further enhanced with the additional loading of soluble NKT ligand KBC009. An Ad-k35HM-transduced, KBC009-loaded B-cell vaccine efficiently suppressed the in vivo growth of established tumors in a mouse model. Moreover, the vaccine elicited human leukocyte antigen (HLA)-A2 epitope-specific cytotoxic T-cell responses in B6.Cg (CB)-Tg (HLA-A/H2-D) 2Enge/Jat mice. These findings indicated that the Ad-k35 could be appropriate for the preclinical and clinical development of B-cell-based anticancer immunotherapies.

(2011) <I>European Journal of Cancer Care</I><B>20</B>, 803–809</P><P><B>Satisfaction with mammography in the National Cancer Screening Programme participants of age 40s in Korea</B></P><P>The aim of this study was to evaluate satisfaction with the National Cancer Screening Programme of mammography in Korea and to examine the association between subscales of satisfaction and general satisfaction. We conducted a cross‐sectional telephone survey for women who had obtained a National Cancer Screening Programme mammographic screening at general hospitals between May and October 2008. The present study included 2005 women in their forties. We performed multivariate linear regression using dependent variable as general satisfaction and independent variables as subscales of satisfaction, such as pre‐screening information transfer, staff interpersonal skills, physical surroundings and results reporting. Participants were stratified according to the result of their mammogram as negative or positive. Mean score of satisfaction was above 2.5 of 4 for all subscales. Women who received positive results were less satisfied with all of subscale factors. Staff interpersonal skills were the most important factor that contributed to general satisfaction. Future efforts such as staff training programme of communication/attitude skills, ensuring privacy and explanation of possible discomfort of the screening would be needed.</P>

        COLD PERFORMANCE OF VARIOUS BIODIESEL FUEL BLENDS AT LOW TEMPERATURE

        This study examines the cold performance of biodiesel blends in a passenger car and a light duty truck at -16 oC and -20 oC. Six different types of biodiesels derived from soybean oil, waste cooking oil, rapeseed oil, cottonseed oil, palm oil and jatropha oil were blended with different volume ratios (B5 (5 vol. % biodiesel - 95 vol. % diesel), B10 and B20). The cold filter plugging point (CFPP) and the cloud point had an effect on the startability and driveability of both the passenger car and the light duty truck. The startability and driveability of the passenger car with all biodiesel blends (B5) were generally good at -20 oC. In the light duty truck, biodiesel blends (B10 and B20) of soybean, waste cooking, rapeseed and jatropha tended to be good at -20 oC in the startability and driveability tests than the biodiesel blends (B10 and B20) of cottonseed and palm. In particular, the palm biodiesel blend (B10) failed at -20 oC, and the palm biodiesel blend (B20) also failed at -16 oC in the startability test. The cold flow properties of biodiesel dictate that the length of the hydrocarbon chains and the presence of unsaturated structures significantly affect the low temperature properties of biodiesel.

        한우 암소의 성장곡선 모수에 영향을 미치는 요인

        Some growth curve models were used to fit individual growth of 1,083 Hanwoo cows born from 1970 to 2001 in Daekwanryeong branch, National Livestock Research Institute(NLRI). The effects of year-season of birth and age of dam were analyzed. In analysis of variance for growth curve parameters, the effects of birth year-season were significant for mature weight(A), growth ratio(b) and maturing rate(k)(P<.01). The effects of age of dam were significant for growth ratio(b) but not significant for mature weight(A) and maturing rate(k). The linear term of the covariate of age at the final weights was significant for the A(P<.01) and k(P<.01) of Gompertz model, von Bertalanffy model and Logistic model. For the growth curve parameters fitted on individual data using Gompertz model, von Bertalanffy model and LOgistic model, rusulting the linear contrasts(fall-spring), Least square means of A in three nonlinear models were higher cows born all fall and A of Logistic model was significant(P<.05) between the seasons. According to the results of the least square menas of growth curve parameters by age of dam, least square means of mature weight(A) in Gompertz model was largest in 6 year and smallest estimating for 3 and 8 years of age of dam. The growth ratio(b) was largest in 2 year of age of da and smallest estimating in 8 year. The A and k were not different by age of dam(p>.05), On the other hand, the b was different by age of dam.(p<.01). The estimate of A in von Bertalanffy model was largest in 6 year and smallest in 8 and 9 years of age of dam. The b was largest in 2 year and tend to decline as age of dam increased. The A and k were not different by age of dam(p>.05), On the other hand, the b was highly significant by age of dam(p<.01).

        한우 암소의 개체별 성장곡선 모수 추정

        Weight records of Hanwoo cows from birth to 36 months of age collected in Daekwanryeong branch, National Livestock Research Institute(LNRI) were fitted to Gompertz, von Bertalanffy and Logistic functions. For the growth curve parameters fitted on individual records using Gompertz model, the mean estimates of mature weight(A), growth ratio(b) and growth rate(k) were 383.42±97.29㎏, 2.374±0.340 and 0.0037±0.0012, respectively, and mean estimates of body weight, age and daily gain rate at inflection were 141.05±35.79㎏, 255.63±109.09 day and 0.500±0.123㎏, respectively. For von Bertalanffy model, the mean estimates of A, b and k were 410.47±117.98㎏, 0.575±0.057 and 0.003±0.001, and mean estimates of body weight, age and daily gain at inflection were 121.62±34.94㎏, 211.02±105.53 and 0.504±0.124㎏. For Logistic model, the mean estimates of A, b and k were 347.64±97.29㎏, 6.73±0.34 and 0.006±0.0018, and mean estimates of body weight, age and daily gain at inflection were 173.82±37.25㎏, 324.47±126.85 and 0.508±0.131㎏. Coefficients of variation for the A, b and k parameter estimates were 25.3%, 14.3% and 32.4%, respectively, for Gompertz model, 28.7%, 9.9% and 33.3% for von Bertalanffy model, and 27.9%, 5.0% and 30.0% for Logistic model.

      • ZNF509S1 downregulates PUMA by inhibiting p53K382 acetylation and p53-DNA binding

        Expression of the POK family protein ZNF509L, and -its S1 isoform, is induced by p53 upon exposure to genotoxic stress. Due to alternative splicing of the ZNF509 primary transcript, ZNF509S1 lacks the 6 zinc-fingers and C-terminus of ZNF509L, resulting in only one zinc-finger. ZNF509L and -S1 inhibit cell proliferation by activating p21/CDKN1A and RB transcription, respectively. When cells are exposed to severe DNA damage, p53 activates PUMA (p53-upregulated modulator of apoptosis) transcription. Interestingly, apoptosis due to transcriptional activation of PUMA by p53 is attenuated by ZNF509S1. Thus we investigated the molecular mechanism(s) underlying the transcriptional attenuation and anti-apoptotic effects of ZNF509S1. We show that ZNF509S1 modulation of p53 activity is important in PUMA gene transcription by modulating post-translational modification of p53 by p300. ZNF509S1 directly interacts with p53 and inhibits p300-mediated acetylation of p53 lysine K382, with deacetylation of p53 K382 leading to decreased DNA binding at the p53 response element 1 of the PUMA promoter. ZNF509S1 may play a role not only in cell cycle arrest, by activating RB expression, but also in rescuing cells from apoptotic death by repressing PUMA expression in cells exposed to severe DNA damage.

        Ce<sub>11</sub>Ge<sub>3.73(2)</sub>In<sub>6.27</sub>: Solid-state synthesis, crystal structure and site-preference

        <P>A novel intermetallic compound of Ce11Ge3.73(2)In6.27 has been synthesized through the high-temperature solid-state reaction using Nb-ampoules. A batch of well grown block-/short bar-shaped single crystals has been obtained, and the crystal structure of the title compound has been characterized by single-crystal X-ray diffraction analyses. Ce11Ge3.73(2)In6.27 adopts the Ho11Ge10-type structure belonging to the tetragonal space group 14/mmm (Z=4, Pearson symbol tI84) with nine crystallographically unique atomic positions in the asymmetric unit. The lattice parameters are a = 12.0163(1) angstrom and c= 16.5396(2) angstrom. The overall crystal structure can simply be depicted as an assembly of three different types of co-facial cationic polyhedra centered by anions, which is further enclosed by the three-dimensional (3-D) cage like anionic framework. The extra amount of In is observed in one of three isolated anionic sites resulting in introducing the Ge/In-mixed site at the Wyckoff 4e site. This unique site-preference of In substitution for Ge at the 4e site has been enlightened via the atomic size-aspect which was fully supported and rationalized by the site- and bond-energies analyses using tight-binding linear muffin-tin orbital (TB-LMTO) calculations. Energy-dispersive X-ray spectroscopy (EDS), density of states (DOS), crystal orbital Hamilton population (COHP), and electron localization function (ELF) analyses for the title compound are also presented. Magnetic susceptibility measurement proves that an antiferromagnetic ordering of Ce atoms at a low temperature with a paramagnetic Curie temperature of -23.2 K. (C) 2015 Elsevier Inc.       • Absorption of carbon dioxide in O/W emulsion absorbent: Kinetics of absorption in N-methylcyclohexylamine and 2,6-dimethylpiperidine emulsion

        Speciation and kinetics of carbon dioxide absorption by N-methylcyclohexylamine (MCHA) and 2,6-dimethylpiperidine (DMPD) emulsion absorbents were evaluated. The physical solubility, diffusivity, and carbon dioxide absorption rate were measured as a function of temperature (293, 303, 313, and 323K) and amine concentration (10, 20, 30, and 40wt%) by using a semibatch reactor and wetted wall column (WWC). The results showed that the rate of carbon dioxide absorption by the MCHA and DMPD emulsion absorbents increased with the carbon dioxide partial pressure and amine concentration. Using the physical data and absorption rate, the rate constants for carbon dioxide absorption by MCHA and DMPD were evaluated. Based on the pseudo-first-order reaction, the rate constants were determined to be k<SUB>2,MCHA</SUB>=2.698x10<SUP>7</SUP>exp(-2458/T) and k<SUB>2,DMPD</SUB>=1.002x10<SUP>6</SUP>exp(-1511/T). Notably, the activation energies for carbon dioxide absorption by the emulsion absorbents containing MCHA or DMPD were lower than those of alkanolamine solutions.

      • K-means 및 헝가리안 알고리즘기반 중소형 유통사업장 배송지역 균등할당 알고리즘 개발

        최근 COVID-19 로 언택트 시대를 맞이하여 유통산업의 물류량은 급진적으로 증가하고 있는 추세이다. 따라서 유통산업은 효율적인 물류관리를 위해 배송 최적화에 노력하고 있다. 하지만 중소형 유통업체는 대형 유통업체에 비해 인력 및 비용의 한계로 배송 최적화에 어려움이 있다. 중소형 유통업체의 배송지 최적화의 이슈는 고객으로부터 접수한 주문의 배송지를 균등하게 할당하는 것이다. 따라서 본 연구는 중소형 유통업체의 배송 최적화를 위해 배송 인력과 배송 이동거리를 고려하여 배송 지역 균등할당 알고리즘을 개발한다. 배송 지역 균등할당 알고리즘은 배송지 밀집도의 특징을 고려하여 배송지를 가용 배송기사에게 균등 할당하는 것이다. 배송지 밀집도 특징을 도출하기 위해 밀도기반 클러스터링 기법인 DBSCAN 으로 배송지의 밀집도가 높은 권역과 낮은 권역으로 나누었다. 밀집도가 높은 권역은 배송 출발지에서 가까우며 배송지 수가 많은 특징을 가지고, 밀집도가 낮은 권역은 배송 출발지에서 멀리 떨어져 있으며 배송지 수가 적은 특징을 가진다. 밀집도가 낮은 권역은 이동거리의 편차가 더 커질 수 있기 때문에 K-means 알고리즘의 Elbow Method 를 사용하여 배송지를 할당한다. 밀집도가 높은 권역은 K-means 알고리즘으로 배송지 그룹을 나누고 그룹의 중심점과 그룹 간 배송지의 거리가 최소가 되게 헝가리안 알고리즘을 적용하여 배송지를 할당한다. 실 데이터를 사용하여 비교한 결과 본 연구에서 제안한 균등알고리즘은 이동거리의 편차와 배송지 수의 편차를 감소시킬 수 있는 것으로 나타났다.
