        Fabrication of Remarkably Bright QD Densely-Embedded Silica Nanoparticle

        We developed a synthesis method for silica-coated quantum dot (QD)-densely-embedded silica nanoparticle (Si@D-QD@Si NP) with remarkably brighter fluorescence by increasing the number of QDs on silica NPs. Si@D-QD@Si NPs exhibited a narrow size distribution and a high signal reproducibility. Red, green, and blue Si@D-QD@Si NPs were successfully fabricated and characterized. The Si@D-QD@Si NP carried 2.4 times more QDs than previously reported silica-coated QD-embedded silica NP (Si@QD@Si NP). The quantum yield of Si@D-QD@Si NPs was slightly higher than Si@QD@Si NPs. The red Si@D-QD@Si NPs showed approximately 26.6%, the green Si@D-QD@Si NP 36.2%, and the blue Si@D-QD@Si NP 39.3% higher fluorescence intensity compared to the corresponding Si@QD@Si NPs. This result implied that the particles can be used at bio-field with a higher sensitive detection.

        SERS (Surface-enhanced Raman scattering) promises as an efficient tool for identification of diverse targets because it has stability against photobleaching and unique signal patterns with narrow band width. To obtain powerful SERS signals, the structure of metal substrate is crucial. Until now, varied approaches have been attempted to sensitively detect targets in low concentration. Herein, we have synthesized magnetic silica nanoparticles-embedded silver nanoshell that has both magnetic and SERS properties for SERS detection. The bumpy silver nanoshell could produce ‘Hot Spots’, guiding to powerful SERS signals. Furthermore, magnetic-induced aggregation made SERS signals further stronger.

        The present inventors report a synthesis method capable of providing monodispersed (104.2 nm ± 2.4 nm) silica coated magnetic nanoparticles (MNP@SiO2 NPs) in gram scale. MNP@SiO2 NP showed superparamagnetism and stored for 3 months, and no aggregation was detected. Plasmon and magnetic materials using NPN (MNP@SiO2 Au NPs) were also prepared by simply adding Au NP to magnetic NPs and showed 500 % higher light scattering than gold NPs of the same size.

      • Water-Soluble Multi-Shell Quantum Dot with Stability High and Quantum Yield

        Semiconductor quantum dots (QDs) have many advantages including a high quantum yield (QY), high fluorescent signal, photo-stability and narrow emission spectrum, compared to a traditional fluorescent dye. However, After the surface modification of QDs for bio-applications, QY is getting significantly lower. In this study, we describe the water-soluble multi shell QDs with high quantum yield. This multi-shell QDs show a strong fluorescence signal compared to established single shell QDs. Besides, multi shell QDs indicate a high stability in a strong pH range. After the surface modification for bio-applications, multi-shell QDs maintain slightly reduced QY. In comparison with single shell QDs, multi shell QDs were discovered a strong fluorescence as feature in vitro and in vivo. This water-soluble multi shell QDs, which surface modified possible one, have a great potential for practical bio-imaging.

      • Ag nanoparticles onto thin silica coating of graphene oxide for SERS substrate

        Ag nanoparticles onto thin silica coating of graphene oxide for SERS substrate was prepared and used an as a surface-enhanced Raman scattering (SERS) substrate. Graphene oxide was coated with silica layer on their surface by using physical absorption of sodium silicate as a binder combine with sol-gel process of of 3-mercaptopropyl trimethoxysilane. The thickness of silica is ~6 nm. Ag NPs were embedded on the silica-coated graphene oxide by the reduction of Ag+ by octylamine in the presence of ethylene glycol to prepare GO@ SiO2@Ag. The Raman signal of GO@SiO2@Ag was 1.8-fold as strong as than that of GO without a silica coating. For application, the GO@SiO2@Ag was applied to detect 4-mercaptobenzoic acid (4-MBA). The limit of detection of 4-MBA was 0.74 μM, indicating that the GO@SiO2@Ag can be utilized as a highly sensitive SERS substrate for detection of trace amounts of chemicals in various fields.

        염생식물인 갯끈풀과 갯메꽃 추출물의 HaCaT 세포에서 피부 보습 및 피부 장벽 기능에 미치는 영향

        노화가 진행될수록 활성산소종으로 인하여 피부 보습은 떨어지고 피부 장벽은 붕괴되어 피부가 손상된다. 본 연구에서는 인천 동막 해변에 서식하는 염생식물인 갯끈풀(Spartina anglica; SAE)과 갯메꽃(Calystegia soldanella; CSE)을 70% 에탄올(EtOH)로 추출하여 피부 보습 및 피부 장벽 기능 강화에 대한 효능을 평가하였다. 이 추출물들에 대한 피부 각질형성세포(HaCaT cell)에서 세포독성을 WST-8 assay를 이용하여, 세포 생존율이 90% 이상을 보이는 농도를 선별하여 추가 실험을 진행하였다. ABTS 라디칼 소거능을 통해 항산화 효과를 확인한 결과, SAE와 CSE는 높은 라디칼 소거능을 보였다. 피부 보습과 관련된 인자들인 filaggrin (FGL), aquaporin 3 (AQP3), hyaluronan synthase 2 (HAS2)과 피부 장벽 기능과 연관 있는 transglutaminase 1 (TGM1)과 involucrin (INV)의 유전자 수준에서의 발현 변화를 측정한 결과, SAE에 의해 AQP3, HAS2, TGM1의 발현이 증가하였으나, CSE는 변화가 없는 것을 확인할 수 있었다. SAE에 의한 세포 내 신호전달 경로를 확인하기 위해 western blot 분석을 수행하였다. Extracellular signal regulated kinase (ERK), c-Jun N-terminal kinase (JNK) and p38 mitogen activated protein kinase의 활성이 SAE에 의하여 상향 조절되었음을 확인하였다. 이러한 결과는 갯끈풀 추출물이 피부 보습 및 피부 장벽 기능 강화를 위한 화장품의 기능성 소재로 사용될 수 있음을 시사한다

        Evaluation of cytotoxicity, genotoxicity, and zebrafish embryo toxicity of mixtures containing Hyssopus officinalis, Morus alba, Engraulis japonicus, and 27 other extracts for cosmetic safety assessment

        Background Although various natural ingredients and formulated reagents are used as ingredients in cosmetics, it is important to have alternatives to animal tests, such as in vitro tests of toxicity, as animal testing of cosmetics is prohibited in many countries. Objectives Here, cytotoxicity, genotoxicity, and embryotoxicity on zebrafish embryos were evaluated to confirm the safety of ingredients formulated with 30 extracts from natural resources used as cosmetic ingredients. The blended ingredients were tested by dividing them into mixtures of 25 natural products (NMA), 30 natural products (NMB), and concentrated NMB (NMC), depending on the number of blended natural components and their application. Results First, in vitro, HaCaT cells were treated with concentrations of 0 to 20% for 24 h to measure cell viability. DNA damage within the IC20 value was measured by comet assay. In the comet assay, 10 mM H2O2 was used as a positive control. Additionally, zebrafish embryos were used as a substitute for conventional animal testing to observe natural mixture toxicity. The olive tail moment (OTM) were not affected by NMA, NMB, or NMC in comparison with the positive control versus three representatives within the IC20 value concentration of each mixture. Zebrafish embryo results were analysed in a manner similar to the cytotoxicity test results. NMA mixtures showed no cytotoxicity, genotoxicity, or toxicity on zebrafish embryos. Other mixtures showed non-toxicity within the representatively concentrated range. Conclusion Therefore, the 3 mixtures containing Hyssopus officinalis, Morus alba, Engraulis japonicus, and 27 other extracts were considered to be not significantly toxic in HaCaT cell and zebrafish, so it is suggested that they could be used as cosmetic ingredients.       • Thin Silica Shell Coated Ag Assembled Nanostructures for Expanding Generality of SERS Analytes: Simultaneous Quantitative Detection of Aniline and 4-Aminothiophenol

        In this study, we demonstrated thin silica-coated and assembled Ag NPs (SiO2@Ag@SiO2 NPs) for simultaneous and quantitative detection of chemicals that have different intrinsic affinities to silver metal. These NPs were detected each SERS peak of aniline or 4-aminothiophenol from the mixture with limits of detection of 93 ppm and 54 ppb, respectively. E-field distribution based on interparticle distance was simulated using discrete dipole approximation calculation to gain insight into enhanced scattering of these thin silica coated Ag NP assemblies. These NPs were successfully applied to detect aniline in river water and tap water. Results suggest that SiO2@Ag@SiO2 NP-based SERS detection systems can be used as a simple and universal detection tool for safety.

        Angiotensin-Converting Enzyme Inhibitor(ACE Inhibitor)에 의해 유발된 안면부 맥관부종(angioedema) 치험례

        Angioedema is a localized transient swelling of sudden onset that can occur in the face, lips, tongue, hand, feet, respiratory system and gastrointestinal system. Angioedema is classified as allergy, hereditary, idiopathic or induced by medication as like aspirin, nonsteroidal anti-inflammatory agents, opiates, antibiotics, and angiotensin-converting enzyme. Angiotensin-converting enzyme inhibitors are widely prescribed for hypertension and heart failure. This drug is commonly associated with angioedema which may be potentially life threatening. We experienced a case of angioedema induced by ACE inhibitor (angiotensin-converting enzyme inhibitor) in a 74-year-old patient who took ACE inhibitor to control hypertension during hospitalization. We thought the angioedema in the face had relation to myenzhong (面腫, mienjong) in oriental medicine. Weiling-tang (Wiryung-tang) was effective for angioedema in the face. As a result the symptoms disappeared rapidly. After 6 days, the patient's symptoms had notably improved.
