제주지역 양식 넙치(Paralichthys olivaceus)를 대상으로 한 스쿠티카증(Scuticociliatosis) 모니터링

Scuticociliatosis can occur from juveniles to adult olive flounder (Paralichthys olivaceus) and the infection usually causes mass mortality. There is no clear way to treat it once the parasite has invaded the fish's internal organs. Therefore, periodic monitoring of scuticociliatosis for surveillance and prompt response is a crucial part of reducing damage to olive flounder farms. In this study, we conducted scuticociliatosis monitoring of olive flounder farms in jeju island to investigate the detection rate of the causative agents (Miamiensis avidus and Uronema marinum), and determine the phylogenetic classification of the detected pathogens. Disease monitoring was conducted on a total of 3,282 olive flounder and the detection rate was highest in August (15.83%) during the investigation period. The sequences of all 4 strains obtained from olive flounder farms in 4 different regions, were 100% homologous with SSU rRNA gene of M. avidus deposited in GenBank.

In this study, a research was conducted on hematological analyses and comparisons of the changes in non-specific immune response with regards to emaciated and non-emaciated fishes that are causing severe damages to olive flounder aquafarms in Jeju. First, the researcher selected a sample of raised olive flounder that exhibited typical symptoms of emaciation in three aquafarms in Jeju for experimental use, and then selected healthy-looking olive flounder from one aquafarm that had no past record of emaciation disease, and checked for possibilities of infection through PCR. Averagely, in the hematological analysis shows that haematocrit (Ht), glutamic oxaloacetic transaminase (GOT), and glucose (GLU), emaciated fish groups (Farm A, B, and C) displayed higher experimental values than control group, and in the glutamic pyruvic transaminase (GPT), total cholesterol (Tchol), and total protein (TP), exhibited lower experimental values than control group. As a result of measuring lysozyme, nitro-blue-tetrazolium (NBT) and myeloperoxidase (MPO), which are markers for non-specific immune responses, lower experimental values were observed in the three experiments. Thus, it can be concluded that a high mortality rate was exhibited due to stress and internal organ damages that resulted from the destruction of the primary defense mechanism that protects fishes from getting infected by emaciation agents.

No established method can be used to select effective antibiotics in antibiotic susceptibility tests for fish bacterialpathogens quickly and accurately. Here, we established the optimal conditions for determining the minimal inhibitoryconcentration (MIC) of major fish bacterial pathogens (Streptococcus spp., Edwardsiella tarda, Vibrio spp., Aeromonasspp., and Pseudomonas spp.) using the KRAQ1 and CAMPY2 panels. The MIC panel used 18 antibiotics of twotypes and we conducted experiments to establish the optimal culture medium and temperature for each species. Theoptimal conditions for incubating Streptococcus spp. were in cation-adjusted Mueller-Hinton broth with TES buffer(CAMHBT) at 28°C, using 5% lysed horse blood (LHB) as recommended by the Clinical Laboratory StandardsInstitute. For Vibrio spp., the optimal culture conditions were 28°C in CAMHBT supplemented with 1% NaCl. Theoptimal conditions for culturing E. tarda, Aeromonas spp., and Pseudomonas spp. were in CAMHBT at 28°C
양식 넙치에서 분리한 어병세균의 lincomycin에 대한 내성 유전자의 분포

Lincomycin as one of the lincosamides antibiotics have been mainly used in human and livestock fields, but have not been used in aquaculture. In this study, the distribution of minimum inhibitory concentration (MIC) values against lincomycin and the detection of the macrolide-lincosamide-streptogramin (MLS) resistance gene were confirmed in bacterial pathogens isolated from cultured olive flounder (Paralichthys olivaceus). Of the 107 strains isolated from Jeju, 36 strains of Gram-positive bacteria and 71 strains of Gram-negative bacteria were identified. Most of Streptococcus spp. was found to have a MIC value of less than or equal to 0.5 ㎍/mL, and Edwardsiella piscicida was found to have a MIC value higher than 1,024 ㎍/mL. V. harveyi and V. alginolyticus mostly showed MIC values of 256 ㎍/mL, but V. scophthalmi displayed values of 8~64 ㎍/mL. In the detection of MLS resistance gene, erm(B) was detected in 9 strains of Streptococcus spp., and erm(A) was confirmed in one strain.
