        농업용 석유기관의 연료소비율에 관한 연구

        This study was attempted to investigate the changes of specific fuel consumption, compression pressure and power output, consequently to obtain basic data on farm kerosene engine. The samples which are used in this study are a 4 cycle water cooled korosene engine for the use of K6-CT83 power tiller and a 4 cycle air-cooled kerosene engine for the use of G5L-3A water pump. The Korean Industrial Standards (K.S)KS-B 6002 "Test code of small internal combustion engine" was referred in carrying out this study, and its results are as follows. 1. According to load increasing, the speific fuel consumption of the engines generally decreases, however, in case of 10% over-loading it increases. 2. As a result of full load consecutive operation, according to passing of operating time, the amount of wear generally increases, consequently the speific fuel consumption also increases, and inversly the compression pressure decreases. 3. The changes of specific fuel consumption and compression pressure were closely related with time of piston ring exchange, and periodically about 100 hours the engines show the increase of specific fuel consumption and the decrease of compression pressure. 4. After about 300 hours, although the engine had new piston rings, the specific fuel consumption increase, consequently the engine needs boring. In actual use, it is impossible to operate consecutively on full load, therefore the boring time of engine is expected to come later.

        Effects of Gypenosides on Dopaminergic Neuronal Cell Death in 6-Hydroxydopamine-lesioned Rat Model of Parkinson's Disease with Long-term L-DOPA Treatment

        The goal of this study was to determine whether gypenosides (GPS) exert protective effects against dopaminergic neuronal cell death in a 6-hydroxydopamine (OHDA)-lesioned rat model of Parkinson's disease (PD) with or without long-term 3,4-dihydroxyphenylalanine (L-DOPA) treatment. Rats were injected with 6-OHDA in the substantia nigra to induce PD-like symptoms; 14 days after injection, groups of 6-OHDA-lesioned animals were treated for 21 days with GPS (25 or 50 mg/kg) and/or L-DOPA (20 mg/kg). Dopaminergic neuronal cell death was assessed by counting tyrosine hydroxylase (TH)-immunopositive cells in the substantia nigra and measuring levels of dopamine, norepinephrine, 3,4-dihydroxyphenylacetic acid (DOPAC), and homovanillic acid (HVA) in the striatum. Dopaminergic neuronal cell death induced by 6-OHDA lesions was ameliorated by GPS treatment (50 mg/kg). L-DOPA treatment exacerbated 6-OHDA-induced dopaminergic neuronal cell death; however, these effects were partially reversed by GPS treatment (25 and 50 mg/kg). These results suggest that GPS treatment is protective against dopaminergic neuronal cell death in a 6-OHDA-lesioned rat model of PD with long-term L-DOPA treatment. Therefore, GPS may be useful as a phytotherapeutic agent for the treatment of PD.

        Berberine이 백서의 6-Hydroxydopamine-유도 파킨슨병 모델에서의 L-DOPA 요법에 미치는 영향

        Isoquinoline compounds including berberine enhance L-DOPA-induced cytotoxicity in PC12 cells. In this study, the effects of berberine on L-DOPA therapy in unilateral 6-hydroxydopamine (6-OHDA)-induced rat models of parkinsonism were investigated. Rats were prepared for the models of Parkinson's disease by 6-OHDA-lesioning for 14 days and then treated with L-DOPA (10 mg/kg) with or without berberine (5 and 30 mg/kg, i.p.) for 21 days. Treatment with berberine (5 and 30 mg/kg, i.p.) showed a dopaminergic cell loss in substantia nigra of 6-OHDA-lesioned rats treated with L-DOPA: 30 mg/kg berberine was more intensive neurotoxic. The levels of dopamine were also decreased by berberine (5 and 30 mg/kg) in striatum-substantia nigra of 6-OHDA-lesioned rats treated with L-DOPA. These results suggest that berberine aggravates cell death of dopaminergic neurons in L-DOPA-treated 6-OHDA-lesioned rat models of Parkinson's disease. Therefore, the long-term L-DOPA therapeutic patients with isoquinoline compounds including berberine may need to be checked for the adverse symptoms.

        Rotenone- 및 MPTP-유도 파킨슨병 동물 모델에서 돌외 에탄올 추출물의 Dopamine 신경세포 보호작용

        The neuroprotective effects of herbal ethanol extract (GP-EX) from Gynostemma pentaphyllum on dopamine neu- rons in animal model of Parkinson's disease (PD) were investigated. Rats and mice were administered with rotenone (2.5 mg/kg) for 28 days and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP, 30 mg/kg) for 5 days for the PD models, respectively and the animals were simultaneously treated with GP-EX (30 mg/kg, daily). After preparing the PD models, the animals were also administered with L-DOPA (10 mg/kg) for 14 days with or without GP-EX treatment. Treatment with GP-EX (30 mg/kg) inhibited the rotenone- and MPTP-induced neurotoxic effects in dopamine neurons of rats or mice, which was determined by the numbers of tyrosine hydroxylase-immunohistochemical staining survival cells, as well as the levels of dopamine, 3,4-dihy- droxyphenylacetic acid and homovanillic acid. GP-EX (30 mg/kg) also showed the protective effects on neurotoxicity which was induced by long-term administration of L-DOPA (10 mg/kg) in rotenone- and MPTP-induced animal model of PD. The used doses of GP-EX (30 mg/kg) did not produce any signs of toxicity, such as weight loss, diarrhea, or vomiting, in rats and mice during the treatment periods. These results suggest that GP-EX has the protective functions against chronic L-DOPA-induced neurotoxic reactions in dopamine neurons of rotenone- and MPTP-induced animal model of PD. Therefore, the natural GP-EX may be beneficial in the prevention of PD progress and L-DOPA-induced neurotoxicity in PD patients.

        돌외 에탄올 추출물 엑스의 만성 스트레스-유도 불안작용에 대한 개선작용

        The effects of herbal ethanol extract from Gynostemma pentaphyllum (GP extract) on chronic stress-induced anxiety in mice were investigated. The animals were treated with GP extracts (50 and 100 mg/kg/day, p.o.) for 21 days before exposure to electric footshock (EF; duration and interval 10 sec for 3 min, 2 mA) for chronic stress once a day. The ambulatory locomotor activity was reduced by chronic EF stress and it was recovered by 12.9-15.1% in GP extract-treated groups. The grip strength was also significantly decreased by chronic EF stress, however, the EF-stressed groups treated with GP extract increased grip strength from 13.9% to 56.8% compared to EF-stressed groups. In addition, the serum levels of corticosterone were significantly elevated by chronic EF stress to 197% of the control levels, which was reduced to 73.1% by treatment with GP extract (100 mg/kg). In contrast, the brain levels of dopamine and serotonin were reduced to 67.6% and 63.1% by chronic EF stress, which was recovered to 90% of the control levels by treatment with GP extract. These results indicate that GP extract shows the ameliorating effects on chronic EF stress-induced anxiety in mice and it can be developed as the promising anti-anxiety agent.

        Berberine이 백서의 6-hydroxydopamine-유도 파킨슨병 모델에 미치는 영향

        Many isoquinoline alkaloids including berberine lower dopamine content by reducing tyrosine hydroxylase (TH) activity and aggravate L-DOPA-induced cytotoxicity in PC12 cells. In this study, the effects of berberine on 6-hydroxydopamine (6-OHDA)-induced cytotoxicity in PC12 cells and on unilateral 6-OHDA-lesioned rat models were investigated. Berberine at 10-30 µM did not affect cell viability in PC12 cells. However, berberine at concentrations higher than 50 µM caused cytotoxicity at 24 h. Berberine (10-50 µM) also enhanced 6-OHDA (10-50 µM)-induced cytotoxicity at 24 h compared to 6-OHDA alone with an apoptotic process. In addition, treatment with berberine (5 and 30 mg/kg, i.p.) for three weeks showed a dopaminergic cell loss in substantia nigra of 6-OHDA-lesioned rats: 30 mg/kg berberine was more intensive cytotoxic. The levels of dopamine were also decreased by berberine (5 and 30 mg/kg) in the ipsilateral substantia nigra of 6-OHDA-lesioned rats. These results suggest that berberine aggravated 6-OHDA-induced cytotoxicity in PC12 cells and treatment with berberine (5 and 30 mg/kg) in 6-OHDA-lesioned rats also enhanced the degeneration of dopaminergic cell death and the decrease in dopamine levels in substantia nigra. Therefore, the long-term L-DOPA therapeutic patients with isoquinoline compounds including berberine may need to be checked for the adverse symptoms.

        Effects of (-)-Sesamin on Memory Deficits in MPTP-lesioned Mouse Model of Parkinson's Disease

        This study investigated the effects of (-)-sesamin on memory deficits in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-lesioned mouse model of Parkinson's disease (PD). MPTP lesion (30 mg/kg/day, 5 days) in mice showed memory deficits including habit learning memory and spatial memory. However, treatment with (-)-sesamin (25 and 50 mg/kg) for 21 days ameliorated memory deficits in MPTP-lesioned mouse model of PD: (-)-sesamin at both doses improved decreases in the retention latency time of the passive avoidance test and the levels of dopamine, norepinephrine, 3,4-dihydroxyphenylacetic acid, and homovanillic acid, improved the decreased transfer latency time of the elevated plus-maze test, reduced the increased expression of N-methyl-D-aspartate (NMDA) receptor, and increased the reduced phosphorylation of extracellular signal-regulated kinase (ERK1/2) and cyclic AMP-response element binding protein (CREB). These results suggest that (-)-sesamin has protective effects on both habit learning memory and spatial memory deficits via the dopaminergic neurons and NMDA receptor-ERK1/2-CREB system in MPTP-lesioned mouse model of PD, respectively. Therefore, (-)-sesamin may serve as an adjuvant phytonutrient for memory deficits in PD patients.
