<P>Histone deacetylases (HDACs) have been shown to play important roles in the regulation of chromatin remodeling by histone deacetylation, and their expression is induced in several types of cancer. In addition, they are known to be associated with resistance to anticancer drugs. However, the relevance of HDAC4 in chemoresistance remains unclear. Therefore, we investigated the interaction between HDAC4 expression and chemoresistance in breast cancer cells. We found that increased HDAC4 expression in MDA-MB-231 cells was associated with resistance to the anticancer drug 5-fluorouracil (5-FU). To verify these results, a cell line stably overexpressing HDAC4 was generated using MCF-7 cells (HDAC4OE). This cell line displayed increased 5-FU resistance, and HDAC4 knockdown in HDAC4OE cells restored 5-FU sensitivity. Consequently, we concluded that HDAC4 is a critical gene associated with 5?FU chemoresistance. Further investigation using a microarray approach revealed that 355 genes were differentially expressed following HDAC4 overexpression. Based on functional annotation of the array results, HDAC4 overexpression was found to downregulate genes related to the transforming growth factor (TGF) β signaling pathway, including SMAD4, SMAD6, bone morphogenetic protein 6, inhibitor of DNA binding 1 and TGFβ2. We also found that HDAC4 expression regulates SMAD4 expression by inducing deacetylation of histone H3 in the SMAD4 promoter region. In addition, SMAD4 knockdown in MCF?7 cells increased 5-FU resistance. In summary, our data suggest that HDAC4?mediated deacetylation of the SMAD4 promoter may lead to 5-FU resistance in breast cancer cells.</P>

This study examined the prevalence of oral microbes in the saliva of oncological patients and healthy subjects. PCR was used to assess the frequency of oral microbes including 3 cariogenic bacteria, 5 periodontopathic bacteria and 4 Candida species in the saliva of 104 oncological patients and 52 healthy subjects. Among these microorganims, Streptococcus mutans, Fusobacterium nucleatum and Candida albicans were most frequently detected in both groups. There were no significant differences in the prevalence of cariogenic bacteria between the patient and healthy groups, whereas significant differences in the frequency of Porphyromonas gingivalis and Tannerella forsythia were observed between the two groups (p < 0.05). The prevalence of all five periodontopathogens was higher in the healthy group than in the patient group. The prevalence of C. albicans in patients was significantly higher than that of healthy group (p < 0.05). In conclusion, there were significant differences in the prevalence of P. gingivalis, T. forsythia and C. albicans between the oncological patient group and healthy group.

The essential oil of Chrysanthemum boreale Makino was analyzed by means of GC and GC-MS. Eighty-seven constituents were identified representing 94.13% of the total oil and the major components were camphor, α-thujone, cis-chrysanthenol, 1,8-cineole, α-pinene, and β-caryophyllene. This essential oil exhibited antibacterial activity against Streptococcus mutans, Streptococcus sanguis, Fusobacterium nucleatum and Porphyromonas gingivalis.

The current design practice for concrete bridge decks requires a top mat and a bottom mat of reinforcing bars. The chloride attack of the top mat of reinforcing bars is a major cause of deterioration of bridge decks. Empirical evidence has indicated that the top transverse reinforeing bars can essentially be eliminated without jeopardizing the structural integrity of a deck. A simplified method for bridge deck analysis considering the effect of girder deflection is developed based on a closed-form analytical solution which is fine tuned with results of finite element analysis. Finite element analysis and a simplified method results confirm the fact that a properly designed bridge deck does not require top transverse reinforcement for sustaining the negative bending moment induced by traffic loads.

Current design specifications prescribe that the upper and lower reinforcement mat is required in the same amount to resist negative and positive moment in bridge decks. This design concept is primarily based on the unrealistic assumption that the girder plays a role of rigid support against deck deflection. In reality, however, girders are flexible and the deflection of girders affect the behavior of deck slabs. In the present study, an analytical method was developed to take the effect of the girder flexibility on the deck behavior into account. The method was formulated based on the slope-deflection equations of plates and harmonic analysis. Unlike the conventional finite element analysis, the input and output schemes are simple and convenient. The validity of the presented study was verified by a series of comparative studies with finite element analyses and experimental tests. It was shown from the analyses that the negative transverse moments of decks were significantly reduced in many cases when the girder flexibility were appropriately taken into consideration whereas the positive moments tend to increase. This poses a strong need to improve the conventional design concept of decks on rigid girders to those on flexible girders.

The purpose of this research was drawing relative importance of core competency factors in council of Sports for All members. Based on this research it contributes Sports for All administration and instruction, providing effective information. For this purpose, the Delphi survey was carried out by five gurus and an hierarchy structure was established with agreed survey questions in the second round. Based on this, pair-wise comparison questionnaires were developed. The research targeted Kyungkiㆍcapital area, ChoongchungㆍJeonllaㆍcapital area, and KangwonㆍKyungsangㆍcapital area’s Sports for All’s staff and instructs. Through a consistency test, samples with the lack of reliability were excluded and SPSS WIN Ver. 18.0, Expert Choice 2000 and Microsoft Office Excel 2007 were used for data process. The following results came from the research. First regarding to whole sample, an organization of Sports for All’s core competency evaluation field’s analysis result, it was evaluated that job competency should be preferentially considerable, and organization harmony, self-development, customer orientation was appeared in order. Second, It evaluate that the ‘business preparation’ was important with job competency, in organization harmony, ‘teamwork and cooperation’, self-development, ‘development of business(instruct) ability’ and customer orientation, ‘speedy service’.

현재 현장에서 건설되어지는 콘크리트교량의 바닥판은 상부와 하부에 동일한 철근을 배근한다. 바닥판 상부철근의 부분적인 제거는 구조적인 측면에서 볼 때, 안전성을 위협하지 않는다는 경험적인 연구 결과들이 발표되어왔다. 판이론에 근거하여 강판형교의 3거더와 4거더시 바닥판의 구조적인 거동을 수식적으로 표현하는 간략화 된 방법을 유도하였다. 거더간 상대처짐을 고려한 간략화 된 방법은 유한요소해석의 결과치와 유사한 값을 얻을 수 있었다. 본 연구에서는, 바닥판 슬래브의 새로운 설계개념이 간략화 된 방법을 통해서 제안되었다. 간략화 된 방법의 변수는 하중위치, 형상비, 상대강성비 그리고 지간 길이 등을 선정하였다. 이 새로운 설계접근방법은 바닥판 상부 철근량을 감소시켜줄 것이다. 바닥판 상부 철근량의 감소는 건설비용의 감소와 철근의 부식문제를 최소화 시켜줄 수 있을 것이다. The current specifications for bridge decks requires the same amount of upper and lower reinforcement mats. There have been many empirical activities that the partial elimination of upper reinforcing bars was not caused the structural integrity of a deck. A simplified method is derived based on thin plate theory for three and four-girder-span bridge decks. A simplified method for bridge deck considering the effect of girder deflection is proposed based on a closed-form solution that shows good agreement with the results of finite element models. In this research, a new design approach for deck slabs is proposed based on the simplified method. The negative bending moments in a deck can be evaluated with the simplified method based on the position of a wheel load, the aspect ratio and relative stiffness and the span length. This new approach can lead to a significant reduction of the quantity of the top reinforcing steel bars in a deck. Reducing the quantify of the top reinforcement not only reduces the construction costs for bridge decks, but also reduces the corrosion of reinforcement to a minimum.

Accumulation of tissue macrophages is a significant cha-racteristic of disease-associated chronic inflammation, and facilitates the progression of disease pathology. However, the functional roles of these bone marrow-derived macrophages (BMDMs) in aging are unclear. Here, we identified age-dependent macrophage accumulation in the bone marrow, showing that aging significantly increases the number of M1 macrophages and impairs polarization of BMDMs. We found that age-related dysregulation of BMDMs is associated with abnormal overexpression of the anti-inflammatory interleukin-10. BMDM dysregulation in aging impairs the expression levels of pro-inflammatory cytokines and genes involved in B-cell maturation and activation. Phagocytosis of apoptotic Jurkat cells by BMDMs was reduced because of low expression of phagocytic receptor CD14, indicating that increased apoptotic cells may result from defective phagocytosis of apoptotic cells in the BM of aged mice. Therefore, CD14 may represent a promising target for preventing BMDM dysregulation, and macrophage accumulation may provide diagnostic and thera-peutic clues. [BMB Reports 2017; 50(1): 43-48]
