        Study on the Stability of Accumulated Layer Landslide under the Coupling Action of Earthquake and Rainfall

        Tectonic movement, heavy rainfall and other environmental factors are important factors causing the slope instability of accumulation body. Aiming at jiangdingya H1 landslide in Bailong River Basin, Zhouqu County, Gansu Province, the deformation law and stability of accumulation landslide under the coupling condition of earthquake and rainfall were systematically analyzed by means of laboratory physical model test, numerical simulation and theoretical derivation. The results show that: 1) The crack development law of landslide model test under the coupling effect of earthquake and rainfall is analyzed, and the stability characteristics of landslide deformation and failure in three stages are obtained. 2) According to the model test and numerical simulation results, it is found that the coupling effect has the greatest influence on the stability of the slope toe of the accumulation landslide, and the failure mode of the accumulation landslide changes from “pull-shear” failure to “compression-shear” failure, and the failure of the accumulation landslide occurs in a local location. 3) Based on the unbalanced thrust method and GA model, a landslide stability calculation model of loose accumulation body under the coupling effect of earthquake and rainfall was improved by comprehensively considering the effects of complex factors such as seismic inertia force and seepage force.The research results can provide reference for landslide prevention, monitoring and early warning in Zhouqu area of Gansu Province.

        Hair follicles transcriptome profiles in Bashang long-tailed chickens with different plumage colors

        Despite the rich variety in plumage color found in nature, genetic studies on how hair follicles affect pigmentation are often limited to animals that have black and white pigment. To test how gene expression influences plumage color, transcriptomes of chicken hair follicles with white, black, hemp, reed catkins, silvery grey, and landscape plumage colors were generated using Illumina sequencing. We generated six RNA-Seq libraries with over 25 million paired-end clean reads per library with percentage of paired-end clean reads ranging from 96.73 to 96.98%. 78% of the reads mapped to the chicken genome, and approximately 70% of the reads were mapped to exons and 6% mapped to introns. Transcriptomes of hair follicles producing hemp and land plumage were similar, but these two showed moderate differences compared with gray and reed colored plumage. The black and white follicle transcriptomes were most divergent from the other colors. We identified several candidate genes, including GPNMB, PMEL, TYRP1, GPR143, OCA2, SOX10, SLC45A2, KRT75, and TYR . All of these genes are known to induce pigment formation in mice. White feathers result from the lack of pigment formation, and our results suggest that the white chickens due to the recessive insertion mutation of TYR . The formation of black area size and color depth may be due to the expression levels of GPNMB, PMEL, TYRP1, GPR143, OCA2, SOX10, SLC45A2, KRT75, and TYR . The GO analysis of the differentially expressed genes (DEGs) revealed that DEGs in our transcriptome analysis were enriched in cytoskeleton and cell structure related pathways. The black plumage transcriptome showed significant differences in melanogenesis, tyrosine metabolism, and riboflavin metabolism compared with transcriptomes of other plumage colors. The transcriptome profiles of the different chicken plumage colors provide a valuable resource to understand how gene expression influences plumage color, and will be an important resource for identifying candidate genes in breeding programs.

        Porcine parvovirus nonstructural protein NS1 activates NF-κB and it involves TLR2 signaling pathway

        Background: Porcine parvovirus (PPV) is a single-stranded DNA virus that causes porcine reproductive failure. It is of critical importance to study PPV pathogenesis for the prevention and control of the disease. NS1, a PPV non-structural protein, is participated in viral DNA replication, transcriptional regulation, and cytotoxicity. Our previous research showed that PPV can activate nuclear factor kappa B (NF-κB) signaling pathway and then up-regulate the expression of interleukin (IL)-6. Objectives: Herein, the purpose of this study is to determine whether the non-structural protein NS1 of PPV also has the same function. Methods: Real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR), enzyme-linked immunosorbent assay, western blot, immunofluorescence assay and small interfering RNA (siRNA) were used. Results: Our findings demonstrated that PPV NS1 protein can up-regulate the expression levels of IL-6 and tumor necrosis factor-alpha in a dose-dependent manner. Moreover, PPV NS1 protein was found to induce the phosphorylation of IκBα, then leading to the phosphorylation and nuclear translocation of NF-κB. In addition, the NS1 protein activated the upstream pathways of NF-κB. Meanwhile, TLR2-siRNA assay showed TLR2 plays an important role in the activation of NF-κB signaling pathway induced by PPV-NS1. Conclusions: These findings indicated that PPV NS1 protein induced the up-regulated of IL-6 expression through activating the TLR2 and NF-κB signaling pathways. In conclusion, these findings provide a new avenue to study the innate immune mechanism of PPV infection.

        Investigation on Galloping of D-Shape Iced 6-Bundle Conductors in Transmission Tower Line

        The first comprehensive test base of electrical test transmission line in China has been established. Galloping response characteristics of real test tower-line system covered with D-shape ice model under natural wind load has been recorded. According to the structural details of the test tower-line, the detailed numerical model of the three spans tower-line system has been set up by nonlinear finite element method (FEM). Then, the digital stochastic wind field is presented. Galloping behaviors of the D-shape iced 6-bundle conductors in the test line under stochastic wind field is numerically simulated. Finally, the dynamic response of displacements, galloping traces, galloping modes, and frequencies are obtained. The numerical simulation results are consistent with the measured galloping characteristics results of the test transmission tower-line, which verifies the efficiency of the numerical model. It is demonstrated that the established FEM numerical method can be applied to investigate galloping behaviors of tower-line. Meanwhile, based on the numerical simulation results, the variation of stress and displacement of tower during galloping is also discussed.

        Extended State Observer Based Adaptive Backstepping Nonsingular Fast Terminal Sliding-mode Control for Robotic Manipulators with Uncertainties

        This paper proposes a novel robust finite-time tracking control scheme for robotic manipulators with uncertainties and external disturbances. In order to reduce the chattering phenomenon and enhance the robustness of the system, we design a novel extended state observer with filter function for estimating the lumped term of system uncertainties and external disturbances. We adopt the backstepping mechanism to design an adaptive backstepping nonsingular fast terminal sliding mode controller, achieve finite time convergence of the tracking errors while avoiding the singularity problem. An adaptive update law is designed for the switching gain, which further weakens the chattering problem. The finite-time convergence of tracking errors and the stability of the closed-loop system are ensured by the Lyapunov criterion. In comparison with other control methods, numerical simulation on a two-link rigid robot manipulator demonstrates the effectiveness and superiority of the proposed method.

        Temporal expression profiling of long noncoding RNA and mRNA in the peripheral blood during porcine development

        Objective: We investigated the temporal expression profiles of long noncoding RNA (lncRNA) and mRNA in the peripheral blood of pigs during development and identified the lncRNAs that are related to the blood-based immune system. Methods: Peripheral blood samples were obtained from the pigs at 0, 7, 28, and 180 days and 2 years of age. RNA sequencing was performed to survey the lncRNA and mRNA transcriptomes in the samples. Short time-series expression miner (STEM) was used to show temporal expression patterns in the mRNAs and lncRNAs. Gene ontology and Kyoto encyclopedia of genes and genomes analyses were performed to assess the genes' biological relevance. To predict the functions of the identified lncRNAs, we extracted mRNAs that were nearby loci and highly correlated with the lncRNAs. Results: In total of 5,946 lncRNA and 12,354 mRNA transcripts were identified among the samples. STEM showed that most lncRNAs and mRNAs had similar temporal expression patterns during development, indicating the expressional correlation and functional relatedness between them. The five stages were divided into two classes: the suckling period and the late developmental stage. Most genes were expressed at low level during the suckling period, but at higher level during the late stages. Expression of several T-cell-related genes increased continuously during the suckling period, indicating that these genes are crucial for establishing the adaptive immune system in piglets at this stage. Notably, lncRNA TCONS-00086451 may promote blood-based immune system development by upregulating nuclear factor of activated T-cells cytoplasmic 2 expression. Conclusion: This study provides a catalog of porcine peripheral blood-related lncRNAs and mRNAs and reveals the characteristics and temporal expression profiles of these lncRNAs and mRNAs during peripheral blood development from the newborn to adult stages in pigs.

        Effect of lateral differential settlement of high-speed railway subgrade on dynamic response of vehicle-track coupling systems

        A difference in subgrade settlement between two rails of a track manifests as lateral differential subgrade settlement. This settlement causes unsteadiness in the motion of trains passing through the corresponding area. To illustrate the effect of lateral differential subgrade settlement on the dynamic response of a vehicle-track coupling system, a three-dimensional vehicletrack- subgrade coupling model was formulated by combining the vehicle-track dynamics theory and the finite element method. The wheel/rail force, car body acceleration, and derailment factor are chosen as evaluation indices of the system dynamic response. The effects of the amplitude and wavelength of lateral differential subgrade settlement as well as the driving speed of the vehicle are analyzed. The study reveals the following: The dynamic responses of the vehicle-track system generally increase linearly with the driving speed when the train passes through a lateral subgrade settlement area. The wheel/rail force acting on a rail with a large settlement exceeds that on a rail with a small settlement. The dynamic responses of the vehicle-track system increase with the amplitude of the lateral differential subgrade settlement. For a 250-km/h train speed, the proposed maximum amplitude for a lateral differential settlement with a wavelength of 20 m is 10 mm. The dynamic responses of the vehicle-track system decrease with an increase in the wavelength of the lateral differential subgrade settlement. To achieve a good operation quality of a train at a 250-km/h driving speed, the wavelength of a lateral differential subgrade settlement with an amplitude of 20 mm should not be less than 15 m. Monitoring lateral differential settlements should be given more emphasis in routine highspeed railway maintenance and repairs.

        Enhanced anticorrosion of methyl acrylate by covalent bonded N-alkylpyridinium bromide for X70 steel in 5 M HCl

        Methyl acrylate connects covalently with alkylpyridinium bromide to build N-alkyl-3-(2-methox-ycarbonyl-vinyl)pyridinium bromide (MPA-n, n = 8–14). MPA-n inhibited X70 steel from corrosion in5 M HCl with inhibition efficiency of 98% and the inhibition capacity was in the order of MPA-14 > MPA-12 > MPA-10 > MPA-8. Quantum chemical calculation unveiled that MPA-n adsorbed on iron surface byO atom in MA moiety offering electrons to d orbital of iron and by p* orbital of pyridinium acceptingelectrons from iron, and MPA-n with longer alkylchain possessed higher EHOMO, lower DE, largermolecular volume and dipole moment, all suggesting the same corrosion inhibition order as that inexperiments

        Genome-wide identification of polyphenol oxidase (PPO) family members in eggplant (Solanum melongena L.) and their expression in response to low temperature

        Browning of fresh-cut eggplant (Solanum melongena L.) reduces its sensory and nutritional qualities and further influences consumption. Polyphenolic oxidases (PPOs) are key enzymes involved in browning, but the mechanisms that regulate the expression of PPO genes are still unclear. Here, 12 SmPPO genes were identified and phylogenetic analysis clustered these genes into four branches. Protein and cis-regulatory element analyses showed that the SmPPO gene family has a conserved gene structure and diverse functions. Gene expression analysis in different tissues showed that the expression of SmPPO2, SmPPO3, SmPPO6, SmPPO7, and SmPPO10 was higher in the flesh of the browning-sensitive inbred line ‘36’ than in the flesh of the browning-resistant line ‘Fu’. Furthermore, almost all SmPPO genes in ‘36’ were upregulated at 4 °C and 36 °C compared with those in ‘Fu’, and the expression increased earlier after harvest. In addition, SmPPO1, SmPPO6, SmPPO7, and SmPPO10 expression was significantly elevated in ‘36’ after 2 days at 36 °C. These results suggest that SmPPOs are key modulators of eggplant browning and provide candidate genes for further research on the mechanisms regulating fruit browning.

        Diffusion mechanism of deep shale gas and its carbon isotope fractionation: a combined simulated and mathematical analysis

        Unlike the release pressure process in shale gas exploitation, in this study, the concentration diffusion process is deeply discussed under constant temperature-pressure conditions, which aims to evaluate the preservation conditions of shale gas reservoirs in combination with isotopic fractionation characteristics. The isobaric diffusion experiment emphasized that decreasing pressure can lead to enhanced diffusion and isotope fractionation. The established mathematical model not only confirms the results of the simulating experiment, but also suggests that the contributions of Fick, Knudsen and surface diffusion to the migration of methane (including 12CH4 and 13CH4) vary in the geological evolution of shale reservoirs, which are mainly controlled by the pore system and gas pressure. Based on the analysis of specific samples of Longmaxi shale (r = 11.7 nm) and Niutitang shale (r = 1.3 nm), we propose that: 1) high pressure condition (> 20 MPa) can significantly limit Knudsen diffusion, which is the reason for reducing the total diffusion coefficient (DTotal) and isotope fractionation, while low pressure (< 10 MPa) and small pore radius (ca. 1 nm) can strengthen the fractionation; 2) pore connectivity and heterogeneity can lead to an order of magnitude change in DTotal, while the influence of temperature is relatively small. According to the mathematical model, it can be expected that enhanced dissipation intensity and isotope fractionation of shale gas may occur in the process of basin uplift and pressure relief, which can potentially be used for the evaluation of reservoir preservation.
