Background: Ginsenoside compound K (CK) is a promising drug candidate for rheumatoid arthritis. This study examined the impact of polymorphisms in NR1I2, adenosine triphosphate-binding cassette (ABC) transporter genes on the pharmacokinetics of CK in healthy Chinese individuals. Methods: Forty-two targeted variants in seven genes were genotyped in 54 participants using Sequenom MassARRAY system to investigate their association with major pharmacokinetic parameters of CK and its metabolite 20(S)-protopanaxadiol (PPD). Subsequently, molecular docking was simulated using the AutoDock Vina program. Results: ABCC4 rs1751034 TT and rs1189437 TT were associated with increased exposure of CK and decreased exposure of 20(S)-PPD, whereas CFTR rs4148688 heterozygous carriers had the lowest maximum concentration ($C_{max}$) of CK. The area under the curve from zero to the time of the last quantifiable concentration ($AUC_{last}$) of CK was decreased in NR1I2 rs1464602 and rs2472682 homozygous carriers, while $C_{max}$ was significantly reduced only in rs2472682. ABCC4 rs1151471 and CFTR rs2283054 influenced the pharmacokinetics of 20(S)-PPD. In addition, several variations in ABCC2, ABCC4, CFTR, and NR1I2 had minor effects on the pharmacokinetics of CK. Quality of the best homology model of multidrug resistance protein 4 (MRP4) was assessed, and the ligand interaction plot showed the mode of interaction of CK with different MRP4 residues. Conlusion: ABCC4 rs1751034 and rs1189437 affected the pharmacokinetics of both CK and 20(S)-PPD. NR1I2 rs1464602 and rs2472682 were only associated with the pharmacokinetics of CK. Thus, these hereditary variances could partly explain the interindividual differences in the pharmacokinetics of CK.
The inter-connection of sports bra design attributes and elderly women's perceptions

As a sports facility category, a sports bra is regarded as an enabling technology garment with advanced functions and new cultural meanings. Women, particularly elderly women, are encouraged to wear a sports bra during physical activity because wearing a sports bra can not only prevent them from exercise-induced injuries but also increase the acceptance of their bodies. However, most studies usually targeted younger women. The requirements and expectations of elderly women on sports bras differ from younger individuals. Hence, this study aims to advance and explore the design demands, providing insights into age-friendly sports bra design by using more permeable physical and psychological boundaries based on conceptual frameworks. The sports bra evaluation words were extracted and calculated to construct the network. The appearance frequency of keywords, closeness centrality, betweenness centrality, and interconnection between every pair of words were illustrated and analyzed by using semantic-based network analysis. Network analysis deduced that the discomfort of sports bras was closely related with the support, tightness, cup design, and strap design. Beside, thermal management contributes significantly to the comfort. The outlook of a sports bra was associated with cup design. The Page-rank result showed that cup, tightness, hot, straps and quality were the top five important design attributes. The unobservable mechanisms underlying the keywords provide an alternative model, which can help to explore the key improvements for sports bra design.

BACKGROUND: This study aims to explore the potential mechanism of action of the newly discovered hsa_- circ_0128899 (circSPEF2) in diffuse large B-cell lymphoma (DLBCL). METHODS: circSPEF2, miR-16-5p and BTB and CNC homologue 2 (BACH2) expression patterns in DLBCL patients and cell lines were studied by RT-qPCR. The biological function of circSPEF2 in vitro and in vivo was investigated by function acquisition experiments. The proliferation activity of lymphoma cells was detected by MTT. Bax, Caspase-3, and Bcl-2 were determined by Western Blot. Apoptosis and the ratio of CD4 to Treg of immune cells in the co-culture system were analyzed by flow cytometry. The mechanism of action of circSPEF2 in DLBCL progression was further investigated by RIP and dual luciferase reporter experiments. RESULTS: circSPEF2 was a circRNA with abnormally down-regulated expression in DLBCL. Increasing circSPEF2 expression inhibited the proliferative activity and induced apoptosis of lymphoma cells in vitro and in vivo, as well as increased CD4?T cells and decreased Treg cell proportion of immune cells in the tumor microenvironment. Mechanically, circSPEF2 was bound to miR-16-5p expression, while BACH2 was targeted by miR-16-5p. circSPEF2 overexpressionmediated effects on lymphoma progression were reversible by upregulating miR-16-5p or downregulating BACH2. CONCLUSIONS: circSPEF2 can influence DLBCL progression by managing cellular proliferation and apoptosis and the proportion of immune cells Treg and CD4 through the miR-16-5p/BACH2 axis.
Profit Model and Financial Analysis of Internet Medical Care: Taking JD Health as an Example

Purpose - Increasing health awareness and the rapid growth of Internet technology has aided the development of Internet medical treatment which can relieve the pressure on medical resources and improve the imbalance to some extent. Although the Internet medical industry is rapidly developing, many enterprises are non-profit making, despite growth in operating revenue. Therefore, exploring a profit model suitable for the development of Internet medical enterprises is crucial. Design/Methodology/Approach - Based on the theory of profit model elements and core competitiveness, we selected JD Health as the research object. By elaborating on the relationship between the profit model, financial analysis, and impact path of profit model components on financial data, we found that profitability problems exist in the model and proposed optimization suggestions. Findings - This study identified some problems in JD Health’s profit model: the overdependence of profit point on the pharmaceutical e-commerce business, the low stickiness of profit sources, and weak profit barriers. We proposed the following optimization suggestions: strengthen traditional businesses, improve and expand new businesses; optimize product quality and service to increase customer stickiness; continue to optimize profit leverage; and improve core competitiveness and strengthen profit barriers. Research Implications - This study provides new insights for studying the Internet medical industry and a theoretical reference for studying its profit model. Simultaneously, it can help peer companies find profit models that suit them and promote the steady development of the entire industry.
