      • Normobaric Oxygen Exposure Promote the Sensitivity of Paclitaxel in Paclitaxel Resistant Lung Cancer Cells by CHOP Activation and Disregulation of Autophagy

        Purpose Hyperbaric oxygen therapy (HBO) to increase anticancer effect, despite several positive Results, has been suggested to have a toxicity problem in other tissues, so further studies on the conditions and application of oxygen therapy are needed. The purpose of this study is to evaluate the anticancer effect of paclitaxel (PTX) in lung cancer cells under normobaric oxygen exposure. Methods Non-small cell lung cancer (NSCLC) cell line A549 and the paclitaxel resistant cell line A549/PR were used. Cell viability, ability of migration and proliferation were evaluated in room air (RA) or normobaric oxygen (NBO) air condition for 48hs. The expression apoptosis related proteins was detected. For elucidation of the cell death in PTX treated A549/PR cells by NBO, the expression level of the C/EBP homologous protein (CHOP) and autophagy related proteins were analyzed. Results NBO exposure attenuated cell viability, ability of migration and proliferation ability in A549 cells and A549/PR cells therby increased apoptotic cell death by PTX. CHOP expression of A549/PR cells was increased by NBO exposure compare to RA condition. And conversion of LC3B-I to LC3B-II by PTX was reduced in A549 and A549/PR cells in NBO exposure (Figure 1-3). Conclusions Our Results show that NBO exposure synergistically increased the apoptotic cell death of A549 and A549/PR cells by paclitaxel. Also, cell death of A549/PR cells under NBO condition was induced by CHOP-mediated disregulation of autophagy. These findings suggest that the NBO exposure can promote the sensitivity of paclitaxel in lung cancer cells by CHOP activation and disregulation of autophagy.

        국내 발암물질 배출량 특성과 암발생 추이에 관한 연구

        Objectives: The purposes of the study were to investigate hazardous pollutant emissions changes among group 1 carcinogens. The emissions characteristics were compared with national cancer registration statistics. Methods: A survey of group 1 carcinogen hazardous pollutant emissions (trichloroethylene, benzene, vinyl chloride, formaldehyde, 1,3-butadiene, ethylene oxide, chromium and its compounds, 3,3 -dichloro-4,4-diaminodiphenylmethane, chloromethyl methyl ether, arsenic and its compounds, cadmium and its compounds, o-toluidine) was conducted through a homepage for 2001-2015. The emission of hazardous chemicals and the cancer trend analysis for 2001-2015 were performed using the Korean statistical information service through its homepage as a reference. Results: Emissions of more than 95% of the substances listed as group 1 carcinogens over the last five years were made up of trichloroethylene, benzene, vinyl chloride, formaldehyde, 1,3-butadiene, and ethylene oxide. As a result of the comparison of emission results and cancer incidence rates, carcinogen pollutant emissions showed a tendency to decrease continuously. In addition, the incidence of cancer tended to increase, but showed a tendency to decrease from 2012. Conclusion: The results indicate hazardous pollutant emissions have continued to increase. However, no association between emissions and health effects was shown and more research is needed.

        공사중단 방치건축물 정비 제도개선을 위한 실증연구: 충청남도를 중심으로

        우리나라는 경제성장과 위기, 사업성 부족 등 다양한 이유로 공사중단 방치건축물이 꾸준히 발생하고 있다. 공사중단 방치건축물은 다양한 형태와 규모로 분포하고 있으며 이로 인해 도시경관을 악화, 주거안정성을 저해하고, 화재 ·범죄·악취 등 다양한 도시문제를 야기하고 있다. 정부는 관련법을 제정·시행하고 있지만 시행과정에서 현실과 거리 있는 많은 문제점이 나타나고 있다. 이에, 본 연구는 충청남도 방치건축물을 대상으로 현지 실태조사, 건축관계인 및 이해관계인 심층인터뷰, 정비사업 시행자(LH), 도 및 시군 담당공무원 FGI를 통해 방치건축물 정비사업 추진의 문제점과 개선사항을 실증적으로 도출하였다. 그리고 이를 바탕으로 향후 방치건축물의 예방, 원활한 정비사업 추진을 위한 과제와 실효성 높은 제도개선사항을 제시하였다. South Korea is renowned as one of the countries which have experienced high economic growth in the past years. However since Global Financial Crisis in 2008, its economic growth has remained slow and majority cities have faced various issues in property sector such as declining property price, the increase of uncompleted building sites, abandoned land and disproportionate rate of vacant buildings. These abandoned sites in various scales and locations, particularly nearby residential areas have negative impacts on the urban environment such as crime and vandalism. To tackle these issue, in May 2013, the "Special Act on the Maintenance of Long-Term abandoned Buildings" was enacted and has been put into operation since May 2014. Subsequently, a base maintenance plan was established in 2016 by the Ministry of Land, Infrastructure and Transport. Nevertheless the plan has not been implemented properly due to the limitation of financial and human resources, lack of expertise and ambiguity of laws and guidelines. This study examines the challenges and barriers of the implementation of the Act and recommendations to resolve the urban issues are drawn. This study is a case study of Chungcheongnam-do and the main methods used are in-depth interviews and field surveys with a wide range of stakeholders including the main developer (LH), architects, surveyor and local authority officers. The findings suggest several improvement in the 'Special Act on the maintenance of long-term abandoned buildings'.

        Effects of aging on accompanying intermittent hypoxia in a bleomycin-induced pulmonary fibrosis mouse model

        Background/Aims: Obstructive sleep apnea (OSA) is prevalent in older patients with idiopathic pulmonary fibrosis (IPF); however, it is underrecognized. OSA is characterized by intermittent hypoxia (IH) and sleep fragmentation. In this study, we evaluated the effects of IH in an older mouse model of bleomycin-induced lung fibrosis. Methods: Bleomycin-induced mice (C57BL/6, female) were randomly divided into four groups of young vs. old and room air (RA)-exposed vs. IH-exposed. Mice were exposed to RA or IH (20 cycles/h, FiO2 nadir 7 ± 0.5%, 8 h/day) for four weeks. The mice were sacrificed on day 28, and blood, bronchoalveolar lavage (BAL) fluid, and lung tissue samples were obtained. Results: The bleomycin-induced IH-exposed (EBI) older group showed more severe inflammation, fibrosis, and oxidative stress than the other groups. The levels of inflammatory cytokines in the serum and BAL fluid increased in the EBI group. Hydroxyproline levels in the lung tissue increased markedly in the EBI group. Conclusions: This study demonstrates the possible harmful impact of OSA in an elderly mouse model of lung fibrosis. This study further suggests that older patients with IPF and OSA may be more of a concern than younger patients with IPF. Further research is required in this area.
