<P>Visfatin (Nampt/PBEF) plays a pivotal role in the salvage pathway for NAD(+) biosynthesis. Its potent inhibitor, FK866, causes cellular NAD(+) levels to decline, thereby inducing apoptosis in tumor cells. In an effort to improve the solubility and binding interactions of FK866, we designed and synthesized IS001, in which a ribose group is attached to the FK866 pyridyl ring. Here, we report the crystal structure of rat visfatin in complex with IS001. Like FK866, IS001 is positioned at the dimer interface, and all of the residues that interact with IS001 are involved in hydrophobic or pi-pi-stacking interactions. However, we were unable to detect any strong interactions between the added ribose ring of IS001 and visfatin, which implies that a bulkier modifying group is necessary for a tight interaction. This study provides additional structure-based information needed to optimize the design of visfatin inhibitors.</P>
Crystal structure of <i>Helicobacter pylori</i> MinE, a cell division topological specificity factor

<P>In Gram-negative bacteria, proper placement of the FtsZ ring, mediated by nucleoid occlusion and the activities of the dynamic oscillating Min proteins MinC, MinD and MinE, is required for correct positioning of the cell division septum. MinE is a topological specificity factor that counters the activity of MinCD division inhibitor at the mid-cell division site. Its structure consists of an anti-MinCD domain and a topology specificity domain (TSD). Previous NMR analysis of truncated <I>Escherichia coli</I> MinE showed that the TSD domain contains a long α-helix and two anti-parallel β-strands, which mediate formation of a homodimeric α/β structure. Here we report the crystal structure of full-length <I>Helicobacter pylori</I> MinE and redefine its TSD based on that structure. The N-terminal region of the TSD (residues 19–26), previously defined as part of the anti-MinCD domain, forms a β-strand (βA) and participates in TSD folding. In addition, <I>H. pylori</I> MinE forms a dimer through the interaction of anti-parallel βA-strands. Moreover, we observed serial dimer–dimer interactions within the crystal packing, resulting in the formation of a multimeric structure. We therefore redefine the functional domain of MinE and propose that a multimeric filamentous structure is formed through anti-parallel β-strand interactions.</P>
A교육청 조직문화에 대한 구성원 인식 탐색 연구: Bolman과 Deal의 조직 프레임을 중심으로

본 연구의 목적은 Bolman과 Deal(2013)의 네 가지(구조적, 인간자원적, 정치적, 상징적) 조직 프레임에 근거하여 A교육청 조직문화의 주요한 현상과 특징을 구성원들의 인식을 바탕으로 다차원적으로 분석하는 데 있다. 일반직, 전문직, 교사 등 다양한 집단을 대상으로 포커스 그룹 면담을 실시한 결과, 구조적 관점에서 교육청은 명확히 구분되지 않는 업무로 인해 부서간의 업무 떠넘기기(업무 핑퐁)가 심한 것으로 나타났고, 인간자원적 관점에서 교육청은 교육청의 구성원들이 대부분 승진 등과 같은 개인의 이익에 대한 욕구가 높은 높았으나, 전문성 개발에 관해서는 일반직과 전문직의 실천 방법이 다른 것으로 나타났다. 정치적 관점에서 교육청은 조직 내부의 직렬간, 직렬내, 부서간 구성원간의 갈등 뿐 만 아니라 조직 외부와의 갈등이 존재하였다. 상징적 관점에서 A교육청은 우리나라 교육을 선도하는 교육청이라는 데 이견은 없었으나 조직 내부에서 비전에 대한 자발적 실천은 부족하다고 지적하였다. 본 연구의 연구 결과를 바탕으로 앞으로 교육청 조직문화 개선을 위해서는 우선 구조적 차원에서 추진 정책과 사업들 간의 정합성을 높여야 할 것이며, 그동안 관행으로 인정되어온 각종의 관료적 일처리 방식과 부서 간 소통을 가로막은 지침들을 새롭게 재편하여야 할 것이다. 인간자원적 차원에서는 구성원들의 역량(전문성) 개발을 위한 각종의 훈련과 연수를 지원하고 각종 정책의 추진과정에 전 구성원이 적극 참여할 수 있는 규범을 만들어야 할 것으로 보이고, 정치적 차원에서는 편가르기식 행정관행을 원점에서 놓고 재검토하고, 생산적인 갈등 관리 방법이 도출될 수 있도록 노력하여야 할 것이다. 마지막으로, 상징적 차원에서 구조적, 인간자원적, 정치적 차원에서 발생하는 문제들을 문화적으로 치유하기 위한 방편으로서 다양한 의례와 문화적 표상들을 다수 창출해야 할 것이다. The purpose of this study is to analyze the characteristics in the organizational culture of “A” Provincial Office of Education based on Bolman and Deal’s four (structural, human resource, political, symbolic) frames. The results are as follows. From the perspective of the structural frame, it has been found that there are severe shift of responsibility between the departments due to the tasks that are not clearly distinguished. From the frame of human resource, most of the members have a specific desire for personal interests such as promotion. From the political frame, there is a conflict between the members in the office and outside the organization. From the symbolic frame, the voluntary practice of vision is lacking. In order to improve the organizational culture of the office of Education, it should increase the coherence between the implementation policy and the projects. Also, it is necessary to reorganize the guidelines to prevent communication between departments. Furthermore, it is necessary to support training for the development of competence of the employees and to make a tradition that all the members can actively participate in the decision making process. Further, it is necessary to reform the one-sided administrative practices and we need to derive productive conflict management methods. And rituals and symbols should be created so that it can become the Office of Education leading the organizational culture.

Pseudohypoparathyroidism (PHP) is a rare clinical state of hypoparathyroidism that results from target tissue resistance to parathyroid hormone, associated with a secondary, hypocalcemia-induced increase in parathyroid gland function. Classically PHP have clinical (tetany and seizures) and biochemical (hypocalcemia and hyperphosphatemia) features of hypoparathyroidism and AHO (Albright's hereditary osteodystrophy), characterized by several distinctive skeletal and developmental defects. But other variants were reported according to clinical and biochemical manifestation. The biochemical hallmark of PHP is PTH resistance. In some patients with PHP, the guanyl nucleotide regulatory protein (G-protein) is decreased by 50 percent in red blood cells in such patients. This defect appears to produce resistance to several other hormones that apparently exert their actions by stimulating the increased production of cellular cyclic AMP. Pseudohypoparathyroidism is classified to Type Ia, Type Ib and Type Ⅱ according to clinical and biochemical manifestations. We experienced recently three pseudohypoparathyroidism patients admitted to St. Mary's Hospital. One was classic PHP (type Ia), manifestated by tetany, hypocalcemia, hyperphosphatemia and AHO.The others were PHP without AHO (PHP type Ⅱ). Here we confirmed these patients to specific PHP subtype with the urinary cAMP response to PTH (J Kor Soc Endocrinol 6:170~178, 1991).
Benzene and toluene concentrations in a hemodialysis room in a medium sized South Korean city

Background/Aims: The current study was designed to determine whether the indoor air pollution in a hemodialysis room (HD) was different from that of other comparable areas in a hospital. Methods: Five air monitor samplers were hung on the ceiling and placed on the table in both the HD and general ward nursing stations, respectively. In addition, five samplers were placed in the nurse`s breathing zone of the HD and the general ward, respectively. Ten air monitor samplers were also placed on the edge of the bed in the HD, which represented the patient`s breathing zone. The levels of benzene and toluene were analyzed by GC/MS. Results: In the general ward, the toluene concentration was significantly higher in the nurse breathing zone than that for the ceiling or table samples (p=0.001). The benzene concentration was also significantly higher in the general ward nurse breathing zone than that in the HD (p=0.006). In addition, the benzene concentrations on the table were higher at the general ward as compared to the HD (p=0.028), but there was no significant difference between the ceiling, general ward station and HD. Conclusions: Both the benzene and toluene concentrations in the HD appear to be more affected by the outdoor atmospheric conditions than by any potential indoor internal sources.
ANALYSIS BY SYNTHESIS FOR ESTIMATION OF DOSE CALCULATION WITH gMOCREN AND GEANT4 IN MEDICAL IMAGE

The use of GEANT4 simulation toolkit has increased in the radiation medical field for the design of treatment system and the calibration or validation of treatment plans. Moreover, it is used especially on calculating dose simulation using medical data for radiation therapy. However, using internal visualization tool of GEANT4 detector constructions on expressing dose result has deficiencies because it cannot display isodose line. No one has attempted to use this code to a real patient's data. Therefore, to complement this problem, using the result of gMocren that is a three-dimensional volume-visualizing tool, we tried to display a simulated dose distribution and isodose line on medical image. In addition, we have compared cross-validation on the result of gMocren and GEANT4 simulation with commercial radiation treatment planning system. We have extracted the analyzed data of dose distribution, using real patient's medical image data with a program based on Monte Carlo simulation and visualization tool for radiation isodose mapping.

<P><B>Abstract</B></P> <P>Coagulation factors (II, VII, IX, X, and particularly XIa) remaining in high concentrations in intravenous immunoglobulin (IVIG) preparations can form thrombi, causing thromboembolic events, and in serious cases, result in death. Therefore, manufacturers of biological products must investigate the ability of their production processes to remove procoagulant activities. Previously, we were able to remove coagulation factors II, VII, IX, and X from our IVIG preparation through ethanol precipitation, but factor XIa, which plays an important role in thrombosis, remained in the intermediate products. Here, we used a chromatographic process using a new resin that binds with high capacity to IgG and removes procoagulant activities. The procoagulant activities were reduced to low levels as determined by the thrombin generation assay: <1.56 mIU/mL, chromogenic FXIa assay: <0.16 mIU/mL, non-activated partial thromboplastin time (NaPTT): >250 s, FXI/FXIa ELISA: <0.31 ng/mL. Even after spiking with FXIa at a concentration 32.5 times higher than the concentration in normal specimens, the procoagulant activities were below the detection limit (<0.31 ng/mL). These results demonstrate the ability of our manufacturing process to remove procoagulant activities to below the detection limit (except by NaPTT), suggesting a reduced risk of thromboembolic events that maybe potentially caused by our IVIG preparation.</P>
