        오이/흑종호박 접목에서 오이녹반모자이크바이러스의 비기주 대목인 흑종호박으로 이동

        Cucumber green mottle mosaic virus (CGMMV) was not infected in figleaf gourd by sap inoculation. However CGMMV was detected by RT-PCR from the figleaf gourd collected from a field growing cucumber grafted onto figleaf gourd in Cheonan, Chungcheongnam Province in 2008. Which field showed 100% infection rate of the virus disease. In the experiment grafted with cucumber onto figleaf gourd, transportation of CGMMV through cucumber to figleaf gourd was confirmed by RT-PCR when the virus was mechanically inoculated on the leaves of the cucumber. The amplified DNA concentration of the virus on electrophoresis gel was much higher in the cucumber than in the figleaf gourd. However, the virus particles from the figleaf gourds were not observed under electron microscopy, also sap of the figleaf gourds was not transmittable to Nicotiana benthamiana. To identify the existence of CGMMV particle, the virus was purified from figleaf gourd and cucumber growing together in the graft system. CGMMV solution extracted from the cucumber represented a typical absorption spectrum of the virus but that from the figleaf gourd did not. Only a few CGMMV particles were observed in the purified preparation from the figleaf gourd. These results confirmed that CGMMV only passed through figleaf gourd in the grafting system.         경북지역 오이에 발생하는 주요 바이러스 종류 및 발생실태

        Cucumber is high valued cash crop, for it is grown during the winter season in plastic house. Recently, virus disease spread widely in cucumber growing area and cause severe income loss. Therefore, occurrence of virus disease on cucumber were surveyed from 2004 to 2006 in Sangju and Gunwi area, Gyeongbuk province. The rate of plastic house which has infected plants was 55.0~88.6%. Infection rate was the highest at Sangju in 2006 than others and ranged from 15 to 90.0% per plastic house. The 217 samples showing virus symptom were analyzed by RT-PCR using appropriate detection primer. Zucchini yellow mosaic virus (ZYMV) has the highest infection rate (detected over 85%) and followed by Cucumber green mottle mosaic virus (CGMMV). But Watermelon mosaic virus-2 (WMV-2) was not detected in our survey. Therefore, we conclude that ZYMV is major pathogene of virus disease on cucumber.         친환경오이와 일반오이의 이화학적 특성 및 소비자 기호도

        Environment-friendly agricultural products are consumed widely, but little is known about consumers' acceptability. The objectives of this study were to 1) determine consumers' acceptability of cucumber when consumed raw, 2) determine differences in appearance liking between environment-friendly and conventionally grown cucumber samples, and 3) analyze the physicochemical properties of cucumber. White dadagi, gasi, non-pesticide mini cucumber, organic gasi, and organic Joseon cucumber samples were evaluated for consumers' acceptability using a nine-point hedonic scale; White dadagi, organic Joseon, and organic gasi cucumber samples showed the highest acceptability scores. However, there were three consumer segmentations differing their degree of cucumber liking and preference. Consumers liked the appearance of organic gasi best. Consumers' preference of cucumbers did not change upon evaluation of the acceptability of the appearance of whole fruit with and without a label indicating an environment-friendly status. Additionally, the physicochemical properties of the cucumbers-including color (skin and flesh), length, circumference, weight, water activity, brix, and texture (hardness and fracturability)-were measured. All measurements showed significant differences among samples, with the exception of water activity and hardness. White dadagi was 3.8 brix, which was the highest among cucumbers, followed by non-pesticide mini cucumber and organic Joseon. White dadagi, organic Joseon, and organic gasi showed greater fracturability than the other cucumber samples. 친환경농산물이 많이 소비되고 있는 반면에 소비자의 기호도에 관한 연구는 부족한 실정이다. 본 연구의 목적은 국내에서 시판되는 오이를 생식으로 섭취할 경우 소비자의 기호도에 차이가 있는지를 알아보고, 친환경오이와 일반오이에 대한 소비자 외관 기호도 차이가 있는지, 그리고 친환경오이와 일반오이의 물리화학적 특성을 연구하고자 하였다. 9점척도법을 이용하여 백다다기오이, 가시오이, 무농약미니오이, 유기농가시오이, 유기농조선오이에 대한 소비자의 기호도를 조사하였고, 전반적인 기호도에서는 백다다기오이, 유기농조선오이, 유기농가시오이를 선호하였다. 그러나 오이에 대한 기호도와 선호도가 다른 소비자의 소그룹이 있었다. 외관 기호도를 평가하였을 때, 친환경농산물 여부에 관계없이 유기농가시오이를 가장 선호하였다. 물리화학적 특성으로 껍질과 과육의 색도, 오이의 길이, 둘레, 무게, 수분활성도, 당도, 조직감(경도, 부서지는 정도)을 측정한 결과, 수분활성도와 경도를 제외한 모든 특성에서 시료간에 유의적인 차이가 있었다(p<0.05). 백다다기오이의 당도는 3.8 brix로 오이 시료 중 가장 높았고, 그 다음으로는 무농약미니오이와 유기농조선오이의 순이었다. 백다다기오이, 유기농조선오이, 유기농가시오이는 부서지는 정도가 다른 시료보다 높았다. 명도는 백다다기오이가 가장 높았으며, 무농약미니오이, 유기농가시오이, 가시오이 사이에는 유의적인 차이가 없었다.

        취청오이, 백다다기오이, 노각의 조리방법에 따른 영양 성분 및 잔존율 연구

        본 연구는 오이의 종류와 조리방법에 따라 일반성분 및 무기질 함량을 비교하고자 하였다. 오이는 취청오이, 백다다기오이, 노각 3종류를 생재료와 삶기, 찌기, 굽기, 로스팅, 볶기, 튀기기, 전자레인지 7가지 조리방법을 사용하였다. 이를 통해 일반성분과 무기성분의 잔존율과 각각의 함량에 대한 종류 및 조리방법의 상관성을 조사하였다. 실험에 사용된 오이는 모두 조리과정에서 중량이 감소하였다. 대부분의 일반성분과 무기질은 종류와 조리방법의 영향을 모두 받았지만 단백질은 영향을 받지 않았고, 철 함량은 조리방법에 의해 영향을 받지 않았으며, 구리 함량은 종류에 대한 영향을 받지 않는 것으로 나타났다. 일반성분 중 단백질, 지방, 회분은 튀기기 했을 때 함량이 높았고 수분은 원재료, 삶기, 찌기를 했을 때 높았다. 무기질은 로스팅과 전자레인지 조리를 한 취청오이의 함량이 높았고, 백다다기오이와 노각은 튀기기를 했을 때 전반적인 함량이 높았다. 단백질, 지방, 수분, 회분의 잔존율은 각각 30~100%, 10~100%, 35~90%, 30~100% 범위로 나타났다. 무기질은 로스팅, 전자레인지, 삶기를 한 취청오이가 높은 잔존율을 보였고, 백다다기오이와 노각은 튀기기를 했을 때 높은 잔존율을 보였다. 취청오이는 찌기를 했을 때, 백다다기오이와 노각은 삶기와 찌기를 했을 때 잔존율이 낮았으며, 특히 노각의 경우 20~45%로 낮은 잔존율을 보였다. 이러한 결과들을 종합해 보았을 때 오이에 대한 조리 전후에 대한 기초 자료로 활용이 가능할 것으로 생각되며, 다양한 오이 종류에 대한 추가적인 연구가 필요할 것으로 생각된다. General component and mineral contents according to species of Capsicum annuum were investigated. In addition, chuichung cucumber, white dadagi cucumber, and yellowish overripe cucumber were cooked by using seven different methods (boiling, steaming, pan-cooking, roasting, pan-frying, deep-frying, and microwaving), and retention rates of general components and minerals according to species were evaluated. Weight loss was observed in all cucumber species using all cooking methods. Most general component and mineral contents of C. annuum samples were significantly different according to species, and cooking method (P<0.05), whereas Fe and Cu contents were not significantly affected by species and cooking method, respectively. As a result, the highest protein, fat, ash contents were detected in deep-fried C. annuum, and high moisture contents were detected in raw, boiled, and steamed C. annuum. High mineral contents were detected in roasted and microwaved chuichung cucumber, deep-fried white dadagi cucumber and yellowish overripe cucumber. Retention rates of C. annuum by cooking method, ranged from 30∼100% for protein, 10∼100% for fat, 35∼90% for moisture, and 30∼100% for ash. High retention rates were observed for minerals in boiled, roasted, and microwaved chuichung cucumber, and deep-fried white dadagi cucumber and yellowish overripe cucumber. However, in yellowish overripe cucumber, mineral retentions rate were remarkably lowered by boiling and steaming, resulting in rates of 20∼45%. These results were the data used as the basis for development of cucumbers.

        Effects of different algae in diet on growth and interleukin (IL)-10 production of juvenile sea cucumber Apostichopus japonicus

        The experiment was conducted to investigate the effects of different algae in diet on growth, survival, and interleukin-10 productions of sea cucumber. At first, a 9-week feeding trail was conducted to evaluate the growth performance and survival of the sea cucumber fed one of the six experimental diets containing ST (Sargassum thunbergii), UL (Ulva lactuca), UP (Undaria pinnatifida), LJ (Laminaria japonica), SS (Schizochytrium sp.), and NO (Nannochloropsis oculata) in a recirculating aquaculture system. The result showed that survival was not significantly different among the dietary treatments, and the specific growth rate (SGR) of sea cucumber fed the UL diet (1.58% d−1) was significantly higher than that of sea cucumber fed the other diets (P < 0.05), except for the LJ and NO diets. Secondly, interleukin (IL)-10 gene expression was determined where mice splenocytes were stimulated with 10 μg ml−1 of sea cucumber extracts for 2 h. The result showed that IL-10 gene expression levels were significantly increased in UL, LJ, and NO diets fed sea cucumber extracts compared to other experimental diets. The results suggest that dietary inclusion with Ulva lactuca, Laminaria japonica, and Nannochloropsis oculata algae may improve the growth of juvenile sea cucumber and could upregulate IL-10 gene expression in mice splenocytes. Such detailed information could be helpful in further development of more appropriate diets for sea cucumber culture.

      • Shelter material and shape preferences of the sea cucumber, <i>Apostichopus japonicus</i>

        <P><B>Abstract</B></P> <P>The purpose of this study was to determine the physical properties of shelter design that could enhance sea cucumber cultivation. The structural material (PVC, cement mortar, and porous cement mortar), hole shape (triangle, square, and circle), and hole size (0.0014, 0.0025, and 0.0079 m<SUP>2</SUP>) of shelters were analyzed to determine sea cucumber preferences. Ninety sea cucumbers were used (30 individuals per body size group) and the initial weights were 20.56 ± 4.97 g, 60.80 ± 6.05 g, and 115.80 ± 9.13 g for small, medium, and large sea cucumbers, respectively. Changes in behavioral characteristics in relation to body weight at 10, 15, and 20 °C were also analyzed. Additionally, movement characteristics on surfaces of varying degrees of roughness (a. PVC [0.68–0.9 mm], b. cement mortar [1.5–1.8 mm], and c. porous cement mortar [2.0–2.2 mm]) were determined using particle image velocimetry (PIV) and 2D velocity fields. By day, we observed that sea cucumbers were attached to the inside of circular holes (<I>P</I> < 0.05). Sea cucumbers were attached to shelters made of cement mortar (average 10) > porous cement mortar (average 8) > PVC (average 6), regardless of water temperature or body weight. The preference for sheltering was higher in the small- and medium-size sea cucumber groups than in the group of large-size sea cucumbers. Most sea cucumbers attached to the shelters were in the weight range 20–60 g. The average distances moved by the small, medium, and large sea cucumbers were 1.501, 1.475, and 1.310 m, respectively, and their movement speeds (×10<SUP>−3</SUP> m/s) were 0.181, 0.289, and 0.269, respectively, regardless of water temperature. The movement speed of the sea cucumbers was the fastest (0.373 × 10<SUP>−3</SUP> m/s) at 20 °C. The PIV measurements showed that different velocities and forces were generated when the sea cucumbers moved to shelter materials of varying degrees of surface roughness (force: PVC > porous cement mortar > cement mortar). The sea cucumbers produced the greatest amount of force when moving over smooth surfaces, such as PVC. The smaller the sea cucumber, the less they were affected by temperature change.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The effects of different shelter properties on the behavior of sea cucumbers were investigated. </LI> <LI> The movement distance and speed of sea cucumbers were measured using cameras, software, and particle image velocimetry. </LI> <LI> The shelter preference was higher for small and medium sea cucumbers than for large ones. </LI> <LI> Different forces were generated when the sea cucumbers moved over shelter materials of varying roughness. </LI> </UL> </P>

        Optimal growth conditions and economic analysis of sea cucumber releasing

        We tried to find the optimal growth conditions of sea cucumber and to analyze the economic effectiveness of the sea cucumber seedling release project in Korea. We first examined the optimal growth conditions of sea cucumber in the relating literatures. Then, we analyzed the economic effectiveness of the sea cucumber seedling release project of the Woncheon fishing village union of Gyeongnam Province in 2016–2018 by using the cost benefit analysis method. The net income of the release project of the Woncheon fishing village union was 69,850 Korean won. The benefit to cost ratio of the sea cucumber seedling release project of the Woncheon fishing village union was estimated to be 1.7, indicating that the project was economically feasible. In order to improve the economic feasibility of the sea cucumber release project, as we see in the case of the Woncheon fishing village union, it is necessary to manage the purchase of the sea cucumber seedling, to improve the recapture rate of sea cucumber, and to manage marketing of sea cucumber.

        Whole-Body Microbiota of Sea Cucumber (Apostichopus japonicus) from South Korea for Improved Seafood Management

        Sea cucumber (Apostichopus japonicus) is a popular seafood source in Asia, including South Korea, and its consumption has recently increased with recognition of its medicinal properties. However, because raw sea cucumber contains various microbes, its ingestion can cause foodborne illness. Therefore, analysis of the microbiota in the whole body of sea cucumber can extend our understanding of foodborne illness caused by microorganisms and help to better manage products. We collected 40 sea cucumbers from four different sites in August and November, which are known as the maximum production areas in Korea. The microbiota was analyzed by an Illumina MiSeq system, and bacterial amounts were quantified by real-time PCR. The diversity and bacterial amounts in sea cucumber were higher in August than in November. Alpha-, Beta-, and Gammaproteobacteria were common dominant classes in all samples. However, the microbiota composition differed according to sampling time and site. Staphylococcus warneri and Propionibacterium acnes were commonly detected potential pathogens in August and November samples, respectively. The effect of experimental Vibrio parahaemolyticus infection on the indigenous microbiota of sea cucumber was analyzed at different temperatures, revealing clear alterations of Psychrobacter and Moraxella; thus, these shifts can be used as indicators for monitoring infection of sea cucumber. Although further studies are needed to clarify and understand the virulence and mechanisms of the identified pathogens of sea cucumber, our study provides a valuable reference for determining the potential of foodborne illness caused by sea cucumber ingestion and to develop monitoring strategies of products using microbiota information.
