        위점막에서 조직화학적 및 면역조직화학적 염색을 이용한 Helocibaxter pylori 의 검출

        logic test (ELISA, BIO-RAD GAP method), histochemical stain (Warthin-Starry silver stain), immunohistochemical stain (Avidin biotin conjugate method, ABC method) were performed to identify the presence of H. pylori infection. H. pylori infection was defined that 2 of 3 studies were positive. Results: Positive result to H. pylori infection by serologic test, Warthin-Starry silver stain and immunohistochemical stain were 82% (28/34 cases), 65% (22/34 cases), 68% (23/34 cases). Sensitivity and specificity were 68%, 20% in serologic test, 95%, 83% in Warthin-Starry silver stain, 100%, 100% in immunohistochemical stain. Correlation among these tests was best between Warthin-Starry silver stain and immunohistochemical stain. Conclusion: By the above results, we can conclude that histochemical stain and immunohistochemical stain are the most accurate diagnostic method to identify the infectivity of R. pylori.

        전 암컷 3배체 넙치(Paralichthys olivaceus)에 대한 효율적인 세포유전학 분석법

        We cytogenetically analyzed a triploid King-Nupchi strain of the olive flounder Paralichthys olivaceus to define the simplest, most rapid, and most effective method of ploidy analysis in aquaculture farms. Female triploidy of the flounder King-Nupchi strain was induced by cold shock (3 min post-fertilization at 2-4°C for 45 min). Triploid induction was confirmed by erythrocyte measurement (nuclear volume, 29.15±2.10 μm3); flow cytometry (2.14±0.03 pg/cell); chromosome count (3N=72); Ag-NOR banding; and silver staining. Silver staining of finned cells obtained using a solid tissue technique was the most effective method of ploidy verification.

        겔스캐너를 이용한 변성아크릴아마이드 겔의 형광 DNA 검출

        형광 염료와 레이저 겔스캐너 장비를 이용하여 변성아크릴 아마이드 겔에서 전기 영동된 DNA를 신속하고 간편한 방법으로 기존의 은염색법과 비슷한 감도로 검출하고자 하였다. 변성아크릴아마이드 겔을 형광 염료인 SYBR Green (Molecular Probes)이나 Vistra Green (Amersham Bioscience) 0.01 X 희석액 (pH 8)으로 염색한 후 480nm 레이져, 520nm filer 옵션으로 스캔하여 DNA를 검출하였으며, 검출감도는 기존의 은염색법과 비슷하면서 염색 단계를 한 단계로 줄일 수 있었다. The denature polyacrylamide gel stain silver nitrate is used for routine nucleic acid detection. More sensitive stains, such as Vistra Green, SYBR Green are available to address a broad range of DNA applications requiring lower detection limits in polyacrylamide gel formats. Gel Scanners, laser-scanning instruments, provide sensitive fluorescence detection of DNA gel stains. We established one step fluorescent impregnation enhanced sensitivity with simple, rapid and low cost. We have applied this fluorescent staining procedure for the routine analysis of DNA profiles generated by SSR amplification.

        Time Course of Remote Neuropathology Following Diffuse Traumatic Brain Injury in the Male Rat

        Traumatic brain injury (TBI) can affect different regions throughout the brain. Regions near the site of impact are the most vulnerable to injury. However, damage to distal regions occurs. We investigated progressive neuropathology in the dorsal hippocampus (near the impact) and cerebellum (distal to the impact) after diffuse TBI. Adult male rats were subjected to midline fluid percussion injury or sham injury. Brain tissue was stained by the amino cupric silver stain. Neuropathology was quantified in sub-regions of the dorsal hippocampus at 1, 7, and 28 days post-injury (DPI) and coronal cerebellar sections at 1, 2, and 7 DPI. The highest observed neuropathology in the dentate gyrus occurred at 7 DPI which attenuated by 28 DPI, whereas the highest observed neuropathology was at 1 DPI in the CA3 region. There was no significant neuropathology in the CA1 region at any time point. Neuropathology was increased at 7 DPI in the cerebellum compared to shams and stripes of pathology were observed in the molecular layer perpendicular to the cerebellar cortical surface. Together these data show that diffuse TBI can result in neuropathology across the brain. By describing the time course of pathology in response to TBI, it is possible to build the temporal profile of disease progression.       • Microglial Contribution to Glioma Progression: an Immunohistochemical Study in Eastern India

        Human glioma, arising from glial cells of the central nervous system, accounts for almost 30%of all brain tumours, neoplasms with a poor prognosis and high mortality rates worldwide. In the present study we assessed tissue architectural modifications associated with macrophage lineage cells, controversial major immune effector cells within the brain, in human glioma tissue samples from eastern India. Ethically cleared post-operative human glioma samples from our collaborative neurosurgery unit with respective CT/MRI and patient history were collected from the Nodal Centre of Neurosciences in Kolkata, over 9 months. Along with conventional histopathology, samples were subjected to silver-gold staining and fluorescence tagged immunophenotyping for the detection of electron dense brain macrophage/microglia cells in glioma tissue, followed by immune-phenotyping of cells. With higher grades, CD11b+/Iba-1+ macrophage/microglia architecture with de-structured boundaries of glioma lesions indicated malfunction and invasive effector state. Present study documented a contribution of microglia to glioma progression in Eastern India.

        Microsatellite 대립유전자 분석을 통한 개에서의 친자감별

        Microsatellite allele analysis has been used for individual identification and paternity test. In the present study, the biological father of three puppies was determined by using microsatellite allele amplification analysis. The mother bitch of the litter was a Poongsan dog. The three stud dogs that could have inseminated the bitch, by being in the same residence, were a white Poosan dog, a mixed breed, and a white Jindo dog. DNA was obtained from all the relevant dogs by buccal swabbing. Four loci of tetranucleotide repeat microsatellite were PCR-amplified, and analyzed by polyacrylamide gel electrophoresis and silver staining. The results of genotyping unambigously assigned the Poongsan dog as the biological father. There was no evidence of superfecundation. Therefore, the present study demonstrated the usefulness of microsatellite allele analysis as a simple, efficient method of paternity test in dogs.

        Purpose: Information on the status of human epidermal growth factor receptor 2 (HER2) is important for treatment planning in breast cancer; therefore, equivocal results of HER2 overexpression obtained using immunohistochemical staining (IHC) require additional evaluation regarding HER2 amplification, which can be obtained by other techniques such as fluorescence in situ hybridization (FISH) assay. The silver-enhanced in situ hybridization (SISH) technique is known to be comparable with FISH and can be performed under a bright field microscope. In this study, the authors evaluated the concordance between the results obtained by IHC and SISH used for determining HER2 status. Methods: Paraffin-embedded tissues from 511 breast cancer patients were prepared and the medical records of the patients were reviewed. HER2 amplification status was evaluated by SISH assay by determining the HER2/chromosome 17 ratio or HER2 copy number status. The slides were blindly reviewed without IHC information. Results: Among the collected tissues, invasive ductal carcinoma was detected in 440 patients (86.1%) and 27 (5.3%) showed ductal carcinoma in situ. The concordance rate between the results obtained by IHC and SISH was 90.6% (κ=0.723). Two cases of negative IHC results had equivocal SISH results. Conclusion: SISH was found to be a useful method for evaluating HER2 status in case of an equivocal result of IHC.

        A Research on the Relationship between Chemical Composition of Nanoparticles and the Color of Medieval Stained Glass

        Stained glass windows are an important elementin both religious and civil architecture. Stainedglass windows are fascinating art crafts becausethey are multifunctional, e.g., mainly acting asfilters for light and as iconographic supports. Theart of stained flourished in the time period of 1100and 1500, when magnificent windows were createdfor great cathedrals. The stained glass windowswere the major pictorial art form in northernFrance, Germany and England during theflourishing period. It is known that there are therelationship between color and colorants. The colorsin the stained glass are attributed to differentchemical compounds that were added to the moltenglass during processing. The addition of metallicoxides called pot-metal glass changed the color ofthe windows-gold for red, copper for green, cobaltfor blue and soon. Most of the properties of traditional ceramicsoriginated from the fact that their raw materials(natural clays) are nanosized. The practical metalnanoparticles used for the pottery and glass arecopper, silver and gold dispersed in an opticallyclear matrix. Incorporation of metallic silver at theglass surface has remained the usual method toobtain yellow stained glass windows. The electron cloud at the surface of a nanoparticleresonates with different wavelengths of lightdepending upon their frequency at the nanoscale. Depending on the size of the nanoparticle, theelectron cloud resonates with a particular wavelengthof light and absorb that wavelength. A nanoparticleof about 90 nm in size will absorb colors on thered and yellow end of the color spectrum toachieve the nanoparticle appear blue-green.
