        아밀로이드 베타 단백질에 의해 유도된 신경세포 독성에 대한 백합과 숙근류의 억제 효과 탐색

        The amyloid β-protein (Aβ) is the principal component of the senile plaques characteristic of Alzheimer's disease (AD) and elicits a toxic effect on neurons in vitro and in vivo. Many environmental factors including antioxidants and proteoglycans modify Aβ toxicity. In this study, we have investigated the effects of methanol-extracts of Liliaceae root on Aβ-induced oxidative cell death in cultured rat pheochromocytoma (PC12) cells. For this study, we used three different Aβ fragments (Aβ25-35, Aβ1-42 Aβ1-43) for the induction of cytotoxicities to PC12 cells. We previously measured cytotoxicities of the extracts of Liliaceae to both human neuronal SK-N-MC and PC12 cells. Fifteen out of the 19 Liliaceae extracts at 100μg/ml showed no cytotoxic effects to both SK-N-MC and PC12 cells. Three different Aβ fragments (Aβ25-35, Aβ1-42 Aβ1-43) exhibited cytotoxicities to both SK-N-MC and PC12 cells, each possessing 1, 5, and 5μg/ml IC50 values, respectively. Among the Liliaceae roots, methanol-extracts of Polygonatum falcatum, Ophiopogon japonicus, Liriope platyphylla, and Hemerocallis fulva strongly attenuated cytotoxicities induced by the three Aβ fragments. Especially methanol-extracts of Ophiopogon japonicus was the most effective in protecting PC12 cells from the Aβ25-35-induced oxidative cell death. These results suggest that extracts of Liliaceae root may contain the protective component (s) against Aβ-induced cell death in PC12 cells. (Cancer Prev Res 10, 242-250, 2005)

        얼레지 인경의 성분에 관한 연구

        From the bulbs of Erythronium japonicum Decaisne(Liliaceae), ${\beta}-sitosterol$ and its $3-O-{\beta}-D-glucoside$ together with fatty acids were isolated. All compounds were identified on the basis of spectral data. Campesterol and stigmasterol were also identified as minor components. Paimitic acid was identified as a major component and stearic, oleic, arachidic, behenic, tricosanoic and lignoceric acids were also characterized as minor ones.

        밀나물 지하부의 Steroid saponin 성분

        Four steroids have been isolated from the subterranean parts of Smilax nipponica Miquel (Liliaceae). The structures of these compounds were characterized on the basis of chemical and speectral data. They were $\beta$-sitosterol, $\beta$-sitosterol-3-O-$\beta$-D-glucopyranoside, neotigogenin-3-O-$\beta$-D-glucopyranoside and a new steroidal saponin designated as smilanippin A which was elucidated as neotigogenin-3-O-$\beta$-D-fucopyranosyl(1$\rightarrow$6)-$\beta$-D-glucopyranos ide.

      • 청미래덩굴의 뿌리에서 추출한 조분획의 항균활성

        청미래덩굴(Smilax china L.)의 뿌리에서 추출한 methanol, n-hexane, chloroform 및 수용성 추출물의 항균활성을 Agrobacterium rhizogenes, Agrobacterium tumefaciens, Candida utilis, Saccharomyces cerevisiae, Bacillus megaterium, Bacillus natto, Bacillus subtilis 및 Escherichia coli에 대하여 disc diffusion과 broth dilution method로 조사하였다. Methanol추출물은 A. rhizogenes, A. tumefaciens, B. megaterium 및 B. subtilis 대하여 항균활성이 나타났고, 수용성 추출물은 B. megaterium와 B. subtilis에 대하여 항균활성을 보였으나 hexane과 chloroform 분획물은 항균활성이 없었다. 또한 수용성 추출물은 800ppm 농도에서 현탁배양중인 A. rhizogenes, A. tumefacience, B. megaterium 및 B.subtilis에 대하여 각각 80, 40, 20, 81%의 성장 억제를 보였고, 특히, 동일 농도의 수용성 추출물을 재첨가한 경우 B. subtilis의 성장이 완전히 억제되었다. Gas chromatography/mass spectrometry에 의한 추출물의 분석 결과, 그 주요 구성 성분은 5-(hydroxymethyl)-2-furancarboxaldehyde(46.95%), 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one(21.08%) 및 2-furancarboxaldehyde(5.85%)였다. Antimicrobial activity of four extract fractions from the root of Smilax china L.(Liliaceae) was investigated by diffusion and broth dilution methods against several microorganisms, Agrobacterium rhizogenes, Agrobacterium tumefaciens, Candida utilis, Saccharomyces cerevisiae, Bacillus megaterium, Bacillus natto, Bacillus subtilis and Escherichia coli. Methanol extract exhibited potent antimicrobial activity against A. rhizogenes, A. tumefaciens, B. megaterium and B. subtilis. However, hexane and chloroform extracts did not show antimicrobial activity against the test microorganisms. Aqueous extract inhibited growth of the tested microbial strains. The relative growth inhibition of A. rhizogenes, A. tumefaciens, B. megaterium and B. subtilis were 80, 40, 20, 81% at 800㎍/mL, respectively. Methanol extract was analyzed by gas chromatography/mass spectrometry(GC/MS). Major components were determined to be 5-(hydroxymethyl)-2-furancarboxaldehyde(46.95%), 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one(21.08%) and 2-furancarboxaldehyde(5.85%).

        Fritillaria walujewii의 Alkaloid 성분

        Four alkaloids were isolated from the n-BuOH extract of the bulbus of Fritillaria walujewii (Liliaceae). On the basis of spectroscopic evidences, the structures of these compounds were established as Alkaloid fr-5(11-deoxo-6-oxo-5${\alpha}$,6-dihydrojervine), delavinone, delavine and petilidine.

        A New Steroidal Glycoside from Allium macrostemon Bunge

        A phytochemical investigation of Allium macrostemon Bunge (Liliaceae) afforded the new pregnane steroidal glycoside, named allimacroside F (1), along with three known glycosides, benzyl-O-a-L-rhamnopy- ranosyl-(1→6)-b-D-glucopyranoside (2), phenylethyl-O-a-L-rhamnopyranosyl-(1→6)-b-D-glucopyranoside (3), (Z)-3-hexenyl-O-a-L-rhamnopyranosyl-(1→6)-b-D-glucopyranoside (4). The identification and structural elucidation of a new compound (1) was carried out based on spectral data analyses (1H-NMR, 13C-NMR, 1H-1H COSY, HSQC, HMBC, and NOESY) and HR-FAB-MS.

        Action of Asparagus racemosus Against Streptozotocin-Induced Oxidative Stress

        Antidiabetic treatment with tubers of Asparagus racemosus (Liliaceae) extract (100 and 250 mg/kg body weight) for three weeks showed significant reduction in thiobarbituric acid reactive substances (TBARS) and glutathione reductase (GSH-R) in both liver and kidney. The treatment with A. racemosus significantly altered the glutathione (GSH) and GSH-R to be comparable with the control group. A. racemosus and glibenclamide treated rats showed decreased lipid peroxidation that is associated with increased activity of superoxide dismutase (SOD) and Catalase (CAT). The ability of A. racemosus on tissue lipid per oxidation and antioxidant status in diabetic animals has not been studied before. The result of this study thus shows that though, A. racemosus possesses moderate antidiabetic activity, but it exhibits potent antioxidant potential in diabetic conditions.

        Further studies have been done on the constituents of the tubers of Liriope spicata L_(OUR) (Liliaceae). Four steroidal glycosides, tentatively designated as compounds Ⅰ, Ⅱ, Ⅲ and Ⅳ, were isolated from the n-BuOH soluble fraction of this plant. The structures of these glycosides were established as β-sitosterol glucoside, prosapogenin Ⅱ of spicatoside A, ophiopogonin B, and prosapogenin Ⅲ of spicatoside A.

        Major Lineages of the Genus Lilium (Liliaceae) Based on nrDNA ITS Sequences, with Special Emphasis on the Korean Species

        We present most comprehensive phylogenetic analysis of 196 accessions of Lilium representing 83 species and 14 varieties of Lilium and three outgroup genera (Cardiocrinum, Notholirion, and Fritillaria) to investigate infrageneric relationships within Lilium as well as to determine the origin and evolution of Korean species of Lilium. We used the internal transcribed spacer sequences of nuclear ribosomal DNA and phylogenetic analysis using maximum parsimony and Bayesian inference identified several major lineages within Lilium. Only one section,Martagon, turned out to be monophyletic in the study. Three sections, Archelirion, Liriotypus, and Pseudolirium,are not monophyletic because two, one, and two species in each section were placed in other lineage, respectively. Two major lineages of section Leucolirion were confirmed in this study, and as several previous studies suggested,section Sinomartagon is highly polyphyletic. The origin of Lilium hansonii, a Korean endemic to Ullung Island, is perplexing given the fact that it has ribotype of Martagon,while its cpDNA haplotype is similar to Sinomartagon. The origin of another endemic, Lilium amabile, is equally elusive and additional phylogenetic and phylogeographic studies will shed light on their evolutions in Korea. We determined that Lilium callosum var. flavum originated from L. callosum in Southern Korea.
