        Understanding intestinal health in nursery pigs and the relevant nutritional strategies

        In the modern pig production, pigs are weaned at early age with immature intestine. Dietary and environmental factors challenge the intestine, specifically the jejunum, causing inflammation and oxidative stress followed by destruction of epithelial barrier and villus structures in the jejunum. Crypt cell proliferation increases to repair damages in the jejunum. Challenges to maintain the intestinal health have been shown to be related to changes in the profile of mucosa-associated microbiota in the jejunum of nursery pigs. All these processes can be quantified as biomarkers to determine status of intestinal health related to growth potential of nursery pigs. Nursery pigs with impaired intestinal health show reduced ability of nutrient digestion and thus reduced growth. A tremendous amount of research effort has been made to determine nutritional strategies to maintain or improve intestinal health and microbiota in nursery pigs. A large number of feed additives have been evaluated for their effectiveness on improving intestinal health and balancing intestinal microbiota in nursery pigs. Selected prebiotics, probiotics, postbiotics, and other bioactive compounds can be used in feeds to handle issues with intestinal health. Selection of these feed additives should aim modulating biomarkers indicating intestinal health. This review aims to define intestinal health and introduce examples of nutritional approaches to handle intestinal health in nursery pigs.         Effects of Yarrowia lipolytica supplementation on growth performance, intestinal health and apparent ileal digestibility of diets fed to nursery pigs

        Objective: The objective was to evaluate the efficacy of increasing supplementation of Yarrowia lipolytica (YL) up to 3.0% replacing 1.6% poultry fat and 0.9% blood plasma for growth performance, intestinal health and nutrient digestibility of diets fed to nursery pigs. Methods: Twenty-four pigs weaned at 24 d of age (initial body weight at 7.2±0.6 kg) were allotted to three dietary treatments (n = 8) based on the randomized complete block. The diets with supplementation of YL (0.0%, 1.5%, and 3.0%, replacing poultry fat and blood plasma up to 1.6% and 0.9%, respectively) were fed for 21 d. Feed intake and body weight were recorded at d 0, 10, and 21. Fecal score was recorded at every odd day from d 3 to 19. Pigs were euthanized on d 21 to collect proximal and distal jejunal mucosa to measure intestinal health markers including tumor necrosis factor-alpha, interleukin-8, immunoglobulin A and immunoglobulin G. Ileal digesta was collected for apparent ileal digestibility (AID) of nutrients in diets. Data were analyzed using Proc Mixed of SAS. Results: Supplementation of YL (1.5% and 3.0%) replacing poultry fat and blood plasma did not affect growth performance, fecal score and intestinal health. Supplementation of YL at 1.5% did not affect nutrient digestibility, whereas supplementation of YL at 3.0% reduced AID of dry matter (40.2% to 55.0%), gross energy (44.0% to 57.5%), crude protein (52.1% to 66.1%), and ether extract (50.8% to 66.9%) compared to diets without supplementation. Conclusion: Yarrowia lipolytica can be supplemented at 1.5% in nursery diets, replacing 0.8% poultry fat and 0.45% blood plasma without affecting growth performance, intestinal health and nutrient digestibility. Supplementation of YL at 3.0% replacing 1.6% poultry fat and 0.9% blood plasma did not affect growth performance and intestinal health, whereas nutrient digestibility was reduced.         Intestinal Alkaline Phosphatase: Potential Roles in Promoting Gut Health in Weanling Piglets and Its Modulation by Feed Additives - A Review

        The intestinal environment plays a critical role in maintaining swine health. Many factors such as diet, microbiota, and host intestinal immune response influence the intestinal environment. Intestinal alkaline phosphatase (IAP) is an important apical brush border enzyme that is influenced by these factors. IAP dephosphorylates bacterial lipopolysaccharides (LPS), unmethylated cytosine-guanosine dinucleotides, and flagellin, reducing bacterial toxicity and consequently regulating toll-like receptors (TLRs) activation and inflammation. It also desphosphorylates extracellular nucleotides such as uridine diphosphate and adenosine triphosphate, consequently reducing inflammation, modulating, and preserving the homeostasis of the intestinal microbiota. The apical localization of IAP on the epithelial surface reveals its role on LPS (from luminal bacteria) detoxification. As the expression of IAP is reported to be downregulated in piglets at weaning, LPS from commensal and pathogenic gram-negative bacteria could increase inflammatory processes by TLR-4 activation, increasing diarrhea events during this phase. Although some studies had reported potential IAP roles to promote gut health, investigations about exogenous IAP effects or feed additives modulating IAP expression and activity yet are necessary. However, we discussed in this paper that the critical assessment reported can suggest that exogenous IAP or feed additives that could increase its expression could show beneficial effects to reduce diarrhea events during the post weaning phase. Therefore, the main goals of this review are to discuss IAP's role in intestinal inflammatory processes and present feed additives used as growth promoters that may modulate IAP expression and activity to promote gut health in piglets.

        쥐 모델에서 고지방사료로 악화된 대장 건강에 대한 콩의 개선 효과

        Obesity is associated with impaired intestinal epithelial barrier function, which contribute to host systemic inflammation and metabolic dysfunction. Korean traditional foods, fiber-rich bean products, have been various biological activities in antiinflammatory responses, but has not reported the large intestinal health. In this study, we investigated the intestinal health promoting effect of cooked soybeans (CSB) on high fat diet (HFD)-induced obesity model. SD rat were fed either a HFD or HFD supplemented with 10.6% CSB (HFD+CSB) for animal experimental period. CSB treatment significantly decreased the HFD-induced weights of body and fat. Also, CSB treatment improved HFD-reduced tight junction components (ZO-1, Claudin-1, and Occludin-1) mRNA expression in large intestine tissue. Additionally, histopathological evaluation showed that CSB treatment attenuated the HFD-increased inflammatory cells infiltration and epithelial damages in large intestine tissue. At the genus level, effects of CSB supplement not yet clear, while dietary effects showed differential abundance of several genera including Lactobacillus, Duncaniella, and Alloprevotella. NMDS analysis showed significant microbial shifts by HFD, while CSB did not shift gut microbiota. CSB increased the abundance of the genera Anaerotignum, Enterococcus, Clostridium sensu stricto, and Escherichia/Shigella by linear discriminant analysis effect size analysis, while reduced the abundance of Longicatena and Ligilactobacillus. These findings indicate that CSB supplement improves HFD-deteriorated large intestinal health by the amelioration of tight junction component, while CSB did not shift gut microbiotas. 비만은 숙주의 전신 염증 및 대사 기능 장애에 기여하는 장 상피 장벽 기능 저하와 관련이 있다. 한국의 전통 식품으로 식이섬유가 풍부한 콩 제품은 항염증 반응을 비롯한 다양한 생물학적 활성을 나타내어 왔으나 대장 건강에 대해서는 보고된 바가없다. 본 연구에서는 고지방 식이(HFD)를 섭취한 비만 모델에서 콩(CSB)에 대한 장 건강 증진 효과를 조사하였다. SD 쥐에게 동물 실험 기간 동안 HFD 또는 10.6% CSB가 함유된HFD (HFD + CSB)를 제공하였다. CSB의 섭취는 HFD로 유발된 체중과 지방 축적 증가를 현저하게 감소시켰다. 또한, CSB 의 섭취는 대장 조직에서 HFD에 의해 감소된 밀착 결합 지표(ZO-1, Claudin-1 및 Occludin-1)의 mRNA 발현을 개선시켰다. 또한, 조직병리학적 평가에서도 CSB 섭취는 대장 조직에서HFD에 의해 증가된 염증 세포 침윤과 대장 상피 조직 붕괴를개선하는 것으로 나타났다. Genus 수준에서 HFD 섭취에 의해 Lactobacillus, Duncaniella, Alloprevotella 등 미생물 종의 abundance 차이는 확인되었으나, CSB 섭취로 인한 영향은 명확하게 나타나지 않았다. NMDS 분석에서 HFD 섭취에 의해 유의적인 장내미생물 생태 이동을 보여주었지만 CSB 섭취는 차이가 없었다. HFD와 CSB 간 유의적으로 차이가 나타나는 genera를 조사하기 위해 LEfSe를 수행한 결과, CSB는 Anaerotignum, Enterococcus, Clostridium sensu stricto 및 Escherichia/Shigella 속의 풍부함을 증가시킨 반면Longicatena 및 Ligilactobacillus의 풍부함을 감소시켰다. 이러한 결과는 CSB 섭취는 긴밀한 접합 성분을 개선하여HFD로 악화된 대장 건강을 개선하는 반면 장내미생물생태에긍정적인 효과를 미치는지에 대해서는 명확하지 않았다.

        비오비타 3균의 장 건강 유용성과 배합의의에 대한 고찰

        BIOVITA 3 bacterial species (Clostridium butyricum, Lactobacillus sporogenes, and Bacillus subtilis) are spore forming probiotic bacteria that are blended as granules. Unlike lactic acid bacteria, which account for most of the available probiotics, the BIOVITA 3 bacterial species are easy to store and stably transit to the intestine, where they promote intestinal health owing to the unique features of the individual strains. The BIOVITA 3 bacterial species differ in their proliferation in different regions of the intestinal tract because of their differing oxygen demands. The aerobic Bacillus subtilis mainly proliferates in the upper small intestine, which has a high oxygen concentration. The facultative anaerobe Lactobacillus sporogenes proliferates well in the small intestine. The anaerobic Clostridium butyricum grows best in the large intestine, which has a relatively low oxygen concentration. In addition, each bacterial species displays positive effects on promoting the growth of bacteria resident in the lower intestine. The differential proliferation of the BIOVITA 3 bacterial species influences intestinal regulation throughout the intestine from the small to the large intestine. The species also improve intestinal regulation through mutual interactions that increase the growth of other beneficial bacteria.

        Health-seeking behaviour, views and preferences of adults with suspected increased intestinal permeability: A cross-sectional survey of Australian adults

        Background: The public health consequence of increased intestinal permeability (IP) is currently limited by the lack of patient-centred research. This study aims to describe the health-seeking behaviour of Australian adults with suspected IP. Methods: A cross-sectional survey of 589 Australian adults who have been diagnosed with IP or have suspected (undiagnosed) IP. Results: The majority (56.2%) of participants with suspected IP reported self-diagnosing their condition, with the majority (56.7%) of these participants preferring to be assessed using an accurate method by a general practitioner or naturopath. On average, Australian adults with suspected IP spent 11.1 (95% CI: 9.5, 12.8) years between first suspecting IP and receiving a formal diagnosis. Over the previous 12 months, participants spent an average of $699 on consultation fees, $2176 on dietary supplements for the treatment of IP, and an average of $287 on the assessment of IP. Furthermore, participants who find it difficult to live on their available household income spent significantly more (mean=$2963) on dietary supplements compared to participants who find it easy to live on their available household income ($1918) (p=0.015). Conclusion: The investigation of Australian adults with suspected IP found the majority of participants experienced a considerable length of time between first suspecting IP and receiving a diagnosis of IP. The out-of-pocket expenditure associated with the management of IP suggests a financial burden for people with suspected IP. The results of this study provide novel patient-centred considerations that can be used to inform a clinical practice guideline for the management of IP.

        장내 세균총과 위장관 질환

        본 연구에서는 최근에 연구되어 온 장내 세균총이 특히 주요 장질환의 발병에 있어서, 어떠한 역할을 하는지 보고된 여러 문헌들을 중심으로 연구 결과들을 요약 하였다. 예를 들자면, 면역체계를 매개하여 병이 악화되는 여러 질환에서 정상일 때와 달리 바뀐 장내 세균총을 통해 악화된다고 보고된 바 있다. 장내 세균총의 역할에 대해 많이 연구된 장질환 중에서, 많이 연구된 3 개의 질환은 과민성 장 증후군, 염증성 장질환, 대장암이다. 그러나, 사람의 장내에 존재하는 세균총은 몸에 이로우며, 비타민A 합성, 단사슬지방산의 생산, 담즙산 대사과정과 같은 장내 생리적 기능 매개를 통해 장내 항상성을 유지한다고 알려져 있다. 이와 같이, 장내에 존재하는 이로운 세균 군집과 해로운 세균 군집의 균형은 장내 건강에 주요한 영향을 미친다. 장질환을 포함한 여러 질환의 발병 및 진행에서 장내 세균총의 변화가 주요 원인으로 추측되고 있는 실정이다. 현재까지 보고된 많은 연구 결과에도 불구하고, 어떤 장내 세균총 구성이 몸에 가장 이로운 지학계에서도 의견이 분분한 상태이다. 본 논문에서는, 주요 장질환으로 알려진 과민성 장 증후군, 염증성 장질환, 대장암과 장내세균총과의 관계에 대해 연구한 논문들에 대해 연결 지어 요약하였다. 마지막으로, 장내 세균총을 매개로 악화되는 장질환을 완화하며, 장내 건강을 지키기 위한 수단으로 천연물을 이용한 치료 전략을 제시하고자 한다. The gut microbiome has been studied extensively over the past decade with most scientific reports focused on the adverse role of the gut microbiome on gastrointestinal diseases. For example, the altered gut microbiome exacerbates the development of immune system-mediated damage in many diseases. The most studied pathologies include irritable bowel syndrome, inflammatory bowel diseases, and colitis-associated cancer. On the other hand, intestinal microflora is also beneficial and contributes to the intestinal physiology by the synthesis of vitamins, production of short chain fatty acids and bile acid metabolism, thereby maintaining gut homeostasis. Therefore, the balance between commensal and pathogenic bacteria populations influences mainly the maintenance of intestinal health. Changes in the intestinal microflora have been suspected to be the underlying causes of multiple diseases. Despite the immense amount of published data, the optimal gut microbiome composition is still controversial. This review briefly outlines the connection between the gut microbiome and critical gastrointestinal diseases focusing on three prominent intestinal disorders: irritable bowel syndrome, inflammatory bowel diseases, and colitis-associated cancer disorders. Finally, intervention strategies using natural products for the alleviation of these diseases and the maintenance of a health gut microbiome are suggested.

        Balancing Act of the Intestinal Antimicrobial Proteins on Gut Microbiota and Health

        The human gut houses a diverse and dynamic microbiome critical for digestion, metabolism, and immune development, exerting profound effects on human health. However, these microorganisms pose a potential threat by breaching the gut barrier, entering host tissues, and triggering infections, uncontrolled inflammation, and even sepsis. The intestinal epithelial cells form the primary defense, acting as a frontline barrier against microbial invasion. Antimicrobial proteins (AMPs), produced by these cells, serve as innate immune effectors that regulate the gut microbiome by directly killing or inhibiting microbes. Abnormal AMP production, whether insufficient or excessive, can disturb the microbiome equilibrium, contributing to various intestinal diseases. This review delves into the complex interactions between AMPs and the gut microbiota and sheds light on the role of AMPs in governing host-microbiota interactions. We discuss the function and mechanisms of action of AMPs, their regulation by the gut microbiota, microbial evasion strategies, and the consequences of AMP dysregulation in disease. Understanding these complex interactions between AMPs and the gut microbiota is crucial for developing strategies to enhance immune responses and combat infections within the gut microbiota. Ongoing research continues to uncover novel aspects of this intricate relationship, deepening our understanding of the factors shaping gut health. This knowledge has the potential to revolutionize therapeutic interventions, offering enhanced treatments for a wide range of gut-related diseases.

        복합미생물발효 제제식품이 인체건강 및 장내미생물균총에 미치는 영향

        The purpose of this study was to investigate the effects of “H” fermented food product of Bayo Co., Ltd., on human health and intestinal microflora, and their relationships. Waist size was measured for the 90 applicants from Seongsangu, Changwon before and after taking the “H” product for 15 days. They were also surveyed to identify the demographic, physical and personal characteristics, and health characteristics. Microbial community structures were analyzed by a pyrosequencing method for the stools collected for some representative applicants. The greatest waist size reduction of 3.3 cm observed in the 40s. Overall satisfaction for the formula food was above average. Especially relief of atopy symptoms was highest in the 20s, constipation relief in the 40s, and digestion promotion in the 60s. In addition, the beneficial enteric bacteria (Faecalibacterium sp., Bifidobacterium sp. and Alistipes sp.) appeared to increase overall after the ingestion by all these ages. The microbial community structure of the 60s was significantly different from those of the 20s and 40s. Ingestion of this fermented food appeared to cause beneficial microbial community structures that could maintain a good intestinal health. Development of various intestinal health indicators based on this study will greatly contribute to the development and utilization of customized functional fermented foods.

        Current status of global pig production: an overview and research trends

        Global pig production has increased by 140% since the 1960s. The increase in global population, coupled with improving socioeconomic conditions of many countries has led to an increased consumption of meat globally, including pork. To keep up with demand and capitalize on economic opportunities, the countries of China, the United States (US), and the European Union (EU) have become the top 3 pork producers globally. China is of particular interest, as it is the both the largest country in pork production and pig numbers, as well as being the largest importer of pork from other countries. Globally, the efficiency of pork production has improved, in relation to the integration of pig production and the dramatic increase in research efforts in pig nutrition and production. Through integration, large producers can consolidate resources and maximize profits and efficiency. The increased research interest and efforts in pig production have given scientists and producers the opportunity to collaborate to adapt to challenges and identify possible solutions to issues brought on by a volatile global market. Intestinal health (23%), general nutrition and growth (23%), and amino acid nutrition (15%) were the top 3 areas (61%) leading research trends in pig nutrition and production. Major dietary interventions with feed additives evaluated include functional amino acids, feed enzymes, pre-/pro-/post-biotics, and phytobiotics with a common goal to improve the growth efficiency by enhancing nutrient utilization and intestinal health. With increasing global issues with environment, pig producers and the supporting scientists should continue their efforts to improve the production efficiency and to reduce the environmental footprint from pig production.
