        도시농업용 쑥부쟁이의 활용에 있어 근경의 냉장기간과 용토에 관한 연구

        도시민들이 전통나물 자원식물 식물을 쉽게 구입 및 재배하는데 필요한 자료 축적 측면에서 쑥부쟁이 근경의 유통기술 개발과 이용성을 조사하였다. 쑥부쟁이는 전남 광양산으로 2년차 근경을 3월 25일에 굴취하여 이용하였다. 쑥부쟁이 근경을 식재한 후 초장 10cm를 기준으로 수확기를 조사한 결과 아파트 베란다에서 재배한 것은 4월 21일경, 정원과 옥상에서 재배한 것은 5월 17일 경이었다. 근경을 굴취하여 4±0.5℃에서 저장한 것을 8월 1일에 출고하여 수태에 적층하여 비닐포장 한 다음 상온의 암상태에 4일간 두어도 84% 이상의 생존율을 나타내었다. 쑥부쟁이 근경을 굴취하여 4±0.5℃에서 저장해 둔 다음 6.27일에 출고한 것은 식재 후 18일이 되어야 10cm 이상 자랐으며, 7월 30일에 출고한 것은 24일이 되어야 10cm 이상 자랐다. 4±0.5℃에 저장한 근경을 물에서 20일간 싹기름한 결과 8.2cm 자랐다. 200mL 물에 식물용 영양제 6mL를 첨가하여 재배한 결과 85일이 되어도 생육이 좋았다. 이와 같은 결과를 고려할 때 쑥부쟁이는 2년차 근경을 냉장하면서 출하기를 조절할 수 있고, 실내에서 물재배도 용이하므로 도시농업, 학습교구 및 원예치료 프로그램 용 등으로 활용성이 높을 것으로 판단되었다. This study was conducted to accumulate the data necessary to easily purchase and cultivation of traditional herbs in a respect for promoting the utilization of plant resources of citizen and developing the marketing techniques. Aster yomena was used 2-year rhizomes growing in Gwangyang, Chonnam and it collected on March 25. We examined the harvest time on the basis of on 10cm of plant height after planting the rhizome of Aster yomena. Aster yomena was harvested on April 21 in the veranda of apartment and harvested on May 1 in the garden and the rooftop. We used rhizome of aster stored at 4±0.5℃ and rhizome of Aster yomena was delivered on Aug. 1. The survival rates of rhizome were over 84% in the condition to be stored in room temperature in the dark room for 4 days after wrapped and piled up with sphagnum moss, and packed with plastic film. We used rhizome of Aster yomena stored at 4±0.5℃ and rhizome of Aster yomena was delivered on June 27. rhizome of Aster yomena at 18 days after planting were grown over 10cm and at July 30 were grown over 10cm at 24 days after planting. Aster yomena sprout of rhizome stored at 4±0.5℃ was 8.2cm at 20 days after sprouting in the water. Aster yomena grown with the supplement of 6mL of plant nutrition agent into the 200mL water had maintained good growth during 85 days after planting. Therefore, we suggest that Aster yomena can be effectively used in the aspects of urban agriculture, teaching tools and horticultural therapy program, because rhizome of aster was can be stored at the cold temperature, and adjusted freely by the5)people and grown well in the water culture.

      • Anti-allergic effect of <i>Aster yomena</i> on ovalbumin-sensitized mouse and RHL-2H3 cells via Th1/Th2 cytokine balance

        <P><B>Abstract</B></P> <P>Recently, the incidence of allergies is increasing worldwide. Immunoglobulin E (IgE)-mediated hypersensitivity is central to the pathogenesis of asthma, and other allergic diseases. <I>Aster yomena</I> (AY) have been valued for their medical properties including anti-inflammation, anti-pyresis and analgesia properties. In this study, we investigated the anti-allergic effects of AY and isoquercitrin, a functional compound of AY, using a mouse allergy model and cells. In mice with ovalbumin (OVA)-induced allergic rhinitis, oral administration of AY or isoquercitrin reduced secretion of IgE and histamine in the plasma. AY or isoquercitrin regulated both Th1 and Th2 cytokines levels in the plasma. In addition, AY and isoquercitrin showed dose-dependent inhibition of β-hexosaminidase and reduction in mRNA levels of Th2 cytokines in RBL-2H3 cells. Thus, our results demonstrate that AY or isoquercitrin suppresses cytokine production of the allergy response pathway, resulting in prevention or alleviation of allergy symptoms.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>Aster yomena</I> extracts inhibit the production of IgE and histamine in OVA-induced mice. </LI> <LI> <I>Aster yomena</I> extracts suppressed Th2 cytokines and increased Th1 cytokines production in OVA-induced mice. </LI> <LI> <I>Aster yomena</I> extracts reduced the number of Th2 cells and increased the number of Th1 cells. </LI> <LI> <I>Aster yomena</I> extracts inhibit the secretion of β-hexosaminidase of RBL-2H3 cells. </LI> <LI> <I>Aster yomena</I> extracts exerted anti-allergy activity. </LI> </UL> </P>

        쑥부쟁이 제품 개발을 위한 소비자 인식도 및 요구도 분석

        This paper provides basic data for product developers by investigating the consumption status, consumer perception, andconsumer demand on products using Aster Yomena. Two hundred ninety-five people over 20 years of age were analyzedaccording to gender and age. In the purchase of Aster Yomena products, men were the higher purchasers except for powder. In age, noodles, wild vegetables, jangajji, tteok, and powder were consumed by those older than 30 years. In an analysisof purchase when developing Aster Yomena products, the male purchase intention was higher in all items except forkalguksu in the staple, and male purchase intention was higher in only bibimbapseasoning among aster powder additionsauce and all items in dessert. The purchase intention of those older than 30 years was higher in all items of staples, asterpowder addition sause and sause The purchase intention of those older than 30 yearse was higher in green vegetables juiceand milk tea among drink, and all items except ice cream among dessert. An analysis of the popularization of Aster Yomenarevealed higher perceptions of men in the medium (TV, internet, online cafe, agro-fishery market), experience program (areatourism, exposition), and product development (convenience food, recipe development). The medium (agro-fisherymarket), experience program, and product development were more recognized in their 30s or more. The products with asteryomana require the most effective marketing to men 30 years and older, and require a strategy that will interest those in their20s.

        쑥부쟁이를 활용한 나물김부각 제조 및 품질특성

        본 연구는 김부각(KB), 쑥부쟁이 나물부각(SB), 쑥부쟁이 나물김부각(NB)을 제조하여 품질특성을 조사하였다. NB는 쑥부쟁이 나물을 잘게 썰어 들깨 양념하여 김에 부착시켜 찹쌀풀을 발라 부각으로 제조 한 후 동결 건조하였다. KB보다 SB와 NB의 기름 흡유율이 높아 나물의 기름 흡수가 많았다. SB의 DPPH radical 소거능이 가장 높게 측정되었으며 KB 보다는 NB가 높게 측정되어 나물이 김보다 항산화능이 높았다. 저장기간이 길어질수록 수분함량은 증가하는 반면 최대하중, 항복응력 및 경도는 감소하는 경향이었다. 전반적인 기호도는 NB>KB>SB 순이었고, NB와 SB 간 유의한 차이가 있었다. $25^{\circ}C$에서 40일 저장한 NB의 산가 및 과산화물가는 점차 증가하는 경향이었으나, 산패의 기준에는 못 미쳤다. 따라서, $25^{\circ}C$ 이하에서 40일까지는 저장 가능할 것으로 판단되며, 쑥부쟁이뿐만 아니라 기능성이 입증된 다양한 나물을 첨가한 부각을 제조한다면 다양한 소비자 기호도를 충족시킬 수 있는 부각제조가 가능할 수 있을 것으로 본다. In this study, kim (dried laver) bugak ("KB"), Aster yomena bugak ("SB"), and namul (vegetables)-kim bugak (combination of Aster yomena and kim) ("NB") were prepared and their quality characteristics were investigated. NB was made by slicing Aster yomena, seasoning it with perilla seeds, attaching it to kim and applying glutinous rice paste, followed by freeze-drying. SB had a higher oil absorption rate than KB, reflecting the high oil absorption rate of namul (here, Aster yomena). DPPH radical scavenging activity was highest for SB; whereas, NB had higher DPPH radical scavenging activity than KB, indicative of the higher antioxidant capacity of namul, as compared to kim. With longer storage period, moisture content tended to be increased, while maximum load, yield stress and hardness tended to be decreased. Overall preference was in the order of NB>KB>SB, without statistical significance. Acid value and peroxide value of NB, stored at $25^{\circ}C$ for 40 days, tended to be increased gradually but did not meet the criteria for rancidity. Therefore, NB could be stored at $<25^{\circ}C$ for > 40 days. Furthermore, it would be possible to make functional bugak using a variety of namul, as well as Aster yomena.

        Neuroprotective effect of Aster yomena ethanolic extract in HT-22 and SK-N-MC cells based on antioxidant activity

        The antioxidant potentials of ethanolic extracts derived from Aster yomena (A. yomena) were evaluated by assessing their total phenolic and flavonoid contents and radical scavenging activities. Our findings revealed that the 60% ethanolic extract of A. yomena exhibited the most robust antioxidant properties among all extracts tested. Specifically, the IC50 values for the 2,2'- azino-bis (3-ethyl benzothiazoline-6-sulfonic acid) and 1,1-diphenyl-2-picrylhydrazyl radical scavenging activities of the 60% ethanolic extract from A. yomena were determined to be 1,640.30 μg/mL and 2,655.10 μg/mL, respectively. Moreover, the inhibitory effect on malondialdehyde increased with the 60% ethanolic extract from A. yomena. To assess the neuroprotective effects, we examined the impact of the 60% ethanolic extract from A. yomena against H2O2-induced cytotoxicity in HT-22 (mouse hippocampal neuronal cell line) and SK-N-MC (human neuroblastoma cell line) cells. The results demonstrated a significant improvement in cell viability and reduced intracellular oxidative stress. Furthermore, the major bioactive compounds present in the 60% ethanolic extract from A. yomena were identified as chlorogenic acid and rutin through high-performance liquid chromatography (HPLC) analysis.

      • Effect of apigenin isolated from <i>Aster yomena</i> against <i>Candida albicans</i>: apigenin-triggered apoptotic pathway regulated by mitochondrial calcium signaling

        <P><B>Abstract</B></P> <P><B>Ethnopharmacological relevance</B></P> <P> <I>Aster yomena</I>, a perennial herb that grows mainly in South Korea, has been employed in the traditional temple food for antibiotic efficacy. Recently, it was reported that apigenin isolated from <I>A. yomena</I> has a physical antifungal mechanism targeting membrane against <I>Candida albicans</I>.</P> <P><B>Aim of the study</B></P> <P>Our study aimed to investigate the biochemical responses underlying the antifungal activity of apigenin isolated from <I>A. yomena</I> due to lack studies reporting the investigation of intracellular responses of apigenin in <I>C. albicans</I>.</P> <P><B>Materials and methods</B></P> <P>Apigenin was isolated from the aerial parts of <I>A. yomena</I>. To evaluate apigenin-induced inhibitory effects and membrane damages, the measurement of the cell viability assay and the flux of cytosolic components were performed with at various concentrations. Intracellular external potassium and calcium levels were assayed by an ion-selective electrode meter, Fura2-AM and Rhod2-AM, respectively. Mitochondrial dysfunctions were analyzed by using JC-1, Mitotracker Green FM, and MitoSOX Red dye. H<SUB>2</SUB>DCFDA, glutathione, and MDA assay were used to detect oxidative damage. Also, flow cytometry was carried out to detect apoptotic hallmarks using Annexin V-PI, TUNEL, and FITC-VAD-FMK staining. Tetraethylammoniumchloride (TEA), Ruthenium red (RR), and <I>N</I>-acetylcysteine (NAC) were used as a potassium channel blocker, mitochondrial calcium uptake inhibitor, and reactive oxygen species (ROS) scavenger, respectively.</P> <P><B>Results</B></P> <P>We confirmed that there was no decrease of cell survival percentages in crude extracts of <I>A. yomena</I> treatment, however, only isolated apigenin has the antifungal effect in <I>C. albicans</I>. Apigenin triggered a dose-dependent mitochondrial calcium uptake followed by mitochondrial dysfunction, loss of the membrane potential and an increase in the mitochondrial mass and ROS. Apigenin also induced intracellular redox imbalance as indicated by the ROS accumulation, glutathione oxidation, and lipid peroxidation. Interestingly, NAC failed the restore the mitochondrial calcium levels and thus alleviate the mitochondrial damages, however, RR reduced the apigenin-induced redox imbalance. Furthermore, apigenin induced apoptosis activation marked by the phosphatidylserine exposure, DNA fragmentation, and caspase activation. The pro-apoptotic effect of apigenin was counteracted by RR and NAC pretreatment. In particular, RR significantly reduced the pro-apoptotic responses.</P> <P><B>Conclusions</B></P> <P>Apigenin isolated from <I>A. yomena</I> induced mitochondrial-mediated apoptotic pathway, and mitochondrial calcium signaling is main factor in its pathway in <I>C. albicans</I>.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Phenolic compounds with IL-6 inhibitory activity from Aster yomena

        A new biflavonoid, named asteryomenin (1), as well as six known phenolic compounds, esculetin (2), 4-O-${\beta}$-D-glucopyranoside-3-hydroxy methyl benzoate (3), caffeic acid (4), isoquercitrin (5), isorhamnetin-3-O-glucoside (6), and apigenin (7) were isolated from the aerial parts of Aster yomena. The structures of compounds (1-7) were identified based on 1D and 2D NMR, including $^1H-^1H$ COSY, HSQC, HMBC and NOESY spectroscopic analyses. Compounds 2-7 were isolated from this plant for the first time. For these isolates, the inhibitory activity of IL-6 production in the $TNF-{\alpha}$ stimulated MG-63 cell was examined. Among these isolates, compounds 4 and 7 appeared to have potent inhibitory activity of IL-6 production in the TNF-${\alpha}$ stimulated MG-63 cell, while compounds 1-3 and 5-6 showed moderate activity.

        쑥부쟁이 분말 첨가 식빵의 품질 특성

        쑥부쟁이는 국화과에 속하는 다년생 초본으로 독특한 향기가 있어 식욕을 촉진하는 것으로 알려져 산채나물로 이용되어 왔다. 산채나물은 항산화, 항암, 항염증 등 다양한 생리활성 효과로 기능성 식품으로의 관심이 높아져 다양한 소재로 이용되고 있다. 최근 쑥부쟁이 에탄올 추출물이 간세포 내 지방축적 유전자 발현 억제 및 지방축적 억제 효과, 항비만효능이 있는 것으로 보고되어 기능성 측면에서 활용가치가 높다. 쑥부쟁이에 대한 연구가 일부 보고되었으나 이를 활용한 가공연구는 전무하다. 본 연구는 우리밀 가루에 쑥부쟁이 분말 첨가량을 0, 0.5, 1.0, 1.5, 2.0%로 달리하여 제조한 식빵의 품질 특성과 항산화 활성을 측정하였다. 그 결과 쑥부쟁이 분말을 0.5~2.0% 첨가한 식빵은 대조구와 비교 시외관, 향, 맛, 조직감, 전반적인 기호도면에서 통계 수치적으로 유의적인 차이가 없어 소비자에게 거부감 없는 제빵이 가능할 것으로 보인다. 또한 쑥부쟁이 분말 함량이 증가할수록 총 폴리페놀 및 DPPH 라디칼 소거 등의 기능성이 증가하므로 기능성을 높인 제과제빵 및 면류 개발이 가능할 것으로 본다. Aster yomena is a perennial plant that belongs to the Asteraceae family. Seasoned wild vegetables are commonly used as functional ingredients because of their bioactive effects against oxidation, cancer, and inflammation. A recent report showed that ethanol extracts from Aster yomena effectively inhibited gene expression related to lipid accumulation within interstitial cells to prevent obesity, further raising awareness of its usefulness as a highly functional ingredient. Several studies have investigated Aster yomena, but none have investigated the effects of processing on its use. Therefore, this study investigated the quality characteristics and antioxidative activity of breads in which refined salt was replaced with Aster yomena powder at 0, 0.5, 1.0, 1.5, or 2.0%. Bread containing any amount of Aster yomena powder did not differ significantly from the control in terms of appearance, aroma, taste, texture, and overall preference. In addition, higher levels of added Aster yomena powder were associated with greater 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity indicating the potential for production of highly functional bread and noodle products using this material.

        Anti-Asthmatic Activities of an Ethanol Extract of Aster yomena in an Ovalbumin-Induced Murine Asthma Model

        Aster yomena is used in traditional remedies to treat cough, asthma and insect bites; however, its therapeutic mechanism is not completely understood. To elucidate the anti-asthmatic effect of A. yomena, we investigated the anti-asthmatic characteristics of an alcohol extract of A. yomena in an ovalbumin (OVA)-induced murine asthma model. In this study, we showed that A. yomena extract inhibited the overall pathophysiological features of asthma by suppressing Th2 responses and enzymes associated with the production of inflammatory mediators. This suppression resulted in decreased Th2 type cytokines and eosinophils in the bronchoalveolar lavage fluid and OVA-specific IgE in serum. Additionally, A. yomena extract significantly decreased airway hyperresponsiveness and abrogated the histopathological changes in the lungs, which reached normal levels in the OVA-challenged mice treated with A. yomena extract. These findings suggest that A. yomena could be a promising natural agent for treating bronchial asthma in humans.
