청소년 고혈압 관련 유전자의 연관성 분석: Kangwha Study

        서일,남정모,김성주,신동직,허남욱,강대룡,Suh, Il,Nam, Chung-Mo,Kim, Sung-Joo,Shin, Dong-Jik,Hur, Nam-Wook,Kang, Dae-Ryong 대한예방의학회 2006 예방의학회지 Vol.39 No.2

Objectives : In this study we examined the association between the genetic markers ACE (A-240T, C-93T, I/D, A2350G), AGT (M235T), AT1R (A1166C), CYP11B2 (T344C, V386A), REN (G2646A), ADRB2 (G46A, C79G, T47C, T1641), GNB3 (C825T) and ADD1 (G460W) and the presence of essential hypertension in adolescents. Methods : The Kangwha Study is an 18-year prospective study that is aimed at elucidating the determinants of the blood pressure level from childhood to early adulthood. For this study, we constructed a case-control dataset of size of 277 and 40 family trios data from the Kangwha Study. For this purpose, we perform a single locus-based case-control association study and a single locus-based TDT (transmission/disequilibrium test) study. Results : In the case-control study, the single locus-based association study indicated that the ADD1 (G460W) (p=0.0403), AGT (M235T) (p=0.0002), and REN (G2646A) (p=0.0101) markers were significantly associated with the risk of hypertension. These results were not confirmed on the TDT study. This study showed that genetic polymorphisms of the ADD1, AGT and REN genes might be related to the hypertension in Korean adolescents. Conclusions : This study provided useful information on genetics markers related to blood pressure. Further study will be needed to confirm the effect of the alpha adducin gene, the angiotensinogen gene and the renin gene on essential hypertension.

Demographic and Genome Wide Association Analyses According to Muscle Mass Using Data of the Korean Genome and Epidemiology Study

        Gim Jeong-An,Lee Sangyeob,Kim Seung Chan,Baek Kyung-Wan,Yoo Jun-Il 대한의학회 2022 Journal of Korean medical science Vol.37 No.50

Background: Sarcopenia is commonly found in the elderly due to a decline in muscle mass. Many researchers have performed genome-wide association studies (GWAS) to find genetic risk factors of sarcopenia. Although many studies have discovered sarcopenia associated single nucleotide polymorphisms (SNPs), most of them are studies targeting Caucasians. The purpose of this study was to evaluate genetic correlation according to muscle mass in middle aged Koreans using data of the Korean Genome and Epidemiology Study (KOGES), a large population-based genomic cohort study. Methods: Baseline participants were 10,030 subjects aged 40 to 69 years who were from Ansan or Anseong in Gyeonggi-do, South Korea. Among them, 9,351 subjects with laboratory data available were included in this study. To identify sarcopenia associated variants, those in the top 30% and bottom 30% of muscle mass index (MMI) were compared. A total of 7,452 people with an MMI of 30-70% were excluded. A total of 1,004 people were also excluded due to missing data. Finally, 895 people were selected for this study. The Korea Biobank Array generated 500,568 SNPs for this dataset. Results: When subjects were divided into top 30% and bottom 30% of MMI, the top 30% had 169 men and 308 women and the bottom 30% had 220 men and 198 women. In men, age, body mass index (BMI), waist and hip were significantly (P < 0.005) different between top 30% and bottom 30% MMI groups. In women, age, BMI, waist, hip, and hypertension history were significantly different between the two MMI groups. There were 13 significant SNPs in men and 14 significant SNPs in women. Genes associated with variants in men based on the single-nucleotide polymorphism database (dbSNP) were LRP1B containing rs11679458 and RGS6 containing rs11848300. A gene associated with variants in women was Pi4K2A, which contained rs1189312 as a variant. In addition, rs11189312 was associated with expression quantitative trait loci (eQTL) of ZFYVE27 in skeletal muscles and other SNPs of ZFYVE27 (rs10882883, rs17108378, rs35077384) known to be associated with spastic paraplegia. The eQTL analysis revealed that rs11189312 was a variant associated with SNPs of ZFYVE27. Conclusions: In the demographic study, significant results were found in BMI, waist, hip, history of hyperlipidemia, and sedentary life status in male group, and significant results were found in BMI, waist, hip, and hypertension history in female group. Variant rs11189312 was found to be a novel variant affecting ZFYVE27 expressed in skeletal muscles, suggesting that rs11189312 might be related to sarcopenia as a novel discovery of this study. Further study is needed to determine the association between sarcopenia and ZFYVE27 known to be associated with spastic paraplegia.

A Genome-Wide Association Study Identifies UTRN Gene Polymorphism for Restless Legs Syndrome in a Korean Population

        ChulHyun Cho,JiHye Choi,SeungGul Kang,HoKyoung Yoon,YoungMin Park,JoungHo Moon,KiYoung Jung,JinKyu Han,HongBum Shin,HyunJi Noh,YongSeo Koo,Leen Kim,HyunGoo Woo,HeonJeong Lee 대한신경정신의학회 2017 PSYCHIATRY INVESTIGATION Vol.14 No.6

Objective-Restless legs syndrome (RLS) is a highly heritable and common neurological sensorimotor disease disturbing sleep. The objective of study was to investigate significant gene for RLS by performing GWA and replication study in a Korean population. Methods-We performed a GWA study for RLS symptom group (n=325) and non-RLS group (n=2,603) from the Korea Genome Epidemiology Study. We subsequently performed a replication study in RLS and normal controls (227 RLS and 229 controls) to confirm the present GWA study findings as well as previous GWA study results. Results-In the initial GWA study of RLS, we observed an association of rs11645604 (OR=1.531, p=1.18×10-6) in MPHOSPH6 on chromosome 16q23.3, rs1918752 (OR=0.6582, p=1.93×10-6) and rs9390170 (OR=0.6778, p=7.67×10-6) in UTRN on chromosome 6q24. From the replication samples, we found rs9390170 in UTRN (p=0.036) and rs3923809 and rs9296249 in BTBD9 (p=0.045, p=0.046, respectively) were significantly associated with RLS. Moreover, we found the haplotype polymorphisms of rs9357271, rs3923809, and rs9296249 (overall p=5.69×10-18) in BTBD9 was associated with RLS. Conclusion-From our sequential GWA and replication study, we could hypothesize rs9390170 polymorphism in UTRN is a novel genetic marker for susceptibility to RLS. Regarding with utrophin, which is encoded by UTRN, is preferentially expressed in the neuromuscular synapse and myotendinous junctions, we speculate that utrophin is involved in RLS, particularly related to the neuromuscular aspects.

A genome-wide association study of the association between single nucleotide polymorphisms and brachial-ankle pulse wave velocity in healthy Koreans

        Xu, EnShi,Shin, Jinho,Lim, Ji Eun,Kim, Mi Kyung,Choi, Bo Youl,Shin, Min-Ho,Shin, Dong Hoon,Lee, Young-Hoon,Chun, Byung-Yeol,Hong, Kyung-Won,Hwang, Joo-Yeon Korean Society of Medical Genetics and Genomics 2017 대한의학유전학회지 Vol.14 No.1

Purpose: Pulse wave velocity (PWV) is an indicator of arterial stiffness, and is considered a marker of vascular damage. However, a genome-wide association study analyzing single nucleotide polymorphisms (SNPs) associated with brachial-ankle PWV (baPWV) has not been conducted in healthy populations. We performed this study to identify SNPs associated with baPWV in healthy populations in Korea. Materials and Methods: Genomic SNPs data for 2,407 individuals from three sites were analyzed as part of the Korean Genomic Epidemiologic Study. Without replication samples, we performed multivariable analysis as a post hoc analysis to verify the findings in site adjusted analysis. Healthy subjects aged between 40 and 70 years without self-reported history or diagnosis of hypertension, diabetes, hyperlipidemia, heart disease, cerebrovascular disease and cancer were included. We excluded subjects with a creatinine level >1.4 mg/dL (men) and 1.2 mg/dL (women). Results: In the site-adjusted association analysis, significant associations (P<$5{\times}10^{-8}$) with baPWV were detected for only 5 SNPs with low minor allele frequency. In multivariable analysis adjusted by age, sex, height, body mass index, mean arterial pressure, site, smoking, alcohol, and exercise, 11 SNPs were found to be associated (P<$5{\times}10^{-8}$) with baPWV. The 5 SNPs (P<$5{\times}10^{-8}$) linked to three genes (OPCML, PRR35 and RAB40C) were common between site-adjusted analysis and multivariable analysis. However, meta-analysis of the result from three sites for the 11 SNPs showed no significant associations. Conclusion: Using the recent standard for genome-wide association study, we did not find any evidence of significant association signals with baPWV.

        재일 조선불교유학생들의 활동

        조아담(A Dam Jo) 동국대학교 불교문화연구원 2008 佛敎學報 Vol.0 No.48

The first Korean Buddhist students studying abroad are known to be Kim Beop-ryong and Kim Seung-beop, who started their studies in 1910s. The number of Korean Buddhist students in Japan increased in 1920s and it seems to be caused by an awakening to the need of learning `new knowledge` in stimulating the Korean Buddhism and a growing favor to the Japanese Buddhism after the lifting of entrance ban to the monks. The Japanese intriguing policy, to abolish independent spirit of Korean people and to merge Korean Buddhism into Japanese Buddhism, also contributed to the increase of the Korean Buddhist students in Japan. The association of Jo-seon Buddhist Students in Japan, founded in 1920, took part in activities like the prevention of trying to unite the order Korean order to Rinzai, the Korean Buddhists led by Yi Hwoi-guang. However, it was dissolved as it changed its name. The Jo-seon Buddhist Youth Association in Japan was founded in 1921 and showed a strong will to join with the Buddhist youth movement in Korean in order to reform the Korean Buddhism. Its passion came to take shape through various activities so it took the initiative in spreading new knowledge and in enlightening people, published its own journal, the Geumgangjeo(which means Diamond Mace) and worked for spreading Buddhism by establishing organizations like the Association in Commemoration of Great Master Wonhyo and the Association for Tipitaka Studies. In 1931, the Jo-seon Buddhist Youth Association in Korea changed its name to General Association of the Jo-seon Buddhist Youth. Accordingly, each of the Buddhist youth associations, scattered over the country, dissolved its organization and became an affiliate of the General Association. Following the general trend, the Jo-seon Buddhist Youth Association in Japan became a member of the General Association, as Tokyo affiliate. The Tokyo affiliate carried its own undertakings, so it held meetings like regular assembly, welcoming and farewell parties, organized the Research Group to study Buddhist teachings and continued to publish its journal, the Geumgangjeo. It also worked for maintaining close relation with the Buddhist movements in Korea, so it actively conveyed their words when important matters became issues since the early 1930s, matters like the execution of the Constitution of the Jo-gye Order, the amendment of Temple Laws, an oppressive measure by Japanese and the abolition of the Jung-ang Buddhist College. However, the Tokyo affiliate was dissolved in 1934 due to the inactivity of the Buddhist youth movement, caused by problems like internal conflicts in the General Association. The Man-dang, a secret anti-Japanese Buddhist organization founded in 1930, had also an affiliate in Tokyo. The Tokyo affiliate of the Man-dang was organized in 1932 by Kim Beop-lin, a student who was actively taking part in the Buddhist youth movement. They played an essential role in the Buddhist youth movement and was known to be dedicated to various activities from the years before studying in Japan to the troubled times after the Liberation. The Association of the Jo-seon Buddhist Students Studying in Tokyo upheld the traditions of the early Buddhist students associations in Japan and built its own hall, supported by the Buddhists organizations in Korea. However, it showed sympathy with Japanese policy of aggression in 1940s. Most of the students, who studied in Japan, were monks and some returned to the laity after their returning to Korea, or married according to the Japanese Buddhist tradition. They were also involved in the vehement discussion on the matter of having a wife and eating meats. Many of them became the leading members of the Business Affairs of the Order or a chief monk of a temple and showed compromising attitudes with the Japanese policy. However, those monks, who distinguished themselves in Buddhist studies with outstanding academic abilities, dedicated themselves to teaching and research at monasteries

Association between urinary arsenic concentration and genetic polymorphisms in Korean adults

        Lee Seul-Gi,Eom Sang-Yong,Lim Ji-Ae,Choi Byung-Sun,Kwon Ho-Jang,Hong Young-Seoub,Kim Yong-Dae,Kim Heon,Park Jung-Duck 한국독성학회 2024 Toxicological Research Vol.40 No.1

Arsenic (As) is a human carcinogen widely distributed in the environment. This study evaluated the association between the urinary As concentration and single nucleotide polymorphisms (SNPs) in Korean adults to determine the genetic factors related to As concentration. The study included 496 participants for the genome-wide association study (GWAS) and 1483 participants for the candidate gene approach study. Participants were 19 years and older. The concentrations of total As (Tot As) and total As metabolites (Tmet As, the sum of inorganic As and their metabolites; arsenite, arsenate, monomethylarsonic, and dimethylarsinic acid) in the urine were analyzed. The GWAS identified four SNPs (rs1432523, rs3776006, rs11171747, and rs807573) associated with urinary Tot As and four SNPs (rs117605537, rs3776006, rs11171747, and rs148103384) significantly associated with urinary Tmet As concentration (P < 1 × 10– 4). The candidate gene study identified two SNPs (PRDX2 rs10427027 and GLRX rs3822751) in genes related to the reduction reaction associated with urinary Tot As and Tmet As. This study suggests that genetic factors may play a role in regulating As metabolism in the human body, affecting both exposure levels and its potential health risks in the general Korean population, even at low exposure levels.

Genome-Wide Association Study Identifies Two Novel Loci with Sex-Specific Effects for Type 2 Diabetes Mellitus and Glycemic Traits in a Korean Population

        Go, Min Jin,Hwang, Joo-Yeon,Park, Tae-Joon,Kim, Young Jin,Oh, Ji Hee,Kim, Yeon-Jung,Han, Bok-Ghee,Kim, Bong-Jo Korean Diabetes Association 2014 Diabetes and Metabolism Journal Vol.38 No.5

Background

Until recently, genome-wide association study (GWAS)-based findings have provided a substantial genetic contribution to type 2 diabetes mellitus (T2DM) or related glycemic traits. However, identification of allelic heterogeneity and population-specific genetic variants under consideration of potential confounding factors will be very valuable for clinical applicability. To identify novel susceptibility loci for T2DM and glycemic traits, we performed a two-stage genetic association study in a Korean population.

Methods

We performed a logistic analysis for T2DM, and the first discovery GWAS was analyzed for 1,042 cases and 2,943 controls recruited from a population-based cohort (KARE, n=8,842). The second stage, de novo replication analysis, was performed in 1,216 cases and 1,352 controls selected from an independent population-based cohort (Health 2, n=8,500). A multiple linear regression analysis for glycemic traits was further performed in a total of 14,232 nondiabetic individuals consisting of 7,696 GWAS and 6,536 replication study participants. A meta-analysis was performed on the combined results using effect size and standard errors estimated for stage 1 and 2, respectively.

Results

A combined meta-analysis for T2DM identified two new (rs11065756 and rs2074356) loci reaching genome-wide significance in CCDC63 and C12orf51 on the 12q24 region. In addition, these variants were significantly associated with fasting plasma glucose and homeostasis model assessment of β-cell function. Interestingly, two independent single nucleotide polymorphisms were associated with sex-specific stratification in this study.

Conclusion

Our study showed a strong association between T2DM and glycemic traits. We further observed that two novel loci with multiple diverse effects were highly specific to males. Taken together, these findings may provide additional insights into the clinical assessment or subclassification of disease risk in a Korean population.
