Differential expression of nitric oxide synthase in human stomach cancer

The level of expression and cellular localization of isoenzymes of nitric oxide synthase (NOS) was detected in human stomach tumor tissues. Tumor tissues showed 70% higher activity of NOS than that of normal tissues (P<0.01). Poorly differentiated adenocarcinoma tend to have higher activity (P<0.05) than well differentiated and moderately differentiated tumor tissues. Aminoguanidine (AG), 2-amino-5,6-dihydro-6-methyl-4H-1,3-thiazine (AMT), N^G-monomethyl-L-arginine (L-NMMA), and N^ω-nitro-L-arginine (L-NNA) inhibited NOS activity in tumor tissues by 18,14,11 and 13%, respectively. The TNF-α mRNA expression was correlated with the inducible NOS(iNOS) level, which was high in adenocarcinomas and low in normal tissues. Tumor tissues showed higher expression of iNOS in gland epithelial cells but the level of eNOS was significantly decreased with an exception of concentrated localization in the proliferation capillary endothelium. These results revealed that isoforms of NOS might contribute differentially to growth and progression of human stomach tumor. ⓒ 1999 Elsevier Science Ireland Ltd. All rights reserved.

자궁경술은 자궁강내를 직접 보고 생검하면서 수술 등을 할 수 있는 장점이 있어 부인과영역에서 진단 및 치료에 보다 더 정확하고 안전한 시술로써 인정받고 있으며 그 영역이 점차 확대되고 있다. 최근 20년간 내시경 영상과 수술기술, 수술도구의 발전으로 많은 진보가 있어 왔으며 이로 인해 office hysteroscopy가 가능하게 되었다. Office hysteroscopy는 입원하지 않고 당일 시술을 받을 수 있어 환자가 간편하고 안전하게 받을 있는 매우 유용한 술기로 여겨진다. 저자들은 1995년 9월부터 2005년 3월까지 시행한 자궁경술 140명 중 office hysteroscopy 18명을 경험하였기에 문헌고찰과 함께 보고하는 바이다. Background: Hysteroscopy is considered to be the gold standard not only for visualizing the cervical canal and the uterine cavity, but also for treating many different types of benign pathologies localized to those regions. The advent and evolution of endoscopic imaging and surgery during the last two decades has added new dimensions to the armamentarium of a gynecologist to combat intrauterine lesions. Office hysteroscopy is increasingly being used as a fast line investigation for abnormal uterine bleeding and other diseases involving the uterine cavity. The aim of our study is to assess the diagnostic and operative efficacy of office hysteroscopy. Materials and Methods: In our department, 140 patients underwent a hysteroscopy examination and 18 of these underwent an office based hysteroscopy examination from September 1995 to March 2005. The cases who underwent an office based hysteroscopy examination were reviewed in order to assess the clinical usefulness and significance in the management of intrauterine lesions. Results: Major indication was abnormal uterine bleeding(12 cases, 66.7%). The others were a missed IUD and infertility. The hysteroscopic findings were a normal uterine cavity(6 cases, 33.3%), lUD in situ, polyp, submucosal myoma, endometrial hyperplasia and a placenta remnant. Conclusion: Office hysteroscopy is a safe, quick and effective method for making an intrauterine evaluation. In addition, it provides immediate results, offers the capacity of direct targeted biopsies of suspicious focal lesions, and offers the direct treatment of some intrauterine conditions.

저자들은 영남대학교 의과대학 부속병원 산부인과에서 임신과 동반된 자궁부속기 염전 1례를 경험하였기에 간단한 문헌적 고찰과 함께 보고하는 바이다. Adnexal torsion is not a common surgical emergency in obstetrics and gynecology, and is not observed frequently during pregnancy with its incidence being one out of about 5,000 pregnancies: pregnancy is a factor that encourages torsion. The traditional treatment for ischemic and twisted adnexa consists of oophorectomy or salpingo-oophorectomy. With early diagnosis, a conservative approach is recommended in cases of young women to assure future fertility. With a brief review of literature, we report a case of unilateral salphingo-oophorectomy during pregnancy due to adnexal torsion with the delivery of a normal baby.

Background: Current cancer research using massive parallel sequencing technologies primarily focuses on somatic changes in the cancer genome. However, it is anticipated that individual germline variations might contribute to tumorigenesis, as exemplified by several well-known examples, such as BRCA2 variations in breast cancer. Thus, it is also important to explore crosstalk between germline variation and somatic mutation in cancer. Methods: We performed whole-exome sequencing (WES) of 80 acute myeloid leukemia (AML) samples and 180 non-small cell lung cancer (NSCLC) samples. First, we searched for cancer hotspots in germline DNA of these samples. Also, we called somatic variants using in-house pipeline Adiscan. Germline hotspot alteration was validated in a separate AML cohort. Results: Non-synonymous changes including MLH1 (Chr3;37067240, T>A), KIT (Chr4;55593464, A>T), and MET (Chr 7;116340262, A>C) were observed in germline of both AML and NSCLC patients. MLH1 germline change was not found in 18 healthy control samples. Frequency of MLH1, KIT, and MET germline changes were similar in AML and NSCLC patients. In contrast to AML, some of NSCLC patients had PTEN, TP53 and STK11 germline changes. When we focused on the crosstalk between germline change and somatic mutation, MET germline alteration seemed to have coupling with IDH2 somatic mutation, while MLH1 germline mutation seemd to have coupling with TTN somatic mutation. When validated in a separate cohort, AML patients with MET germline mutation had good prognosis compred to MET wild-types suggesting prognostic impact of MET germline alteration. Conclusions: Cancer hotspot variations are present in germline of cancer patients, and their frequency might be different across various cancer subtypes. Germline alteration per se may have prognostic impact, and coupling between certain germline alteration and somatic mutation would exist.

N/A Objective : The TDX analyzer, an automated fluorescence polarimeter, has been utilized to assess surfactant content in amniotic fluid. Amniostat-FLM is an immunologic semiquantitative agglutination test for determining the presence of phosphatidylglycerol in human amniotic fluid at concentration indicative of fetal lung maturity. We evaluated the usefulness of the recently introduced TDx-FLM assay in determining fetal lung maturity. Methods : Seventy-three samples of amniotic fluid were analyzed. Among them, 18 samples were compared with Amniostat-FLM assay. Results : Surfactant/albumin ratio using TDx-FLM assay increased with longer gestational age. And the concordance rate between Amniostat-FLM assay and TDx-FLM assay was 66.9%. Conclusion : The TDx-FLM assay appears to be a useful test in the field of fetal lung maturity testing.
