A Simple Method for the Detection and Identification of Korean Brown Cattle(Hanwoo) using DHPLC

Denaturing high-performance liquid chromatography(DHPLC) is used in a wide variety of genetic applications and is an efficient method for detection of mutations involving one or a few nucleotides. We developed a high-throughput DHPLC method for identifying polymorphisms in the MC1R gene that are characteristic of Hanwoo cattle. We compared 10 tissue samples from Hanwoo cattle, 10 samples from Holstein cattle and 10 samples from Hanwoox Holstein crossbred cattle to determine whether DHPLC analysis can be used to distinguish between these genotypes. Samples obtained from Hanwoo cattle had a unique profile of peaks that could be used as a molecular fingerprint for this breed. We also analyzed two hundred samples in a trial in which we were blinded to the genotype of the samples and correctly identified the breed-of-origin of 594 out of 600 sequence variations(99%).

<P><B>Abstract</B></P> <P>Influenza virus A (H5N1) clade 2.3.2.1 has recently caused widespread outbreaks of disease in domestic poultry and wild birds in Eastern Asia. In the current study, the antigenicity and pathogenicity of three clade 2.3.2.1 viruses (Ck/Kr/Gimje/08, Ws/Mongolia/1/09, and Ws/Mongolia/7/10) were investigated in domestic ducks and mice. The H5N1 influenza viruses in this study were antigenically similar to each other (<I>r</I>-values of 0.35–1.4). The three viruses replicated systemically in all tissues tested in domestic ducks, indicating high pathogenicity. However, the viruses produced different clinical signs and mortality rates: Ck/Kr/Gimje/08 and Ws/Mongolia/1/09 resulted in 100% mortality with severe neurological signs, whereas Ws/Mongolia/7/10 resulted in 50% mortality with relatively mild neurological signs. In mice, infection with Ck/Kr/Gimje/08 and Ws/Mongolia/7/10 resulted in weight loss that peaked at 4 days post-infection (22.3% and 20.8%, respectively), same MLD<SUB>50</SUB> (2.2 Log<SUB>10</SUB> EID<SUB>50</SUB>) and systemic replication. The three viruses had K deletion at the −2 position of the HA1-connecting peptide (PQRERRRK-R), which is associated with increased virulence in domestic ducks and harbored NA stalk deletion, NS1 deletion and mutation of P42S in NS1, and full length (90aa) in PB1-F2, which confer increased virulence in mice. Our study shows that clade 2.3.2.1 viruses from Korea and Mongolia are antigenically similar and highly pathogenic in both domestic ducks and mice. Moreover, we provide molecular determinants of the clade 2.3.2.1 viruses associated with the pathogenicity in domestic ducks and mice, respectively.</P>

Various cytokines including IL-1β, IL-2, IL-4, IL-10, IL-12p40, IFN-γ, and TNF-α have been recently reported to play important roles in both host and donor cell activations during xenotransplantation. Interleukin (IL)-18, a proinflammatory cytokine, can induce IL-4 and IL-13 production in T cells, NK cells, B cells, mast cells, and basophils. IL-18 has the capacity to stimulate innate immunity and both Th1-and Th2- mediated responses, but little is known to involve in the xenograft rejection. In order to investigate the role of IL-18 in xenograft rejection, we transplanted the pig kidney (PK15) cells to C57BL/6 mouse with or without intraperitoneal injection of mouse recombinatant IL-18. It was analyzed the population of T cell, B cell and NK cell in the mice transplanted with PK 15 cells and recombinant IL-18 by flow cytometry. We found that splenic CD3+ T cells were increased in mice injected PK15 cells with recombinant IL-18. These results suggest that recombinant IL-18 is critical in the xenograft rejection by increasing T cell population of the recipients.

오디 추출물(Morus alba L.)의 혈행개선 효과

본 연구에서는 다양한 생리활성물질을 다량 함유하고 있는 국내산(전라북도 부안군) 오디의 혈액순환장애의 개선 효과를 확인하기 위하여 in vitro 및 고지혈증 동물모델을 이용한 실험을 진행하였다. MBE가 DPPH 라디칼을 50% 소거하는 SC50값은 89.36±5.46 μg/mL로 정제되지 않은 복합물질임을 감안하면 다른 식물추출물에 비해 항산화능이 우수함을 알 수 있었다. RAW 264.7 세포 및 HepG2 세포에 대한 MBE의 세포독성은 2,500 μg/mL 이하의 농도를 처리한 경우 세포독성을 나타내지 않았다. In vitro 실험에서 COX-2의 활성저해 효과를 측정한 결과 MBE의 IC50값은 215.94±18.15이었고, human LDL의 산화억제 효과는 최종반응 농도가 200 및 400 μg/mL일 경우 유의적인 감소(P<0.05)효과가 있었다. 또한 TNF-α에 의하여 활성화된 HUVECs에서 MBE가 증가된 ICAM-1 및 VCAM-1의 발현을 감소(P<0.05)시켰으며 LPS에 의하여 활성화된 RAW 264.7 세포에서 MBE가 증가된 COX-2 및 5-LO의 발현을 감소(P<0.05)시킴을 알 수 있었다. 고지혈증 동물모델을 이용한 실험에서 MBE의 일부 실험군(200 mg 및 400 mg/kg 체중/day, 14 days)에서 혈청 total cholesterol의 수치는 음성대조군에 비해 유의적으로 감소(P<0.05)시켰으나 혈청C-reactive protein의 수치는 모든 실험군에서 음성대조군 대비 유의적으로 감소하지 않았다. 이러한 MBE의 항산화능, COX-2 활성억제 및 COX-2와 5-LO의 발현억제를 통한 염증억제, 혈중 total cholesterol 감소를 통한 이상지질혈증의 개선 효과 및 ICAM-1과 VCAM-1의 발현억제를 통한 혈관협착 방지의 효과들을 종합해 보면 오디추출물인 MBE가 혈액순환장애를 유발하거나 확대시키는 다양한 핵심요소들을 다각도로 개선시킴으로써 혈액순환장애의 예방과 치료에 긍정적인 효과가 있을 것으로 사료된다. The objective of this study was to investigate the beneficial effects of mulberry extract (MBE) on blood flow improvement. The SC50 value for the DPPH radical scavenging activity of MBE was 89.36±5.46 μg/mL. Analysis of the cellular toxicity of MBE on RAW 264.7 and HepG2 cells showed no toxicity under a concentration of 2,500 μg/mL. We found that MBE inhibited the enzyme activity of cyclooxygenase (COX)-2 as well as oxidation of human LDL. Western blotting analysis showed that MBE inhibited protein expression of COX-2 and 5-lipoxygenase in RAW 264.7 cells. In addition, MBE inhibited protein expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1 in human umbilical vein endothelial cells. Furthermore, MBE reduced the serum levels of total cholesterol and C-reactive protein in a concentration-dependent manner. These results both in vitro and in vivo suggest that MBE can be employed for the improvement of blood flow.

This study is to verify the structural relationship among 3 typical factor relational benefits, relational commitment, and relational continuance intention in service corporation. To test each measures; predictive utility in this context with AMOS. Sampling in this study is limited to the department store, beauty parlor, dental clinic and hotel restaurant in Busan, South Korea. The responders included 632. Data were analyzed by frequency, factor analysis, and AMOS. As the results, social benefit is major to influence on the relational commitment and relational continuance intention. Confidence benefit is major to influence on the relational continuance intention, while isn`t major determinant to influence on the relational commitment. Special treatment benefit is major to influence on the relational commitment, while isn`t major determinant to influence on the relational continuance intention. Social benefit is major to influence on the relational commitment in all of 4 service corporations. While confidence benefit isn`t major determinant to influence on the relational commitment in all of 4 service corporations. Special treatment benefits is major to influence on the relational commitment in beauty parlor, dental clinic and hotel restaurant.
