Statistical inference of acupoint specificity: forward and reverse inference

Background: The acupoint specificity has been considered important issue in acupuncture research. In clinical aspects, it is essential to identify which acupoints are associated specifically with a particular disease. The present study aimed to identify the specificity of acupoint selection (forward inference) and the specificity of acupoint indication (reverse inference) from the online virtual diagnosis experiment. Methods: Eighty Korean Medicine doctors conducted the virtual medical diagnoses provided for 10 different case reports. For forward inference, the acupoints prescribed for each disease were quantified and the data were normalised among 30 frequently used acupoints using Z-scores. For reverse inference, diseases for each acupoint were quantified and the data were normalized among 10 disease using Z-scores. Results: Using forward inference we demonstrated the specificity of acupoint selection in each disease. Using reverse inference we identified the specificity of acupoint indication in each acupoint. In general, a certain acupoint can be selected specifically for a particular disease, and it has a specific indication for the disease. However, the specificity of acupoint indication and the specificity of acupoint selection are not always identical. Conclusions: The selection of an acupoint for a particular disease does not imply that the acupoint has specific indications for that disease. Inferring the specificity of acupoint indication from clinical observations should be considered.

Purpose : The purpose of this study was to apprehend the relationship among adversity quotient, self-leadership and stress associated with clinical experience on clinical competence of nursing students and to provide the influential factors to clinical competence. Methods : The participants in this study were 184 nursing students in the third and fourth grade who experienced clinical practice and data was collected through structured questionnaires. Results : The result of the study showed that clinical experience on clinical competence was 3.51, adversity quotient was 3.35, self-leadership was 3.60 and stress associated with clinical experience was 3.01 respectively. Factors influencing nursing students’ clinical competence were shown in the order of satisfaction on clinical practice, adversity quotient, self-leadership, grade. Among them, satisfaction on clinical practice was the greatest influence on clinical competence and the explanatory power was 37.3%. Conclusion : The findings showed that in order to enhance clinical competence of nursing students, it is necessary to develop education programs by increasing satisfaction of nursing major and adversity quotient.
PHGDH: a novel therapeutic target in cancer

Serine is a key contributor to the generation of one-carbon units for DNA synthesis during cellular proliferation. In addition, it plays a crucial role in the production of antioxidants that prevent abnormal proliferation and stress in cancer cells. In recent studies, the relationship between cancer metabolism and the serine biosynthesis pathway has been highlighted. In this context, 3-phosphoglycerate dehydrogenase (PHGDH) is notable as a key enzyme that functions as the primary rate-limiting enzyme in the serine biosynthesis pathway, facilitating the conversion of 3-phosphoglycerate to 3-phosphohydroxypyruvate. Elevated PHGDH activity in diverse cancer cells is mediated through genetic amplification, posttranslational modification, increased transcription, and allosteric regulation. Ultimately, these characteristics allow PHGDH to not only influence the growth and progression of cancer but also play an important role in metastasis and drug resistance. Consequently, PHGDH has emerged as a crucial focal point in cancer research. In this review, the structural aspects of PHGDH and its involvement in one-carbon metabolism are investigated, and PHGDH is proposed as a potential therapeutic target in diverse cancers. By elucidating how PHGDH expression promotes cancer growth, the goal of this review is to provide insight into innovative treatment strategies. This paper aims to reveal how PHGDH inhibitors can overcome resistance mechanisms, contributing to the development of effective cancer treatments.
코로나-19 상황에서 대학생들의 스트레스와 주관적 안녕감의 관계: 가족유연성과 응집력 및 불안통제성의 매개효과

본 연구에서는 코로나-19 상황에서 대학생들의 지각된 스트레스와 주관적 안녕감의 관계에서 가족유연성과 응집력, 불안통제성의 매개효과를 가정한 구조모형을 설정하고, 모형의 적합도와 개별 변수의 관련성을 살펴보았다. 이를 위해, 전국에 소재한 4년제 대학에 재학 중인 291명의 대학생들로부터 자료를 수집하였다. 자료분석을 위해서 구조모형 분석을 실시하였다. 연구결과 본 연구에서 설정한 매개모형은 대학생들의 자료를 적절히 설명하는 것으로 나타났으며, 스트레스, 가족유연성과 응집력, 불안통제성은 주관적 안녕감의 70%를 설명하였다. 각 변인들의 관련성을 살펴보면, 스트레스는 가족유연성과 응집력 및 불안통제성, 주관적 안녕감에 유의한 부적영향을 미치는 것으로 나타났다. 한편, 가족유연성과 응집력, 불안통제성은 주관적 안녕감에 유의한 정적영향을 미쳤으며 가족유연성과 응집력, 불안통제성이 높을수록 대학생들의 주관적 안녕감이 높아졌다. 가족유연성과 응집력, 불안통제성의 매개효과를 살펴본 결과 대학생들이 지각하는 스트레스와 주관적 안녕감의 관계를 두 변인이 유의하게 매개함을 알 수 있었다. 본 연구 결과를 바탕으로 대학생들을 대상으로 하는 상담 및 교육에의 주요함의점을 논하였다. This study examined the role of family cohesion and adaptability and anxiety control in the relation between stress and subjective well-being. For this study, data were collected from 291 college students attending four-year universities. As a result of analyzing data through structural equation modelling, the mediating model showed that the data of college students were properly explained, and stress, family cohesion and adaptability, and anxiety control explained 70% of subjective well-being. As for the relationship between each variable, stress had a significant negative effect on family cohesion and adaptability, anxiety control, and subjective well-being. On the other hand, family cohesion and adaptability, and anxiety control had a significant positive effect on subjective well-being and the higher family cohesion and adaptability, and anxiety control, the higher subjective well-being of college students. As a result of examining the mediating effects of family cohesion and adaptability, and anxiety control, it was found that the two variables significantly mediated the relationship between stress and subjective well-being perceived by college students. Based on the results of this study, the main implications of counseling and education for college students were discussed.

The cytoplasmic polyadenylation element (CPE)-binding protein (CPEB) binds to CPE containing mRNAs on their 3' untranslated regions (3'UTRs). This RNA binding protein comes out many important tasks, especially in learning and memory, by modifying the translational efficiency of target mRNAs via poly (A)tailing. Overexpressed CPEB has been reported to induce the formation of stress granules (SGs), a sort of RNA granule in mammalian cell lines. RNA granule is considered to be a potentially important factor in learning and memory. However, there is no study about RNA granule in Aplysia. To examine whether an Aplysia CPEB, ApCPEB1, forms RNA granules, we overexpressed ApCPEB1-EGFP in Aplysia sensory neurons. Consistent with the localization of mammalian CPEB,overexpressed ApCPEB1 formed granular structures,and was colocalized with RNAs and another RNA binding protein, ApCPEB, showing that ApCPEB1 positive granules are RNA-protein complexes. In addition,ApCPEB1 has a high turnover rate in RNA granules which were mobile structures. Thus, our results indicate that overexpressed ApCPEB1 is incorporated into RNA granule which is a dynamic structure in Aplysia sensory neuron. We propose that ApCPEB1granule might modulate translation, as other RNA granules do, and furthermore, influence memory.
Development of chinese cabbage cell plant factory for biopharmaceutical production

EpCAM is a tumor-related antigen and is a cell surface glycoprotein expressed in colon cancer. EpCAM has been considered a potential target for tumor vaccines. In this study, Agrobacterium-mediated transformation was applied to produce genetically modified Chinese cabbage plants to express EpCAM-IgM FcK. To avoid the plant-specific N-glycosylation process occurring in Golgi devices, the KDEL ER sequence was attached to the C-terminal of Fc to refair the glycoprotein of ER. The recombianat protein productivity was maximized by developing a safe and efficient plant cell factory system using a genetically modified plant cell line expressing EpCAM-IgM FcK protein. In addition, plant seedlings expressing both EpCAM-IgM FcK and J-chain K were obtained by crossing. PCR confirmed that the EpCAM-IgM FcK gene present in the seedlings. Western Blot screened seedlings with high expression. Chinese cabbage protoplasts were obtained by successfully optimizing the protoplast separation protocol to perform single cell transcription analysis. Single cell transcription analysis was performed to reveal the cell types that highly express EpCAM-IgM FcK protein. Cell lines with high expression will be applied to plant cell factory systems to produce EpCAM-IgM FcK.
트레이닝 데이터 생성과 의사 결정 트리를 이용한 계통수 생성 방법

전통적인 동물 계통수(系統樹)는 초기발생 혈질에 기초하여 몸 구조가 단순한 것에서 복잡한 것으로 동물문(animal phylum)들을 배열하는 것이다. 현재 활발하게 연구 진행되는 분자수준에서의 분자계통 분류학(Molecular Systematics) 연구들이 이런 경향을 재평가하고 새로운 계통과 진화의 의미를 제시하고 있다. 본 논문에서는 한 염기서열로부터 획득할 수 있는 특성 값들을 추출하여 트레이닝 데이터를 생성하고, 생성된 데이터를 기반으로 데이터마이닝 기법중의 하나인 분류기법(classification) 을 사용하여 계통수를 생성하였다. 실험용 데이터는 미토콘드리아 염기서열을 사용하였으며 생물학분야에서 사용하는 분석 프로그램인 MEGA 프로그램을 사용하여 이를 증명하였다. 비록 마이닝을 수행한 결과는 생물학적 실험을 거쳐 정확성을 검증 받아야 하지만 인터넷상에 떠다니는 무수한 유전체들에 대한 유효한 분류기준을 제시할 수 있고 계통수 제작을 위한 실험에 소요되는 많은 시간과 노력들을 줄일 수 있다. The traditional animal phylogenetic tree is to align the body structure of the animal phylums from simple to complex based on the initial development character. Currently, molecular systematics research based on the molecular, it is on the fly, is again estimating prior trend and show the new genealogy and interest of the evolution. In this paper, we generate the training set which is obtained from a DNA sequence ans apply to the classification. We made use of the mitochondrial DNA for the experiment, and then proved the accuracy using the MEGA program which is anaysis program, it is used in the biology field. Although the result of the mining has to proved through biological experiment, it can provede the methodology for the efficient classify and can reduce the time and effort to the experiment.
