      • 탄수화물의 종류가 IN VITRO 반추위박테리아에 의한 OLEIC ACID 의 HYDROGENATION 및 INCORPORATION 에 미치는 효과

        본 시험은 첨가하는 탄수화물의 종류가 반추위 박테리아에 의한 발효특성, 박테리아 성장 및 oleic acid(C_(18:1))의 hydrogenation과 박테리아 lipid로의 incorporation을 조사하고자 in vitro 방법으로 실시 되었다. 200㎖ non-selective basal broth medium에 dextrose, starch 또는 cellulose powder를 0.20%로(w/v) 첨가하였으며, 탄수화물이 첨가되지 않은 control을 포함한 4처리 각각을 위해 nylon 천(2×3㎝)을 이용하여 평균 87.4㎎의 C_(18:1) 과 I^-(14)C_(18:1) 2.09uCi를 흡착시켰다. Oleic acid와 각각의 탄수화물이 첨가된 broth medium에 8겹의 cheese cloth로 여과시킨 반추위액 3㎖를 첨가하여 혐기적인 방법으로 39℃의 진탕배양기에서 24시간 배양하였다. 이와는 별도로 반추위액만이 포함된 broth medium(blank)을 동일한 조건에서 배양하였다. 탄수화물의 첨가는 blank와 control에 비하여 배양 12시간 이후 배양액(broth medium)의 pH를 낮추었으며 배양 6시간에서는 dextrose 첨가구에서 가장 낮았다(p<0.0001). 배양액 암모니아 농도의 경우 12시간의 배양 이후 탄수화물 첨가구의 pH보다 blank와 control에서 더 증가되었으나(p<0.0111) 첨가한 탄수화물 종류에 따른 뚜렷한 차이는 없었다. 배양 종료시 배양액의 총 VFA 농도는 blank 및 control에 비하여 3종류의 탄수화물 첨가구 모두에서 현저히(p<0.0002) 증가되었다. 또한 배양시간이 경과함에 따라 acetate(C₂)의 조성 비율이 점차 감소되었던 반면 propionate(C₃)의 조성 비율은 점차 증가되었다. 배양 12시간에서는 starch와 cellulose 첨가구에서 C₂의 조성비율이 낮았으나(p<0.0002), 종료시점인 24시간에서는 blank와 control의 경우에 비하여 모든 탄수화물 첨가구에서 더 낮은 것으로 나타났다 (p<0.0001). Propionate의 경우 C₂와는 반대의 경향을 보였다. Butyrate 조성 비율은 24시간에 dextrose와 staph 첨가구에서(p<0.0001) 더 높았다. 한편, 24시간의 배양 후 박테리아 수는 dextrose 첨가구에서 가장 높았으나(p<0.0001) 다른 처리구 간에는 차이가 없었다. Nylon 천에서 배양액으로 유출된 C_(18:1)의 bydrogenation 율(%)은 배양 6시간 및 12시간에서 상대적으로 pH가 낮은 dextrose나 starch 첨가구에서 높았던 반면 cellulose 첨가구에서 낮은 경향을 보였다. 또한 C_(18:1)이나 stearic acid의 형태로 박테리아 lipid로의 incorporation된 율은 탄수화물 첨가구 중 pH가 가장 높았던 cellulose 첨가구에서 11.1%로 가장 증가된 경향이었다. 그러나 hydrogenation 및 incorporation 모두 탄수화물 첨가여부 또는 탄수화물의 종류에 따른 현저한 차이는 없었다. An in vitro study was conducted to examine the effect of carbohydrate sources(dextrose, starch or cellulose) added to the broth medium on fermentation characteristics, bacterial growth, hydrogenation of oleic acid(C_(18:1)) and direct incorporation of C_(18) fatty acids by mined ruminal bacteria in vitro. Carbohydrates were added to 200㎖ non-selective basal broth medium at the level of 0.2%(w/v). Oleic acid(87.4㎎) and 1-^(14)C_(18:1)(2.09uCi) were absorbed into the nylon cloth(2 × 3㎝), and the two pieces of nylon cloth were placed to 200㎖ broth medium for each treatment. Three mls of rumen fluid strained through 8 layers of cheese cloth were added to each broth medium, then was incubated anaerobically in the shaking incubator of 39℃ for 24 hour. The broth media of control which do not contain the carbohydrates and blank which contain rumen fluid only were also incubated. Addition of carbohydrates to the broth medium tended to decrease the pH of broth media after 12h and 24h incubations compared to those of blank and control, and the lowest(p<0.0001) pH was observed from the dextrose addition at the 6h incubation. Ammonia concentrations in the broth media of blank and control after 12h incubation slightly increased(p<0.0111) compared to those in the carbohydrates added media, but there were no differences in pH among carbohydrate sources. Volatile fatty acid concentration in the carbohydrates added broth media increased(p<0.0002) compared to those in blank and control after 24h incubation. As incubation time passed molar proportion of acetate(C₂) decreased gradually but propionate(C₃) proportion increased. Proportions of C₂ were lower (p<0.0002) for the starch and cellulose added media at 12h incubation while increased C₂ proportions were observed from all the carbohydrate added media compared to those from blank and control when the incubation was terminated(24h). Opposite results to the C₂ were observed from C₃. Increased(p<0.0001) molar proportions of butyrate were found from the dextrose and starch added media after 24h incubation. Highest number of viable bacteria was observed from the dextrose added medium after 24h incubation. Percent hydrogenation of C_(18:10) tended to increase for the dextrose and starch added media which were relatively low in pH after 6h and 12h incubation while incorporation(%) of C_(18:1) or C_(18:0) into the bacterial lipids tended to increase for the celluiose addod medium which was high in pH.

        Survival and Performance of Two Cellulose-Degrading Microbial Systems Inoculated into Wheat Straw-Amended Soil

        A cellulose-degrading composite microbial system containing a mixture of microbes was previously shown to demonstrate a high straw-degrading capacity. To estimate its potential utilization as an inoculant to accelerate straw biodegradation after returning straw to the field, two cellulose-degrading composite microbial systems named ADS3 and WSD5 were inoculated into wheat straw-amended soil in the laboratory. The microbial survival of the inoculant was confirmed by a denaturing gradient gel electrophoresis (DGGE) analysis, whereas the enhancement of straw degradation in soil was assessed by measuring the mineralization of the soil organic matter and the soil cellulase activity. The results indicated that most of the DGGE bands from ADS3 were detected after inoculation into straw-amended autoclaved soil, yet only certain bands from ADS3 and WSD5 were detected after inoculation into straw-amended non-autoclaved soil during five weeks of incubation; some bands were detected during the first two weeks after inoculation, and then disappeared in later stages. Organic matter mineralization was significantly higher in the soil inoculants ADS3 and WSD5 than in the uninoculated controls during the first week, yet the enhanced degradation did not persist during the subsequent incubation. Similar to the increase in soil organic matter, the cellulase activity also increased during the first week in the ADS3 and WSD5 treatments, yet decreased during the remainder of the incubation period. Thus, it was concluded that, although the survival and performance of the two inoculants did not persist in the soil, a significant enhancement of degradation was present during the early stage of incubation.

        동영상 기반 학습에서 하위목표(Sub-goal) 제시와 자기모니터링(self-monitoring)이 학습동기 및 몰입, 학습효과에 미치는 효과

        비대면 학습의 일상화와 함께 동영상 학습에 관한 관심이 급격하게 높아지고 있다. 이에 본 연구는 하위목표와 자기모니터링 원리를 적용하여 동영상 콘텐츠를 개발하였고 효과를 살펴보았다. 설계된 동영상 콘텐츠의 효과를 살펴보기 위하여 학습자의 동영상 몰입, 수업 흥미도, 수업 두려움, 자기효능감, 학습 이해도의 사전 점수와 사후 점수를 실험 전과 후로 자기보고식 설문조사를 실시하였다. S 대학에 재학 중인 20명의 학습자를 대상으로 수집된 자료를 바탕으로 기술통계 분석과 대응표본 t검증(Paired Samples t-test)을 실시하여 동영상 콘텐츠의 효과를 분석하였다. 이에 따른 연구결과는 첫째, 설계된 동영상 콘텐츠 학습을 통하여 동영상 몰입, 수업 흥미도, 자기효능감 요인에서 사전 검사 점수는 사후 검사 점수보다 향상되었고, 수업 두려움 점수는 줄어들었다. 둘째, 하위목표와 자기모니터링 원리를 활용한 동영상 콘텐츠는 학습자의 학습 이해도 향상에 효과를 미치는 것으로 확인되었다. 이러한 연구결과를 바탕으로 온라인 동영상 기반학습에서 동영상 설계를 위한 시사점을 제언하였다. The popularity of video learning is rising quickly as non-face-to-face learning becomes more commonplace. So, using the concepts of subgoals and self-monitoring, this study created video content and evaluated the success of it. A self-report survey measuring the pre-scores and post-scores of learners' video engagement, class interest, class anxiety, self-efficacy, and academic achievement was carried out before and after the experiment to assess the efficacy of the intended video material. The efficiency of the video content was examined using descriptive statistics and Paired Samples t-test on the information gathered from 20 students enrolled in University S. The findings revealed two main trends. First, the pretest scores for video engagement, class interest, and self-efficacy factors improved in comparison to the posttest scores, and the class anxiety score reduced because of learning the prepared video content. Second, it was discovered that using subgoals and self-monitoring concepts in video content improved learners' understanding of what they were learning. These conclusions lead to suggestions for video design in online video-based learning.

      • Biotoxins for Cancer Therapy

        In recent times, a number of studies have provided evidence that biotoxins present great potential as antitumor agents, such as snake venom, bee venom, some bacteria toxins and plant toxins, and thus could be used as chemotherapeutic agents against tumors. The biodiversity of venoms and toxins make them a unique source from which novel anticancer agent may be developed. Biotoxins, also known as natural toxins, include toxic substances produced by plants, animals and microorganisms. Here, we systematically list representative biological toxins that have antitumor properties, involving animal toxins, plant toxins, mycotoxins as well as bacterial toxins. In this review, we summarize the current knowledge involving biotoxins and the active compounds that have anti-cancer activity to induce cytotoxic, antitumor, immunomodulatory, and apoptotic effects in different tumor cells in vivo or in vitro. We also show insights into the molecular and functional evolution of biotoxins.

      • The Influence of VR Tourism’s Perceived Benefits on Value Experience and Intention to Visit: The Moderation Effect of Involvement

        Purpose - The current study focuses on the effects of different degrees of attention to information provided by VR tourism. The purpose of this study was to explore the impact of VR tourism on potential tourists’ intention to visit, and provided an analysis of how information about a tourist destination moderates VR users’ perceived value and intention to visit. Design/Methodology/Approach - The study uses the SPSS 26.0 and AMOS 26.0 statistical packages to analyze a sample of 328 respondents are analyzed using structural equation modeling. Findings - The results suggest that usefulness, ease of use, enjoyment, interactions among the perceived benefits of VR tourism has a positive effect on perceived value except for Presence. Perceived value is positively correlated with intention to visit intention. Perception of value experience has a significant positive impact on the intention to visit a tourist destination. However, while the overall intention to visit is higher with high involvement, increasing experience with tourism has little influence on the intention to visit. Research Implications - The study found that the perceived value of the VR experience has a direct and significant positive impact on the intention to visit, such that a good perception of value in the experience is a driving factor in users’ visit a tourism destination. The implications of this study for future research are discussed.

        TV홈쇼핑의 소비가치가 만족도 및 재구매의도에 미치는 영향: 확장된 기술수용모델(ETAM)을 기반으로

        본 연구는 소비자의 TV홈쇼핑의 재이용여부를 예측하기 위하여 Davis가 개발한 기술수용모델을 변형 ·확정하여 소비가치가 지각된 이용용이성, 유용성, 만족도와 재구매의도에 미치는 영향을 파악하며, 소비가치와 수용, 만족도와 재구매의도 간의 관계를 탐색해 보았다. 설문조사를 실시하여 최종 501부를 분석에 사용하였으며 구조방정식모형을 통하여 연구가설을 검증하였다. 연구 결과를 요약하면, TV홈쇼핑에 대한 세 가지 소비가치는 지각된 이용용이성과 유용성에 정적인 영향을 미치며, 편의적 가치가 지각된 이용 용이성에 미치는 영향력이 가장 크고, 경제적 가 치는 지각된 유용성에 미치는 영향력이 가장 높았다. 또한 지각된 이용용이성과 지각된 유용성은 만족도에 정적인 영향을 주고, 유용성과 만족도는 재구매의도에 정적인 영향을 주었으므로 TV홈쇼핑의 시스템 개선을 통하여 소비자의 수용, 만족과 재구매의도를 높일 수 있을 것이다. In order to predict the consumers`` repurchasing behavior on TV home shopping, we expanded Davis``s technology acceptance mode (TAM) by using the consumers`` perceived ease of use, perceived usefulness, satisfaction, and repurchase intentions. The present research demonstrated the consumption value is a multidimensional definition construct, and could be divided into three elements, namely economic value, convenience value and hedonic value. Five-hundred and one Korean adult consumers, who purchased products or service on TV home shopping last twelve months, were invited to take part in the online investigation. Data analyses were conducted by descriptive statistical analysis, reliability test, confirmatory factor analysis (CFA) and structural equation modeling (SEM) with SPSS ver. 21.0 and AMOS ver. 21.0. Empirical verification through verifying seven major hypotheses indicate that the data offer substantial supports. First, all the three components of consumption value represent the positive influence on perceived ease of use, and the convenience value has the strongest influence. Only economic value and hedonic value showed significant effect on perceived usefulness, and economic value was much more influential. Second, the perceived ease of use influenced the perceived usefulness positively, and both of which show positive effect on the satisfaction, the perceived usefulness and satisfaction affected the repurchase intentions directly.
