        Molecular characterization of chemosensory protein genes in Bactrocera minax (Diptera: Tephritidae)

        Chemoreception is of great importance for survival of insects. Chemosensory proteins (CSPs) are believed to be involved in the perireceptor events of chemosensory system in many insect species, but this has not been clarified in the citrus fruit fly, Bactrocera minax, a serious dipteran pest of citrus crops. Previous studies indicated that four CSP genes were identified in many fly speices in Diptera. In the present study, we also identified four CSP genes in B. minax,namely BminCSP1–4, from a transcriptome database. All CSP proteins encoded by these genes bear the typical hallmarks of the CSP family: an N-terminal signal peptide and the four highly conserved cysteine residues. Phylogenetic analysis comparing with other dipteran CSPs indicated that dipteran CSPs may evolved from three ancestral CSP genes, and revealed the sequence diversities of BminCSPs and showed that BminCSP1–4 are clustered in separate groups, indicating the possibility of their contrasting function in B. minax. Furthermore, the tissue distribution of the four CSP genes in adult B. minax was analyzed by real-time quantitiative PCR. The results demonstrated that BminCSP3 are significantly transcriptionally enriched in antennae; BminCSP4 expressed primarily in heads; BminCSP1 and BminCSP2 showed high expressions in different tissues, such as antennae, abdomens and wings. Based on these findings, the different implications for the functions of BminCSPs are discussed. This study will offer a significant indication for further functional studies of the CSPs in B. minax

        Effect of the Thickness of Al<sub>2</sub>O<sub>3</sub>-TiO<sub>2</sub> Composite Oxide Films on the Electrical Properties of Anode Al Foils

        <P>High-voltage-etched Al foils with TiO2 coating were fabricated via vacuum infiltration. Specimens subjected to different number of coating cycles (n = 0, 1, 2, 4, and 8) were annealed at 550 °C, and subsequently anodized at different voltages (100, 300, and 500 V). The microstructures of the specimens were observed by field emission scanning electron microscopy and field emission transmission electron microscopy. The results showed that uniform TiO2 films (30 nm per coating cycle) were successfully formed on the high-voltage-etched Al foils. Its tunnels showed a dual-layer structure comprising an Al2O3 outer layer and an Al-Ti composite oxide inner layer after anodizing at 500 V. All the TiO2-coated specimens anodized at different voltages could withstand the corresponding voltage. The thicker is the Al-Ti composite oxide layer, the higher is the specific capacitance of the anode Al foils. Compared to the specimens without the TiO2 coating, the increasing ratios of the specific capacitances of the specimens coated 4 times reached the maximum values, which were 60.2%, 34.0%, and 30.2%, when anodized at 100 V, 300 V, and 500 V, respectively. These results suggest that the Al-Ti composite oxide layer is not only effective in enhancing the specific capacitance but also has a significant effect on the properties of the anode Al foils.</P>

        Ginsenoside Rh2 reduces depression in offspring of mice with maternal toxoplasma infection during pregnancy by inhibiting microglial activation via the HMGB1/TLR4/NF-kB signaling pathway

        Background: Maternal Toxoplasma gondii (T. gondii) infection during pregnancy has been associated with various mental illnesses in the offspring. Ginsenoside Rh2 (GRh2) is a major bioactive compound obtained from ginseng that has an anti-T. gondii effect and attenuates microglial activation through toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-kB) signaling pathway. GRh2 also alleviated tumor-associated or lipopolysaccharide-induced depression. However, the effects and potential mechanisms of GRh2 on depression-like behavior in mouse offspring caused by maternal T. gondii infection during pregnancy have not been investigated. Methods: We examined GRh2 effects on the depression-like behavior in mouse offspring, caused by maternal T. gondii infection during pregnancy, by measuring depression-like behaviors and assaying parameters at the neuronal and molecular level. Results: We showed that GRh2 significantly improved behavioral measures: sucrose consumption, forced swim time and tail suspended immobility time of their offspring. These corresponded with increased tissue concentrations of 5-hydroxytryptamine and dopamine, and attenuated indoleamine 2,3-dioxygenase or enhanced tyrosine hydroxylase expression in the prefrontal cortex. GRh2 ameliorated neuronal damage in the prefrontal cortex. Molecular docking results revealed that GRh2 binds strongly to both TLR4 and high mobility group box 1 (HMGB1). Conclusion: This study demonstrated that GRh2 ameliorated the depression-like behavior in mouse offspring of maternal T. gondii infection during pregnancy by attenuating the excessive activation of microglia and neuroinflammation through the HMGB1/TLR4/NF-kB signaling pathway. It suggests that GRh2 could be considered a potential therapy in preventing and treating psychiatric disorders in the offspring mice of mothers with prenatal exposure to T. gondii infection.

        Design and Test of the MEMS Coupled Piezoelectric- Electromagnetic Energy Harvester

        This paper researches on the design and test of the output performance of double-end clamped MEMS coupled piezoelectric–electromagnetic energy harvester. It establishes the theoretical output model of the double-end clamped rectangular beam and trapezoidal beam piezoelectric–electromagnetic energy harvester, and optimizes the structure parameters of piezoelectric and electromagnetic unit with simulation analysis. It also respectively realizes the processing of piezoelectric and electromagnetic unit by MEMS and flexible PCB technology, and completes the performance test of structure prototype through the experimental system. The result showed that the capacity of MEMS coupled piezoelectric–electromagnetic energy harvester, which taked four coil piezoelectric with integrated electromagnetic in series, was 12.23 times higher than that of piezoelectric energy harvester. Also the output voltage and power of coupled trapezoidal beam energy harvester were respectively increased 18.89% and 2.26%, compared with coupled rectangular beam energy harvester.

        Evaluation of In-Vitro Dissolution and In-Vivo Absorption for Two Different Film-Coated Pellets of Clarithromycin

        The aim of this study was to compare two formulations of film-coated pellets containing clarithromycin after single oral dose study in healthy male volunteers. Two formulations with different coating polymers were prepared: formulation-1 (F-1) was prepared by incorporating three kinds of pH-dependent gradient-release coated pellets into capsules and formulation-2 (F-2) was prepared by coated with an insoluble semiosmotic film. Release profiles of filmcoated pellets were evaluated using paddle method under different conditions. Pharmacokinetic profiles of these formulations were obtained in three healthy male volunteers and compared to commercially available immediate release (IR) tablets. The relative bioavailability based on the AUC0-24 h was found to be 96.2% and 58.7% for F-1 and F-2 compared with IR, and the Tmax was delayed.

        Ginsenoside Rh2 attenuates microglial activation against toxoplasmic encephalitis via TLR4/NF-kB signaling pathway

        Background: Ginsenoside Rh2 (GRh2) is a characterized component in red ginseng widely used in Korea and China. GRh2 exhibits a wide range of pharmacological activities, such as anti-inflammatory, antioxidant, and anticancer properties. However, its effects on Toxoplasma gondii (T. gondii) infection have not been clarified yet. Methods: The effect of GRh2 against T. gondii was assessed under in vitro and in vivo experiments. The BV2 cells were infected with tachyzoites of T. gondii RH strain, and the effects of GRh2 were evaluated by MTT assay, morphological observations, immunofluorescence staining, a trypan blue exclusion assay, reverse transcription PCR, and Western blot analyses. The in vivo experiment was conducted with BALB/c mice inoculated with lethal amounts of tachyzoites with or without GRh2 treatment. Results and conclusion: The GRh2 treatment significantly inhibited the proliferation of T. gondii under in vitro and in vivo studies. Furthermore, GRh2 blocked the activation of microglia and specifically decreased the release of inflammatory mediators in response to T. gondii infection through TLR4/NF-kB signaling pathway. In mice, GRh2 conferred modest protection from a lethal dose of T. gondii. After the treatment, the proliferation of tachyzoites in the peritoneal cavity of infected mice markedly decreased. Moreover, GRh2 also significantly decreased the T. gondii burden in mouse brain tissues. These findings indicate that GRh2 exhibits an antieT. gondii effect and inhibits the microglial activation through TLR4/NF-kB signaling pathway, providing the basic pharmacological basis for the development of new drugs to treat toxoplasmic encephalitis.

        Decision-Making of Determining the Start Time of Charging / Discharging of Electrical Vehicle Based on Prospect Theory

        The moment when Electrical Vehicle (EV) starts charging or discharging is one of the most important parameters in estimating the impact of EV load on the grid. In this paper, a decisionmaking problem of determining the start time of charging and discharging during allowed period is proposed and studied under the uncertainty of real-time price. Prospect theory is utilized in the decision-making problem of this paper for it describes a kind of decision making behaviors under uncertainty. The case study uses the parameters of Springo SGM7001EV and adopts the historical realtime locational marginal pricing (LMP) data of PJM market for scenario reduction. Prospect values are calculated for every possible start time in the allowed charging or discharging period. By comparing the calculated prospect values, the optimal decisions are obtained accordingly and the results are compared with those based on Expected Utility Theory. Results show that with different initial Stateof-Charge ( 0 SoC ) and different reference points, the optimal start time of charging can be the one between 12 a.m. to 3 a.m. and optimal discharging starts at 2 p.m. or 3p.m. Moreover, the decision results of Prospect Theory may differ from that of the Expected Utility Theory with the reference points changing.

      • Effect of Cisplatin on the Frequency and Immuno-inhibitory Function of Myeloid-derived Suppressor Cells in A375 Melanoma Model

        Background: To investigate the change of frequency and immuno-inhibitory function of myeloid-derived suppressor cells (MDSCs) after treatment of cisplatin (DDP) in A375 human melanoma model. Materials and Methods: BALB/c nude mice were inoculated with A375 cells to establish the human melanoma model and randomly divided into control group given normal saline (NS) and experimental group treated with DDP (5 mg/kg). The percentages of MDSCs in the tumor tissue and peripheral blood after DDP treatment were detected by flow cytometry. The proliferation and interferon-${\gamma}$ (IFN-${\gamma}$) secretion of T cells co-cultured with MDSCs were analyzed through carboxyfluorescein succinimidyl ester (CFSE) labeling assay and enzyme-linked immunospot (ELISPOT) assay, respectively. Results: In A375 human melanoma model, DDP treatment could significantly decrease the percentage of MDSCs in the tumor tissue, but exerted no effect on the level of MDSCs in peripheral blood. Moreover, DDP treatment could attenuate the immuno-inhibitory function of MDSCs. T cells co-cultured with DDP-treated MDSCs could dramatically elevate the proliferation and production of INF-${\gamma}$. Conclusions: DDP can decrease the frequency and attenuate immuno-inhibitory function of MDSCs in A375 melanoma model, suggesting a potential strategy to augment the efficacy of combined immunotherapy.
