        PHF2 histone demethylase acts as a tumor suppressor in association with p53 in cancer

        Plant homeodomain finger 2 (PHF2) has a role in epigenetic regulation of gene expression by demethylating H3K9-Me2. Several genome-wide studies have demonstrated that the chromosomal region including the PHF2 gene is often deleted in some cancers including colorectal cancer, and this finding encouraged us to investigate the tumor suppressive role of PHF2. As p53 is a critical tumor suppressor in colon cancer, we tested the possibility that PHF2 is an epigenetic regulator of p53. PHF2 was associated with p53, and thereby, promoted p53-driven gene expression in cancer cells under genotoxic stress. PHF2 converted the chromatin that is favorable for transcription by demethylating the repressive H3K9-Me2 mark. In an HCT116 xenograft model, PHF2 was found to be required for the anticancer effects of oxaliplatin and doxorubicin. In PHF2-deficient xenografts, p53 expression was profoundly induced by both drugs, but its downstream product p21 was not, suggesting that p53 cannot be activated in the absence of PHF2. To find clinical evidence about the role of PHF2, we analyzed the expressions of PHF2, p53 and p21 in human colon cancer tissues and adjacent normal tissues from patients. PHF2 was downregulated in cancer tissues and PHF2 correlated with p21 in cancers expressing functional p53. Colon and stomach cancer tissue arrays showed a positive correlation between PHF2 and p21 expressions. Informatics analyses using the Oncomine database also supported our notion that PHF2 is downregulated in colon and stomach cancers. On the basis of these findings, we propose that PHF2 acts as a tumor suppressor in association with p53 in cancer development and ensures p53-mediated cell death in response to chemotherapy.

        Breath hydrogen and methane are associated with intestinal symptoms in patients with chronic pancreatitis

        Backgrounds: The bacterial overgrowth might be associated with chronic pancreatitis. This study was to evaluate the prevalence and characteristics of small intestinal bacterial overgrowth (SIBO) in patients with chronic pancreatitis. Methods: 36 patients with chronic pancreatitis and 49 healthy controls undergoing the hydrogen (H<SUB>2</SUB>)-methane (CH<SUB>4</SUB>) lactulose breath test (LBT) were reviewed. The LBT positivity (+) indicating the presence of SIBO, gas types, bowel symptom questionnaire, laboratory and radiologic results were surveyed. The LBT+ was (1) an increase in the breath H<SUB>2</SUB> (≥20 ppm) or CH<SUB>4</SUB> (≥10 ppm) over the baseline or (2) a baseline H<SUB>2</SUB> (≥20 ppm) or CH<SUB>4</SUB> (≥10 ppm) within 90 min after lactulose load. Results: LBT+ was significantly higher in the patients (17/36) than in controls (1¾9) (47.2% vs. 26.5%, P < 0.05). During LBT, the H<SUB>2</SUB> levels between 0 and 105 min were significantly higher in patients than in controls. Among LBT+ patients, 11 (64.7%), 1 (5.9%), 5 (29.4%) were in the LBT (H<SUB>2</SUB>)+, (CH<SUB>4</SUB>)+, (mixed)+ groups, respectively. The LBT+ group had significantly higher scores of flatus than those of the LBT- group. Considering the subtypes of LBT, the LBT (mixed)+ group had higher symptom scores of significance or tendency in hard stool, strain, urgency, and flatus than LBT- group The laboratory and radiologic features were not significantly different between LBT+ and LBT- groups. Conclusions: SIBO is common in patients in chronic pancreatitis. Especially, excretions of mixed H<SUB>2</SUB> and CH<SUB>4</SUB> appear to be related with deterioration of intestinal symptoms.

      • Substrate and pretreatment dependence of Cu nucleation by metal-organic chemical vapor deposition

        The nucleation of copper (Cu) with (hfac)Cu(VTMS) organometallic precursor is investigated for Si, SiO2, TiN, and W2Nsubstrates. As the deposition temperature is increased, the dominant growth mechanism is observed to change from the nucleationof Cu particles to the clustering of Cu nuclei around 180.C independent of the employed substrates. It is also observed that the2 sub-strates. Dimethyldichlorosilane treatment is found to passivate the surface of TiN substrate, contrary to the generally acceptedbelief, when the TiN substrate is cleaned by H2O2 solution before the treatment.. 2002 Elsevier Science B.V.         , The purpose of this study was carried out to know the responses to the climatic factors on the flowering time and bud burst of Forsythia Koreana NAKAI in the Southern island, Jindo, Wando, and Sinan. Monthly average air temperature, and precipitation were compared with the responses of flowering time and bud burst among islands so that the line of equal flowering time and bud burst were demonstrated on the map. The distribution of monthly average temperature and precipitation showed high in order of Wando, Jindo and Sinan island especially from Winter to Spring. Also the same trend were appeared in the flowering time and bud burst of Forsythia Koreana NAKAI.

        In designing and evaluating manipulators, it is important to understand the capability of kinematic and static performances. Both workspace and manipulability can be considered as such performance measures. In general, the measure of manipulability is related to the kinematics for serial type manipulators while it is related to the statics for parallel type manipulators. In the development of a new definition of manipulability measure emphasis has been placed on the invariant properties with change of the origin and dimension. In this paper, the mathematical and physical meaning of manipulability suggested in some research works have been examined, and more meaningful measure of manipulability using the ratio of ?? to ?? has been suggested, which has been applied to a planar serial redundant robot.

        Studies on the pathogenesis of hemorrhagic fever with syndrome(HFRS) in patients have been seriously hindered by the abscence of appropriate animal model. The major pathologic findings in patient with HFRS are generalized vasculopathy. However, the pathogenesis of disseminated intravascular coagulation (DIC) and bleeding tendency in patients with HFRS are not well understood. It has been reported that the specific viral antigens are usually demonstrated in the endothelium of glomerular capillaries, and resulted in application of primary cultured human umbilical vein endothelial of glomerular capillaries, and resulted in application of primary cultured human umbilical vein endothelial cells (HUVEC) to study on the pathogenesis of HFRS. In this study, HUVEC were infected with Hantaan virus. The outcome Hantaan virus infection of HUVEC were assessed by flurescence activated cell scanning(FACS). The result of FACS assay indicated that 11% of the cells contained veiral antigens on 3 days postinfection. The proportion of virus-containing cells increased to 85% by the fifth days, adn 96% by the seventh, respectively. These observed no CPE development in the HUVEC cells infected with Hantaan virus and observed up to 13 days. The sensitivity of immunohistochemical method used for the viral antigens appeared similar to that of indirect immunofluorescent antibody technique. The monocyte adherence rate of infected HUVEC was much higher that than that of uninfected HUVEC, The monocyte adherence rate of infected HUVEC was increased by addition of convalescent serum of patient with HFRS. Infection with Hantaan virus HUVEC stimulated the production of superoxide radical.

        High-level wastes contain significant quantities of platinum group metals such as Pd(II), Rh(III), and Ru(III). There are many techniques such as solvent extraction, precipitation, and adsorption methods for the separation of Rh(III). In this paper, batch experiments were carried out to investigate ion exchange characteristics of Rh(III) from a nitric acid solution by commercial cation and anion exchange resins such as Dowex 50W, IRN 78 and Dowex 1x8. The effects of ionic group of resins, solution temperature, and the concentration of nitric acid were investigated. And the elution characteristic of Rh(III) by various eluents was studied.

      • Characterization of Siberian wild rye grass EsHsp16.9 Gene and Their Expression under Various Environmental Conditions

        Small heat shock proteins (Hsps) are one of most conserved molecular chaperones that protect stress-inducible denaturation of substrates in living organisms. Small Hsps consist of a large subfamily categorized by subcellular localization ranging in size from 12 to 40 kDa. Here, we identified and characterized a small Hsp 16.9 gene (EsHsp16.9) from Siberian wild rye (Elymus sibiricus L.). EsHsp16.9 is a 456-bp cDNA with an open reading frame predicted to encode a 151-amino acid protein. It possesses a conserved ɑ-crystallin domain, which is a unique domain for small Hsps; shares high sequence similarity with cytosolic class I small Hsps among the small Hsp subfamily in Arabidopsis; and is close (96% similarity) to small Hsp in wheat. Northern blot analysis showed that EsHsp16.9 transcripts were enhanced by heat, drought, arsenate, methyl viologen, and H2O2 treatments. Moreover, we expressed and purified recombinant EsHsp16.9 proteins in Escherichia coli to confirm its activity as a molecular chaperone. We found that recombinant EsHsp16.9 exhibits effective molecular chaperone activity, as determined by inhibition of thermal aggregation of malate dehydrogenase (MDH), which is broadly used as a model substrate.
