Objective: Apoptosis of ovarian granulosa cells (GCs) affects mammalian follicular development and fecundity. This study aimed to explore the regulatory relationship between microRNA-26a (miR-26a) and the 3β-hydroxysteroid-Δ24-reductase gene (DHCR24) gene in porcine follicular granular cells (pGCs), and to provide empirical data for the development of methods to improve the reproductive capacity of pigs. Methods: The pGCs were transfected with miR-26a mimic, miR-26a inhibitor and DHCR24-siRNA in vitro. The cell apoptosis rate of pGCs was detected by the flow cytometry. The secretion levels of estradiol (E2) and progesterone (P) in pGCs were detected by enzyme-linked immunosorbent assay. Double luciferase validation system was used to detect the binding sites between miR-26a and DHCR24 3'-UTR region. Qualitative real-time polymerase chain reaction and Western blotting were used to verify the DHCR24 mRNA and protein expression in pGCs, respectively, after transfecting with miR-26a mimic and miR-26a inhibitor. Results: Results showed that enhancement of miR-26a promoted apoptosis, and inhibited E2 and P secretion in pGCs. Meanwhile, inhibition of DHCR24 also upregulated the Caspase-3 expression, reduced the BCL-2 expression, promoted pGCs apoptosis, and inhibited E2 and P secretion in pGCs. There were the binding sites of miR-26a located within DHCR24 3'-UTR. Up-regulation of miR-26a inhibited DHCR24 mRNA and protein expression in pGCs. Conclusion: This study demonstrates that miR-26a can promote cell apoptosis and inhibit E2 and P secretion by inhibiting the expression of DHCR24 in pGCs.

This study fabricated nano-CeO2/biodiesel carbon soot (BCS) composites using the raw material BCS,which is the byproduct of biodiesel combustion and cerium salt and used as a novel lubricant additiveto PAO6 for steel/steel contact. Oleylamine modified nano-CeO2/BCS composites comprise abundant lipophilicgroups, and thus nano-CeO2/BCS composites exhibit good dispersion in PAO6. The tribologicalresults showed that nano-CeO2/BCS composites significantly improved the lubricating performances ofPAO6. In case of 0.1 wt% nano-CeO2/BCS composites dispersed in PAO6, the friction coefficient and wearvolume were reduced by 17.3 and 35.2%, respectively. The improved tribological properties were attributedto the composites rolling and forming of a lubricating film on the friction surface and the presence ofBCS and CeO2 in composites exerting a synergistic lubricating effect, which simultaneously improved thetribological properties of PAO6. This discovery contributes to the conversion of BCS into valuables andpromotes the high value utilization of BCS as a lubricating additive.
Simulation of an X-ray Fresnel Zone Plate with Nonideal Factors

Fresnel zone plates have been widely used in many applications, such as x-ray telescopes, microfluorescence, and microimaging. To obtain an x-ray Fresnel zone plate, many fabrication methods, such as electron-beam etching, ion-beam etching and chemical etching, have been developed. Fresnel zone plates fabricated by these methods will inevitably lead to some nonideal factors, which have an impact on the focusing characteristics of the zone plate. In this paper, the influences of these nonideal factors on the focusing characteristics of the zone plate are studied systematically, by numerical simulations based on scalar diffraction theory. The influence of the thickness of a Fresnel zone plate on the absolute focusing efficiency is calculated for a given incident x-ray’s wavelength. The diffraction efficiency and size of the focal spot are calculated for different incline angles of the groove. The simulations of zone plates without struts, with regular struts, and with random struts are carried out, to study the effects of struts on the focusing characteristics of a zone plate. When a Fresnel zone plate is used to focus an ultrashort x-ray pulse, the effect of zone-plate structure on the final pulse duration is also discussed.

In continuation of our endeavor to develop new, potent, selective and less toxic antiviral agents, a novel series of 2-(2-amino/chloro-4-(2,4-dibromophenyl) thiazol-5-ylthio)acetamide derivatives was synthesized via an expeditious route and evaluated for their anti-HIV activities against wild-type virus and clinically relevant mutant strains, and for their anti-influenza virus activities against influenza A (H1N1 and H3N2) and influenza B in cellular assays. The selected active compounds were also assayed for their enzymic inhibitory activities. The results showed that some 2-chloro substituted thiazolylthioacetamide derivatives possessed potent activity against wild type HIV-1 and several key mutant strains (E138K, K103N, L100I) of HIV-1 in MT-4 cells with EC50 values in micromolar range. Two 2-amino substituted thiazole derivatives 8a7 and 8a8 displayed significant potency against influenza A/H1N1 in MDCK cells with EC50 values much lower than that of oseltamivir carboxylate, ribavirin, amantadine and rimantadine. Though the mechanism of actions is still unclear, these novel thiazolylthioacetamides might serve as original leads for further pharmacological investigations as potential therapeutic agents against HIV-1 or influenza virus.
Overexpression of IbMPK3 increases low-temperature tolerance in transgenic sweetpotato

Sweetpotato is an important crop that is very sensitive to low temperatures. Mitogen-activated protein kinase (MAPK) is involved in plant growth and development and is responsive to many environmental stresses. IbMPK3 is strongly regulated by low-temperature stress. We studied the function of IbMPK3 and how it may enhance the adaptability of sweetpotato plants to low-temperature stress. Transgenic sweetpotato plants overexpressing IbMPK3 were generated, and three trans- genic lines with the highest expression level of IbMPK3 were used for low-temperature tests. Transgenic plants exposed to low-temperature stress had less damage associated with higher photosynthesis efficiency and less cell membrane damage. IbMPK3-overexpressing transgenic plants could modulate reactive oxygen species (ROS) metabolism with lower levels of O2− and H2O2 accumulation and higher enzymatic activities than WT plants. Transcript expression levels of some ROS- related genes (IbCAT, IbCu-ZnSOD, and IbCAT) and stress-responsive genes (IbP3B and IbCOR27) were significantly upregulated in transgenic plants compared to WT. These results indicate that IbMPK3 has an important role in sweetpotato response to low temperatures.
