        Characterization and functional study of juvenile hormone diol kinase gene in Nilaparvata lugens (Stål) (Hemiptera: Delphacidae)

        The brown planthopper, Nilaparvata lugens (Stål) is an important pest in rice. It has been widely recognized that the juvenile hormone (JH) is regulated by its hydrolase, which includes juvenile hormone esterase (JHE), juvenile hormone epoxide hydrolase (JHEH) and juvenile hormone diol kinase (JHDK). In this paper, we cloned the gene of Jhdk and the gene expression at different stages of N. lugens was analysed, and the relationship with Jhe and Jheh was studied after silencing the jhdk gene of N. lugens (Nljhdk) through double-stranded RNA (dsRNA) feeding. We also explored the expression of the three JH hydrolase after indoxacarb treatments. RT-PCR was used to amplify the full length Jhdk cDNA, and the Nljhdk gene was expressed throughout all the development periods tested and showed the lowest level at the 4th instar and the highest in the 5th instar. The expression level of Nljhdk in male adults was higher than that of female adults. Through feeding, dsRNA against Nljhdk successfully knocked down the target gene, which had no significant effect on the expression of the jhe gene of N. lugens (Nljhe), while the expression of Nljheh was upregulated. Indoxacarb could inhibit N. lugens reproduction, and the expression level of Nljhe and Nljhdk increased with the increasing of indoxacarb concentration, but the expression of the jheh gene of N. lugens (Nljheh) was reduced. These studies provide a line of experimental evidence in N. lugens to support that Nljhdk encodes the functional protein involved in JH degradation and further showed the relationship of the three hydrolases and the mechanism of indoxacarb inhibition of the fecundity of N. lugens.

        Experimental study on heat transfer characteristics of supercritical carbon dioxide natural circulation

        An experimental study has been conducted to investigate the heat transfer characteristics of supercriticalcarbon dioxide (sCO2) uniformly heated in the horizontal circular smooth tube. The results illustratedthat there was a significant difference in heat transfer between the top wall and bottom wall due to thebuoyancy. Bulk flow acceleration cannot be negligible in the high heat flux region, which leads to heattransfer deterioration. A new heat transfer correlation is proposed, in which the buoyancy parameter andbulk flow acceleration have been taken into account. The new correlation and six classic correlations forsCO2 are examined in horizontal tubes. The comparison indicates that the new correlation has a betterperformance for sCO2 flowing through a horizontal heating tube under natural circulation conditions. Forexample, 94.9% of the calculated results using the new heat transfer correlation were within ±30% of theexperimental results while only 87.9% of that using the Jackson correlation (the best of the six) werewithin the same error bands.

        A novel HB-SC-MCCNN model for intelligent fault diagnosis of rolling bearing

        The incompleteness and lack of bearing fault data have become important problems in bearing fault diagnosis. This paper presents an intelligent fault diagnosis method for rolling bearings based on a similarity clustering multi-channel convolution neural network with the hierarchical branch (HB-SC-MCCNN). First, the relevant features are extracted by MCCNN, and combined with the similarity clustering principle, the accurate binary classification is realized in the case of insufficient labeled data. Second, the similarity clustering module and additional loss are added to the SC-MCCNN network to form a hierarchical-branch network, which simplifies the problem of fault multi-classification into binary classification with multiple steps, and to reduces the dependence on the amount of label data in multi-classification. Finally, based on the self-learning characteristics of HB-SC-MCCNN, the unlabeled data and the missing fault types in the training set are re-labeled to realize the re-training of the network. On the benchmark dataset, the comparison experiment results with several salient deep learning models show that the method proposed in this paper successfully realizes the hierarchical diagnosis of bearing faults and presents more substantial competitiveness in the case of insufficient labeled data and missing fault types.

        Expression of Prostaglandin E2 Receptors in Acquired Middle Ear Cholesteatoma

        Objectives. To investigate the expression of prostaglandin E2 receptor subtypes, E-prostanoid (EP) 1–4 receptors, in acquired cholesteatoma and its possible role in the pathologic process of this disorder. Methods. Specimens of human acquired cholesteatoma were obtained from 29 patients and 19 skin biopsies of normal external auditory canal were as controls. The mRNA and protein expression of EP receptors was assessed by quantitative real-time polymerase chain reaction, immunohistochemistry and Western blot. Results. In acquired cholesteatoma, EP1–EP4 receptors were mainly expressed on squamous epithelium and subepithelial infiltrated inflammatory cells. In external auditory canal skin, EP1–EP4 receptors were mainly expressed on squamous epithelium and glandular epithelium. The expression of EP4 receptor on mRNA and protein levels were significant lower in acquired cholesteatoma compared with controls. EP1–EP3 receptors had no significant difference between the experimental and control group. Conclusion. Low expression of EP4 may play a crucial role in the pathologic process of inflammation reaction and bone destruction in acquired cholesteatoma, but not EP1, EP2, or EP3 receptors.

        ssc-miR-185 targets cell division cycle 42 and promotes the proliferation of intestinal porcine epithelial cell

        Objective: microRNAs (miRNAs) can play a role in a variety of physiological and pathological processes, and their role is achieved by regulating the expression of target genes. Our previous high-throughput sequencing found that ssc-miR-185 plays an important regulatory role in piglet diarrhea, but its specific target genes and functions in intestinal porcine epithelial cell (IPEC-J2) are still unclear. We intended to verify the target relationship between porcine miR-185 and cell division cycle 42 (CDC42) gene in IPEC-J2 and to explore the effect of miR-185 on the proliferation of IPEC-J2 cells. Methods: The TargetScan, miRDB, and miRanda software were used to predict the target genes of porcine miR-185, and CDC42 was selected as a candidate target gene. The CDC42-3' UTR-wild type (WT) and CDC42-3'UTR-mutant type (MUT) segments were successfully cloned into pmirGLO luciferase vector, and the luciferase activity was detected after co-transfection with miR-185 mimics and pmirGLO-CDC42-3'UTR. The expression level of CDC42 was analyzed using quantitative polymerase chain reaction and Western blot. The proliferation of IPEC-J2 was detected using cell counting kit-8 (CCK-8), methylthiazolyldiphenyl-tetrazolium bromide (MTT), and 5-ethynyl-2'-deoxyuridine (EdU) assays. Results: Double enzyme digestion and sequencing confirmed that CDC42-3'UTR-WT and CDC42-3'UTR-MUT were successfully cloned into pmirGLO luciferase reporter vector, and the luciferase activity was significantly reduced after co-transfection with miR-185 mimics and CDC42-3'UTR-WT. Further we found that the mRNA and protein expression level of CDC42 were down-regulated after transfection with miR-185 mimics, while the opposite trend was observed after transfection with miR-185 inhibitor (p<0.01). In addition, the CCK-8, MTT, and EdU results demonstrated that miR-185 promotes IPEC-J2 cells proliferation by targeting CDC42. Conclusion: These findings indicate that porcine miR-185 can directly target CDC42 and promote the proliferation of IPEC-J2 cells. However, the detailed regulatory mechanism of miR-185/CDC42 axis in piglets' resistance to diarrhea is yet to be elucidated in further investigation.

        A key ABA hydrolase gene, OsABA8ox3 is involved in rice resistance to Nilaparvata lugens by affecting callose deposition

        Abscisic acid (ABA) is an important plant hormone in regulating abiotic and biotic stresses. OsABA8ox3 is the key gene in ABA hydrolase genes, and plays an important role in controlling ABA level, but little is known in rice resistance to insects. We used rice osaba8ox3 T-DNA insertion mutant (knocking down the OsABA8ox3 gene) to elucidate rice resistance to the insect. There were obvious phenotype differences between the osaba8ox3 T-DNA insertion mutant and wild-type (WT), and the relative expression of synthetase genes in the osaba8ox3 mutant was higher, while the relative expression of hydrolase genes was lower than that of WT, respectively. The electrical penetration graph (EPG) recording indicated that the osaba8ox3 mutant had the less sucking phloem sap duration compared with WT, which indicated a significant increase in rice resistance to brown planthopper (Nilaparvata lugens; BPH). The callose deposition in the osaba8ox3 mutant increased by 60.39%, 52.2%, 26.6% and 31.7% than that of WT after BPH feeding for 0, 24, 48, and 72 h, respectively. These results showed OsABA8ox3 gene played an important role in rice resistance to BPH, and also provided new insights into the mechanism of callose deposition regulation in response to the piercing-sucking pest.

        Effects of dietary Acremonium terricola culture supplementation on the quality, conventional characteristics, and flavor substances of Hortobágy goose meat

        This study aimed to determine the effect of dietary supplementation with Acremonium terricolaculture (ATC) on the quality, conventional characteristics, and flavor substances of Hortobágygoose meat. A total of 720 one-day-old goslings were divided into four dietary treatments, eachconsisting of six cages of 30 goslings. The dietary conditions consisted of the control group andthree treatment groups supplemented with 3, 5, or 7 g/kg ATC. In male geese, supplementationwith 3 g/kg ATC elevated the crude ash (CA) content of the thigh muscle compared to the controlgroup, and the CA content of the pectoralis major was significantly elevated when geesewere supplemented with 5 g/kg ATC (p < 0.05). In females, compared with the control group,supplementation with 7 g/kg ATC enhanced the crude protein (CP) content of the pectoralismajor. Supplementation with 7 g/kg ATC also increased the crude fat (CF) content of the pectoralismajor in females as well as in both sexes; moreover, this supplementation dose increasedthe inosinic acid content of the thigh muscle in males and in both sexes. In contrast, supplementationwith 5 g/kg ATC decreased the pH of the thigh muscle at 12 h postmortem (p < 0.01). No significant changes in meat color, water loss rate, shear force, moisture content or aminoacid (AA) levels were observed after ATC supplementation (p > 0.05). Levels of saturated fattyacids (SFAs) and polyunsaturated FAs (PUFAs) in the pectoralis major and levels of SFAs,monounsaturated FAs (MUFAs), and PUFAs in the thigh muscle were not affected by the supplementation. Overall, ATC supplementation had positive effects on the pH, and CA, CP, CF,inosinic acid contents as well as on the FA composition of gosling meat. The optimal level ofATC supplementation was 7 g/kg in goslings from 1 to 70 days of age.

        IRE1α protects against osteoarthritis by regulating progranulin-dependent XBP1 splicing and collagen homeostasis

        Osteoarthritis (OA) is a full-joint, multifactorial, degenerative and inflammatory disease that seriously affects the quality of life of patients due to its disabling and pain-causing properties. ER stress has been reported to be closely related to the progression of OA. The inositol-requiring enzyme 1α/X-box-binding protein-1 spliced (IRE1α/XBP1s) pathway, which is highly expressed in the chondrocytes of OA patients, promotes the degradation and refolding of abnormal proteins during ER stress and maintains the stability of the ER environment of chondrocytes, but its function and the underlying mechanisms of how it contributes to the progression of OA remain unclear. This study investigates the role of IRE1α/ERN1 in OA. Specific deficiency of ERN1 in chondrocytes spontaneously resulted in OA-like cartilage destruction and accelerated OA progression in a surgically induced arthritis model. Local delivery of AdERN1 relieved degradation of the cartilage matrix and prevented OA development in an ACLT-mediated model. Mechanistically, progranulin (PGRN), an intracellular chaperone, binds to IRE1α, promoting its phosphorylation and splicing of XBP1u to generate XBP1s. XBP1s protects articular cartilage through TNF-α/ERK1/2 signaling and further maintains collagen homeostasis by regulating type II collagen expression. The chondroprotective effect of IRE1α/ERN1 is dependent on PGRN and XBP1s splicing. ERN1 deficiency accelerated cartilage degeneration in OA by reducing PGRN expression and XBP1s splicing, subsequently decreasing collagen II expression and triggering collagen structural abnormalities and an imbalance in collagen homeostasis. This study provides new insights into OA pathogenesis and the UPR and suggests that IRE1α/ERN1 may serve as a potential target for the treatment of joint degenerative diseases, including OA.
