        Transcriptome analysis of flower colour reveals the correlation between SNP and differential expression genes in Phalaenopsis

        Background Phalaenopsis is an important ornamental plant that has great economic value in the world flower market as one of the most popular flower resources. Objective To investigate the flower colour formation of Phalaenopsis at the transcription level, the genes involved in flower color formation were identified from RNA-seq in this study. Methods In this study, white and purple petals of Phalaenopsis were collected and analyzed to obtained (1) differential expression genes (DEGs) between white and purple flower color and (2) the association between single nucleotide polymorphisms (SNP) mutations and DEGs at the transcriptome level. Results The results indicated that a total of 1,175 DEGs were identified, and 718 and 457 of them were up- and down-regulated genes, respectively. Gene Ontology and pathway enrichment showed that the biosynthesis of the secondary metabolites pathway was key to color formation, and the expression of 12 crucial genes (C4H, CCoAOMT, F3’H, UA3’5’GT, PAL, 4CL, CCR, CAD, CALDH, bglx, SGTase, and E1.11.17) that are involved in the regulation of flower color in Phalaenopsis. Conclusion This study reported the association between the SNP mutations and DEGs for color formation at RNA level, and provides a new insight to further investigate the gene expression and its relationship with genetic variants from RNA-seq data in other species.

        A BIO-INSPIRED POLYDOPAMINE APPROACH TO PREPARATION OF GOLD-COATED Fe3O4 CORE-SHELL NANOPARTICLES: SYNTHESIS, CHARACTERIZATION AND MECHANISM

        A biomimetic and facile approach for integrating Fe3O4 and Au with polydopamine (PDA) was proposed to construct gold-coated Fe3O4 nanoparticles (Fe3O4@Au–PDA) with a core–shell structure by coupling in situ reduction with a seed-mediated method in aqueous solution at room temperature. The morphology, structure and composition of the core–shell structured Fe3O4@Au–PDA nanoparticles were characterized by transmission electron microscopy (TEM), X-ray powder diffraction (XRD) and X-ray photoelectron spectrometry (XPS). The formation process of Au shell was assessed using a UV-Vis spectrophotometer. More importantly, according to investigating changes in PDA molecules by Fourier transform infrared spectroscopy (FTIR) and in preparation process of the zeta-potential data of nanoparticles, the mechanism of core–shell structure formation was proposed. Firstly, PDA-coated Fe3O4 are obtained using dopamine (DA) self-polymerization to form thin and surface-adherent PDA films onto the surface of a Fe3O4 "core". Then, Au seeds are attached on the surface of PDA-coated Fe3O4 via electrostatic interaction in order to serve as nucleation centers catalyzing the reduction of Au3+ to Au0 by the catechol groups in PDA. Accompanied by the deposition of Au, PDA films transfer from the surface of Fe3O4 to that of Au as stabilizing agent. In order to confirm the reasonableness of this mechanism, two verification experiments were conducted. The presence of PDA on the surface of Fe3O4@Au–PDA nanoparticles was confirmed by the finding that glycine or ethylenediamine could be grafted onto Fe3O4@Au–PDA nanoparticles through Schiff base reaction. In addition, Fe3O4@Au–DA nanoparticles, in which DA was substituted for PDA, were prepared using the same method as that for Fe3O4@Au–PDA nanoparticles and characterized by UV-Vis, TEM and FTIR. The results validated that DA possesses multiple functions of attaching Au seeds as well as acting as both reductant and stabilizing agent, the same functions as those of PDA.

        Recent advances of Na3V2(PO4)3 as cathode for rechargeable zinc-based batteries

        Rechargeable zinc-based batteries (RZBs) with the advantages of high safety, low cost, abundant resources and environmental friendliness, are considered as advanced secondary battery systems that can be applied to large-scale energy storage. As an important cathode material for RZBs, NASICON-type Na3V2(PO4)3 (NVP) possesses three-dimensional and large-scale ion channels that facilitate the rapid diffusion of Zn2+, and has a higher average operating voltage compared with other vanadium-based compounds, thus exhibiting the possibility of realizing RZBs with high energy density. However, NVP still has some problems, such as poor electronic conductivity and spontaneous dissolution in aqueous solution. The sluggish kinetics of Zn2+ (de)intercalation in NVP and dendritic growth on the Zn anode also contribute to the poor rate performance and short cycle life of the batteries. In this review, optimization strategies for the electrochemical performance of RZBs with NVP as cathode are systematically elaborated, including modification of NVP cathode and optimization of electrolyte. Several mainstream energy storage mechanisms and analysis methods in this battery system are sorted out and summarized. On this basis, the development direction of NVP–RZB system is further prospected.

        A quantitative analysis on the rheological characteristics of polyurea thickeners with the geometry of helical fiber and nanotube

        Understanding the thickening mechanisms of grease thickeners can provide critical insight into thedesign of lubricating greases. Polyurea (PU), as an important thickening system with diverse structures,the role of thickener structure in the thickening mechanism is not clearly elucidated. In this study, twotypes of PU thickeners, which have the structures of helical fibers and nanotubes, were synthesized,respectively. Via a quantitative analysis in the rheological characteristics of the two PU thickeners, itwas found that the PU with helical fibers structure possess higher thickening capacity and more obviousshear-thinning effect, while the one with nanotubes structure is more effective to improve the elasticityand structural stability of grease. Based on the rheological characteristics and structural features of thetwo PU thickeners, the role of structure in the thickening mechanism of PU was discussed. The proposedmechanisms explain why the commercial PU greases commonly have a complex thickening system thathas both fibrous clusters and nanotube structures.

      • A long-term tunnel settlement prediction model based on BO-GPBE with SHM data

        The new metro crossing the existing metro will cause the settlement or floating of the existing structures, which will have safety problems for the operation of the existing metro and the construction of the new metro. Therefore, it is necessary to monitor and predict the settlement of the existing metro caused by the construction of the new metro in real time. Considering the complexity and uncertainty of metro settlement, a Gaussian Prior Bayesian Emulator (GPBE) probability prediction model based on Bayesian optimization (BO) is proposed, that is, BO-GPBE. Firstly, the settlement monitoring data are analyzed to get the influence of the new metro on the settlement of the existing metro. Then, five different acquisition functions, that is, expected improvement (EI), expected improvement per second (EIPS), expected improvement per second plus (EIPSP), lower confidence bound (LCB), probability of improvement (PI) are selected to construct BO model, and then BO-GPBE model is established. Finally, three years settlement monitoring data were collected by structural health monitoring (SHM) system installed on Nanjing Metro Line 10 are employed to demonstrate the effectiveness of BO-GPBE for forecasting the settlement.

        Vitamin E improves antioxidant status but not lipid metabolism in laying hens fed a aged corn-containing diet

        Objective: The objective of this study was to determine whether a dietary vitamin E (VE) supplement could alleviate any detrimental effects of aged corn on lipid metabolism and antioxidant status in laying hens. Methods: The experiment consisted of a 2×3 factorial design with two corn types (normal corn and aged corn (stored for 4 yr) and three concentrations of VE (0, 20, and 100 IU/kg). A total of 216 Lohmann laying hens (50 wk of age) were randomly allocated into six treatment diets for 12 wk. Each treatment had 6 replicates of 6 hens per replicate. Results: The results show that aged corn significantly decreased the content of low-density lipoprotein cholesterol (p<0.05), and reduced chemokine-like receptor 1 (CMKLR1) mRNA expression (p<0.05) in the liver compared to controls. Diet with VE did not alter the content of crude fat and cholesterol (p>0.05), or acetyl-CoA carboxylase, lipoprotein lipase, fatty acid synthase or CMKLR1 mRNA expression (p>0.05) in the liver among treatment groups. Aged corn significantly increased the content of malondialdehyde (MDA) (p<0.05) and decreased superoxide dismutase (SOD) activity (p<0.05) in the liver. The VE increased the content of MDA (p<0.05) but decreased glutathione peroxidase (GSH-Px) activity in serum (p<0.01) and in the ovaries (p<0.05). Adding VE at 20 and 100 IU/kg significantly increased GSH-Px activity (p<0.05) in liver and in serum (p<0.01), 100 IU/kg VE significantly increased SOD activity (p<0.05) in serum. Aged corn had no significant effects on GSH-Px mRNA or SOD mRNA expression (p<0.01) in the liver and ovaries. Addition of 100 IU/kg VE could significantly increase SOD mRNA expression (p<0.01) in the liver and ovary. Conclusion: Aged corn affected lipid metabolism and decreased the antioxidant function of laying hens. Dietary VE supplementation was unable to counteract the negative effects of aged corn on lipid metabolism. However, addition of 100 IU/kg VE prevented aged corninduced lipid peroxidation in the organs of laying hens.

        Full-scale testing and modeling of the mechanical behavior of shield TBM tunnel joints

        For shield TBM (Tunnel Boring Machine) tunnel lining, the segment joint is the most critical component for determining the mechanical response of the complete lining ring. To investigate the mechanical behavior of the segment joint in a water conveyance tunnel, which is different from the vehicle tunnel because of the external loads and the high internal water pressure during the tunnel's service life, full-scale joint tests were conducted. The main advantage of the joint tests over previous ones was the definiteness of the loads applied to the joints using a unique testing facility and the acquisition of the mechanical behavior of actual joints. Furthermore, based on the test results and the theoretical analysis, a mechanical model of segment joints has been proposed, which consists of all important influencing factors, including the elastic-plastic behavior of concrete, the pre-tightening force of the bolts and the deformations of all joint components, i.e., concrete blocks, bolts and cast iron panels. Finally, the proposed mechanical model of segment joints has been verified by the aforementioned full-scale joint tests.

        Experimental and modeling study of viscoelastic behaviors of magneto-rheological shear thickening fluids

        Nowadays, both Magneto-rheological Fluid (MRF) and Shear Thickening Fluid (STF) have separately attracted considerable interest due to the fast reversible response to either external magnetic field or abrupt shearing loading. In this paper, we fabricated a combined phase of Magneto-rheological Shear Thickening Fluid (MRSTF), where the 25 wt% STF is applied as medium phase with the addition of varied fractions of iron particle. The investigation of the dynamic behavior of this novel material under oscillatory shear was launched in a parallel-plate rheometer. The relevance of the dynamic behavior to strain amplitude, frequency and external magnetic field were investigated and discussed. A four-parameter viscoelastic model was applied to reconstruct the mechanical behavior of the MRSTF under different working conditions, and the parameters were identified within the Matlab optimization algorithm. The comparison between the experimental data and the model prediction results indicated that the four-parameter model could predict viscoelastic material with desired accuracy. The MRSTF exhibits features of both components, while prone more to MRF with the inception of external field excitations.

        Numerical investigation of tip clearance effects on the performance of ducted propeller

        Tip clearance loss is a limitation of the improvement of turbomachine performance. Previous studies show the Tip clearance loss is generated by the leakage flow through the tip clearance, and is roughly linearly proportional to the gap size. This study investigates the tip clearance effects on the performance of ducted propeller. The investigation was carried out by solving the Navier-Stokes equations with the commercial Computational Fluid Dynamic (CFD) code CFX14.5. These simulations were carried out to determine the underlying mechanisms of the tip clearance effects. The calculations were performed at three different chosen advance ratios. Simulation results showed that the tip loss slope was not linearly at high advance due to the reversed pressure at the leading edge. Three type of vortical structures were observed in the tip clearance at different clearance size.

        Chicken serum uric acid level is regulated by glucose transporter 9

        Objective: Glucose transporter 9 (GLUT9) is a uric acid transporter that is associated with uric absorption in mice and humans; but it is unknown whether GLUT9 involves in chicken uric acid regulation. This experiment aimed to investigate the chicken GLUT9 expression and serum uric acid (SUA) level. Methods: Sixty chickens were divided into 4 groups (n = 15): a control group (NC); a sulfonamide-treated group (SD) supplemented with sulfamonomethoxine sodium via drinking water (8 mg/L); a fishmeal group (FM) supplemented with 16% fishmeal in diet; and a uric acid-injection group (IU), where uric acid (250 mg/kg) was intraperitoneally injected once a day. The serum was collected weekly to detect the SUA level. Liver, kidney, jejunum, and ileum tissues were collected to detect the GLUT9 mRNA and protein expression. Results: The results showed in the SD and IU groups, the SUA level increased and GLUT9 expression increased in the liver, but decreased in the kidney, jejunum, and ileum. In the FM group, the SUA level decreased slightly and GLUT9 expression increased in the kidney, but decreased in the liver, jejunum, and ileum. Correlation analysis revealed that liver GLUT9 expression correlated positively, and renal GLUT9 expression correlated negatively with the SUA level. Conclusion: These results demonstrate that there may be a feedback regulation of GLUT9 in the chicken liver and kidney to maintain the SUA balance; however, the underlying mechanism needs to be investigated in future studies.
