      • Effects of antioxidants on the neuronal cell death

        증추신경흥분꼭로 낭용피고 있는 methamphetamine(METH)은 doparnine 신경세포에서 dopandne유리를 증가시킴으로서 흥분작움, 약물의존성 등의 효과를 나타낼 뿐 아니라, dopamine 신경계에 작용하여 세포손상을 유도한다. dopamine 신경세포주인 CATH.a 세포배양을 이용한 지난해의 in uifro 연구결과 MEH에 의한 신경세포사는 apoptosis에 의한 것으로 산화성 스트레스가 관여하며, MEHfl의해 세폰밖으로 유리틴 dopamine의 산화에 의한 것으로 확인되었다. 본 실험에서는 항산화 효능을 가진 것으로 알려져 있는 물질 중 인삼의 성븐 중 하나인 ginsenos펀e Rbl, 록차의 성분인 opjgallocatechin g패ate 및 selenium, aminoguanidine과 JV-acetyl cysteine이 METH과 6-hy강oxydopamine(6-·OHDA)에 의한 신경세포독성에 보호효과를 가지는지 여부에 대하여 살펴보았다. ginsenoside Rbl의 전처치레 의하여 6-0딘DA 및 METH 투여에 의한 도파민 신경세포손상이 유의성 있게 감소되었듬을 착인하고, 녹차의 성분 중 하나인 (-)eP구allocatechin 토allate 처리에 의하여 금-OHDA에 의한 도_파민 신경세포손상이 통계적으로 유의성 있게 감소되었음을 착인하였다. iNO즈 inhibitor인 aminoguanidine이 UfETH 투여에 의한 신경세포손상을 감소시킨 결과를 얻어, METH에 의한 신경세포독성을 」:1해하는 ginsenoside Rbl의 효과가 nitric oxide 기전과 관련이 있는지 여부를 살펴럴았다. 그러나 끄nsenoside Rbl 투여가 UfETH에 의하여 증가되는 NO 유리 및 iNO를 발현 증가를 억제하지는 않f=것으로 관찰되었다. 이러한 결과는, 도파민신경세포에 대한 MEH 및 6-OHDA의 손상효과를 확인하고, finsenoside Rbl, catechins, selenium 등 천연물 유래성분과 aminoguanidine,rf-acetylcystelne이 이들 신경세포손상에 미치는 영향을 살펴보고 그 기전을 검토함으로써 도파민신경 기능장애 및 보호 기전연구의 기초자료를 제공할 수 있을 것으로 사료된다. The methamphetamine(METH) Is commonly abused for its rewarding effects, which are thought to result from dopamine release in the nucleus accumbens, and is amonoaminergic toxi(:ant that destroys dopamine terminals in uivo. Previous study from ourlaboratory showed dopamine-dependent oxidative stress was the initial event of METH-inducedneurotoxicity, by using an in uifro model of immortaEzed neuronal cetl tine, CATH.a(CNSadrenergic tyrosine hydroxylase(THf expressing). In the present study, effects of possibleantioxidants, ginsenoside Rbl, epigallocatechin 2allate, selenillm, ardnoguanidine and N-acetylcysteine were exarfed whether they have protective effort from neurotoxicity induced byafETH or 6-OHDA. Pretreatment of ginsenoside Rbl significantly reduced neurotoxicity inducedby METH and 6-OHDA. Epigallocatechin gallate also sigaificantly reduced 6-OHDA inducedneurotoldcy, whereas it did not show protective effect from TvfETH-induced toxicity in CATH.acelT- Pretreatment of aminoguanidine, a specific iNOS inhibitor, decreased cytotoxicity inducedby METH. Ginsenoside Rbl, however, Od not show any difference in METH-induced increasein iNOS nor it did affect nftric oxide release. Aminoguanidine itself also did not affect ftricoxide release nor a10s induction induced by afETH. These results suggest possible role ofginsenoside Rbl..3hd epigallocatechin gallate in protectiHg dopandnergic neurotoxicity induced byMETH or 6-OHDA and that more investigations should be done to claffy the involvement ofnitric oxide as a possible mechanism of ginsenoside Rbl against bfETH-induced nevrotoxirity.

        새로운 물질이 개발되어 신약으로 탄생하기까지 많은 시간과 노력, 예산이 필요하다. 최근, 우리 나라에서도, 신약캐발이 어릴고 힘든 분야이지만 새로운 신약개발을 총해 얻어지는 분가가치가 막대함을 인식, 연구에 박차를 가하 있는 실정이다. 본 연구는, 의약품 개발 선진국인 미국 FDA에서 발행하는 참고자료 중 항균제 임상편가에 대한 가이드라인 및 주요 임상적응증별 항균제 기발(지역사획회득성 폐렴, 원내감염성 계렴, 급성기관지염의 2차세균감염, 만성기지염의 급성세균성 악화, 연쇄구균에 의한 인두영 및 편도염 , 단순성 요로감염증, 복합성 요로감염증 및 신우신영, 급성 세균성 부비동옆, 항 바이러스제 개발시 전임상단계에서 고려사항, 항 바이러스제 허가와 된련 잉상적 고려사항, 카레타 괸련 혈류감염)에 있어서의 파이드라인 등의 자료를 소개함으로져, 향후 제약사의 항균제 신약개발 및 허가 신청된 의악품의 전임상차료 및 잉상시험자료의 검토 평가업무에 참고자료로 활용될 수 있을 것이다. Changing or unclear interpretations of clinical trial data needed to demonstrate the effectiveness and safety of antimicrobial drug products have a times led to confusion and frustration on the part of both applicants and division of these drug products reviewers. The FDA published guidance on desing clinical protocols, implementing, and analyzing data from clinical trials for antimicrobial drug products has been presented and additional companion guidances introduced specific issues to individual infections. This document provides applicants and reviewers with minimal information appropriate for the clinical development of routine antimicrobial drug products and identifies issues common to many antimicrobial drug applications. The agency can use the kind of information to determine whether the antimicrobial drug product under study is safe and effective in the treatment of the specific infection studied.

      • DNA-Base Guanine as Hydrogen Getter and Charge-Trapping Layer Embedded in Oxide Dielectrics for Inorganic and Organic Field-Effect Transistors

        <P>DNA-base small molecules of guanine, cytosine, adenine, and thymine construct the DNA double helix structure with hydrogen bonding, and they possess such a variety of intrinsic benefits as natural plentitude, biodegradability, biofunctionality, low cost, and low toxicity. On the basis of these advantages, here, we report on unprecedented useful applications of guanine layer as hydrogen getter and charge trapping layer when it is embedded into a dielectric oxide of n-channel inorganic InGaZnO and p-channel organic heptazole field effect transistors (FETs). The embedded guanine layer much improved the gate stability of inorganic FETs gettering many hydrogen atoms in the gate dielectric layer of FET, and it also played as charge trapping layer to which the voltage pulse-driven charges might be injected from channel, resulting in a threshold voltage (<I>V</I><SUB>th</SUB>) shift of FETs. Such shift state is very ambient-stable and almost irrevocable even under a high electric-field at room temperature. So, Boolean logics are nicely demonstrated by using our FETs with the guanine-embedded dielectric.       • Electron transport mechanism of bathocuproine exciton blocking layer in organic photovoltaics

        <P>Efficient exciton management is a key issue to improve the power conversion efficiency of organic photovoltaics (OPVs). It is well known that the insertion of an exciton blocking layer (ExBL) having a large band gap promotes the efficient dissociation of photogenerated excitons at the donor-acceptor interface. However, the large band gap induces an energy barrier which disrupts the charge transport. Therefore, building an adequate strategy based on the knowledge of the true charge transport mechanism is necessary. In this study, the true electron transport mechanism of a bathocuproine (BCP) ExBL in OPVs is comprehensively investigated by in situ ultraviolet photoemission spectroscopy, inverse photoemission spectroscopy, density functional theory calculation, and impedance spectroscopy. The chemical interaction between deposited Al and BCP induces new states within the band gap of BCP, so that electrons can transport through these new energy levels. Localized trap states are also formed upon the Al-BCP interaction. The activation energy of these traps is estimated with temperature-dependent conductance measurements to be 0.20 eV. The Al-BCP interaction induces both transport and trap levels in the energy gap of BCP and their interplay results in the electron transport observed.</P>

      • Primary Cecal Diffuse Large B-Cell Lymphoma Presenting as a Subepithelial Tumor

        We report on a unique case of diffuse large B-cell lymphoma of the cecum, which presented as a subepithelial tumor. A 62- year-old woman with abdominal pain underwent a colonoscopy, resulting in detection of a subepithelial tumor measuring 2 cm in the cecum. We performed a right hemicolectomy with diagnostic and curative intent; histological examination and immunohistochemical staining of the resected tumor indicated a diagnosis of diffuse large B-cell lymphoma. The patient achieved complete remission with combination chemotherapy.

      • 혈액 투석을 받는 두경부암 환자의 동시화학방사선요법에서 Cisplatin의 약력학 조사 1예

        목 적 :투석을 받고 있는 진행된 두경부암 환자에서 Cisplatin동시 화학방사선 요법의 타당성과 약물역동학에 대해 연구 하였다. 방 법 :57세 말기 신장병을 앓고 있던 남자환자가 임상 병기 3기의 외이도 암을 진단 받고 종양 완전 절제술을 시행하였다. 수술 후 6개월에 환자는 국소 부위 재발하였다. 절단술과 술후 5200cGy, 26fx의 방사선 치료 시행에도 불구하고 다시 국소 부위 재발 보여 3주기동안 Cisplatin 기반의 방사선 화학 병합 요법 치료를 하였다. 총 3주간 Cisplatin 20 mg/m 2 를 투여했으며 화학 요법 1일째 30분 동안 Cispla-tin을 정주하고 30분 후 혈액투석을 4시간 동안 시행하였다. 화학요법 3일째와 5일째에도 혈액투석을 시행하였다. 환자의 혈장 시료는 Cisplatin 정주 후 특정한 시간에 채취되었다. 결 과 :Cisplatin 동시 화학방사선요법 3차 시행 후 환자의 종 괴 크기는 현저하게 감소하였다. Cisplatin의 최대 혈장 농도는 총 platinum은 0.74μg/ml였고 free platinum은 0.37μg/ml로 이었다. AUC 값은 총 platinum은 94.7μg·h/ml, free platinum은 11.3 μg·h/ml이었다. 결 론 :투석을 받는 진행된 두경부암 환자에서 Cisplatin 동시 화학방사선요법을 시행한 증례를 보고하며 이들 환자에서 정용량의 Cisplatin 동시 화학방사선요법이 감내할 만하다고 제안한다. Objectives :We study the feasibility and pharmacokinetics of cisplatin concurrent chemoradiation for ad-vanced head and neck cancer patient undergoing hemodialysis. Materials and Methods :A 57-year old male with end stage renal disease developed stage III external auditory canal cancer. Complete resection surgery was done. Postoperative 6 months, local recurrence was occurred. De-spite excision and adjuvant radiotherapy, local tumor was recurred. We decided to treat a cisplatin concurrent chemoradiotherapy. Cisplatin was administered at a dose of 20mg/m 2 for 30 min. Hemodialysis was started 30 min after completion of the cisplatin infusion and performed for 4 hours. Hemodialysis was performed on day 3 and 5 of chemotherapy. Plasma samples were collected at specified times after administration of cisplatin. Result :At the end of the third cycle of cisplatin concurrent chemoradiotherapy, the tumor size was mark-edly decreased. The maximum plasma concentrations of plasma platinum and free platinum were 0.74 and 0.37μg/ml respectively. The area under the curve of plasma platinum and free platinum were 94.7 and 11.3μg·h/ml respectively. Conclusion :We report a case of Cisplatin concurrent chemoradiation for hemodialysis patient with advanced head and neck cancer and suggest full dose cisplatin concurrent chemoradiotherpay is tolerable for these patients
