      • Novel Methylation Biomarker for Non-invasive Diagnostics in Lung Cancer

        To identify aberrantly hypermethylated DNA in lung cancer cells we established a genome-wide analysis for hypermethylation sites, namely Methyl DNA Isolation and Amplification (MeDIA) coupled-CpG microarray analysis. In the comprehensive methyaltion profiling analysis between human lung cancer, A549 cells and normal NHBE cells, we observed that several clusters of genes show a significant level of aberrancy in CpG island methylation pattern in cancer cells compared to normal cells. We further identified PCDHGA12 gene as a new marker of non-invasive diagnostics for lung cancer based on followings. 1) Transcription of PCDHGA12 gene is reactivated after treatment of A549 cells with demethylating agent. 2) Bisulfide clonal-sequencing reveals that CpG island of PCDHGA12 shows a distinctive differential methylation between two cell lines. 3) Pyrosequencing-based quantitative methylation assay for such region in tumor and non-tumorous tissues from lung cancer patients shows aberrant hypermethylation in 37 (92%) of the 40 tumor tissues. In clinical validation by pyrosequencingin induced-sputum of lung cancer patients (n=87) and healthy controls (n=51), we observed aberrant hypermethylation incident at significantly elevated level in samples derived from lung cancer patients. According to the optimal threshold calculated by ROC curve analysis, sensitivity and specificity of PCDHGA12 was 86.2% and 82.4%, respectively. PCDHGA12 methylation status could be a potential methylation biomarker alone or combined with others for the screen and the detection of relapse of lung cancer.

      • Al_(0.3)Ga_(0.7)As/GaAs HEMT 구조에서의 Photoreflectance에 관한 연구

        The optical characteristics of Al_(0.3)Ga_(0.7)As/GaAs HEMT structure were investigated by using the photoreflectance method. From the PR measurement, 2DEG transition energy related signal were obtained by between GaAs buffer and Al_(0.3)Ga_(0.7)As/GaAs spacer layer at the temperature of below about 150 K. And the transition energy is shift to low energy about 21 meV. We obtained that 2DEG transition energy, interface field and carrier concentration are 1.455 eV, 5.26 × 10⁴ V/cm and 7.18 × 10⁴cm^(-3), respectively.

        낙동강 물 관개논의 농업환경 특성과 질소, 인산 요구량

        낙동강 물을 관개하여 벼를 재배하는 강 상류의 안동 풍산에서부터 하류의 부산 구포까지 7개 지역 논토양의 농업환경 특성을 1999년부터 2년간 비교 분석하였다. 토양의 중금속 함량은 자연함량 안팎이었으며 가장 높았던 곳은 부산 강서 지역이었고 다음으로는 고령 성산 지역이었다. 그러나 현미중의 중금속은 자연함량 이하를 보였다. 토양의 화학성은 고령 성산지역과 부산 강서지역에서 높았는데, 1999~2000년도의 영농 후 토양에서 고령은 pH (5.9~6.1), EC(0.8~0.9 dS/m), 유효인산(155~201 mg/kg), 치환성 칼슘(6.7~7.4 cmol^(+)//kg), 치환성 마그네슘(1.92~2.50 cmol^(+)/kg), 치환성 칼리(0.18~0.21 cmol^(+)//kg )가 가장 높았고 부산에서는 유기물(23.0~29.1 g/kg), 총질소(1.6~l.8 mg/kg)가 가장 높았다. 낙동강 물의 관개수질과 토양화학성을 고러한 벼의 진단시비량은 질소의 경우 110 kg/ha를 기준으로 안동 21.4%, 상주 11.85, 구미지역에서 8.8% 정도 증시할 필요가 있었으나, 고령에서는 14.9%, 창녕은 4.6%, 밀양 하남은 4.55, 부산 강서지역은 11.5% 내외로 줄일 수 있을 것으로 분석되었다. 그리고 인산질 비료는 45 kg/ha 기준으로 창녕 도천에서만 18.9% 줄여 주고 다른 지역에서는 기본시비량인 30 kg/ha만 시용하여도 무방할 것으로 판단되었다. 논토양에 분포하는 세균과 사상균, 방선균, 중온성 Bacillus, 형광성 Pseudomonas, Biomass C는 유기물과 총 질소, 인산 등의 양분함유량이 많았던 고령 성산과 그 하류지역에서 높았다. Agroenvironmental characteristics in paddy fields irrigated with the water of Nagdong river were analyzed along the river watershed for two years from 1999. The sites monitored from upper reaches of the river were Andong, Sangju, Gumi, Goryeong, Changnyeong, Milyang and Pusan. In paddy soils, the contents of heavy metals such as Cd, Cr, Cu, Pb, Ni, Zn and As were around natural values showing the highest values in Pusan followed by Goryeong. In brown rice, the contents of heavy metals were lower than natural values. Soil chemical properties appeared higher values in the lower reaches including Goryeong than the upper ones. The highest parameters in Goryeong were pH (5.9~6.1), EC (0.8~0.9 dS/m), Av.P₂O_(5) (155~201 mg/kg), exchangeable Ca (6.7~7.4 cmol^(+)/kg), Mg (1.92~2.50 cmol^(+)/kg), K (0.18~0.21 cmol^(+)/kg) and those in Pusan were organic matter (23.0~29.1 g/kg) and T-N (1.6~1.8 mg/kg). In conclusion, the recommended rates of N fertilizer for rice cropping were 21.4%, 11.8% and 8.8% high for Andong, Sangju and Gumi, respectively and 14.9%, 4.6%, 4.5% and 11.5% low for Goryeong, Changnyeong, Milyang and Pusan, respectively reflecting the chemical properties of soils and the quality of irrigation water on the basis of 110 kgN/ha. In the case of phosphorous, the rates were 18.9% and 33.3% low for Changnyeong and others, respectively on the basis of 45 kgP₂O_(5)/ha. The populations of bacteria, fungi, actinomycetes, Bacillus, fluorescent Pseudomonas and Biomass C were high at the lower reaches including Goryeong, which showed relatively much nutrient contents of organic matter, total N and phosphorous etc..

        Pollen Allergic Risk Assessment of Genetically Modified Virus Resistant Pepper and Functional Chinese Cabbage

        Pollen of genetically modified (GM) pepper containing the gene for cucumber mosaic virus (CMV) coat protein (CP) and GM Chinese cabbage with high phenylethylisothiocyanate (PEITC) content was investigated for assessment of allergic risk. Amino acid (AA) sequences of the inserted gene products of GM virus resistant pepper and GM Chinese cabbage with high PEITC content were compared with those of known allergens. No known allergen greater than 35% AA sequence homology, over 80 AA window or more than 8 consecutive identical AA was found. Protein patterns of GM/non-GM pepper and Chinese cabbage pollen extracts in SDS-PAGE analysis showed the same distribution of protein bands among the GM and non-GM pepper or Chinese cabbage, respectively. Sera from pollen allergic patients showed some IgE reactivity via immunoblotting and ELISA; however, no differences were observed between the pollen of GM and non-GM pepper or Chinese cabbage, respectively. Based on these results, we conclude that pollens of the virus resistant GM pepper and GM Chinese cabbage with high PEITC have no differences in their protein composition or allergenicity relative to non-GM pepper and Chinese cabbage.

        The Therapeutic Effect of Human Embryonic Stem Cell-Derived Multipotent Mesenchymal Stem Cells on Chemical-Induced Cystitis in Rats

        <P><B>Purpose</B></P><P>To evaluate the therapeutic effect of human embryonic stem cell (hESC)-derived multipotent mesenchymal stem cells (M-MSCs) on ketamine-induced cystitis (KC) in rats.</P><P><B>Methods</B></P><P>To induce KC, 10-week-old female rats were injected with 25-mg/kg ketamine hydrochloride twice weekly for 12 weeks. In the sham group, phosphate buffered saline (PBS) was injected instead of ketamine. One week after the final injection of ketamine, the indicated doses (0.25, 0.5, and 1×10<SUP>6</SUP> cells) of M-MSCs (KC+M-MSC group) or PBS vehicle (KC group) were directly injected into the bladder wall. One week after M-MSC injection, the therapeutic outcomes were evaluated via cystometry, histological analyses, and measurement of gene expression. Next, we compared the efficacy of M-MSCs at a low dose (1×10<SUP>5</SUP> cells) to that of an identical dose of adult bone marrow (BM)-derived MSCs.</P><P><B>Results</B></P><P>Rats in the KC group exhibited increased voiding frequency and reduced bladder capacity compared to rats of the sham group. However, these parameters recovered after transplantation of M-MSCs at all doses tested. KC bladders exhibited markedly increased mast cell infiltration, apoptosis, and tissue fibrosis. Administration of M-MSCs significantly reversed these characteristic histological alterations. Gene expression analyses indicated that several genes associated with tissue fibrosis were markedly upregulated in KC bladders. However the expression of these genes was significantly suppressed by the administration of M-MSCs. Importantly, M-MSCs ameliorated bladder deterioration in KC rats after injection of a low dose (1×10<SUP>5</SUP>) of cells, at which point BM-derived MSCs did not substantially improve bladder function.</P><P><B>Conclusions</B></P><P>This study demonstrates for the first time the therapeutic efficacy of hESC-derived M-MSCs on KC in rats. M-MSCs restored bladder function more effectively than did BM-derived MSCs, protecting against abnormal changes including mast cell infiltration, apoptosis and fibrotic damage.</P>

        Radiological Characteristics of Peritumoral Edema in Meningiomas

        Objective: The purpose of this study is to evaluate the radiological charactersitics related to the formation of peritumoral edema in meningiomas. Methods: Fifty patients with meningioma were examined by magnetic resonance images and cerebral angiography. The predictive factors associated peritumoral edema, such as, tumor size, peritumoral rim (cerebrospinal fluid cleft), shape of tumor margin, signal intensity of tumor in T2WI, and pial blood supply were evaluated. Results: Tumor size, peritumoral rim and pial blood supply correlated with peritumoral edema on univariate analyses. But in multivariate analyses, pial blood supply was statistically significant as a factor for peritumoral edema in meningioma. Conclusion: In our results, pial blood supply is significant contributing factor for peritumoral edema in meningioma.

        All medicines are poisonous, and there are none free from poison. Thus, all things may be regarded as poisonous. And poison and medicine is the same one of which actions differ from each other. Medicine may be good medicine or poison according to doses, whether chemical medicine or herbal one. Herbal medicine falls into the category of natural materials. From ancient times humankind has Been using animals and plants as food, but on the other hand, chemical medicine is unfamiliar relatively to natural materials. Somecine is unfamilis, absorbed into the human boan, are transformed and help deemicfication. Other medicines protect the gastric mucous membrane and are demulcent, and inhibithemica materials from being absorbed. In some cases, ine is unfamilis function as deemicfiers in combination withhemica materials, and remove the toxicity and side effect caused by drug properties. Herbal medicine causes less side effects, as compared to chemical one, and thus can work effectively. As the case stands, some deem that herbal medicine does not cause problems despite high dose and long-term use because it is nontoxic or low-toxic. However, herbal medicine may be also poisonous though it is used at a very low dose. Even a deadly poison may produce therapeutic results satisfactorily without side effects, on condition that it is used properly.

        Epidemiological and Clinical Characteristics of Hepatitis C Virus Infection in South Korea from 2007 to 2017: A Prospective Multicenter Cohort Study

        Background/Aims: This study aimed to elucidate the epidemiological and clinical characteristics of chronic hepatitis C (CHC) patients in South Korea from 2007 to 2017 and to compare the treatment patterns between two periods before and after the first approval of direct-acting antivirals (DAA) in South Korea in 2015. Methods: This prospective, multicenter cohort enrolled 2,758 patients with hepatitis C virus (HCV) viremia at seven tertiary centers, and clinical data were prospectively collected with questionnaire surveys focused on lifetime risk factors related to HCV infection. Results: The HCV patients had a mean age of 57.3 years (50.8% male). Among them, 14.3% showed a positive history of transfusion before HCV screening and 5.6% reported intravenous drug use (IVDU), with significant differences in these risk factors between men and women. The proportions of patients with chronic hepatitis, liver cirrhosis and hepatocellular carcinoma (HCC) were 69.5%, 18.9%, and 11.5%, respectively. The mean alanine aminotransaminase level was within the upper normal limit at 49.9%, and the major genotypes were 1b (48.2%) and 2 (46.4%). The overall treatment rate was 53.8%, showing a rapid transition from interferon-based therapy to DAA therapy. In the post-DAA-approval era, the untreated group was older, had a higher prevalence of HCC, and had less education than the treated group. Conclusions: More than 90% of CHC patients were over 40 years old, the major genotypes were 1b and 2, and IVDU was observed in less than 6% of CHC patients. Approximately half of the patients underwent antiviral therapy even in the DAA era, showing an unmet need with regard to HCV elimination.
