The Role of Kif4A in Doxorubicin-Induced Apoptosis in Breast Cancer Cells

This study was to investigate the mechanism and role of Kif4A in doxorubicin-induced apoptosis in breast cancer. Using two human breast cancer cell lines MCF-7 (with wild-type p53) and MDA-MB-231 (with mutant p53), we quantitated the expression levels of kinesin super-family protein 4A (Kif4A) and poly (ADP-ribose) Polymerase-1 (PARP-1) by Western blot after doxorubicin treatment and examined the apoptosis by flow cytometry after treatment with doxorubicin and PARP-1 inhibitor, 3-Aminobenzamide (3-ABA). Our results showed that doxorubicin treatment could induce the apoptosis of MCF-7 and MDA-MB-231 cells, the down-regulation of Kif4A and upregulation of poly(ADP-ribose) (PAR). The activity of PARP-1 or PARP-1 activation was significantly elevated by doxorubicin treatment in dose- and time-dependent manners (P < 0.05), while doxorubicin treatment only slightly elevated the level of cleaved fragments of PARP-1 (P > 0.05). We further demonstrated that overexpression of Kif4A could reduce the level of PAR and significantly increase apoptosis. The effect of doxorubicin on apoptosis was more profound in MCF-7 cells compared with MDA-MB-231 cells (P < 0.05). Taken together, our results suggest that the novel role of Kif4A in doxorubicin-induced apoptosis in breast cancer cells is achieved by inhibiting the activity of PARP-1.
Seismic performance of Bujian Puzuo considering scale ratio and vertical load effects

This study investigated the influence of scale ratio and vertical load on the seismic performance of Puzuo joints in traditional Chinese timber structures. Three low-cyclic reversed loading tests were conducted on three scaled specimens of Bujian Puzuo in Yingxian Wooden Pagoda. This study focused on the deformation patterns and analyzed seismic performance under varying scale ratios and vertical loads. The results indicated that the slip and rotational deformations of Bujian Puzuo were the primary deformations. The scale of the specimen did not affect the layer where the maximum interlayer slip occurred, but it did decrease the proportion of slip deformation. Conversely, the reducing vertical load caused the layer with the maximum slippage and the position of the damaged Dou components to shift upward, and the proportion of slip deformation increased. When the vertical load was decreased by 3.7 times, the maximum horizontal bearing capacity under positive and negative loadings, initial stiffness, and energy dissipation of the specimen decreased by approximately 60%, 58.79%, 69.62%, and 57.93%, respectively. The horizontal bearing capacity under positive loading and energy dissipation of the specimen increased by 35.63% and 131.54%, when the specimen scale was doubled and the vertical load was increased by 15 times.

In this paper, decentralized robust H∞ output feedback control problem for large-scale interconnected system with value bounded uncertainties in the state, control input and interconnection matrices is investigated. A new bounded real lemma for the large-scale interconnected systems is derived by Lyapunov stability theory and linear matrix inequality method. Based on the new bounded real lemma, a sufficient condition expressed as matrix inequalities for the existence of a decentralized robust H∞ output feedback controller is obtained. The controller which enables the closed-loop large-scale system robust stable and satisfies the given H∞ performance is designed through a homotopy iterative method. Finally, a numerical example is given to illustrate the effectiveness of the proposed method.

In this paper, the problem of delay-dependent stabilization for singular systems with multiple internal and external incommensurate constant point delays is investigated. The condition when a singular system subject to point delays is regular independent of time delays is given and it can be easily test with numerical or algebraic methods. Based on Lyapunov-Krasovskii functional approach and the descriptor integral-inequality lemma, a sufficient condition for delay-dependent stability is obtained. The main idea is to design multiple memory less state feedback control laws such that the resulting closed-loop system is regular independent of time delays, impulse free, and asymptotically stable via solving a strict linear matrix inequality (LMI) problem. An explicit expression for the desired memory less state feedback control laws is also given. Finally, a numerical example illustrates the effectiveness and the availability for the proposed method.
Proposals for flexural capacity prediction method of externally prestressed concrete beam

Flexural capacity prediction is a challenging problem for externally prestressed concrete beams (EPCBs) due to the unbonded phenomenon between the concrete beam and external tendons. Many prediction equations have been provided in previous research but typically ignored the differences in deformation mode between internal and external unbonded tendons. The availability of these equations for EPCBs is controversial due to the inconsistent deformation modes and ignored secondorder effects. In this study, the deformation characteristics and collapse mechanism of EPCB are carefully considered, and the ultimate deflected shape curves are derived based on the simplified curvature distribution. With the compatible relation between external tendons and the concrete beam, the equations of tendon elongation and eccentricity loss at ultimate states are derived, and the geometric interpretation is clearly presented. Combined with the sectional equilibrium equations, a rational and simplified flexural capacity prediction method for EPCBs is proposed. The key parameter, plastic hinge length, is emphatically discussed and determined by the sensitivity analysis of 324 FE analysis results. With 94 collected laboratory-tested results, the effectiveness of the proposed method is confirmed, and comparisons with the previous formulas are made. The results show the better prediction accuracy of the proposed method for both stress increments and flexural capacity of EPCBs and the main reasons are discussed.

The clearance of activated hepatic stellate cells (HSCs) by apoptosis is critical for the reversibility of hepatic fibrosis. Mitochondrial homeostasis is regulated by mitophagy, which is an efficient way of clearing injured mitochondria that plays an important role in apoptosis. However, the role of mitophagy in apoptosis in HSCs and hepatic fibrosis is still unclear. Here, we show that mitophagy is enhanced in parallel with increased apoptosis in hepatic stellate cells during the reversal of hepatic fibrosis. The inhibition of mitophagy suppressed apoptosis in HSCs and aggravated hepatic fibrosis in mice. In contrast, the activation of mitophagy induced apoptosis in HSCs. Furthermore, we confirmed that BCL-B, which is a member of the BCL-2 family, is a regulator mediating mitophagy-related apoptosis. The knockdown of BCL-B resulted in increased apoptosis and mitophagy in HSCs, while the overexpression of BCL-B caused the opposite effects. BCL-B inhibited the phosphorylation of Parkin (a key regulator of mitophagy) and directly bound phospho-Parkin. Altogether, enhanced mitophagy promotes apoptosis in HSCs during the reversal of hepatic fibrosis. BCL-B suppresses mitophagy in HSCs by binding and suppressing phospho-Parkin, thereby inhibiting apoptosis. Kaiso is a Pox Virus and Zinc Finger (POZ-ZF) transcription factor with bi-modal DNA-binding specificity. Here, we demonstrated that Kaiso expression is inversely correlated with glucocorticoid receptor (GR) expression in breast carcinomas. Knockdown of Kaiso increased GR expression, while overexpression of Kaiso inhibited GR expression in breast cancer cells. Furthermore, Kaiso repressed GR proximal promoter-reporter activity in a dose-dependent manner. Remarkably, ChIP experiments demonstrated that endogenous Kaiso was associated with the GR promoter sequence in a methylation-dependent manner. Since glucocorticoids inhibit chemotherapyinduced apoptosis and have been widely used as a co-treatment of patients with breast cancer, we assessed the role of Kasio in GR-mediated anti-apoptotic effects. We found that overexpression of Kaiso attenuated the anti-apoptotic effects of glucocorticoids in breast cancer cells. Our findings suggest that GR is a putative target gene of Kaiso and suggest Kaiso to be a potential therapeutic target in GC-combination chemotherapy in breast cancer.
