        The purpose of this research is to analyze relationships among the habitual life, stress, and constipation and to help prevent and improve the constipation among women's university students. This is a descriptive interrelated research that has targeted women's university students. The data had been collected for 10 days, from December 22, 2003 to December 31, 2003. The survey was carried out to the 295 women's university students that had been randomly selected by visiting four-year-course colleges and universities in Seoul. The survey consists of 78 questions: five questions on the general trait, 11 questions on the habitual life, 51 questions on the stress, and 11 questions on the constipation. There are seven questions on the dietary life, four questions on exercising, and Shin Jung Ran's survey(2003) and Lee Ji Jung's survey(2001) was used as a reference. Yoon Ho Yul's five grade stress barometer(1996) was also used to measure the amount of stress. The collected data was analyzed by a real number, percentage, the arithmetical mean, the standard deviation, t-test, x²test using SPSS 10.00 for Windows Program. Results are as follow 1. Four to six cups of daily water intake was the highest by 50.0%. Students not have breakfast(55.7%) mostly because they did not have enough time(62.6%). Dinner was the most overeated meal by 72.3% and the most preferred food was meat by 42.0%. In the questions related to exercising, more than half of the students answered that they hardly do not exercise(58%), work out for less than 30minutes(67.3%), and exercised lightly(75.3%). 2. Students were suffering from stresses due to the personal relations(51.36±14.0426), scholastic achievement and personal direction(24.93±8.3037), financial conditions and family matters(20.28±8.1718) and social circumstances(2.7262). 3. In the defection part, the student's reply that they have more than five times of bowel movement on a weekly basis(48.3%), and that it takes three to five minutes to defecate(41.3%) were the highest. More than half of the students answered that their one time excrement quantity was between 30 grams to 60 grams(72.3%) and that it was normal(68.7%). In a question asking how it feels after the defecation(students were allowed to choose more than one answer), relieved and refreshed was the highest by 66.6%, an uncomfortable followed by 42.8%. 4.4% of the students answered that they suffer from strong abdominal pain during the defecation. The rest of the students answered that they do not have stomachache or that it was normal. The students were classified into the constipation group of 103 people(34.9%) and the normal group of 192 people(65.1%) under the devised definition of this research. 4. Students that live in dormitories and do their own cooking were more likely to have constipation than those that live in private residence or in a lodgins. 5. The research showed that there is a mutual relation between water in a day intake and the constipation. It means that the lesser water students drink, the more they suffer from the constipation. The number of times a student had a meal everyday and the constipation was interrelated. If a student had lesser or irregular meals in a day, they had more constipation. Preferred food and constipation was also interrelated. Students that preferred vegetable, fruit, and dairy products were more likely to belong in a normal/general group. 6. By dividing the stress factors into the lower categories, we were able to see that the total stress(t-2.063, p-0.040), personal relations(t-2.316, p-0.021), scholastic achievements and personal directions(t-20127, p-0.034) had mutual relation with the constipation. The constipation group(120.27±30.2708) was suffering from stronger level of stress than the normal group(112.97±28.1466). Through this study, we were able to verify that the few general traits, habitual life, and stress of the women's university students have mutual relations with the constipation.

        신체화장애 환자의 스트레스 대처방식 및 심리적 특성

        Objectives : The purpose of this study was to investigate the stress coping strategies and psychological charactehstics, such as combined psychopathology and tendency of symptom interpretation, in patients with somatization disorder. Methods : Thirty patients meeting the criteria of DSM-IV somatization disorder were compared with thirty controls. We evaluated the subjects using Symptom Checklist-90-Revised (SCL-90-R), Somato-Sensory Amplification Scale (SSAS), Symptom Interpretation Questionnaire (SIQ), and The Ways of Stress Coping Questionnaire (SCQ). Independent t-test and Pearson correlation analysis were used. Results : From the results of SCL-90-R subscales, the scores of somatization, obsession-compulsion, depression, anxiety, and psychoticism were significantly higher in patients with somatization disorder than nonnal controls. Somatization disorder patients had greater amplification of physical sensation in SSAS and significantly higher score in physical interpretation of SIQ compared with psychological or environmental interpretation, In the SCQ of somatization disorder patients, we observed generallylower levels of total coping scores than the control group and significant positive correlation between passive coping style of SCQ and psychological interpretation of SIQ. Conclusion : These results show that patients with somatization disorder have various psychopathology, greater amplification of physical sensation, physical interpretation tendency of symptoms, and insufficient copying strategy.

      • 객체지향 개념 모델링에서 OCL에 의한 상속성 원리

        OOA/D에서는 클래스의 일반화와 특수화의 계층구조 사이에서 상속성의 개념이 어디까지 보장되는지 명확한 규칙이 없다. 예를 들어 상위 클래스의 제약조건이 하위 클래스에도 상속되는지, 상위 클래스의 상태 다이어그램이 하위 클래스의 상태 다이어그램에도 상속되는지, 하위 클래스의 행위는 상위 클래스의 행위를 제한시키는 것인지 확장시키는 것인지 등의 문제가 명확하지 않다. 또한 역할과 하위 클래스 설정 문제도 혼동이 일어난다. 본 논문에서는 Liskov의 치환 규칙에 의하여 정적인 면과 동적인 면을 동시에 고려한 상속성 원리를 OCL을 사용하여 정립한다. 이에 따라 역할, 일반화, 특수화, 집합체와 합성에 관한 개념의 가이드라인도 제시한다. There are no explicit rules in OOA/D to indicate whether or not the generalization and specialization hierarchy on a superclass is inherited by its subclasses. For example, we have relevant questions: ls a constraint on a suerclass is inherited by its subclasses? Does subclasses inherit the state diagram of the superclass? Can it refine this state diagram by adding. removing or redefining states, transitions, or events? Can it restrict the behavior of the generalization classes or extend it both? Many methods do not answer these questions. Also concepts of role and subclass are confused in the problem of structure. In this paper, we give an OCL formalization of inheritance by Liskov's Substitution Principle and propose a general guide-lines of concepts of role, generalization, specialization, aggregation, and composition.

      • Glyco-engineering of Recombinant Expression Systems for Production of Biobetter

        More than 60% of the currently available therapeutic proteins are glycoproteins attached with glycans, which play important roles in the protein folding, therapeutic efficacy, in vivo half-life, and immune reactions. Our group has focused on N-glycosylation pathway engineering of recombinant expression hosts including yeasts and Chinese hamster ovary (CHO) cells for production of therapeutic glycoproteins with improved potency. Different engineering strategies are required depending on the classes of therapeutic proteins because their optimal glycan structures are different. Many therapeutic enzymes used for treatment of lysosomal storage diseases require high contents of mannose-6- phosphate (M-6-P) glycan, which is essential for cellular uptake and targeting to lysosomes. Although yeasts do not have M-6-P glycans, they have some glycans containing mannosylphosphorylated mannose residues which can be converted to M-6-P structure through removal of the outer capping mannose. Recently, we constructed a glyco-engineered Saccharomyces cerevisiae mnn1Δoch1Δ strain overexpressing YlMPO1 gene, which was shown to have very high content of mannosylphosphorylated glycans [1, 2]. As they were efficiently converted to high content of M-6-P glycans through uncapping process, our engineering strategy showed a promise for production of biobetter enzymes with enhanced lysosomal targeting capabilities. For prolonged in vivo half-life of glycoproteins, sialic acids present on non-reducing terminal of glycan are important. We compared several bacterial α(2,6)-sialyltransferases and found that they have inherent siallidase activities, which decreased the amount of sialylated products [3]. These sialidase activities were inhibited by simple treatment of alkaline phosphatase, which desphosphorylates free cytidine monophosphate (CMP) generated from a donor substrate (CMP-N-acetylneuraminic acid) during the sialyltransferase reaction. It greatly enhanced the efficiency of bacterial α(2,6)- sialyltransferase reaction, which resulted in the increased content of bi-sialylated N-glycan up to ~98% without any decrease in prolonged reactions. Our efforts to construct Chinese hamster ovary (CHO) cells for production of glycoproteins with increased content of sialic acids shall be introduced also.

        Preoperative localization of potentially invisible colonic lesions on the laparoscopic operation field: using autologous blood tattooing

        Purpose: Preoperative colonoscopic (POC) localization is recommended for patients scheduled for elective laparoscopic colectomy for early colon cancer. Among the various localization method, POC tattooing localization has been widely used. Several dyes have been used for tattooing, but dye has disadvantages, including foreign body reactions. For this reason, we have used autologous blood tattooing for POC localization. This study aimed to evaluate the safety and efficacy of the autologous blood tattooing method.Methods: This study included patients who required POC localization of the colonic neoplasm among the patients who were scheduled for elective colon resection. The indication for localization was early colon cancer (clinically T1 or T2) or colonic neoplasms that could not be resected endoscopically. POC autologous blood tattooing was performed after saline injection, and 2 hemoclips were applied.Results: A total of 45 patients who underwent autologous blood tattooing and laparoscopic colectomy were included in this study. All POC localization sites were visible in the laparoscopic view. POC localization sites showed almost perfect agreement with intraoperative surgical findings. There were no complications like bowel perforation, peritonitis, hemoperitoneum, and mesenteric hematoma.Conclusion: Autologous blood is a safe and effective agent for localizing materials that can replace previous dyes. However, a large prospective case-control study is required for the routine application of this procedure in early colon cancer or colonic neoplasms.

      • Conjugation Strategies Using Mannose-6-phosphate Glycans Derived from Glyco-engineered Yeast Cell Wall Proteins

        Most of the therapeutic enzymes, which is used for treatment of lysosomal storage diseases, require the glycans containing mannose-6-phosphate (M-6-P). It has been shown that the increased content of M-6-P glycan enhances lysosomal targeting and, therefore, the efficacy of therapeutic enzymes [1]. We have constructed the glyco-engineered yeast harboring very high content of mannosylphosphorylated N-glycans, which can be converted to M-6-P glycan through an uncapping process [2]. In this study, cell wall mannoproteins of the glyco-engineered yeast were extracted by using hot citrate buffer and then digested with pronase, which generated mannosylphosphorylated glycans as attached to asparagine amino acid (Asn). Through the mild acid hydrolysis for uncapping and α(1,2)-mannosidase treatment for trimming, these Asn-linked glycans were converted to Asn-linked M-6-P glycans, which were specifically purified by titanium dioxide resin. The purified Asn-linked M-6-P glycans could be easily conjugated with the crosslinkers containing an amine-reactive group such as a N- hydroxysuccinimide esters which attacks amine group of Asn. Using a chemical group (maleimide, azide or dibenzo-bicyclo-octyne) at the opposite site of the attached cross-linkers, M-6-P glycans were successfully conjugated to target proteins. Proper conjugation of M-6-P glycan to target protein was confirmed by increase of molecular weight in SDS-PAGE and lectin blot using domain 9 of cation-independent M-6-P receptor. Further, M-6-P glycan-conjugated protein was shown to be internalized and targeted to lysosomes of cells.

        구제역 백신의 국가출하승인검정을 통한 품질 평가

        Foot-and-mouth disease (FMD) causes economic problems in livestock industry because of fast spread and inducing low productivity. FMD outbreaks occurred in South Korea over the period from 2000 to 2019. Vaccination is the most practical and effective means of controlling or preventing these outbreaks, and a national vaccination policy has been in place for all FMD-susceptible animals since 2010. To prevent and control of FMD, South Korea has been using vaccines imported from the United Kingdom, Argentina, and Russia. The Animal and Plant Quarantine Agency of South Korea oversees continuous quality control of imported FMD vaccines. FMD vaccines were evaluated characteristics, sterility, pH, inactivation, safety, potency test by Korean FMD vaccine standard assay (Test for National Lot Re-lease). The 6 company vaccines (A∼F) were used Test for National Lot Release by each method. We evaluated quality of each FMD vaccine from 2015 to 2019. All batch of vaccine showed good quality control and were passed the Test for National Lot Release. The serotypes of vaccine are increasingly changing to multiple vaccine because the FMD was outbreak by various serotype virus in South Korea. Furthermore, this data may be useful as a basis for ensuring the quality of FMD vaccines and for base data to manage them. Additional study is required to simple approach for rapid evaluation of quality and antigen content identification in vaccines.
