        Association of DOCK8, IL17RA, and KLK12 Polymorphisms with Atopic Dermatitis in Koreans

        Background: Early-onset and severe atopic dermatitis (AD) in patients increase the probability of the development of allergic rhinitis or asthma. Treatment and prevention strategies in infants and young children with AD are targeted toward treating the symptoms, restoring skin barrier functions, and reducing the absorption of environmental allergens in an attempt to attenuate or block the onset of asthma and food allergy. Objective: Given that the initiating events in AD remain poorly understood, identifying those at risk and implementing strategies to prevent AD is necessary. Methods: Whole-exome sequencing (WES) was performed in a 43 control group and a disease group with 20 AD patients without atopic march (AM) and 20 with AM. Sanger sequencing was carried out to validate found variants in cohorts. Results: DOCK8, IL17RA, and KLK12 single-nucleotide polymorphisms were identified by WES as missense mutations: c.1289C>A, p.P97T (rs529208); c.1685C>A, p.P562G (rs12484684); and c.457+27>C, rs3745540, respectively. A case-control study show that total immunoglobulin E (IgE) level was significantly increased in the AA genotype of DOCK8 compared to the CA genotype in allergic patients. The rs12484684 of IL17RA increased risk of adult-onset AD (odds ratio: 1.63) compared to the control for (A) allele frequency. AD and AM Patients with the IL17RA CA genotype also had elevated IgE levels. rs3745540 of KLK12 was associated with AD in dominant model (odds ratio: 2.86). Conclusion: DOCK8 (rs529208), IL17RA (rs12484684), and KLK12 (rs3745540), were identified using a new WES filtering method. the result suggests that polymorphism of DOCK8 and IL17RA might be related to increase the total IgE level.       • Aberrant expression of interleukin-10 and activation-induced cytidine deaminase in B cells from patients with Behçet's disease

        <P>Despite extensive studies, the pathogenesis of Behçet's disease (BD) remains unclear. In particular, the roles of B cells in patients with BD have not been elucidated. Activation-induced cytidine deaminase (AID) is a critical enzyme for immunoglobulin (Ig) heavy chain class switching and somatic hypermutation in B cells and the abnormal expression of AID in various immune conditions has previously been studied. B10 cells, an interleukin (IL)-10-secreting subset of regulatory B cells, function to downregulate inflammation and autoimmunity. Thus, in the present study, the relevance of B cells in patients with BD was investigated. The plasma levels of IL-10 and IgA and the proportions of cluster of differentiation (CD)43<SUP>+</SUP> B cells, excluding naïve B cells, were measured in 16 patients with BD and 16 age- and sex-matched healthy controls (HCs). Additionally, the mRNA levels of IL-10 and AID were assessed in B cells from fresh peripheral blood samples of the BD patients and HCs. The plasma level of IL-10 in patients with BD did not differ significantly from that in HCs. Similarly, there was no significant difference in the plasma level of IgA, although a slight increase was observed in patients with BD compared with that in HCs. There were no differences in CD43<SUP>+</SUP>CD19<SUP>+</SUP> B cell numbers between patients with BD and HCs. However, IL-10 mRNA levels were significantly reduced (P<0.05), while AID mRNA levels were significantly increased (P<0.01) in the B cells of patients with BD compared with those in HCs. These results provide insight into the role of B cells in patients with BD.</P>

        들깨(Perilla frutescens) 새싹 추출물의 항산화 및 항염 효과

        본 연구는 들깨 새싹 추출물의 항산화, 항염증 및 항부종에 대한 효과를 조사하였다. 들깨 새싹 추출물은 DPPH와 ABTS 라디칼을 효과적으로 제거하는 항산화 활성이 우수하였다. 또한 들깨 새싹 추출물은 활성화된 설치류 유래 대식세포주인 RAW 264.7세포와 인간 유래 HMC-1 세포의 TNF-α와 IL-1β를 효과적으로 억제하였다. 더욱이 마우스의 귀와 발 부종을 억제하는 우수한 효과가 있었다. 이러한 결과는 들깨 새싹 추출물은 항산화제로 사용될 수 있을 뿐만 아니라 항염증과 항부종에 효과적인 물질이라는 것을 제시해주었다. 이와 관련된 들깨 새싹 추출물의 기능성에 대해서는 앞으로 분자생화학적 수준에서 더 연구해야할 필요성이 있는 것으로 사료된다. We investigated the effects of an ethanol extract from Perilla frutescens sprouts (PFSE) as an antioxidant, and its effects on edema and inflammation in RAW 264.7 cells and HMC-1 cells. The antioxidant activities (DPPH and ABTS radical scavenging) of PFSE were similar to those of butylated hydroxytoluene (BHT) and (±)-6-hydroxy-2,5,7,8- tetramethylchromane-2-carboxylic acid (Trolox). We also investigated the anti-inflammatory effects of PFSE on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells and HMC-1 cells stimulated with phorbol 12-myristate 13-acetate (PMA) with the calcium ionophore A23187. TNF-α and IL-1β production, which had been increased by treatment with LPS or PMA plus A23187, were significantly inhibited by PFSE in a dose-dependent manner. Furthermore, PFSE significantly reduced the xylene-induced ear edema and the carrageenan-induced paw edema of ICR mice in a dosedependent manner. The effects of PFSE (200 mg/kg) in reducing ear and paw edema were similar to those of aspirin (50mg/kg). These results suggest that PFSE can be potentially used as a medicine for treating oxidative stress, an edematous and inflammatory disease.

        H₂O₂ 유도 U2OS 세포에서 Pinus sylvestris L.의 산화적 스트레스 및 골관절염 개선 효과

        This study investigated the pharmacological impact of the Pinus sylvestris L. water extract (PS) on hydrogen peroxide (H2O2)-induced oxidative stress in the human osteosarcoma cell line U2OS and a monosodium iodoacetate (MIA)-induced osteoarthritis model in rats. A MitoSoxTM indicator was used to measure mitochondrial reactive oxygen species (ROS) levels, while mitochondrial integrity was assessed using MitoTrackerTM. Moreover, we assessed oxidative stress-related markers, including sirtuin 1 (Sirt1), metal-responsive transcription factor 1 (MTF1), and 4-hydroxynonenal (4-HNE) using immunohistochemistry. We performed serum analysis and western blotting in vivo. The cell viability of U2OS cells was increased in a concentration-dependent manner, independent of H2O2 treatment. PS dramatically reduced the accumulation of mitochondrial ROS, while simultaneously reversing the decreased MitoTrackerTM levels. Additionally, both Sirt1 and MTF1 intensities were significantly elevated, whereas 4-HNE intensity noticeably decreased. In the animal experiment, the MIA treatment resulted in a reduction of hindpaw weight distribution changes. However, PS pretreatment significantly reversed this reduction. The serum levels of tumor necrosis factor-alpha, interleukin (IL)-6, and IL-1β were significantly decreased compared to the control group. The control group, treated with only MIA, exhibited increased levels of three enzymes: Matrix metalloproteinase 13 (MMP- 13), which degrades collagen or proteoglycan, and MMP-2 and MMP-3, which degrade the non-collagen matrix components of the joints. However, the PS treatment significantly inhibited the levels of the enzymes except MMP-3. In conclusion, these results suggest that PS alleviated oxidative stress and regulated inflammatory cytokines and the expression of MMPs.

        Plant synthetic GP4 and GP5 proteins from porcine reproductive and respiratory syndrome virus elicit immune responses in pigs

        <P>Porcine reproductive and respiratory syndrome virus (PRRSV) causes PRRS, the most economically significant disease affecting the swine industry worldwide. However, current commercial PRRSV vaccines (killed virus or modified live vaccines) show poor efficacy and safety due to concerns such as reversion of virus to wild type and lack of cross protection. To overcome these problems, plants are considered a promising alternative to conventional platforms and as a vehicle for large-scale production of recombinant proteins. Here, we demonstrate successful production of recombinant protein vaccine by expressing codon-optimized and transmembrane-deleted recombinant glycoproteins (GP4D and GP5D) from PRRSV in planta. We generated transgenic Arabidopsis plants expressing GP4D and GP5D proteins as candidate antigens. To examine immunogenicity, pigs were fed transgenic Arabidopsis leaves expressing the GP4D and GP5D antigens (three times at 2-week intervals) and then challenged with PRRSV at 6-week post-initial treatment. Immunized pigs showed significantly lower lung lesion scores and reduced viremia and viral loads in the lung than pigs fed Arabidopsis leaves expressing mYFP (control). Immunized pigs also had higher titers of PRRSV-specific antibodies and significantly higher levels of pro-inflammatory cytokines (TNF-alpha and IL-12). Furthermore, the numbers of IFN-gamma(+)-producing cells were higher, and those of regulatory T cells were lower, in GP4D and GP5D immunized pigs than in control pigs. Thus, plant-derived GP4D and GP5D proteins provide an alternative platform for producing an effective subunit vaccine against PRRSV.</P>

        아토피 피부염 모델에 대한 β-1,3/1,6-glucan과 Lactobacillus plantarum LM1004의 면역 조절 효과

        본 연구에서는 아토피 피부염 동물 모델에 대한 β-1,3/1,6-glucan과 L. plantarum LM1004의 면역조절 효과를 확인하고자 하였다. 가려움증의 횟수와 유출된 evans blue, 그리고 혈청 IgE와 histamine의 농도는 β-1,3/1,6-glucan과 L. plantarum LM1004를 섭취한 그룹에서 아토피 피부염 유발그룹에 비해 유의적으로 감소하는 결과를 나타내었다. 아토피 피부염이 유발되면 전사 수준에서 Th2 및 Th17 세포의 전사인자 및 cytokine은 과발현되며, β-1,3/1,6-glucan과 L. plantarum LM1004를 섭취하였을 때 이를 유의적으로 감소되었다. 또한 β-1,3/1,6-glucan과 L. plantarum LM1004는 Th1 및 Treg 세포의 전사인자(T-bet, GATA-3, RORγT, Foxp3) 및 cytokine (INF-γ, IL-4, IL-17, TGF-β)의 발현을 증가시킴으로써 면역 균형을 조절하는 것으로 나타났다. Galectin-9과 filaggrin은 아토피 피부염 유발 처리군에서 유의적으로 가장 낮았으며, β-1,3/1,6-glucan 처리군에서 유의적으로 가장 높게 나타났다. 이와 반대로 TSLP는 아토피 피부염 유발그룹에서 유의적으로 가장 높았으며 β-1,3/1,6-glucan과 L. plantarum LM1004를 섭취한 그룹은 대조군과 유사한 수준이었다. 이러한 결과를 통해 β-1,3/1,6-glucan과 L. plantarum LM1004는 아토피 피부염 동물 모델에서 면역조절 작용 및 아토피 피부염의 개선 효과를 가짐을 알 수 있었다. 따라서 β-1,3/1,6-glucan과 L. plantarum LM1004는 아토피 피부염에 유용한 천연소재로서 사용될 것으로 기대된다. In this study, we examined the efficacy of the immune regulation of β-1,3/1,6-glucan and Lactobacillus plantarum LM1004 on atopic dermatitis models. The oral administration of β-1,3/1,6-glucan and L. plantarum LM1004 on mice significantly decreased the amount of scratching, leakage to evans blue, and concentrations of serum immunoglobulin E (IgE) and histamine compared with the atopic dermatitis–induced group. When atopic dermatitis was induced, the transcription factors (GATA-3, retinoic acid-related orphan receptor γ T [RORγT]) and cytokines (interleukin-4 [IL-4], IL-17) of Th2 and Th17 cells were overexpressed at the transcriptional level, and they significantly decreased with oral administration of β-1,3/1,6-glucan and L. plantarum LM1004. In addition, β-1,3/1,6-glucan and L. plantarum LM1004 were shown to modulate the immune balance by increasing the expression of Th1 and Treg transcription (T-bet, forkhead box p3 [Foxp3]) and cytokines (interferon-γ [IFN-γ], transforming growth factor-β [TGF-β]). Galectin-9 and filaggrin were significantly lower in the atopic dermatitis–induced group and significantly higher in the β-1,3/1,6-glucan-treated group. In contrast, thymic stromal lymphopoietin (TSLP) was highest in the atopic dermatitis–induced group, while mice that were orally administered β-1,3/1,6-glucan and L. plantarum LM1004 showed similar TSLP levels to the control group. These results indicate that β-1,3/1,6-glucan and L. plantarum LM1004 have immunomodulatory effects and atopic dermatitis improvement effects in an animal model of atopic dermatitis. Therefore, it is expected that β-1,3/1,6-glucan and L. plantarum LM1004 can be used as natural materials in the treatment of atopic dermatitis.

        <書齋夜會錄>의 構造와 意味

        This article studied the romantic novel characteristics of <Seojaeyahoirok>, the novel collection of Gwang-han Shin. When looking into the structure and the meaning of <Seojaeyahoirok>, followings were discovered. Firstly, in this article <Seojaeyahoirok> was defined as a romantic novel not as a pseudo-romantic novel (假傳). The combination of multiple genres was named as 'Formatting Phenomenon (樣式化現象)' and addressed this phenomenon would necessarily appear when a certain genre or trend transits into another genre or bend. Secondly, <Seojaeyahoirok> described a romantic event that the hem met ghost in his study, a space of everyday life. It must be noted that the meeting between the hero and the ghost was made not in the dream but in an ordinary space in his sober consciousness. It implies that <Seojaeyahoirok> is more realistic in the aspect of subject material. Third, <Seojaeyahoirok> is very important in that it is the only work that dealt with the meeting between man and ghost among Korean romantic novels. The subject of romantic meetings shown in Korean romantic history was getting more realistic such as 'God → Ghost - Spirit - Man'. Fourthly, the subject of <Seojaeyahoirok> is based on the logic of Confucism.

        <崔生遇眞記>연구 : 傳奇的 人物의 특징과 작가의식을 중심으로

        In the analysis of <Oloisaengujingi (崔生遇眞記)>, followings were discovered. First, it identified the characters of <Choisaengujingi(崔生遇眞記)>had similar characteristics of characters in the 17th century's World Situational(世情) romantic novels. Secondly, it identified that the ideology of <Choisaengujing(崔生遇眞記)> is Taoism and Gwang-han Shin longed for Taoist Ideal World through the frictional hero. Thirdly, it identified that the hero reflected the author himself and his consciousness of 'Scholar'. <Choisaengujing(崔生遇眞記)> is a romantic novel in which the author longs for non-secular ideal world based on Taoism.

        Followings were revealed through the analysis of <Anbingmongyurok>. Firstly, in<Anbingmongyurok>, Mr. Art(安生) recognized the nature of 'Hwaguk-The Country of Flowers' by meeting personified figures. Consequently, he kept distance from flowerbed (negative space) and was absorbed in reading only with clean mind. The recognition of Mr. An was from the comparison between the world of the things and the world of human beings. <Anbingmongyurok>showed the process of dedication to Study of a Confucist by 'Gyeokmulchiji, thorough reasoning of things into truth'('格物致和') of 《Daehak (大學)》. Secondly, the flowerbed in<Anbingmongyurok>had allegorical and historical characteristics. Additionally, the confronting characters had allegorical meanings addition to facial meanings. Gwang-han Shin presented the process of his life dedicating to reading after retiring from the world of politics in this novel.
