        Continuous Cooling Transformation Behavior and Impact Toughness in Heat-Affected Zone of Nb-Containing Fire-Resistant Steel

        Simulated heat-affected zone continuous cooling transformation diagram was developed for advanced fireresistantsteel. Over a wide range of cooling rates, corresponding to t8/5 from 6 s to 150 s, granular bainite was thedominant transformation constituent, while the morphology of less dominant martensite-austenite (M-A) constituentchanged from film-like to block-type constituent; but the hardness remained similar to the average value of 190-205 HV (0.2). The start and finish transformation temperature was high at 700 °C and 500 °C, and is different fromthe conventional high strength low alloy steels. It is believed that the high-content (0.09 wt%) of Nb may promotebainite transformation at relatively high temperatures. Martenistic matrix was not observed at high cooling rateand the film-like M-A constituent and blocky M-A constituent with thin film of retained austenite and lath martensitewere observed on slow cooling. Excellent impact toughness was obtained in the heat-affected zone with15-75 kJ/cm welding heat input.

        Genome constitution and evolution of Elymus atratus (Poaceae: Triticeae) inferred from cytogenetic and phylogenetic analysis

        Background Elymus atratus (Nevski) Hand.-Mazz. is perennial hexaploid wheatgrass. It was assigned to the genus Elymus L. sensu stricto based on morphological characters. Its genome constitution has not been disentangled yet. Objective To identify the genome constitution and origin of E. atratus. Methods In this study, genomic in situ hybridization and fluorescence in situ hybridization, and phylogenetic analysis based on the Acc1, DMC1 and matK sequences were performed. Results Genomic in situ hybridization and fluorescence in situ hybridization results reveal that E. atratus 2n = 6x = 42 is composed of 14 St genome chromosomes, 14 H genome chromosomes, and 14 Y genome chromosomes including two H-Y type translocation chromosomes, suggesting that the genome formula of E. atratus is StStYYHH. The phylogenetic analysis based on Acc1 and DMC1 sequences not only shows that the Y genome originated in a separate diploid, but also suggests that Pseudoroegneria (St), Hordeum (H), and a diploid species with Y genome were the potential donors of E. atratus. Data from chloroplast DNA showed that the maternal donor of E. atratus contains the St genome. Conclusion Elymus atratus is an allohexaploid species with StYH genome, which may have originated through the hybridization between an allotetraploid Roegneria (StY) species as the maternal donor and a diploid Hordeum (H) species as the paternal donor.         SP0454, A Putative Threonine Dehydratase, Is Required For Pneumococcal Virulence In Mice

        Increasing pressure in antibiotic resistance and the requirement for the design of new vaccines are the objectives of clarifying the putative virulence factors in pneumococcal infection. In this study, the putative threonine dehydratase sp0454 was inactivated by erythromycin-resistance cassette replacement in Streptococcus pneumoniae CMCC 31203 strain. The sp0454 mutant was tested for cell growth, adherence, colonization, and virulence in a murine model. The Δsp0454 mutant showed decreased ability for colonization and impaired ability to adhere to A549 cells. However, the SP0454 polypeptide or its antiserum did not affect pneumococcal CMCC 31203 adhesion to A549 cells. The sp0454 deletion mutant was less virulent in a murine intranasal infection model. Real-time RT-PCR analysis revealed significant decrease of the pneumococcal surface antigen A expression in the sp0454 mutant. These results suggest that SP0454 contributes to virulence and colonization, which could be explained in part by modulating the expression of other virulence factors, such as psaA in pneumococcal infection.

        Circulating Low Absolute CD4+ T Cell Counts May Predict Poor Prognosis in Extranodal NK/T-Cell Lymphoma Patients Treating with Pegaspargase-Based Chemotherapy

        Purpose Extranodal natural killer/T-cell lymphoma, nasal type (ENKTL) is a rare subtype of non- Hodgkin lymphoma, and asparaginase-based regimens are the best first-line treatments. Data on the role of specific circulating lymphocyte subsets in the progression of ENKTL are limited. The aim of this study was to investigate the clinical correlation and distribution of circulating absolute CD4+ T-cell counts (ACD4Cs) in ENKTL. Materials and Methods We retrospectively searched medical records for 70 newly diagnosed ENKTL patients treated with pegaspargase-based regimens. Comparison of ACD4Cs as a continuous parameter in different groups was calculated. Univariate and multivariate analyses were used to assess prognostic factors for overall survival (OS) and progression-free survival (PFS). Results Stage III/IV, B symptoms, elevated lactate dehydrogenase, monocytopenia, high-intermediate and high risk International Prognostic Index (IPI) and Korean Prognostic Index (KPI), high risk Prognostic Index of Natural Killer Lymphoma (PINK), and lower lymphocytes were significantly associated with low ACD4C at diagnosis. With a median follow-up time of 32 months, patients who had an ACD4C < 0.30109/L had a worse OS. Median OS was 11 months and median PFS was 5 months in the low ACD4C cohort. There were significant differences in both OS and PFS between the two cohorts. Moreover, multivariate Cox analysis identified ACD4Cs as an independent predictor for OS and PFS. Conclusion Low ACD4Cs were associated with poorer survival and could act as a negative predictor for ENKTL patients treated with asparaginase-based regimens.

        The New Ecosystem of Cross-border E-Commerce among Korea, China and Japan Based on Blockchain

        Purpose - The purpose of the study is to propose a theoretical framework of cross-border e-commerce ecosystems based on blockchain technology. The ecosystem includes five systems, namely, crossborder supply chain intelligent system, cross-border logistics system, cross-border payment system, cross-border product quality traceability system and cross-border customs supervision system. Design/methodology - This study firstly derived the main improvement factors for the new ecosystem based on blockchain through prior research and expert interviews on cross-border e-commerce. Then explored the use of virtue of decentralization, anti-counterfeiting traceability, consensus mechanism, smart contract and other means of the core technology of blockchain to overcome the bottleneck of cross-border e-commerce development among Korea, China, and Japan. Finally, proposed valuable implications in both theoretical and practical perspectives. Findings - As a result, we combined with the problems existing in cross-border e-commerce among Korea, China and Japan, this paper proposes a solution based on blockchain. On this basis, it constructs a cross-border e-commerce ecosystem among these three countries, including five systems. In addition, we discuss the main problems existing in the current blockchain, such as low transaction concurrency, security loopholes, and inconsistent standards, the corresponding countermeasures are proposed from the technical level, security level and industry standards. Originality/value - This study is the first to apply the blockchain technology to solve the cross-border e-commerce problems in Korea, China and Japan, which is of pioneering significance in both literature and practice. Block chain technology is in the ascendency. This study provides technical solutions for promoting the development of cross-border e-commerce import and export trade between Korea, China and Japan.

      • Effect of Poly(butyl acrylate)-Poly(methyl methacrylate) Rubber Particle Texture on the Toughening Behavior of Poly(methyl methacrylate)

        Monodisperse composite latex particles with size of ca. 300 nm, which consist ofn-butyl acrylate as a soft phase and methyl methacrylate as a hard phase with different morphology, were synthesized by seeded multi-stage emulsion polymerization. Three types of composite latex particles including random-, core/shell-, and gradient-type particles were obtained by using different monomer feeding methods during semi-batch emulsion polymerization. Effect of poly(butyl acrylate)-poly(methyl methacrylate) rubber particle morphology on the mechanical and rheological properties of rubber toughened poly(methyl methacrylate) was investigated. Among three different rubber particles, the gradient-type rubber particle showed better toughening effect than others. No significant variation of rheological property of poly(methyl methacrylate)/rubber blends was observed for the different rubber particle morphology.

        Sensory involvement in the SOD1-G93A mouse model of amyotrophic lateral sclerosis

        A subset of patients of amyotrophic lateral sclerosis (ALS) present with mutation of Cu/Zn superoxide dismutase 1 (SOD1), and such mutants caused an ALSlike disorder when expressed in rodents. These findings implicated SOD1 in ALS pathogenesis and made the transgenic animals a widely used ALS model. However, previous studies of these animals have focused largely on motor neuron damage. We report herein that the spinal cords of mice expressing a human SOD1 mutant (hSOD1-G93A), besides showing typical destruction of motor neurons and axons, exhibit significant damage in the sensory system, including Wallerian-like degeneration in axons of dorsal root and dorsal funiculus, and mitochondrial damage in dorsal root ganglia neurons. Thus, hSOD1-G93A mutation causes both motor and sensory neuropathies, and as such the disease developed in the transgenic mice very closely resembles human ALS.         Selection of Reference Genes for Real-time Quantitative PCR Normalization in the Process of Gaeumannomyces graminis var. tritici Infecting Wheat

        Gaeumannomyces graminis var. tritici is a soil borne pathogenic fungus associated with wheat roots. The accurate quantification of gene expression during the process of infection might be helpful to understand the pathogenic molecular mechanism. However, this method requires suitable reference genes for transcript normalization. In this study, nine candidate reference genes were chosen, and the specificity of the primers were investigated by melting curves of PCR products. The expression stability of these nine candidates was determined with three programs-geNorm, Norm Finder, and Best Keeper. TUBβ was identified as the most stable reference gene. Furthermore, the exopolygalacturonase gene (ExoPG) was selected to verify the reliability of TUBβ expression. The expression profile of ExoPG assessed using TUBβ agreed with the results of digital gene expression analysis by RNA-Seq. This study is the first systematic exploration of the optimal reference genes in the infection process of Gaeumannomyces graminis var. tritici.
