        Primer 155(GCTGGCGCTG) was the most useful primer with distinctive polymorphic DNA pattern to distinguish between KI races and KJ races. Primer 173(CAGGCGGCGT) showed polymorphic DNA pattern to distinguish isolates of KI-100 race. The specific DNA fragment of 0.6Kb, generated from 88-62 of KJ-201 race by RAPD with primer 412)TGCGCCGCTG), was promising as a molecular marker to identify the isolate.

        3분과 : 가선집재작업에서의 작업 생산성 및 비용 분석

        본 연구에서는 임목수확작업에 사용되는 집재기계의 작업효율성을 분석하여 경사가 급한(20° 이상) 작업조건에 적합한 가선계 집재기계를 선정하는데 기초 정보를 제공하고자, 타워야더(RME-300T), 트랙터부착형 집재기(일명: 춘천집재기), 트랙터윈치(FARMI) 등에 의한 집재작업에서의 작업생산성 및 비용을 비교 분석하였다. 평균 흉고직경이 20 cm, 집재거리 60 m, 가로집재거리 10 m, 그리고 기계이용률이 70%인 동일한 작업 조건에서 작업이 이루어질 경우, 타워야더의 1일 집재작업 생산성은 33.04 m3/일, 트랙터부착형 집재기는 38.47 m3/일, 그리고 트랙터윈치는 14.17 m3/일으로 분석되었으며, 이에 따른 집재작업비용은 각각 25,105원/m3, 20,520원/m3, 37,835원/m3으로 나타났다. 동일한 작업조건에서 집재거리에 따른 각 집재기계별 작업효율성을 비교하였을 경우, 집재거리가 40 m 이내로 짧을 경우에는 트랙터윈치에 의한 집재작업이 효율적이며, 40~140 m의 집재거리에서는 춘천집재기가, 140 m 이상의 장거리 집재작업에서는 타워야더가 가장 효율적인 것으로 나타났다. This study was conducted to provide field-based harvesting study information which can be used to select an optimal cable system for certain work conditions on steep grounds (>20° ground slope) in Korea. Toaccomplish this study objectives, we evaluated three cable yarding systems (RME-300T tower-yarder, Chuncheontower-yarder, FARMI tractor winch) working in typical work conditions for their yarding productivity and operational efficiency. Those yarders are commonly used for removing logs or trees on steep grounds in Korea. Under the same work conditions (average DBH of tree to be cut, 20 cm; yarding distance, 60 m; lateral yardingdistance, 10 m; and machine utilization rate, 70%), the average productivities were 33.04 m3/day, 38.47 m3/day, and 14.17 m3/day for RME-300T, Chuncheon tower-yarder, and FARMI, respectively. Our standardized cost comparison study also showed that the yarding cost was highest at 37,835 won/m3 with FARMI, followed byRME-300T at 25,105 won/m3 for the same work conditions. We found the lowest yarding cost with the Chuncheon tower-yarder at 20,520 won/m3 which was resulted primarily from high yarding productivity at theyarding distance (60 m). Our analysis suggested that a small machine such as FARMI could be a low-costyarding machine option for a cable yarding job with a short yarding distance (40 m or less). The Chuncheontower-yarder is well suited for a mid-range yarding distance job in Korea, ranged between 40 to 140 m. Ifyarding distance were longer than 140 m, the RME-300T tower yarder appears to be most cost-effective.

        We report a case of microcystic meningioma which showed unusual radiological findings resembling malignant tumor. A 44-year-old woman presented with generalized tonic clonic seizure and right hemiparesis. Computed tomography and magnetic resonance images showed a mass at left frontal, parasagittal area with septa-like enhancement and surrounding edema. 201Tl SPECT showed focal uptake in mass lesion which suggests the malignant tumor. The histopathological findings of this tumor revealed to be a microcystic meningioma and Ki-67 labeling index was 3.8. Although the mass was a subtype of meningioma, it showed a early recurrence 10 months after grossly total resection because of high proliferative index. We suggest that the biological behaviour in addition to morphologic classification should be considered in the management of benign brain tumor, such as meningioma.

        Growth Inhibition of Colon Cancer through Inactivation of STAT3 Pathway by IL-10 and IL-1ra Released from Murine Macrophage

        The objective of this study was to determine the effect of macrophages on growth of human colon cancer cells. The results showed that co-culture of colon cancer cells with macrophages inhibited the growth of colon cancer cells (HCT116 and SW620) depending on the number of macrophages, RAW 264.7 cells, and activated THP-1 cells accompanied by down regulation of pSTAT3 in cancer cells. We also found that expression and release of cancer cell growth inhibitory cytokines, IL-1 receptor antagonist (IL-1ra) and IL-10, was increased in macrophages. Blocking of the STAT3 pathway with specific inhibitor and siRNA of STAT3 abolished the growth of colon cancer cells and expression of IL-1ra and IL-10. In addition, neutralization of IL-1ra and IL-10 with antibodies resulted in reversal of macrophage-induced inhibition of cancer cell growth. These data showed that IL-1ra and IL-10 released from macrophages inhibit growth of colon cancer cells through inhibition of the STAT3 pathway

        Long-term postresection prognosis of primary neuroendocrine tumors of the liver

        Purpose: Primary hepatic neuroendocrine tumor (PHNET) is a very rare neoplasm, requiring strict exclusion of metastasis from possible extrahepatic primary sites for its diagnosis. Methods: We reviewed our clinical experience of 13 patients with primary hepatic NET who underwent liver resection from January 1997 to December 2015. Results: The mean age of the 13 patients (8 males and 5 females) was 51.1 ± 12.8 years; the most common clinical manifestation was vague, nonspecific abdominal pain (n = 9). Of them, 11 patients underwent preoperative liver biopsy, 7 of which correctly diagnosed as neuroendocrine tumor (NET). Ten patients underwent R0 resection, and 3 underwent R1 resection. Diagnosis of PHNET was confirmed both immunohistochemically and by absence of extrahepatic primary sites. All tumors were single lesions, with a mean size of 9.6 ± 7.6 cm and a median size of 4.3 cm; all showed positive staining for synaptophysin and chromogranin. During a mean follow-up period of 95.1 ± 86.6 months, 7 patients died from tumor recurrence, whereas the other 6 remain alive to date, making the 5-year tumor recurrence rate 56.0% and the 5-year patient survival rate 61.5%. When confined to R0 resection, 5-year recurrence and survival rates were 42.9% and 70.0%, respectively. Univariate analysis showed that Ki-67 proliferative index was the only risk factor for tumor recurrence. Conclusion: PHNET is a very rare tumor with no specific clinical features, and its final diagnosis depends primarily on pathology, immunohistochemistry, and exclusion of metastasis from other sites. Aggressive surgical treatment is highly recommended for PHNET because of acceptably favorable postresection outcomes

      • Clinicopathological and molecular analysis of multinodular and vacuolating neuronal tumors of the cerebrum

        <P><B>Summary</B></P> <P>Multinodular and vacuolating neuronal tumor (MVNT) of the cerebrum is a recently recognized rare neuronal tumor, and its pathogenesis is unclear. We analyzed 7 cases of histologically typical MVNT: 6 were adults (mean age, 43.0 years [range, 23-56 years]) and 1 was a child (age, 10 years). The most common symptoms were seizures (n = 4) and headache (n = 2). The tumors were supratentorial (temporal, 5; frontal lobes, 2) in origin as reported. Vacuolated tumor cells were robustly positive for α-INA and Olig2 and at least partly positive for synaptophysin and MAP2, but negative for Neu-N, nestin and CD34. GFAP and vimentin were expressed in reactive astrocytes but not in tumor cells. Negative results were obtained for p53, IDH-1, BRAF<SUP>V600E</SUP>, H3 K27M, EGFR, Lin28A, and L1CAM. ATRX, BRG1, INI-1, and TMHH were retained. The Ki-67 labeling index was very low (<1%), and pHH3 revealed no mitotic figure. Ultrastructural features of tumor cells were comparable with those of immature neuronal cells, with several intracytoplasmic myelin-like autophagosomes and pericellular vacuolization. No <I>IDH1</I>/<I>IDH2</I> and <I>BRAF</I> <SUP> <I>V600E</I> </SUP> mutations were found upon direct sequencing. Whole-exome sequencing revealed <I>FGFR2</I>-<I>ZMYND11</I> gene fusion in 1 case. After gross total resection, all patients were alive without seizures. There was no tumor recurrence during an average period of 68 months (range, 23-101 months). The analysis of 7 typical cases of MVNT suggested that these lesions may be clonal tumors because <I>FGFR2</I>-<I>ZMYND11</I> fusion was found (1 case).</P> <P><B>Highlights</B></P> <P> <UL> <LI> “MVNT” of the cerebrum is a recently recognized rare epileptogenic neuronal tumor. </LI> <LI> Whole-exome sequencing revealed <I>FGFR2</I>-<I>ZMYND11</I> gene fusion in one of our MVNT cases. </LI> <LI> The tumor cell is robustly positive for α-INA and Olig2, but negative for Neu-N. </LI> <LI> Ultrastructurally, tumor cells have autophagolysosomes with pericellular vacuolization. </LI> <LI> There was no tumor recurrence during an average period of 68 months. </LI> </UL> </P>

      • Dietary fat increases solid tumor growth and metastasis of 4T1 murine mammary carcinoma cells and mortality in obesity-resistant BALB/c mice

        <P><B>Introduction</B></P><P>High-fat diets (HFDs) are known to cause obesity and are associated with breast cancer progression and metastasis. Because obesity is associated with breast cancer progression, it is important to determine whether dietary fat <I>per se </I>stimulates breast cancer progression in the absence of obesity. This study investigated whether an HFD increases breast cancer growth and metastasis, as well as mortality, in obesity-resistant BALB/c mice.</P><P><B>Methods</B></P><P>The 4-week-old, female BALB/c mice were fed HFD (60% kcal fat) or control diet (CD, 10% kcal fat) for 16 weeks. Subsequently, 4T1 mammary carcinoma cells were injected into the inguinal mammary fat pads of mice fed continuously on their respective diets. Cell-cycle progression, angiogenesis, and immune cells in tumor tissues, proteases and adhesion molecules in the lungs, and serum cytokine levels were analyzed with immunohistochemistry, Western blotting, and enzyme-linked immunosorbent assay (ELISA). <I>In vitro </I>studies were also conducted to evaluate the effects of cytokines on 4T1 cell viability, migration, and adhesion.</P><P><B>Results</B></P><P>Spleen and gonadal fat-pad weights, tumor weight, the number and volume of tumor nodules in the lung and liver, and tumor-associated mortality were increased in the HFD group, with only slight increases in energy intake and body weight. HF feeding increased macrophage infiltration into adipose tissues, the number of lipid vacuoles and the expression of cyclin-dependent kinase (CDK)2, cyclin D1, cyclin A, Ki67, CD31, CD45, and CD68 in the tumor tissues, and elevated serum levels of complement fragment 5a (C5a), interleukin (IL)-16, macrophage colony-stimulating factor (M-CSF), soluble intercellular adhesion molecule (sICAM)-1, tissue inhibitors of metalloproteinase (TIMP)-1, leptin, and triggering receptor expressed on myeloid cells (TREM)-1. Protein levels of the urokinase-type plasminogen activator, ICAM-1, and vascular cell adhesion molecule-1 were increased, but plasminogen activator inhibitor-1 levels were decreased in the lungs of the HFD group. <I>In vitro </I>assays using 4T1 cells showed that sICAM-1 increased viability; TREM-1, TIMP-1, M-CSF, and sICAM-1 increased migration; and C5a, sICAM-1, IL-16, M-CSF, TIMP-1, and TREM-1 increased adhesion.</P><P><B>Conclusions</B></P><P>Dietary fat increases mammary tumor growth and metastasis, thereby increasing mortality in obesity-resistant mice.</P>

        Hiccups, also known as singultus, are spasmodic involuntary contractions of respiratory muscles that shorten respiration. The characteristic sound is caused by rapid closure of the glottis. In oriental medicine, the mechanism of hiccups is "reversed flow of Ki". Hiccups have been associated with neoplasm, infections, seizures, diabetes. renal failure, alcohol ingestion, various drugs, and ischemic events of the myocardium or central nervous system. We present a case of chronic hiccups after cerebral infarction that was successfully treated using moxibustion therapy.

        Substance P restores normal skin architecture and reduces epidermal infiltration of sensory nerve fiber in TNCB-induced atopic dermatitis-like lesions in NC/Nga mice

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Atopic dermatitis (AD) is a chronic inflammatory skin disorder characterized by intense pruritus and eczematous lesion. Substance P (SP) is an 11-amino-acid endogenous neuropeptide that belongs to the tachykinin family and several reports recently have supported the anti-inflammatory and tissue repairing roles of SP.</P> <P><B>Objective</B></P> <P>In this study, we investigated whether SP can improve AD symptoms, especially the impaired skin barrier function, in 2, 4, 6-trinitrochlorobenzene (TNCB)-induced chronic dermatitis of NC/Nga mice or not.</P> <P><B>Method</B></P> <P>AD-like dermatitis was induced in NC/Nga mice by repeated sensitization with TNCB for 5 weeks. The experimental group designations and topical treatments were as follows: vehicle group (AD-VE); SP group (AD-SP); and SP with NK1R antagonist CP99994 (AD-SP-A) group. Histological analysis was performed to evaluate epidermal differentiation, dermal integrity, and epidermal nerve innervation in AD-like lesions. The skin barrier functions and pruritus of NC/Nga mice were evaluated by measuring transepidermal water loss (TEWL) and scratching behavior, respectively.</P> <P><B>Result</B></P> <P>Topical SP treatment resulted in significant down-regulation of Ki67 and the abnormal-type keratins (K) K6, K16, and K17, restoration of filaggrin and claudin-1, marked reduction of TEWL, and restoration of basement membrane and dermal collagen deposition, even under continuous sensitization of low dose TNCB. In addition, SP significantly reduced innervation of itch-evoking nerve fibers, gelatinase activity and nerve growth factor (NGF) expression in the epidermis but upregulated semaphorin-3A (Sema3A) expression in the epidermis, along with reduced scratching behavior in TNCB-treated NC/Nga mice. All of these effects were completely reversed by co-treatment with the NK1R antagonist CP99994. In cultured human keratinocytes, SP treatment reduced expression of TGF-α, but upregulated TGF-β and Sema3A.</P> <P><B>Conclusion</B></P> <P>Topically administered SP can restore normal skin barrier function, reduce epidermal infiltration of itch-evoking nerve fibers in the AD-like skin lesions, and alleviate scratching behavior. Thus, SP may be proposed as a potential medication for chronic dermatitis and AD.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Topical Substance P treatment reduced AD-like symptoms in TNCB-induced NC/Nga mice. </LI> <LI> Topical Substance P treatment reduced epidermal acanthosis and expressed normal keratin profiles in AD-like mice. </LI> <LI> Topical Substance P treatment regulated both small nerve attraction and repulsion factors in AD-like skin. </LI> <LI> Topical Substance P treatment recovered skin barrier function and reduced pruritus in AD-like mice. </LI> </UL> </P>
