        Candidate of Probiotic Bacteria Isolated from Several Jeotgals: Korean Traditional Fermented Seafoods

        Seventy eight bacterial strains were isolated from several jeotgals using MRS and M 17 agar media. The probiotic properties such as tolerance of extreme growth condition, production of antimicrobial compound, production of hydrogen peroxide, and enzymatic activity of bile salt hydrolase were investigated. DHK 4, 10, 21 and 74 strains showed a strong tolerance property against extreme conditions such as low pH and 0.5% oxgall-supplemented medium. DHK 10 and 47 strains produced hydrogen peroxide on TMB agar plate. DHK 8 and 10 strains produced antimicrobial compounds onto MRS agar against E. facalis. DHK 4, 6, 21, 29, 33, 63 and 87 strains had high activities of bile salt hydrolase. Especially, DHK 10 displayed a strong probiotic candidate; the abilities to produce the antimicrobial compound, hydrogen peroxide, and bile salt hydrolase. All these strains are assumed to be useful probiotic candidates. Among 78, twenty seven strains which have probiotic properties were tentatively identified by 16S rRNA sequencing. Among them, 7 Lactobacillus spp., 6 Leuconosotoc spp., 2 Weisella spp., 1 Pediococcus sp., 1 Staphylococcus sp., 1 Enterococcus sp. and 2 Streptococcus spp. were tentatively identified.

        IFATS Collection: Immunomodulatory Effects of Adipose Tissue-Derived Stem Cells in an Allergic Rhinitis Mouse Model

        <P>Adipose tissue-derived stem cells (ASCs) exhibit immunosuppressive effects in allogeneic transplantation. However, there is no report that evaluates the in vivo immune-modulating effect of ASCs in an experimental allergic rhinitis (AR) model. We investigated whether ASCs migrate to the nasal mucosa in an AR mouse model and evaluated the immune-modulating effect of ASCs in the AR mouse model. Cultured ASCs (2 x 10(6)) were injected i.v. before the first allergen challenge in the AR mouse model. Migration of ASCs to the nasal mucosa was evaluated by immunofluorescence. The immunomodulatory effects of ASCs were evaluated by nasal symptoms, histology, serum ovalbumin (OVA)-specific antibody, and the cytokine profile of the spleen. ASCs migrated to the nasal mucosa in the AR mouse model. ASCs significantly reduced allergic symptoms and inhibited eosinophilic inflammation in the nasal mucosa. ASCs significantly decreased the serum allergen-specific IgE level and the IgG(1)/IgG(2a) ratio and significantly increased the IgG(2a) level in the AR mouse model. ASCs inhibited interleukin (IL)-4 and IL-5 production from OVA-incubated splenocytes, but enhanced interferon-gamma production. In conclusion, ASCs can migrate to the nasal mucosa in the AR mouse model and inhibit eosinophilic inflammation partly via shifting to a T-helper 1 (Th1) from a Th2 immune response to allergens.</P>

        Immunohistochemical study on cytokeratin expression on developing tongue in Korean native goats (Capra hircus)

        Cytokeratin (CK) comprises the intermediate filament cytoskeleton of epithelial cells. Patterns of CK expression can be regarded as a specific marker for epithelial differentiation status. The aim of this study was to identify CK expression on tongues of Korean native goats ranging from 60-day-old fetuses to newborns during prenatal development using immunohistochemistry. The tongues of fetuses were removed from 2- to 4-year-old female Korean native goats by caesarean section performed under general anesthesia. Immunohistochemistry was performed to assess CK expression patterns on developing goat tongues using serial paraffin-embedded sections. Light zones signifying CK immunoreactivity in dorsal lingual epithelia were weakly positive in 60-day-old fetuses. In 90-day-old fetuses, deep areas in dorsal lingual epithelia were strongly positive for CK expression and superficial areas were moderately positive. In 120-day-old fetuses, light zones of lingual epithelia in the vallate papilla were strongly positive for CK expression, whereas ducts of von Ebner’s glands were moderately positive. In neonates, taste buds were positive for CK expression, whereas non-taste epithelial cells and von Ebner’s glands were negative. These findings indicate that goat tongues have different patterns of CK expression during development and provide a morphological basis for studies on the biological mechanism of epithelial differentiation.

        갑상선암에서 갑상선전절제술 후 저칼슘혈증의 예측

        Purpose: Postoperative hypocalcemia is a common complication compared with the other complications following total thyroidectomy due to thyroid carcinoma. We evaluated the incidence of transient and permanent hypocalcemia and the preservation status of the parathyroid glands following total thyroidectomy due to thyroid carcinoma. Methods: We analyzed a total of 145 cases of total thyroidectomy due to thyroid carcinoma. The preservation status of the prarathyroid glands after total thyroidectomy was classified as intact preservation or ischemic change, according to the number of intact preserved parathyroid glands in group 1 (more than 2), group 2 (1) or group 3 (0). Results: Permanent hypocalcemia was not found in group 1. The rate of transient or permanent hypocalcemia was significantly lower in group 2 than that in group 3 (P< 0.05). Conclusion: The parathyroid glands should be preserved to minimize the occurrence of hypocalemia after performing total thyroidectomy. The number of intact preserved parathyroid glands can be a good indicator for predicting the occurrence of post-total thyroidectomy hypocalcemia. (Korean J Endocrine Surg 2008;8:256-259)

        A light microscopic study on tongue development in Korean native goats (Capra hircus)

        The purpose of this study was to identify the composition and organization of lingual tissues underlying the histo-structural change of developing tongue in Korean native goats by light microscopy (LM). Tongues of the fetuses on days 60, 90, 120 and neonate were examined for the morphological development. In the 60-day-old fetuses, the tongue tissues were differentiated into epithelium, lamina propria and muscle layer. Primordia of filiform, conical, lentiform, fungiform and vallate papillae appeared and rudiments of taste bud were observed in the epithelia of the primordia of the gustatory papillae. The dorsal surface of the lingual epithelia showed a weak PAS positive reaction. Collagenous fibers and small blood vessels were shown in the connective tissues. In the 90-day-old fetuses, Von Ebner’s glands were moderately PAS positive while the muscle fibers and connective tissue were strongly positive for PAS. The collagenous fibers increased and came to have a more complex arrangement in the tongue. The muscle fibers were spread out at various directions and developed in striated muscle bundles. In the 120-day-old fetuses, taste buds were observed in the epithelia of the gustatory papillae, and several well-developed tissues visible such as blood vessels, collagenous fibers, muscle fiber bundles and Von Ebner’s glands. In the neonates, many taste buds were found in a transverse section of the vallate papilla. The muscle layers, Von Ebner’s glands, collagenous fibers and blood vessels were more developed than those of the 120-day old fetuses. These findings indicate that goat tongues have a variety of different shapes during prenatal development

        Facilitation of AMPA receptor-mediated steady-state current by extrasynaptic NMDA receptors in supraoptic magnocellular neurosecretory cells

        In addition to classical synaptic transmission, information is transmitted between cells via the activation of extrasynaptic receptors that generate persistent tonic current in the brain. While growing evidence supports the presence of tonic NMDA current (I<sub>NMDA</sub>) generated by extrasynaptic NMDA receptors (eNMDARs), the functional significance of tonic I<sub>NMDA</sub> in various brain regions remains poorly understood. Here, we demonstrate that activation of eNMDARs that generate INMDA facilitates the α-amino-3-hydroxy-5-methylisoxazole-4-proprionate receptor (AMPAR)-mediated steady-state current in supraoptic nucleus (SON) magnocellular neurosecretory cells (MNCs). In low-Mg<sup>2+</sup> artificial cerebrospinal fluid (aCSF), glutamate induced an inward shift in I<sub>holding</sub> (I<sub>GLU</sub>) at a holding potential (V<sub>holding</sub>) of –70 mV which was partly blocked by an AMPAR antagonist, NBQX. NBQX-sensitive IGLU was observed even in normal aCSF at V<sub>holding</sub> of –40 mV or –20 mV. IGLU was completely abolished by pretreatment with an NMDAR blocker, AP5, under all tested conditions. AMPA induced a reproducible inward shift in Iholding (I<sub>AMPA</sub>) in SON MNCs. Pretreatment with AP5 attenuated I<sub>AMPA</sub> amplitudes to ~60% of the control levels in low-Mg <sup>2+</sup> aCSF, but not in normal aCSF at V<sub>holding</sub> of –70 mV. I<sub>AMPA</sub> attenuation by AP5 was also prominent in normal aCSF at depolarized holding potentials. Memantine, an eNMDAR blocker, mimicked the AP5-induced I<sub>AMPA</sub> attenuation in SON MNCs. Finally, chronic dehydration did not affect I<sub>AMPA</sub> attenuation by AP5 in the neurons. These results suggest that tonic INMDA, mediated by eNMDAR, facilitates AMPAR function, changing the postsynaptic response to its agonists in normal and osmotically challenged SON MNCs.

        24시간 보행성 식도내압검사 및 식도 pH 검사를 이용한 역류성 식도염의 병인론에 대한 연구

        Background/Aims: There are still controversies on the pathogenesis of the development of esophagitis in patients with gastroesophageal reflux disease (GERD). The aim of this study was to evaluate the reflux characteristics in esophagitis GERD patients and non-esophagitis GERD patients. Methods: Forty one patients with typical reflux symptoms more than 6 months and an abnormal 24 hour ambulatory pH monitoring were enrolled. Eighteen patients had esophagitis on endoscopic examination and 23 did not. All patients performed 24 hour ambulatory pH and pressure monitoring after standard esophageal manometry. Results: The mean GER pH and the mean peak of GER pH in esophagitis GERD patients (Group A) and non-esophagitis GERD patients (Group B) showed no difference (pH 3.6±0.2, 2.9±0.3 vs pH 3.7±0.3, 3.1±0.5) respectively. The mean duration of GER in Group A showed longer tendency than that of Group B, but failed to show statistical difference (64.4±51.0 seconds vs 43.0±26.9 seconds). The number of reflux episode $gt; 5 min of GER in Group A and B showed no difference (3.78±2.78 vs 3.36±3.86) The mean percent of peristaltic contraction of esophageal body was significantly decreased in Group A compared with Group B (59.7±15.1% vs 67.8±7.8%, p$lt;0.05), and mean percent of isolated contraction was significantly increased in Group A compared with Group B (20.3±17.1% vs 9.9±7.9%, p$lt;0.05). There was no difference between Group A and B in the mean percent of simultaneous contractions, and mean percent of retrograde contractions. The LES pressure of Group A was significantly lower than that of Group B (18.0±7.9 mmHg vs 24.6±9.8 mmHg, p$lt;0.05). There was no difference between Group A and B in the DeMesster score (31.9±14.3 vs 40.6±40). There was no difference between Group A and B in amplitude of esophageal body. Conclusions: The esophagitis GERD patients showed decreased peristaltic contraction and increased isolated contraction of esophageal body compared to non-esophagitis GERD patients. It suggests that motility disorder plays a major role in the pathogenesis of esophagitis in GERD patients.
