중기 다장기 발암성시험법을 이용한 플라스틱 가소제의 전암단계 병변에 미치는 영향

This study was conducted to assess the modifying effects of diisodecyl phthalate (DIDP) and diethylhexyl phthalate (DEHP) in the rat medium-term multi-organ bioassay using five different carcinogens. Seventy five, 6-week-old, male F344 rats were divided into four groups. Animals were sacrificed at week 12. To evaluate modifying effects of preneoplastic lesions of DIDP and DEHP, we measured glutathione S transferase placental form (GST-P) and 5-bromodeoxyuridine (BrdU) expression level in liver, numbers of aberrant crypt foci (ACF) in colon and performed histopathological observation. The body weights of rats given 12,000 ppm DEHP and 8,000 ppm DIDP were significantly decreased compared to those of control group. Relative liver and kidney weights of rats given DIDP were significantly increased compared to those of control group in a dose dependent manner. There was no significance of GST-P positive foci and BrdU labeling indices in liver and numbers of ACF in colon among each group. Although hyperplasia of urinary bladder of rat given DEHP were significantly increased compared to that of control group, there were no significant changes of preneoplastic lesions on all organs of rats given DIDP. These results indicate that DIDP has no modifying effect on preneoplastic lesions in all organs.

Corrosion of reinforcement is a major problem affecting a large number of reinforced concrete structures, this paper presents the results of an experimental study on reinforced concrete beams to determine their residual flexural capacity after undergoing different corrosion degrees (corrosion length, corrosion location, corrosion rate) of reinforcing bars. Corrosion was induced in the laboratory by an accelerated corrosion technique using two sources of external power supply. The results show marked reductions in flexural strength due to reinforcement corrosion.
화학적 간발암모델에서 Bisphenol A가 간암의 전암성 및 종양성 병변에 미치는 영향

Bisphenol A(BPA) was known as an endocrine disrupting chemical. This study was conducted to assess the effect of BPA, weaker estrogen, on the preneoplastic and neoplastic lesions induced by diethylnitrosamine (DEN). One hundred male F344 rats were divided into four groups which were treated with DEN followed by BPA. To make liver tumor early, we conducted that DEN containing osmotic pump implanted into rat abdominal cavity. Then diet containing BPA were fed to the rats. All animals were sacrificed at 10 and 18 weeks. Body weights were significantly decreased in 4000ppm at 18 weeks. Relative Liver weights of 4000 ppm BPA treated group were significantly increased compared to that of DEN alone group at 18 weeks. There were no significant differences of liver tumor incidences. Sum area of GST-P positive foci and BrdU labeling indices of BPA treated group were not significantly different compared to those of control group. These results suggest that BPA have no effects in preneoplastic and neoplastic lesions on DEN-induced hepatocarcinogenesis.
미성숙 랫드 자궁비대반응시험을 이용한 DEHA의 내분비계 장애작용 평가

This study was aimed to investigate the estrogenic activity of Di-(2-ethylhexyl) adipate (DEHA) using immatured type uterotrophic assay. SD rats were treated with DEHA (40, 200, 1000mg/kg/day), estradiol-3-benzoate (EB) $(1{\mu}g/kg/day)$ as positive control on the assay. In immatured-type uterotrophic assay, relative organ weights of kidney and reproductive organs such as ovary at high-dose group were significantly increased compared to those of vehicle control group. DEHA did not influence the levels of serum FSH and LH, and uterine morphological changes such as luminal epithelial height, myometrial thickness and numbers of uterine gland, and BrdU indices. In these results, there was no significant variation by DEHA treatment, suggesting that DEHA appears not to be a endocrine disrupter with estrogenic activity.
06 식품의약품안전처 SESSION : 잔류농약의 일일섭취허용량 자료 연구

현재 식품 중 잔류농약 위해평가 체계에 대한 비교 연구는 거의 전무한 상태이며, 농약의 안전성 자료는 각 나라마다 상이할 수가 있으므로, CODEX, 유럽, 미국, 일본 등 국가별 자료를 비교.분석하여 국내 잔류농약의 위해평가 체계를 확립할 필요가 있다. 본 연구에서는 조사대상 농약 100 종에 대한 식품 중 잔류농약에 대한 코덱스, 일본, 유럽, 미국, 한국 등의 위해성 국제평가기관에서 발표한 안전성 자료를 분석한 후 프로파일을 작성하고, 국외 주요 독성 자료 Data base를 구축하여 식품 중 잔류농약 기준.규격 제.개정의 근거 자료로 활용하고, 안전관리 지원의 과학적 근거를 제공하고자 하였다. 식품을 통해 소량의 농약을 섭취하더라도 인체에서의 대사 반응에 의해 제거될 수 있으며, 일생동안 섭취를 해도 건강에는 영향이 없음을 국제적으로 과학적 근거로 제시하는 것이 ADI(Acceptance Daily Intake, 일일섭취허용량)이다. ADI는 국제기구나 각 국가의 농약관리기관에서 독성전문가들이 농약의 독성자료를 검토하여 위해성평가 전문가회의를 통해서 설정하게 된다. 각국의 안전성 자료를 분석한 결과 설정된 ADI는 주요국가 기관 등의 독성의 endpoint, SF, NOAEL 선정 등 설정지침에 따라 일반증상, 체중변화, 사료섭취량변화, 혈액학적 변화, 조직병리학적변화 등의 독성영향이 있었으며, 설정된 ADI의 근거 endpoint는 각국의 주요 기관의 안전성 자료에서 조직병리학적 판독에 근거한 병리학적 소견이 가장 많았고, 다음으로 생리학적 변화(혈액, 뇨 등)의 사례가 많은 것으로 나타났다. 또한 각국의 주요기관 농약의 안전성 자료에서 ADI값은 kasugamycin 등이 기관별로 상이한 것으로 조사되었다. 하지만 대부분의 경우는 ADI 값이 국가별로 동일하거나 유사하였으며, 주요국가 기관별 ADI 값의 차이는 안전계수의 차이와 실행한 독성시험의 종류가 다른 것으로 조사되었고, 각국 전문가의 독성자료에 대한 견해차이가 있으므로, 추후 농약의 안전성 자료에 대한 자세한 분석이 필요할 것으로 판단되었다.
