Analysis of the Caenorhabditis elegans dlk-1gene expression

C. elegans DLK-1 has been reported to play an important role in synaptogenesis by shaping the structure of presynaptic terminal. In this study, we investigated the expression pattern and regulation of the dlk-1 gene in C. elegans. To determine the expression pattern, we made a dlk-1::gfp fusion construct, named pPDdg1, which consisted of ~2.2kb 5' upstream region, the first exon, the first intron, and a part of the second exon of the dlk-1 gene. By microinjecting this construct into the worm, we observed that the DLK-1::GFP was expressed mainly in neurons. We next examined the regulatory elements of gene expression by deletion analysis of pPDdg1. Removal of a large portion of the 5' upstream region (-361 to -2246) of the gene had little effect on the expression pattern, whereas deletion of the first intron led to elimination of the DLK-1::GFP expression in most of the neurons. Our results suggest that the first intron of the C. elegans dlk-1 gene contains the regulatory element critical for gene expression.
Synthesis of fluorinated epoxy and epoxy acrylate

Recently, Fluorine-contained epoxy resins and epoxy acrylate resins are used in various areas like light curable refractive transfer for optical-fiber device, low dielectric materials for photography signal device and the adhesive for low dielectric PCB and optical device etc. Specially, as the technology of optical transportation develops, there needs the high optical transparence without light scattering in the adhesion of optical passage and the proper refractive index in 1.3~1.5㎛ wave length for optical tranportation. To obtain the optimum properties, fluorine-contained epoxy resins and epoxy acrylate resins have been used. According to applied fields, there needs to reduce the byproducts and make the low molecular weight resin to optimize the properties. In this study, we synthesized the low molecular weight fluorine-contained epoxy resin with changing the catalysts on reaction conditions and synthesized the fluorine-contained epoxy acrylate resin using the synthesized epoxy resin, and characterized their properties.

Because welding residual stress is formidable result in electric resistance spot welding process, and it detrimentally affect to fatigue crack initiation and growth at nugget edge of spot welded la p joints, it should be considered in fatigue analysis. Thus, accurate prediction of residual stress is very important. In this study, nonlinear finite element analysis on welding residual stress generated in process of the spot welding was conducted, and their results were compared with experimental data measured by X-ray diffraction method. By using their results, the maximum principal stress considered welding residual stress at nugget edge of the spot welded lap joint subjected to tension-shear load was calculated by superposition method. And, the $\Delta$P- $N_f$ relations obtained through fatigue, tests on the IB-type spot welded lap joints was systematically rearranged with the maximum principal stress considered welding residual stress. From the results, it was found th2at fatigue strength of the IB-type spot welded lap joints could be systematically and more reasonably rearranged by the maximum principal stress($\sigma$1max-res considered welding residual stress at nugget edge of the spot welding point.

4,4`-Dihydroxy-α-methylstilbene을 주쇄로 하여 액정성을 나타내는 열경화형 에폭시 올리고머의 양말단에 아크릴기를 도입하여 광경화형 에폭시 아크릴레이트 올리고머를 제조하고 액정거동과 광경화 반응성을 검토하였다. 4,4`-Dihydroxy methylstilbene과 epichlorohydrin을 반응시켜 4,4`-dihydroxy methylstilbene diglycidyl ether를 제조한 후, 톨루엔을 반응희석제로 사용하여 상이동촉매 하에서 acrylic acid와 12 h 반응시켜 4,4`-dihydroxy methylstilbene glycerolate diacrylate를 제조하였다. 제조된 에폭시 아크릴레이트 올리고머는 상온에서 49 ℃ 영역까지 smectic 액정상을 나타내었다. 제조된 에폭시 아크릴레이트 올리고머에 반응회석제를 사용하여 광경화 반응을 하였을 경우 소량의 광개시제에서도 높은 전환율의 광경화 반응이 이루어졌다. Liquid crystalline(LC) epoxy acrylate containing the mesongenic core of α-methylstilbene was synthesized. The epoxy oligomers were synthesized with 4,4`-dihydroxy-α-methylstilbene and epichlorohydrin. The epoxy acrylate oligomer was synthesized from epoxy oligomer and acrylic acid in the presence of a reaction medium and a phase transfer catalyst for 12 h. The synthesized epoxy acrylate oligomer showed smectic LC phase from room temperature to near 50 ℃ compared with the nematic epoxy oligomer. UV-photo curing of synthesized eposy acrylate with a reactive diluent photo-initiator was performed. The synthesized epoxy acrylate showed high photo-reactivity.

본 연구에서는 버섯류의 중금속 함량 실태조사를 통해 우리나라 국민이 식품으로써 버섯을 섭취할 경우 중금속 노출수준에 따른 위해성을 평가하였다. 버섯류의 중금속 함량 실태조사를 위해 국내 유통 중인 버섯류 17개 품목, 536건을 수거하였으며 버섯류 중 납 함량을 조사한 결과, 생물버섯은 평균 0.005(큰 느타리)-0.026(송이) mg/kg이었고 건조버섯은 평균 0.014(느타리)-16.411(석이) mg/kg이었다. 카드뮴 함량은 생물버섯의 경우 평균 0.002(팽이)-0.205(노루궁뎅이) mg/kg이었고 건조버섯은 평균 0.021(동충하초)-2.650(신령) mg/kg이었다. 따라서, 국내 유통 중인 버섯류의 중금속 함량은 타 문헌과 유사하거나 낮은 수준이었다. 노출 및 위해성 평가를 위해 버섯별 섭취를 통한 중금속 노출량을 산출한 후, Joint FAO/WHO Expert Committee on Food Additives(JECFA)에서 설정한 PTWI 또는 PTMI값 대비 위해도를 평가하였다. 버섯 섭취로 인한 전체국민의 평균 납 노출량은 0.053 μg/day으로 PTWI 대비 0.03% 수준이었으며, 카드뮴 평균 노출량은 0.168 μg/day으로 PTMI 대비 0.37% 수준이었다. 따라서, 국내 유통되는 버섯을 식품으로써 섭취할 경우 전체국민의 중금속 평균 노출수준은 JECFA의 인체노출안전기준보다 낮은 위해도를 보임으로써, 우리나라 국민은 버섯에 존재하는 중금속의 위해성으로부터 안전한 것으로 사료된다. The present study was carried out in order to assess the dietary exposure as well as the risk for lead (Pb) and cadmium (Cd) exposure through mushrooms among the Korean population. Various samples (n=536) covering 17 kinds of mushrooms were collected from retail outlets and markets in Korea. The contents of Pb were 0.005 (King oyster)-0.026 (Matsutake) mg/kg in raw mushrooms and 0.014 (Oyster)-16.411 (Manna lichen) mg/kg in dry mushrooms, respectively. The contents of Cd were 0.002 (Enoki)-0.205 (Hericium erinaciium) mg/kg in raw mushrooms and 0.021 (Vegetable worms)-2.650 (Agaricus blazei Muill) mg/kg in dry mushrooms, respectively. The mean dietary exposure of Pb for the general population was 0.053 μg/day, taking 0.03% of provisional tolerable weekly intake (PTWI). The mean dietary exposure of Cd for the general population was 0.168 μg/day, corresponding to 0.37% of provisional tolerable monthly intake (PTMI). Therefore, the level of the overall dietary exposure to heavy metals through mushroom for the Korean population was far below the recommended JECFA levels, indicating little possibility of concern.
