        전나무 부산물 추출물의 항산화 및 항박테리아 활성

        Background: The extract of Abies holophylla is used as an ingredient in cosmetics. This study assessed the antioxidant and antibacterial activities of the material remaining after the extract is used. Methods and Results: The 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azinobis (3-ethyl benzothiazoline)-6-sulfonic acid (ABTS) radical scavenging abilities were assessed to determined the free radical scavenging activity. The total phenol and flavonoid contents were determined to measure the antioxidant activity. The DPPH and ABTS radical scavenging activities of the resudual extract were higher (95.61 - 99.42% and 74.26 - 77.98% in water extract respectively) than those of the positive control. In 50% EtOH extract, the total phenol content was 389.84 ㎎·GAE/㎖, and the total flavonoid was 0.15 ㎎·QE/㎖. The minimum inhibition concentration degree for antibacterial activity against Staphylococcus aureus was < 8 to < 125 ㎍/㎖ compared to that of the positive control in all extracts. The clear zone against S. aureus was found to be 12.2 ± 3.8 ㎜. Conclusions: The A. holophylla byproducts were found to have antioxidant and antibacterial activities. Therefore, the materials remaining after the A. holophylla extract is used in cosmetics has potential functional uses.         The Mediating Effect of Job Esteem on The Relationship Between Customer Misbehavior Experienced by Beauty Workers And Their Job Continuity

        This study aims to verify the mediating effect of job esteem on the relationship between customer delinquent behavior experienced by beauty workers, who are subjected to longer and more frequent face-to-face interactions with clients compared to workers in other industries, and their job continuity. Correspondingly, this study surveyed 496 individuals between the ages of 20 and 50, who are currently working in the beauty service industry where building and maintaining customer relationships is crucial in an attempt to confirm whether cultivating job satisfaction and a sense of self-worth has a direct influence on job continuity. As a result of the analysis, customer misbehavior was found to have significant and negative impact on job esteem. The frequency of such experiences is inversely related to the level of job esteem; the more incidents of customer misbehavior, the lower the job esteem of the workers. On the other hand, job esteem was found to have significant and positive effects on job continuity, indicating that the job continuity of beauty workers would decrease due to the negative impact of customer delinquent behavior. However, this study revealed that experiencing customer misbehavior does not have directly impact on the job continuity of beauty workers. Instead, customer delinquent behavior was observed to have a less direct effect on job continuity compared to the influence of job esteem on job continuity, implying that the decrease in job esteem resulting from customer misbehavior affects job continuity. Thus, in order to improve job continuity among beauty workers, measures must be taken to enhance their job esteem. This can be achieved through several means, including fostering job value consciousness, implementing an effective organizational reward system, providing education on emotion control, maintaining a positive organizational culture that values emotional and psychological well-being, and operating human service systems while taking into account the individual's self-efficacy and self-esteem.

        들깨 Limonene 유전자의 담배식물(Nicotiana benthamiana)내 Agroinfiltration에 의한 분자적 특성

        The full-length cDNA encoding Perilla frutescens limonene synthase (PFLS) (603 amino acids, GenBank accession no. D49368) was cloned. To elucidate the role of PFLS in gene regulation, we transiently transformed full-length PFLS into tobacco plants. PFLS mRNA was first detected in the intact leaves of the plants at 6 h, and the LS transcript level increased after 12 h in leaves treated with oxidative stress-related chemicals. The transient overexpression of PFLS resulted in increased transcription of NbPR1 and NbSIP in Nicotiana benthamiana leaves. Thus, our result confirmed that the infiltration of PFLS gene act as a transcriptional regulator of NbPR1 or NbSIP genes in the tobacco.

        맥문동 뿌리줄기의 저장력 향상을 위한 기능성필름 활용

        Background: The objective of this study was to investigate the effect of packaging material on the growth of rootstock of Liriope platyphylla. Methods and Results: This study examined the effects of two types of packaging material, LDPE (low density polyethylene) and functional film on the growth of the tubers of L. platyphylla, at 5℃. During the 16-weeks of storage period, the ratio of loss and decay of the tubers was examined at intervals of 4, 8, and 16 weeks to detect the quality of the plant. After 16 weeks of storage, the treated tubers were own. Subsequently, plant height and the number of leaves were recorded. The results revealed that functional film at 5℃ was the ideal material for the storage of L. platyphylla tubers. The rate of loss was the highest (57.42%) with a onion net and the lowest (22.12%) with a functional film. Similarly, the rate of tuber decay was highest (8.20%) using onion net and the least (4.60%) when the functional film was used. Conclusions: Thus, the use of the functional film proved to be the most effective in the storage of L. platyphylla tubers when compared with the LDPE.

        Agrobacterium 농도가 사과 형질전화 효율에 미치는 영향

        This study was conducted to find optimum bacterial density for improving the efficiency of transformation mediated by Agrobacterium tumefaciens in apples. Regeneration(15%) and transformation frequency(10%) were increased in resuspension-culture density $A_{600}$ 1.3 from preculture density $A_{600}$ 0.7 of Agrobacterium tumefaciens in ′Fuji′. In ′Gala′, 20% regeneration and 16% transformation frequency were observed at optimum bacterial density $A_{600}$ 0.7 form preculture density $A_{600}$ 1.3. ′Mclntosh as well as "Gala" were 25%regeneration and 10% transformation frequency. Hence a frequency optimum condition of bacterial density for the efficient transformation of apple could be depend on apple genotypes.

        바이오에너지 자원으로서 강원도 지역에서 수집된 수수의 작물학적 특성 평가

        1. 수수 성숙기에 간장 길이를 조사한 결과 6개체(12.2%) 가 300 cm 이상의 길이로 바이오매스가 우량한 것으로 파악되었으며, 10개체(20.4%)정도는 200~300 cm 사이의 평균 길이를 나타내었다. 2. 성숙기의 수수 줄기 직경에는 큰 차이를 보여준 개체는 없었으며, 대부분의 수수 수집종들은 2.2±0.1 cm 정도의 직경을 갖는 것으로 드러났다. 3. 본 연구에 시험한 수수 수집종들은 엽장이 60~70 cm 정도의 길이에 속하는 유전자원이 22 개체가 되는 것으로 나타났으며, 7개체는 70 cm 이상되는 것으로 나타났다. 4. 엽형은 49% 정도의 개체가 협단으로 분류되었고, 대부분의 엽맥색은 백색으로 나타났다. 5. 본 연구에 사용된 대부분의 수수의 수형은 반밀수 타원형이 많은 것으로 나타났으며, 수장은 평균적으로 20~30 cm에 달하는 것으로 나타났다. Forty nine accessions of the Sorghum bicolor (L) Moench collected from different agronomical zone of Gangwondo province, Korea were characterized morphologically on field trial. Plant length, stem diameter, tiller number, ear type, ear length, ear width, leaves length, leaves width and leaf color differed significantly in the cultivated accessions. Out of forty nine test accessions, six accessions (12.2%) showed comparatively taller height, measuring more than 300 cm, while, other ten test accessions (20.4%) were measured from 200~300 cm. There were no significant differences in the diameter of stem in the collected test accessions (2~3 cm). Of the total test accession, seven accessions showed leaf length of over 70 cm, and twenty two test accession revealed leaf length between 60-70 cm. Forty nine of the collected test accessions showed narrow and short leaves with white mid vain. These results provided additional information for the breeding programs and conservation of Sorghum bicolor as a bio-energy crop.

      • 열대 약용식물 Clausena excavata의 RNA-seq 및 SSR 분석

        Background : Several members of the genus Clausena have a great potential as a candidate for the identification of new drug lead molecules, but lack of their genomic information can be a hindrance for the verification of the genetic background for future use. To broaden and delve into the genomic features of this genus, Clausena excavata, an important medicinal plant in many Asian countries, was used for RNA-seq analysis. Methods and Results : A ten ㎍ of the total RNA was used for mRNA isolation using oligo-dT beads and random sheared mRNAs were used to prepare a cDNA library for a illumina hiseq 2500 analysis. In total, 17,580,456 trimmed clear reads from the illumina hiseq 2500 were used for de novo assembly using three assemblers, CLC genomics workbench, velvet-oases, and Trinity. A total of 16,638 non-redundant unigenes with an average length of 755 bp were generated by the assembly. The functional categorization of the identified unigenes by a gene ontology (GO) term resulted in 2,305 genes in the cellular component, 5,577 in the biological processes, and 8,056 in the molecular functions, respectively. The top sub-category in biological processes was the metabolic process with 4,374 genes. Among annotated genes, 3,006 were mapped to 123 metabolic pathways by KEGG metabolic pathway analysis tool. The search for simple sequence repeats (SSRs) resulted in 845 SSRs from 749 SSR-containing unigenes and the most abundant SSR motifs was AAG/CTT with 179 occurrences. Twelve SSR markers were tested for cross transferability among five Clausena species; eight of them exhibited polymorphism. Conclusion : In the present study, genomic resources of the genus Clausena were enriched through RNA-seq and SSR markers, which will serve as valuable resources for genomic/genetic studies of the genus Clausena and close relatives.
