<P>The pharmacokinetics and metabolism of 2'-benzoyloxycinnamaldehyde (BCA) was characterized in male Sprague-Dawley rats as part of the preclinical evaluations for developing this compound as an antitumour agent. BCA was not detected in the plasma following either intravenous or oral dose, whereas its putative metabolites 2'-hydroxycinnamaldehyde (HCA) and o-coumaric acid were present at considerable levels. In separate pharmacokinetics studies, HCA exhibited a high systemic clearance and a large volume of distribution, whereas both pharmacokinetic parameters were much lower for o-coumaric acid. The terminal half-life of both metabolites was approximately 2 h. BCA was converted rapidly to HCA in rat serum, liver microsomes and cytosol in vitro; HCA was subsequently converted to o-coumaric acid in a quantitative manner only in the liver cytosol. In addition, the formation of o-coumaric acid was inhibited significantly by menadione, a specific inhibitor for aldehyde oxidase. Taken collectively, the results suggest that the rapid systemic clearance of HCA is likely due mainly to hepatic clearance occurring from aldehyde oxidase-catalysed biotransformation to o- coumaric acid. In conclusion, the present work demonstrates that the anticancer drug candidate BCA is highly likely to work as its active metabolite HCA in the body.</P>
Evaluation of ST2 and NT-proBNP as cardiac biomarkers in dogs with heartworm disease

This study compared serum concentrations of suppression of tumorigenicity 2 (ST2) and N-terminal pro-B-type natriuretic peptide (NT-proBNP) between healthy and heartworm- infected dogs. Eighteen heartworm-infected dogs and five healthy dogs were included in the study. Dogs were diagnosed and categorized by history, clinical signs, and blood assay, thoracic radiography, echocardiography, and commercial ELISA kit results. Serum samples were sent to the IDEXX reference laboratory for NT-proBNP measurement. ST2 was examined by using a canine interleukin 33 receptor ELISA kit with the quantitative sandwich ELISA method. The severely infected group showed significant elevation of NT-proBNP concentration over those of the control (P=0.03) and mildly infected (P=0.04) group. There were no significant difference in ST2 concentrations among the three groups. The usefulness of NT-proBNP as a cardiac biomarker in dogs with severe heartworm disease was confirmed by the results of this study. Further investigations to assess ST2 as a cardiac biomarker are warranted.

Purpose: This study aimed to evaluate the long-term clinical outcomes based on the ligation level of the inferior mesenteric artery (IMA) in patients with rectal cancer.Methods: This was a retrospective analysis of a prospectively collected database that included all patients who underwent elective low anterior resection for rectal cancer between January 2013 and December 2019. The clinical outcomes included oncological outcomes, postoperative complications, and functional outcomes. The oncological outcomes included overall survival (OS) and relapse-free survival (RFS). The functional outcomes, including defecatory and urogenital functions, were analyzed using the Fecal Incontinence Severity Index, International Prostate Symptom Score, and International Index of Erectile Function questionnaires.Results: In total, 545 patients were included in the analysis. Of these, 244 patients underwent high ligation (HL), whereas 301 underwent low ligation (LL). The tumor size was larger in the HL group than in the LL group. The number of harvested lymph nodes (LNs) was higher in the HL group than in the LL group. There were no significant differences in complication rates and recurrence patterns between the groups. There were no significant differences in 5-year RFS and OS between the groups. Cox regression analysis revealed that the ligation level (HL vs. LL) was not a significant risk factor for oncological outcomes. Regarding functional outcomes, the LL group showed a significant recovery in defecatory function 1 year postoperatively compared with the HL group.Conclusion: LL with LNs dissection around the root of the IMA might not affect the oncologic outcomes comparing to HL; however, it has minimal benefit for defecatory function.

<P><B>Background</B></P><P>In template-based modeling when using a single template, inter-atomic distances of an unknown protein structure are assumed to be distributed by Gaussian probability density functions, whose center peaks are located at the distances between corresponding atoms in the template structure. The width of the Gaussian distribution, the variability of a spatial restraint, is closely related to the reliability of the restraint information extracted from a template, and it should be accurately estimated for successful template-based protein structure modeling.</P><P><B>Results</B></P><P>To predict the variability of the spatial restraints in template-based modeling, we have devised a prediction model, Sigma-RF, by using the random forest (RF) algorithm. The benchmark results on 22 CASP9 targets show that the variability values from Sigma-RF are of higher correlations with the true distance deviation than those from Modeller. We assessed the effect of new sigma values by performing the single-domain homology modeling of 22 CASP9 targets and 24 CASP10 targets. For most of the targets tested, we could obtain more accurate 3D models from the identical alignments by using the Sigma-RF results than by using Modeller ones.</P><P><B>Conclusions</B></P><P>We find that the average alignment quality of residues located between and at two aligned residues, quasi-local information, is the most contributing factor, by investigating the importance of input features used in the RF machine learning. This average alignment quality is shown to be more important than the previously identified quantity of a local information: the product of alignment qualities at two aligned residues.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1186/s12859-015-0526-z) contains supplementary material, which is available to authorized users.</P>

<P>We propose the Combined Symbol-based Closed-Loop Orthogonal Space-Time Block Code (CS-CL-OSTBC) for four transmit antennas. In the multiple antenna systems, the CS-CL-OSTBC not only achieves full rate and full diversity with linear maximum-likelihood detection but also obtains higher feedback gain than existing CL-OSTBCs due to more efficient utilization of channel feedback information. In the proposed scheme, all the complex-valued channel coefficients are rotated to positive real values with exact channel phase feedback information. As a result, the channel gain can be expressed as the square of the sum of all positive real values and can obtain the maximum value without any loss. Simulation results on bit error rate performance show that the CS-CL-OSTBC outperforms existing CL-OSTBCs for various modulation schemes.</P>
LDPC-Coded Single Symbol Decodable Quasi-Orthogonal Space-Time Block Code

In this paper, we consider a concatenation scheme of a LDPC (Low-Density Parity-Check) code and a new QOSTBC (Quasi-Orthogonal Space-Time Block Code). The QOSTBC with four transmit antennas generally achieves full rate, a half or full diversity, while its decoding complexity becomes high by using a pair of maximumlikelihood detectors. To improve this decoding complexity, we propose a new QOSTBC scheme named SSD-QOSTBC (Single Symbol Decodable Quasi-Orthogonal Space-Time Block Code). As this scheme converts a non-linear decoding process of the QOSTBC to a linear process, the decoding complexity is equal to the OSTBC (Orthogonal Space-Time Block Code). Moreover, the SSD-QOSTBC can be linearly concatenated with LDPC codes by its linear decoding. Simulation results show that the LDPC-coded SSDQOSTBC is superior in terms of BER performance to the LDPC-coded OSTBC in quasi-static flat Rayleigh fading channels.
