<P>The pharmacokinetics and metabolism of 2'-benzoyloxycinnamaldehyde (BCA) was characterized in male Sprague-Dawley rats as part of the preclinical evaluations for developing this compound as an antitumour agent. BCA was not detected in the plasma following either intravenous or oral dose, whereas its putative metabolites 2'-hydroxycinnamaldehyde (HCA) and o-coumaric acid were present at considerable levels. In separate pharmacokinetics studies, HCA exhibited a high systemic clearance and a large volume of distribution, whereas both pharmacokinetic parameters were much lower for o-coumaric acid. The terminal half-life of both metabolites was approximately 2 h. BCA was converted rapidly to HCA in rat serum, liver microsomes and cytosol in vitro; HCA was subsequently converted to o-coumaric acid in a quantitative manner only in the liver cytosol. In addition, the formation of o-coumaric acid was inhibited significantly by menadione, a specific inhibitor for aldehyde oxidase. Taken collectively, the results suggest that the rapid systemic clearance of HCA is likely due mainly to hepatic clearance occurring from aldehyde oxidase-catalysed biotransformation to o- coumaric acid. In conclusion, the present work demonstrates that the anticancer drug candidate BCA is highly likely to work as its active metabolite HCA in the body.</P>

<P>Pigs with severe combined immunodeficiency (SCID) may provide useful models for regenerative medicine, xenotransplantation, and tumor development and will aid in developing therapies for human SCID patients. Using a reporter-guided transcription activator-like effector nuclease (TALEN) system, we generated targeted modifications of recombination activating gene (<I>RAG</I>) 2 in somatic cells at high efficiency, including some that affected both alleles. Somatic-cell nuclear transfer performed with the mutated cells produced pigs with <I>RAG2</I> mutations without integrated exogenous DNA. Biallelically modified pigs either lacked a thymus or had one that was underdeveloped. Their splenic white pulp lacked B and T cells. Under a conventional housing environment, the biallelic <I>RAG2</I> mutants manifested a “failure to thrive” phenotype, with signs of inflammation and apoptosis in the spleen compared with age-matched wild-type animals by the time they were 4 wk of age. Pigs raised in a clean environment were healthier and, following injection of human induced pluripotent stem cells (iPSCs), quickly developed mature teratomas representing all three germ layers. The pigs also tolerated grafts of allogeneic porcine trophoblast stem cells. These SCID pigs should have a variety of uses in transplantation biology.</P>

Purpose: This study aimed to evaluate the long-term clinical outcomes based on the ligation level of the inferior mesenteric artery (IMA) in patients with rectal cancer.Methods: This was a retrospective analysis of a prospectively collected database that included all patients who underwent elective low anterior resection for rectal cancer between January 2013 and December 2019. The clinical outcomes included oncological outcomes, postoperative complications, and functional outcomes. The oncological outcomes included overall survival (OS) and relapse-free survival (RFS). The functional outcomes, including defecatory and urogenital functions, were analyzed using the Fecal Incontinence Severity Index, International Prostate Symptom Score, and International Index of Erectile Function questionnaires.Results: In total, 545 patients were included in the analysis. Of these, 244 patients underwent high ligation (HL), whereas 301 underwent low ligation (LL). The tumor size was larger in the HL group than in the LL group. The number of harvested lymph nodes (LNs) was higher in the HL group than in the LL group. There were no significant differences in complication rates and recurrence patterns between the groups. There were no significant differences in 5-year RFS and OS between the groups. Cox regression analysis revealed that the ligation level (HL vs. LL) was not a significant risk factor for oncological outcomes. Regarding functional outcomes, the LL group showed a significant recovery in defecatory function 1 year postoperatively compared with the HL group.Conclusion: LL with LNs dissection around the root of the IMA might not affect the oncologic outcomes comparing to HL; however, it has minimal benefit for defecatory function.

본 논문은 미분방정식의 수치해를 메모리와 프로그래밍언의 사용등 많은 제한점을 가진 마이크로 컴퓨터를 이용해서 풀 수 있는 방법을 제시해서 마이크로 컴퓨터의 활용도를 높이고저 한 것이다. 본교가 소유하고 있는 마이크로 컴퓨터, cannon BX-1,는 16KB메모리를 갖고 있는 디스케트를 사용하며 BASIC어를 사용한다. 수치해법중 Runge-Kutta방법 특히 Ralston Runge-Kutta방법을 사용한 프로그래밍을 짜서, 참값과 근사치의 비교를 그래프로 나타냈으며 고계미분방정식을 다루었고 또한 다단계방법(Adams Bashfold method), Predictor-Corrector방법을 다루었다.
Evaluation of ST2 and NT-proBNP as cardiac biomarkers in dogs with heartworm disease

This study compared serum concentrations of suppression of tumorigenicity 2 (ST2) and N-terminal pro-B-type natriuretic peptide (NT-proBNP) between healthy and heartworm- infected dogs. Eighteen heartworm-infected dogs and five healthy dogs were included in the study. Dogs were diagnosed and categorized by history, clinical signs, and blood assay, thoracic radiography, echocardiography, and commercial ELISA kit results. Serum samples were sent to the IDEXX reference laboratory for NT-proBNP measurement. ST2 was examined by using a canine interleukin 33 receptor ELISA kit with the quantitative sandwich ELISA method. The severely infected group showed significant elevation of NT-proBNP concentration over those of the control (P=0.03) and mildly infected (P=0.04) group. There were no significant difference in ST2 concentrations among the three groups. The usefulness of NT-proBNP as a cardiac biomarker in dogs with severe heartworm disease was confirmed by the results of this study. Further investigations to assess ST2 as a cardiac biomarker are warranted.
