Karyopherin-α3 (KPNA3), a karyopherin- α isoform, is intimatelyassociated with metastatic progression via epithelial-mesenchymaltransition (EMT). However, the molecular mechanismunderlying how KPNA3 acts as an EMT inducer remains to beelucidated. In this report, we identified that KPNA3 was significantlyupregulated in cancer cells, particularly in triple-negativebreast cancer, and its knockdown resulted in the suppressionof cell proliferation and metastasis. The comprehensivetranscriptome analysis from KPNA3 knockdown cells indicatedthat KPNA3 is involved in the regulation of numerous EMTrelatedgenes, including the downregulation of GATA3 andE-cadherin and the up-regulation of HAS2. Moreover, it wasfound that KPNA3 EMT-mediated metastasis can be achievedby TGF-β or AKT signaling pathways; this suggests that thenovel independent signaling pathways KPNA3-TGF-β-GATA3-HAS2/E-cadherin and KPNA3-AKT-HAS2/E-cadherin are involvedin the EMT-mediated progress of TNBC MDA-MB-231 cells. These findings provide new insights into the divergent EMTinducibility of KPNA3 according to cell and cancer type.
Inhibition of Wntless/GPR177 suppresses gastric tumorigenesis

Wntless/GPR177 functions as WNT ligand carrier protein and activator of WNT/β-catenin signaling, however, its molecular role in gastric cancer (GC) has remained elusive. We investigated the role of GPR177 in gastric tumorigenesis and provided the therapeutic potential of a clinical development of anti-GPR177 monoclonal antibodies. GPR177 mRNA expression was assessed in GC transcriptome data sets (GSE15459, n = 184; GSE66229, n = 300); protein expression was assessed in independent patient tumor tissues (Yonsei TMA, n = 909). GPR177 expression were associated with unfavorable prognosis [log-rank test, GSE15459 (P = 0.00736), GSE66229 (P = 0.0142), and Yonsei TMA (P = 0.0334)] and identified as an independent risk predictor of clinical outcomes: GSE15459 [hazard ratio (HR) 1.731 (95% confidence interval; CI; 1.103- 2.715), P = 0.017], GSE66229 [HR 1.54 (95% CI, 1.10-2.151), P = 0.011], and Yonsei TMA [HR 1.254 (95% CI, 1.049- 1.500), P = 0.013]. Either antibody treatment or GPR177 knockdown suppressed proliferation of GC cells and sensitized cells to apoptosis. And also inhibition of GPR177 suppresses in vitro and in vivo tumorogenesis in GC cells and inhibits WNT/β-catenin signaling. Finally, targeting and inhibition of GPR177 with antibody suppressed tumorigenesis in PDX model. Together, these results suggest GPR177 as a novel candidate for prognostic marker as well as a promising target for treatment of GC patients. [BMB Reports 2018; 51(5): 255-260]

이 연구의 목적은 캐나다에서 개발된 부모용 유아 놀이 선호도 측정도구(Finegan et al., 1991)를 우리나라에 적용하여 개념적, 통계적 타당화를 검증하는 데 있다. 이 연구의 대상자는 서울지역 중산층인 만 4, 5, 6세 유아의 부모 295명이었다. 문항분석 결과, 부모들의 응답과정을 거친 15문항은 탐색적 요인분석결과 활동-모험 놀이, 운동놀이, 거친 놀이, 그리고 조용한 놀이의 네 가지로 나타났다. 첨도와 왜도는 모두 .57∼-1.10범위 안에, 문항과 전체척도간의 상관관계는 모든 문항이 .40이상의 높은 신뢰도를 갖고 있었다. 부모에 의해 유아의 놀이 선호도를 검사하는 이 검사도구는 유아 놀이 선호정도와 밀접한 연관성이 있음이 증명되었다. 확인적 요인분석결과, 4요인 14문항과 3요인 13문항 모두 적합도지수를 수용하고 있었다(CFI, GFI:.90; RMR: .048: RMSEA; .079). 만약 부모용 유아 놀이 선호도 검사도구의 문항을 3개 요인으로 구성한다면, 활동-모험 놀이와 운동놀이를 한 요인으로 간주할 수 있다. 연구결과, 부모용 유아 놀이 선호도 측정도구는 문항 신뢰도와 타당도가 안정성 있게 검증되었다. This study attempts to conceptually and statistically validate the Play Activity Questionnaire(PQ), a parent report measure of children's play preferences developed in Canada(Finegan et al., 1991). This study surveyed 295 middle class parents of 4-to 6-year-old children. In the result of questionnaire analysis, the 15-item PQ was completed by parents of children, and exploratory factor analysis revealed four play factors: Active and Adventurous, Athletic, Rough-and-Tumble, and Quiet. In addition, both krutosis and skewness have reliability within the range from .57 to -1.10, and interrelationship between questionnaire and all scales has high reliability of more than .40 for all questions. The Play Activity Questionnaire(PQ), a parent report measure of children's play preferences indicates that it has a close connection with children's play preferences. In the result of confirmatory factor analysis, all of 14 questions for 4 factors and 13 questions for 3 factors accepted person-fit index(CFI, GFI:.90; RMR: .048: RMSEA; .079). Accordingly, if PQ is consisted of 3 factor, 'Active and Adventurous' and 'Athletic' can regard as one factor. In the research result, validation of a parent report measure of children's play preferences was verified stably in terms of reliability and validity of questions.

기후변화 대응을 위한 온실가스 감축 수단으로 신재생에너지 기술의 중요성이 날로 부각되고 있다. 해수냉난방시스템은 해수의 열원으로 냉난방을 공급하는 기술로서 국내에서도 곧 실용화가 될 것으로 전망된다. 그러나 이 시스템은 초기 투자비용이 높기 때문에 경제성 확보 방안이 필요하다. CDM은 교토의정서의 온실가스 감축사업 중 하나로 사업 등록 시 온실가스감축 크레딧(CERs)을 발급 받으며 이를 거래할 수 있다. CDM사업 등록을 위한 가장 중요한 문제는 베이스라인 및 모니터링 방법론을 결정하는 것이다. 따라서 본고는 해수냉난방시스템 방법론으로 ACM-I.C.(Version 19)의 적용 가능성을 검토하고, 사례분석을 통하여 방법론 적용에 있어 이슈가 되는 문제들을 고찰하고자 한다. The importance of renewable energy technology is being emphasized in greenhouse gas reduction portfolio for corresponding climate change. The Sea Water Air Conditioning(SWAC) system is technology that supplies heating and cooling as sea water source and the technology will soon be commercialized in Korea. However, we need to look to ways of improving economic feasibility because the system is high initial investment cost. The Clean Development Mechanism(CDM) is one of the flexibility mechanisms defined in the Kyoto Protocol that provides for emissions reduction projects which generate Certified Emission Reduction units(CERs) which may be traded in emissions trading schemes. The most important issue for registering CDM projects is to determine the baseline and monitoring methodology. We reviewed about CDM project application method of SWAC system.

The study was carried out in order to investigate the effects of various factor on regeneration and growth of inflorescence stalk of Hyacinthus orientalis cvs. Carnegie, Delft Blue, Jan Bos, Pink Pearl. Ability of bulblet regeneration of inflorescence stalk tissue was good in 1 mg/L NAA+1 mg/L BA, The ability of bulblets regeneration from inflorescence stalk tissue with inflorescence was better than that without inflorescence and the bulblet regeneration ability and growth according to the physiological age of explant were promoted more in young tissue before anthesis than in the mid-mature tissue during anthesis. Bulblet regenrration and root fomation of inflorescence stalk tissue were effective Murashige and Skoog's medium. The growth of regenerated bulblet was promoted on medium supplemented with 0.1 mg/L NAA, 0.1mg/L NAA+0.1mg/L BA, 0.1mg/L NAA+BA or 1mg/L IBA in inflorescence stalk Culture.
Evaluation of Welding Quality using Servo-Gun Displacement Data for Field Spot Welding

To ensure quality of spot welds, various nondestructive testing methods are used in addition to destructive testing methods, such as tensile shear tests and metallographic examination. Specifically, ultrasonic evaluation is widely used for measuring the nugget size in resistance spot welding. This study focuses on determining the possibility of establishing a standard to evaluate the quality of resistance spot welding in real time using servo-gun displacement data. Two-, three, and four-layered materials, including galvanized steel and hot stamped steel, were used in the experiment. To obtain a welding lobe diagram, we performed resistance spot welding three times under the same conditions, and secured the servo-gun data, which change depending on the expansion and contraction of the material. The strength and fracture mode data obtained from a tensile shear test were compared with the servo-gun displacement data, and the results were analyzed. The results evidently confirmed that the displacement data of the electrode of the medium-sized C gun clearly correlated with the tensile strength data of welding. However, the displacement data of the large C gun exhibited a slightly low correlation, presumably because of a mechanical issue. Furthermore, it was predicted that the results of this study could be used for real-time welding quality evaluation with minimal additional costs.
Efficient Activation of Dendritic Cells with Immuno-modulating Functional Nanoparticles in Hydrogel

Modulation of the immunosuppressive tumor microenvironment (TME) is crucial for effective tumor treatment. Herein, we demonstrate an immune-modulating hydrogel containing functional nanoparticles. These nanoparticles were synthesized by a heat-up method and coated with CpG oligonucleotide capable of activating the dendritic cells. When the CpG-coated nanoparticles were treated to bone marrow-derived dendritic cells (BMDCs), it upregulated activation marker for DCs such as CD80, CD86, CD40, and MHC2 molecules, suggesting the nanoparticles-mediated maturation of BMDCs. For sustained and local delivery of the prepared CpG-coated nanoparticles to the tumor resection site, the functional nanoparticles were embedded in a hydrogel based on methacrylate-modified hyaluronic acid. Finally, the immune-modulating hydrogel was implanted in a fat pad after the surgical-resected breast tumor model in vivo. The implantation of the immune-modulating hydrogel could be able to effectively activate the DCs in the vicinity of the tumor resection site, resulting in the activation of cytotoxic T cells. We believe this strategy has great potential for effective prevention of tumor recurrence and metastasis.
