        Identification of Primary Failure Modes of Tunnel System and Influence of Supporting Structures on Tunnel System Reliability using Multiple Response Surfaces

        A method of calculating the failure probability of the tunnel system combining finite element numerical simulation with multiple response surface method (MRSM) and Monte-Carlo simulation (MCS) is presented. The applicability of the proposed methodology is verified through a subway interval soft rock tunnel in Qingdao, China. The sensitivity of Monte-Carlo sampling number and coefficients of variation for rock masses on the failure probability of tunnel system is conducted. The primary failure modes in tunnel system are identified by reanalyzing the failure samples. The simulation results demonstrate that the failure probability of a tunnel system within soft upper and hard lower surrounding rock mass is mainly attributed to the soft upper part of the surrounding rock. The coefficients of variation of the elastic modulus E1 and the internal friction angle ϕ1 of the pebble layer (soft upper part) have significant effect on the failure probability. The failure probability of tunnel system increases as the coefficients of variation of E1 and ϕ1 increase. Two primary failure modes are found to contribute to the tunnel system reliability. The effect of rock bolt length L and pipe-roof thickness H on tunnel system reliability and two primary failure modes as well are investigated. The simulation results indicate that both the enhancements in L and H tend to be more effective than the enhancement in either L or H if a small target failure probability of tunnel system is expected. The supporting structures design can be performed based on the potential sets of (L, H) satisfying target failure probability.

        Deep sequencing reveals microRNA signature is altered in the rat epididymis following bilateral castration

        Background In the epididymis of bilateral castrated male rat, gene expression profile changed significantly. However, up to date, no study has investigated how these genes were regulated by microRNAs (miRNAs). Objective We investigated the alterations in the miRNA signature of the epididymis from sham-operated and bilaterally castrated rats. Methods By employing deep sequencing technology and qPCR, the global alterations of epididymal miRNA signature between sham-operated (Con-EP library) and bilaterally castrated rats (Cas-EP library) were explored. MiRNA-target interaction networks were annotated by GO and KEGG enrichment. Results We identified 313 and 306 known miRNAs as well as 152 and 114 novel miRNAs in the Con-EP and Cas-EP libraries, respectively. 59 miRNAs were differentially expressed, including 24 up-regulated and 35 down-regulated miRNAs, among which two up-regulated and three down-regulated ones were validated using qPCR. The expression of these miRNAs in the epididymides of rats at different postnatal ages showed regular changes from birth to adult, suggesting they were androgen-regulated. GO analysis showed that many of the miRNA targets were enriched in metabolic processes. KEGG analysis demonstrated that the targets mainly participated in the mitogen-activated protein kinase (MAPK) pathway. Moreover, 3 and 6 functional modules were detected among the up- and down-regulated miRNA target interaction networks, respectively, and these modules were involved in various biological processes. Conclusion This study represents the first systematic investigation of alterations in the miRNA signature of the epididymis from bilaterally castrated rats and will provide useful resources for functional studies of the miRNAs in the male reproductive system.

        Novel Active Voltage Quality Regulator with Adaptive DC-Link Voltage Control

        In this paper, a novel Active Voltage Quality Regulator (AVQR) topology with a thyristor rectifier and an adaptive dclink voltage control strategy are proposed. The proposed AVQR can efficiently mitigate the long duration variations (e.g. undervoltages/overvoltages), voltage imbalances and voltage harmonics. Compared with conventional AVQRs, it can regulate the load voltage very well with a much lower dc-link voltage. This is accomplished by replacing the diode rectifier with a thyristor rectifier. Moreover, its dc-link voltage can vary with the deviations of the supply voltage through the proposed adaptive dc-link voltage control strategy. All of these contribute to its significantly higher efficiency for online operating, which is very important and attractive for many applications. The proposed topology and control strategy are theoretically analyzed in detail. Simulation results are also provided in the paper. Finally, the feasibility and effectiveness of the proposed method are verified by means of experimental results from a 2kVA prototype. Both of the simulation and experimental results show that the proposed AVQR can achieve a much higher efficiency and similar regulation performance when compared with the conventional ones.

        Maternal undernutrition alters the skeletal muscle development and methylation of myogenic factors in goat offspring

        Objective: The effects of maternal undernutrition during midgestation on muscle fiber histology, myosin heavy chain (MyHC) expression, methylation modification of myogenic factors, and the mammalian target of rapamycin (mTOR) signaling pathway in the skeletal muscles of prenatal and postnatal goats were examined. Methods: Twenty-four pregnant goats were assigned to a control (100% of the nutrients requirement, n = 12) or a restricted group (60% of the nutrients requirement, n = 12) between 45 and 100 days of gestation. Descendants were harvested at day 100 of gestation and at day 90 after birth to collect the femoris muscle tissue. Results: Maternal undernutrition increased (p<0.05) the fiber area of the vastus muscle in the fetuses and enhanced (p<0.01) the proportions of MyHCI and MyHCIIA fibers in offspring, while the proportion of MyHCIIX fibers was decreased (p<0.01). DNA methylation at the +530 cytosine-guanine dinucleotide (CpG) site of the myogenic factor 5 (MYF5) promoter in restricted fetuses was increased (p<0.05), but the methylation of the MYF5 gene at the +274,280 CpG site and of the myogenic differentiation (MYOD) gene at the +252 CpG site in restricted kids was reduced (p<0.05). mTOR protein signals were downregulated (p<0.05) in the restricted offspring. Conclusion: Maternal undernutrition altered the muscle fiber type in offspring, but its relationship with methylation in the promoter regions of myogenic genes needs to be elucidated.         Complete chloroplast genome sequence and phylogenetic analysis of Rhododendron molle G. Don, an endangered Ericaceae species located on Dabie Mountains (central China)

        Rhododendron molle G. Don, an endemic species of the Ericaceae family, possesses valuable medicinal and horticultural values. In this research, R. molle cp genome proved to be a typical quadripartite structure with the length of 200,878 bp. In particular, the lengths of large single-copy region (LSC), small single-copy region (SSC), and inverted repeat regions (IR) were 198,019 bp, 629 bp, and 1117 bp, respectively. Among the 149 unique genes, 97 were protein-coding genes, 44 were tRNA genes, and 8 were rRNA genes, respectively. Leucine was the most representative amino acid (10.663%), while Cysteine was the lowest representative (1.178%). A set of 30 codons showed obvious codon usage bias, while 29 were A/U-ending codons. Six gene regions showed high levels of nucleotide diversity (Pi > 0.02). Totally, 273 SSRs were identified. Maximum-likelihood (ML) phylogenetic analysis revealed that R. molle was relatively closed to the R. pulchrum and R. delavayi. High similarity was detected among Ericaceae species, and the coding regions were more conserved than the non-coding regions. Expansion and contraction detected in IR region could be the main length variation in R. molle and related Ericaceae species. This research will supply rich genetic resource for R. molle and related species of Ericaceae.

        CRISPR/Cas9-mediated knockout of the Vanin-1 gene in the Leghorn Male Hepatoma cell line and its effects on lipid metabolism

        Objective: Vanin-1 (VNN1) is a pantetheinase that catalyses the hydrolysis of pantetheine to produce pantothenic acid and cysteamine. Our previous studies have shown that the VNN1 is specifically expressed in chicken liver which negatively regulated by microRNA-122. However, the functions of the VNN1 in lipid metabolism in chicken liver haven’t been elucidated. Methods: First, we detected the VNN1 mRNA expression in 4-week chickens which were fasted 24 hours. Next, knocked out VNN1 via CRISPR/Cas9 system in the chicken Leghorn Male Hepatoma cell line. Detected the lipid deposition via oil red staining and analysis the content of triglycerides (TG), low-density lipoprotein-C (LDL-C), and highdensity lipoprotein-C (HDL-C) after VNN1 knockout in Leghorn Male Hepatoma cell line. Then we captured various differentially expressed genes (DEGs) between VNN1- modified LMH cells and original LMH cells by RNA-seq. Results: Firstly, fasting-induced expression of VNN1. Meanwhile, we successfully used the CRISPR/Cas9 system to achieve targeted mutations of the VNN1 in the chicken LMH cell line. Moreover, the expression level of VNN1 mRNA in LMH-KO-VNN1 cells decreased compared with that in the wild-type LMH cells (p<0.0001). Compared with control, lipid deposition was decreased after knockout VNN1 via oil red staining, meanwhile, the contents of TG and LDL-C were significantly reduced, and the content of HDL-C was increased in LMH-KO-VNN1 cells. Transcriptome sequencing showed that there were 1,335 DEGs between LMH-KO-VNN1 cells and original LMH cells. Of these DEGs, 431 were upregulated, and 904 were downregulated. Gene ontology analyses of all DEGs showed that the lipid metabolism-related pathways, such as fatty acid biosynthesis and long-chain fatty acid biosynthesis, were enriched. KEGG pathway analyses showed that “lipid metabolism pathway”, “energy metabolism”, and “carbohydrate metabolism” were enriched. A total of 76 DEGs were involved in these pathways, of which 29 genes were upregulated (such as cytochrome P450 family 7 subfamily A member 1, ELOVL fatty acid elongase 2, and apolipoprotein A4) and 47 genes were downregulated (such as phosphoenolpyruvate carboxykinase 1) by VNN1 knockout in the LMH cells. Conclusion: These results suggest that VNN1 plays an important role in coordinating lipid metabolism in the chicken liver.

        Support vector machine based estimation of remaining useful life: current research status and future trends

        Estimation of remaining useful life (RUL) is helpful to manage life cycles of machines and to reduce maintenance cost. Support vectormachine (SVM) is a promising algorithm for estimation of RUL because it can easily process small training sets and multi-dimensionaldata. Many SVM based methods have been proposed to predict RUL of some key components. We did a literature review related toSVM based RUL estimation within a decade. The references reviewed are classified into two categories: improved SVM algorithms andtheir applications to RUL estimation. The latter category can be further divided into two types: one, to predict the condition state in thefuture and then build a relationship between state and RUL; two, to establish a direct relationship between current state and RUL. However,SVM is seldom used to track the degradation process and build an accurate relationship between the current health condition stateand RUL. Based on the above review and summary, this paper points out that the ability to continually improve SVM, and obtain a novelidea for RUL prediction using SVM will be future works.
