        The mitochondrial unfolded protein response (UPRmt) protects against osteoarthritis

        The mitochondrial unfolded protein response (UPRmt) is a mitochondrial-to-nuclear signaling pathway that is activated to maintain mitochondrial function when there is an accumulation of misfolded proteins within mitochondria. Mitochondrial function is essential for chondrocyte homeostasis, and mitochondrial dysfunction is a characteristic of osteoarthritis (OA). However, the role of the UPRmt in OA remains unclear. In the present study, the level of the UPRmt was examined in primary mouse chondrocytes subjected to different stresses and in the articular cartilage of OA model mice and OA patients. The relationship between UPRmt activation and OA progression was studied. The UPRmt was induced in primary mouse chondrocytes subjected to diverse stresses and in the cartilage of OA mice. Enhancement of the UPRmt with nicotinamide riboside (NR) significantly improved mitochondrial function, reduced chondrocyte death, attenuated OA pain, and ameliorated OA progression, and the protective effects decreased significantly in chondrocyte-specific Atf5 knockout (ATF5f/fCol2a1-CreERT2) mice. UPRmt induction was also identified in the articular cartilage of OA patients and was associated with reduced chondrocyte death, less severe hip pain, and lower levels of inflammation in synovial fluid. These findings identify the induction of the UPRmt in primary mouse chondrocytes exposed to pathological stresses and in the articular cartilage of OA model mice and OA patients. Enhancement of the UPRmt ameliorates OA progression, suggesting that the UPRmt exerts a protective effect against OA and may be a potential diagnostic and therapeutic strategy for OA.

        Sound Recognition of Harmful Bird Species Related to Power Grid Faults Based on VGGish Transfer Learning

        Bird activities threaten the safe operation of transmission lines and substations. In order to assist differentiated prevention of bird-related faults in power grid, this paper proposes a birdsong recognition method based on VGGish transfer learning. Firstly, according to the information of bird species related to historical power grid faults and the investigation results of bird species around transmission lines, 18 high-risk, 18 low-risk, and 2 harmless bird species were selected to establish a sample set with their song signals. Then, the birdsong signals were preprocessed by framing, windowing, noise reduction and clipping, thus to extract the spectrogram, which was mapped to a 64-order Mel filter banks to get the Mel spectrogram. Aiming at weak generalization ability of traditional birdsong recognition models due to insufficient number of samples, the VGGish transfer learning network pretrained by AudioSet was used as the birdsong feature extractor, and the Mel spectrograms of harmful bird species belong to the training set were taken as inputs to train the network parameters, thus to extract 128-dimensional VGGish deep features for bird recognition. This method was applied to classify 38 kinds of bird species related to power grid faults, and the recognition accuracy reaches 94.43%. The research results can provide references for power grid inspector to carry out intelligent recognition and ecological prevention of bird species.

        First-principles investigation of the monoclinic NaMnO2 cathode material for rechargeable Na-ion batteries

        Using first-principles calculations, we successfully investigate the electrochemical performance of the monoclinic NaMnO2 for the sodium ion batteries. NaMnO2 possesses a voltage window of 3.54–2.52 V and theoretical reversible capacity of 136 mAh g−1. Besides, we find that the metallicity of the monoclinic NaMnO2 gradually increases during Na extraction. Moreover, the computational Na migration energy barrier in the monoclinic NaMnO2 is 0.18 eV, ensuring ideal conductivity and reversible capacity. Although the Jahn-Teller distortion effects limit the enhancement of the reversible capacity of the monoclinic NaMnO2, it is still a right cathode material for the sodium ion batteries. The computational results are well in consistent with the experimental investigations.

        Functional analysis of prv-miR-LLT11a encoded by pseudorabies virus

        Viral-encoded microRNAs (miRNAs) play have vital roles in the regulations of virus replications and host immune responses. The results of previous studies have indicated that miRNA clusters are involved in the replication and virulence of the pseudorabies virus (PRV), which may potentially lead to the immune escape or facilitation of PRV replications. This study's previous research revealed that the prv-mirmiR-LLT11a was differentially expressed during PRV infections. The present study's results have demonstrated that the prv-miR-LLT11a could significantly inhibit PRV replications. It was further determined that SLA-1 was the target gene of the prv-miR-LLT11a, and simultaneously, thate overexpression of prv-miR-LLT11a could down-regulate the mRNA and protein levels of SLA-1 in a dose-independent manner. Furthermore, the present study also found observed that the prv-miR-LLT11a canhad also down-regulated the TAP1 expressions. Our findings provide a better understanding of the molecular mechanism involved in on the effects of prv-miR-LLT11a on SLA-1 and TAP1, as well as and its involvement in a potential immune system evasion of PRV.

        Protective effect and mechanism of ginsenoside Rg2 on atherosclerosis

        Background: Ginsenoside Rg2 (Rg2) has a variety of pharmacological activities and provides benefits during inflammation, cancer, and other diseases. However, there are no reports about the relationship between Rg2 and atherosclerosis. Methods: We used 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) to detect the cell viability of Rg2 in vascular smooth muscle cells (VSMCs) and human umbilical vein endothelial cells (HUVECs). The expression of inflammatory factors in HUVECs and the expression of phenotypic transformation-related marker in VSMCs were detected at mRNA levels. Western blot method was used to detect the expression of inflammation pathways and the expression of phenotypic transformation at the protein levels. The rat carotid balloon injury model was performed to explore the effect of Rg2 on inflammation and phenotypic transformation in vivo. Results: Rg2 decreased the expression of inflammatory factors induced by lipopolysaccharide in HUVECs-without affecting cell viability. These events depend on the blocking regulation of NF-κB and p-ERK signaling pathway. In VSMCs, Rg2 can inhibit the proliferation, migration, and phenotypic transformation of VSMCs induced by platelet derived growth factor-BB (PDGF-BB)-which may contribute to its anti-atherosclerotic role. In rats with carotid balloon injury, Rg2 can reduce intimal proliferation after injury, regulate the inflammatory pathway to reduce inflammatory response, and also suppress the phenotypic transformation of VSMCs. Conclusion: These results suggest that Rg2 can exert its anti-atherosclerotic effect at the cellular level and animal level, which provides a more sufficient basis for ginseng as a functional dietary regulator.

        Regulation of HBV-specific CD8+ T cell-mediated inflammation is diversified in different clinical presentations of HBV infection

        Chronic HBV infection is the leading cause of liver cirrhosis and hepatic cancer, but the individual responses toward HBV infection are highly variable, ranging from asymptomatic to chronic active hepatitis B inflammation. In this study, we hypothesized that the different individual responses to HBV infection was associated with differences in HBV-specific CD8+ T cell-mediated inflammation and cytotoxicity. Blood samples were collected from subjects with asymptomatic HBV-infection, subjects undergoing active chronic HBV flares (active CHB), and subjects with HBV-infected hepatocellular carcinoma (HBV-HCC). By tetramer staining, we found that all three groups had similar frequencies of HBVspecific CD8+ T cells. However, after HBV peptide stimulation, the HBV-specific CD8+ T cells in asymptomatic subjects had significantly stronger interferon gamma (IFN-γ), tumor necrosis factor alpha (TNF-α), and CD107a expression than those in active CHB and HBV-HCC patients. Examination of surface marker expression revealed that the PD-1-Tim-3- double-negative cell population was the main contributor to HBV-specific inflammation. In active CHB patients and HBV-HCC patients, however, the frequencies of activated PD-1-Tim-3- cells were significantly reduced. Moreover, the serum HBV DNA titer was not correlated with the frequencies of HBV-specific CD8+ T cells but was inversely correlated with the frequencies of IFN-g-expressing and CD107a-express cells in response to HBV stimulation. Together, our data demonstrated that the status of HBVspecific CD8+ T cell exhaustion was associated with different clinical outcomes of chronic HBV infection.

        Core–shell structure g-MnO2-PANI carbon fiber paper-based flexible electrode material for high-performance supercapacitors

        Good conductivity is the key to ensuring high electrochemical performance, and the uniformity of theactive material directly affects the electrical conductivity. This paper proposes a novel modified stepwisein situ polymerization method to prepare composite materials, and then uses the K/S value method tocharacterize the uniformity of polyaniline distribution. Specifically, the study presents a papermakingwet-lay technique to prepare a carbonfiber paper substrate (CP) that is light-weight,flexible, andfoldable while also having low internal resistance and high porosity. The g-MnO2 is wrapped with PANI toform a core–shell structure, and a modified stepwise in situ polymerization method is adopted to depositthe core–shell structure uniformly on the CP substrate by physical adsorption. This process yields aflexible electrode material for supercapacitors, called g-MnO2-PANI-CP electrode. Results show thatthe g-MnO2-PANI-CP electrode has a high specific capacitance of 642.5 F g 1 at a current density of 1A g 1, and also has a high energy density of 114.2 Wh kg 1 at a power density of 798.6 W kg 1. In addition,the electrode retains 81.3% of the initial capacitance after 5000 cycles, demonstrating high cyclingperformance.

        <P>We report the plasmon-assisted photocurrent enhancement in Ag-nanoparticles (Ag-NPs) embedded PEDOT:PSS/P3HT:PCBM organic solar cells, and systematically investigate the causes of the improved optical absorption based on a cylindrical Ag-NPs optical model which is simulated with a 3-Dimensional finite difference time domain (FDTD) method. The proposed cylindrical Ag-NPs optical model is able to explain the optical absorption enhancement by the localized surface plasmon resonance (LSPR) modes, and to provide a further understanding of Ag-NPs shape parameters which play an important role to determine the broadband absorption phenomena in plasmonic organic solar cells. A significant increase in the power conversion efficiency (PCE) of the plasmonic solar cell was experimentally observed and compared with that of the solar cells without Ag-NPs. Finally, our conclusion was made after briefly discussing the electrical effects of the fabricated plasmonic organic solar cells.</P>

        Understanding the friction behavior of sulfur-terminated diamond-like carbon films under high vacuum by first-principles calculations

        Generally, the repulsive force was a key factor account for superlow friction of H or F doped diamond-like carbon (DLC) films under high vacuum. As we known, H or F doped DLC usually exhibited superlow friction under high vacuum. However, the superlow friction of S doped DLC under high vacuum was not found so far. This phenomenon was desirable to be well investigated. In this work, S-terminated diamond interfaces also exhibited strong repulsive force, however, the estimated friction coefficient was variable for S-terminated diamond interfaces. The lowest and largest friction coefficient was about 0.003 and 0.4 respectively, which indicated that the superlow friction of S doped DLC could achieve in theory. In order to well probe the unusual friction behavior of S doped DLC under high vacuum, using first-principles method, the repulsive interaction between sliding surfaces was well investigated in order to understand the unusual friction behavior of S doped DLC films.
