        Characteristics and Regulating Role in Thermotolerance of the Heat Shock Transcription Factor ZmHsf12 from Zea mays L.

        Plant heat shock transcription factors (Hsfs) areimportant regulators of heat shock signal transduction pathway. There are 30 members of the Hsf family in maize, only twoof which, ZmHsf06 and ZmHsf12, belong to subclass A1. Our previous work demonstrated that ZmHsf06-overexpressingArabidopsis lines showed improved tolerance to heat anddrought stresses. In this study, we isolated ZmHsf12 fromyoung leaves of maize (Zea mays L.) using homologouscloning methods. The CDS (coding sequence) of ZmHsf12 is1,494 bp and encodes a putative protein consisting of 497amino acids which possesses domains such as DBD (DNAbindingdomain), OD (oligomerization domain), NLS (nuclearlocalization signal), NES (nuclear export signal), and anAHA (activator) motif. The ZmHsf12-GFP fusion protein islocalized to the cell nucleus. ZmHsf12 was expressed inmany maize organs, and its expression was up-regulated byheat shock. Furthermore, we characterized the function ofZmHsf12 in yeast and Arabidopsis. Yeast cells overexpressingZmHsf12 showed enhanced heat tolerance. ZmHsf12-overexpressing Arabidopsis seedlings displayed significantincreases in both basal and acquired thermotolerance. Comparedto WT seedlings, the ZmHsf12-overexpressing lines displayedboth increased chlorophyll contents and higher survivalrates. Also, the expression of AtHsps was increased higher inthe ZmHsf12-overexpressing Arabidopsis lines after heatstress. The results of our study strongly suggested thatZmHsf12 may take part in plant response to heat stress.

        Variability in the Viral Protein Linked to the Genome of Turnip Mosaic Virus Influences Interactions with eIF(iso)4Es in Brassica rapa

        Plants protect against viruses through passive and ac- tive resistance mechanisms, and in most cases charac- terized thus far, natural recessive resistance to potyvi- ruses has been mapped to mutations in the eukaryotic initiation factor eIF4E or eIF(iso)4E genes. Five eIF4E copies and three eIF(iso)4E copies were detected in Brassica rapa. The eIF4E and eIF(iso)4E genes could interact with turnip mosaic virus (TuMV) viral protein linked to the genome (VPg) to initiate virus translation. From the yeast two-hybrid system (Y2H) and bimo- lecular fluorescence complementation (BiFC) assays, the TuMV-CHN2/CHN3 VPgs could not interact with BraA.eIF4E.a/c or BraA.eIF(iso)4E.c, but they could interact with BraA.eIF(iso)4E.a in B. rapa. Further analysis indicated that the amino acid substitution L186F (nt T556C) in TuMV-UK1 VPg was important for the interaction networks between the TuMV VPg and eIF(iso)4E proteins. An interaction model of the BraA. eIF(iso)4E protein with TuMV VPg was constructed to infer the effect of the significant amino acids on the interaction of TuMV VPgs-eIF(iso)4Es, particularly whether the L186F in TuMV-UK1 VPg could change the structure of the TuMV-UK1 VPg protein, which may terminate the interaction of the BraA.eIF(iso)4E and TuMV VPg protein. This study provides new insights into the interactions between plant viruses and trans- lation initiation factors to reveal the working of key amino acids.

        Constant-stress accelerated life test of white organic light-emitting diode based on least square method under Weibull distribution

        It is currently hard to estimate the reliability parameters of organic light-emitting diodes (OLEDs) when conducting a life test at normal stress, due to the remarkably improved life of OLEDs to thousands hours. This work adopted three constant-stress accelerated life tests (CSALTs) to predict the life of white OLEDs in a short time. TheWeibull function was applied to describe the life distribution, and the shape and scale parameters were estimated using the least square method. The experimental test data were statistically analyzed using a self-developed software. The life of white OLEDs predicted via this software is ingood agreement with that reported from the customers. The numerical results indicated that the assumptions of CSALT are correct, and that CSALT can be used to predict the life of white OLEDs. This work confirmed that the life of white OLEDs meets the Weibull distribution, and that the accelerated life equation conforms to the inverse power law. Furthermore, the precise accelerated parameters were shown to be particularly useful in enabling the rapid estimation of white OLEDs’ life.

        Many cultures and ideologies emerged in last century tend to be more emphasized on conflict, but 21 century should be an era of cultural harmony. The cultural harmony is overwhelming to some extent. Its soft-power had been considered as major cause of the normalizations of Sino-Japanese and Sino-Korean relations as well as their continuing friendship and cooperation in recent years. The North-eastern Asian, consisting of China, Japan and Korea, is one of the leading regions in culture and economic development of Asian, even of the world. Essentially, its traditional cultures have emphasized on entireness, equilibrium and harmony. Harmony leads to mutual benefit, mutual benefit boosts the cooperation, cooperation advances the development, the conflict will be solved under the development of each parties. Hence, harmony is the key to win-win strategy. Actually, there are differences among the north-eastern Asian countries despite the common value of them. The Korean fashion, such as film, TV and music etc, are very popular among the youngsters of China in recent years, hence being called as “Han tide” in China. Some Chinese scholars ague that it is due to the approximation of cultures between two countries. The Korean fashion culture, absorbing much from modern western culture, with its traditional heritage still retain, would be easily accepted by people with some extent of acquaintance and curiosity. Nevertheless, some Chinese scholars show anxious of the “Han tide” that many fans have blindly imitated life styles which are so much influenced by the western culture. Moreover, they believe that the over immerging in “Han tide” will make the youngsters lost in the identity of our own culture, thus the pride of nation will be weakened. Some scholars assent to a cultural self-protection action in order to counter against so-called “vulgar pop culture”. As a result, many cities of China, including Shanghai, show a great impetus to cultivating their own cultural industries based on Chinese tradition and its unique style. They think the development of culture is an indispensable brand of a metropolis. The North-eastern Asian countries, including China, Japan and Korea, have the common cultural origin­­­Confucianism. But now, as a sub-branch of Confucianism, Korean pop culture have more influential on China than Chinese one on Korea, with a great impact on the value and aesthetic perception of Chinese youngsters. I believe that the pop culture has its own truth, the “Han tide” blow with its wits of Korean, uncovering the lag of Chinese own pop culture which has long been neglected. It is not the truth that the “Han tide” was attributed only to the chance or the ill-consumption with a blink and superficial manner. Actually, the “Han tide” was not a momentary phenomenon and would not pass away quickly. The truth is that we should have an open mind with it, meanwhile, take a thorough consideration from the depth of our traditional culture. The 21 century should be era of harmony with people oriented. The North-eastern Asian countries have to build their common culture beyond the nation boundaries on a people oriented basis. Only get through the myopic view of cultural imperialism can we create our common oriental culture admired by all the human being. If the Korean movies or other pop cultures have pushed a step for the whole region of East Asia, or even for the world, and cast a light for the world cultures, it would be significant not only for the Korean pop culture itself, but also for the whole of oriental culture, including Chinese culture, to go into the world arena.

        Geochemical record of rapid climate change and chemical weathering in a semi-arid area, northeastern Tibetan Plateau

        Lacustrine deposits can provide insight into chemical weathering and climate change. Based on the analysis of the element concentrations and parameters of the acid-leaching residual fractions (AR) in the lacustrine deposits of the Qaidam Basin (QB), chemical weathering and paleoclimate changes in the northeastern Tibetan Plateau (NETP) have been studied. The results show that the characteristics of trace element concentrations exhibit two kinds of patterns by normalized element arbitrary units (a.u.). The rare earth element (REE) distribution patterns are moderately rich in light rare earth elements (LREEs) with slightly right-tilting and negative Eu anomalies. Triangular plots of the REEs and (La/Yb)n-ΣREE suggest that the sediments have a similar source. The geochemical records of climate proxies indicated warm-wet climate stages during 45.1–31.6 cal. ka BP., further proving that a megalake stage existed in the semi-arid area in marine isotope stage (MIS) 3. Strong evaporation resulted in a high paleolake level history in the northeastern Tibetan Plateau after the late period of MIS 3. Meanwhile, the geochemical records of the Qaidam Basin are synchronous with the global records.

        Alterations in antioxidant function and cell apoptosis in duck spleen exposed to molybdenum and/or cadmium

        To investigate the effects of molybdenum (Mo) and/or cadmium (Cd) on antioxidant function and the apoptosis-related genes in duck spleens. Sixty healthy 11-day-old ducks were randomly divided into six groups of 10 ducks (control, low Mo group, high Mo, Cd, low Mo + Cd, and high Mo + Cd groups). All were fed a basal diet containing low or high dietary doses of Mo and/or Cd. Relative spleen weight, antioxidant indices, apoptosis-related gene mRNA expression levels, and ultrastructural changes were evaluated after 120 days. The results showed that the relative spleen weight decreased significantly in the high Mo + Cd treatment group which compared with control group. Malondialdehyde levels increased and xanthine oxidase and catalase activities decreased in the Mo and/or Cd groups compared with levels in the control group. Bak-1 and Caspase-3 expressions were upregulated in the high Mo + Cd group, while Bcl-2 was downregulated. In addition, mitochondrial crest fracture, swelling, vacuolation, deformed nuclei, and karyopyknosis in both Mo + Cd treated groups were more severe than in the other groups. The results suggest that Mo and/or Cd can induce oxidative stress and apoptosis of spleen via effects on the mitochondrial intrinsic pathway. Moreover, the results indicate the two elements have a possible synergistic relationship.

        Baicalein induces the apoptosis of U251 glioblastoma cell lines via the NF-kB-p65- mediated mechanism

        Glioblastoma (GBM) is the most aggressive cerebral gliomas. Moreover, the overall prognosis of GBM is still little. Baicalein (BA) is a flavonoid derived from the Scutellaria baicalensis root, and has historically been used in anticancer therapies. However, its apoptosis role and related mechanisms in GBM has not yet been researched clearly. Thus, this study aimed to investigate the effects of BA on human GBM U251 cell line. The effects of BA on proliferation of U251 cells were measured by Cell Counting Kit-8 assay. Cellular apoptosis was detected by flow cytometry with annexin V-FITC/propidium iodide staining. The expression of apoptosis-related protein Bcl-2, Bax and cleaved-caspase3 was detected by quantitative real-time PCR and western blot. The expression of nuclear p65 protein, the active subunit of nuclear factor-kappa B (NF-κB), was determined by immunofluorescence and western blot. Our results showed that the viability of U251 cells significantly decreased in a time- and dose-dependent manner after treated with BA, and the apoptotic ratio of BA-treated groups was significantly higher than that of control groups. Furthermore, the expression of NF-kB-p65 in the nucleus was remarkably reduced, and the activity of NF-kB-p65 was remarkably inhibited after BA treatment. Combined treatment with a NF-kB-P65 inhibitor (QNZ) and BA resulted in the synergistic reduction of Bcl-2 expression and then increase of Bax and cleaved-caspase3 expression; and the viability of U251 cells was also inhibited. In conclusion, BA inhibits GBM cells viability and induces apoptosis via inhibit the activity of NF-kB-p65, suggesting that BA is a potential therapeutic agent for GBM.

        PD-type ℓ2-ℓ∞ Intermittent Pinning Synchronization Control of Discrete Time-delay Nonlinear Dynamical Networks

        In this paper, the PD-type `2 − `∞ synchronization control problem is concerned for a class of discrete time-delayed nonlinear dynamical networks. A PD-type (proportional-derivative-type) pinning control protocol is proposed where only a small fraction of network nodes is controlled in order to achieve the global synchronization performance. Meanwhile, for the sake of decreasing the update frequency of the controller, the periodic intermittent control strategy is developed. The main purpose of the addressed problem is to design a PD-type intermittent pinning feedback controller such that the closed-loop system achieves both the exponential synchronization and the prescribed `2 −`∞ performance index. Subsequently, by means of the Lyapunov-Krasovskii functional method, a sufficient condition is established under which the addressed PD-type controller design problem is recast into a linear convex optimization one that can be easily solved via available software packages. Finally, a simulation example is given to show the applicability of the developed theoretical results.
